Abstract
Description
[実施例1]
シロイヌナズナにおけるアルギニン-窒素の分配
野生型のシロイヌナズナを用いて、3mMの硝酸塩および30μMのU-15N(>98% 15N)L-Argを追加し、3.6mMのMES(2N-モルホリノエタンスルホン酸)でpH5.8に緩衝させた、0.65%w/vの寒天(植物寒天、Duchefa Biochemie)、0.5%w/vのショ糖を有する、半分に薄めたムラシゲ・スクーグ(MS)培地(Murashige and Skoog, 1962)を含有する無菌寒天プレートにおいて実験を行った。植物を19日間生長させ、植物体20個を採取し、4つの同じ物の群(replicate)に分けた(すなわち、各群は植物体5個からなる)。苗条と根を分離し、根を、0.5mMのCaCl2溶液中で3回、徹底的にすすいできれいにし、表面から付着化合物を取り除いた。苗条および根を、60℃で一晩中乾燥させ、計量し、ホモジナイズした。最後に、安定同位体比質量分析装置(Europe Scientific Isotope Ratio Mass Spectrometer)を用いて試料を分析して総N含有量および総15N含有量を決定した。結果を図3に表す。
シロイヌナズナおよびヤマナラシにおけるバイオマスおよびグルタミン-Nの分配
野生型のシロイヌナズナを用いて、無菌寒天プレートにおいて分配試験を行った。また、ヤマナラシの場合は、等量の、窒素を含まない、0.8%w/vの寒天(植物寒天、Duchefa Biochemie)および0.5%w/vのショ糖を有し、MES緩衝液を用いてpHを5.8にした、半分に薄めたムラシゲ・スクーグ(MS)培地(Murashige and Skoog, 1962)を含有するプラスチックの箱において分配試験を行った。窒素を、NH4 +とNO3 -の等モル混合物として、またはL-GlnとNO3 -それぞれの50%に相当する等モル混合物としてのいずれかで、どちらの混合物の場合も3mMのNに相当する総添加率で寒天に加えた。4つの標識化処理、すなわち、各N混合物に対して2つずつの標識化処理を行った。したがって、NH4NO3混合物を含むプレートの半分は標識NH4 +を含有し、他の半分は標識NO3 -を含有した。同様に、L-Gln:NO3 -混合物を含むプレートの半分は標識L-Glnを含有し、他の半分は標識NO3 -を含有した。それぞれを標識化処理するために、N供給源の1%を15Nとして投与した。無菌ろ過したL-Glnを、オートクレーブ後に寒天混合物に加えた。シロイヌナズナ植物を生長させ21日後に採取し、ヤマナラシ植物を生長させ28日後に採取した。苗条および根を60℃で一晩中乾燥させ、計量し、ホモジナイズした。最後に、安定同位体比質量分析装置(Europe Scientific Isotope Ratio Mass Spectrometer)を用いて試料を分析して総N含有量および総15N含有量を決定した。異なる植物体部分(すなわち、苗条と根)における、2種のN供給源のいずれか由来のN量を、過剰原子%15Nおよび植物体部分それぞれの総N含有量の値から計算した。2種の異なるN混合物から得た植物の根の割合を、根に存在する総植物バイオマスの百分率として計算した。シロイヌナズナについての結果を図4に表し、ヤマナラシについての結果を図5に表す。
ヨーロッパアカマツの菌根の評価
北スウェーデンにおいて、植物を1生長期間中、屋外で栽培した。窒素供給源として硝酸アンモニウムまたはアルギニンのいずれかを含む複合養分溶剤を植物に与えた。アルギニン栽培の実生には、生長期間中、週に1回施肥し(実生ごとに総計50mgのN)、一方、参照植物には、週に2〜3回施肥した(実生ごとに総計71.5mgのN)。アルギニン投与の実生に対しては、初秋の最終施肥イベントの後にもアルギニンのパルスを受けさせたことを除いて、参照植物と同じ処理をした。このパルスで与えられたアルギニン量は、1つの実生当たり窒素5mgに相当した。根の平均キチン含有量は、図6およびTable2(表2)で見ることができる。値は、平均値±標準評価、n=6で示されている。
典型的な肥料の組成
シロイヌナズナにおける窒素型に影響される生長およびバイオマス分配
シロイヌナズナ(Arabidposis thaliana)の植物を、無菌培養基中、種々の窒素供給源を追加した培地で21日間生長させた。全ての培地の総窒素濃度は6mMであり、他のマクロ養分およびミクロ養分は全て、各処理について同量を与えた。
Claims (10)
- 根の生長を刺激するための窒素供給源として天然L-アミノ酸を含むことを特徴とする肥料の使用。
- 菌根の発達を刺激するための窒素供給源として天然L-アミノ酸を含むことを特徴とする肥料の使用。
- 根の生長および/または菌根の発達を刺激するための窒素供給源としての天然L-アミノ酸が、L-アルギニン、L-アスパラギン、およびL-グルタミンの群から選ばれることを特徴とする、請求項1から2のいずれか一項に記載の肥料の使用。
- 肥料が、硝酸塩またはアンモニウムなどの無機窒素化合物も含むことを特徴とする、請求項1から3のいずれか一項に記載の肥料の使用。
- 肥料中の窒素供給源の少なくとも30%(wt)、好ましくは肥料中の窒素供給源の少なくとも70%(wt)、好ましくは少なくとも85%(wt)、最も好ましくは少なくとも90%(wt)が、L-アミノ酸であり、他の窒素供給源が無機窒素含有化合物であり、それによりL-アミノ酸と無機窒素含有化合物との比に対して、根および苗条の生長の刺激が関連していることを特徴とする、請求項1から4のいずれか一項に記載の肥料の使用。
- L-アミノ酸が、L-アルギニン、L-アスパラギン、グリシン、L-グルタミン酸およびL-グルタミンの群から選ばれることを特徴とする、請求項1から5のいずれか一項に記載の肥料の使用。
- 肥料が、適切な保存料をも含むことを特徴とする、請求項1から6のいずれか一項に記載の天然L-アミノ酸を含む肥料の使用。
- 保存料が、安息香酸カリウムなどの安息香酸塩、酢酸、サリチル酸、プロピオン酸、ソルビン酸、クエン酸、およびアレキシンプラス(alexin plus)の群から選択されることを特徴とする、請求項1から7のいずれか一項に記載の天然L-アミノ酸を含む肥料の使用。
- 肥料が固形であることを特徴とする、請求項1から8のいずれか一項に記載の天然L-アミノ酸を含む肥料の使用。
- 肥料が溶剤であることを特徴とする、請求項1から9のいずれか一項に記載の天然L-アミノ酸を含む肥料の使用。
