Abstract
Description
本出願は、2007年11月2日に出願された仮出願第60/985,094号の優先権の利益を主張するものである。
以下の手順を使用して、5/6腎摘出(Nx)ラットモデルにおいて、カプリン酸ナトリウムのインビボ保護効果の実証を実施した。雄の6週齢ウィスターラットに対して5/6腎摘出または偽手術を行った。ケタミン麻酔(60〜100mg/kg,i,p.)下で、左の腎臓の3分の2を除去した後、7日後に右の一側性腎摘出により腎切除を達成した。偽ラットに対して腎臓の曝露および腎周囲の脂肪の除去を行った。最初の手術から21日後、血清クレアチニンが腎臓の機能障害を示す125μmol/Lを超えた場合、ラットを実験に割り当てた。偽手術を受けた動物は、ビヒクル(生理食塩水)が与えられ、対照として使用された。Nx動物をビヒクルまたはカプリン酸ナトリウムを受ける群に分けた。35日間、1日1回、経胃的強制栄養法により生理食塩水またはカプリン酸ナトリウムを与えた。本末期腎疾患モデルの重症度を評価するために、血清クレアチニンを毎週測定した。Day63にラットを屠殺した。
実施例1に記載する手順を使用して、5/6腎摘出ラットモデルにおいて、インビボにおけるカプリン酸ナトリウムの心保護効果の実証を実施した。つまり、カプリン酸ナトリウムが、重症に罹患した5/6腎摘出ラットにおける心臓の保護を示すことを実証するために、5/6腎摘出ラットにおいて、心圧力をRTBP2000(Kent Scientific)を用いて記録した。カプリン酸ナトリウム処理Nxラットにおいて、血圧の著しい低下が観察された(図8)。
以下の手順を使用して、ドキソルビシン誘発腎毒性モデルにおいて、カプリン酸ナトリウムの経口投与のインビボ保護効果の実証を実施した。C57B1/6マウス(6〜10週齢)をDay3からDay10までカプリン酸ナトリウムで予防的処置、またはDay1からDay10まで、治療的に処置した。腎毒性は、Day0に、10mg/kgのドキソルビシンの静脈内注入により誘発された。Day4、7および11に、血清クレアチニンおよび尿素をモニタした。
実施例3に記載する手順を使用して、ドキソルビシン誘発腎毒性モデルにおいて、トリカプリンの経口投与のインビボ保護効果の実証を実施した。
Claims (39)
- 前記組成物が、中鎖トリグリセリド(MCT)である式Iにより表される少なくとも2つの化合物の混合物を含み、A=B=R1が、それぞれ、直鎖または分岐鎖、飽和または不飽和C7およびC9アルキル基である、請求項1に記載の方法。
- 前記混合物が、2つのトリグリセリドから成り、第1のMCTが式IおよびA=B=R1=CH3(CH2)6−により表され、第2のMCTが式IおよびA=B=R1=CH3(CH2)8−により表される、請求項2に記載の方法。
- 前記組成物が、0.1%から3%の式IおよびA=B=R1=CH3(CH2)4−により表される第3の化合物、ならびに式IおよびA=B=R1=CH3(CH2)10−により表される第4の化合物をそれぞれさらに含む、請求項2に記載の方法。
- 前記組成物が、式IIにより表される1つもしくは複数の化合物を含み、かつXが中鎖脂肪酸であるOHである、請求項1に記載の方法。
