Description
本発明は、Aβペプチドに関連した疾患のための非ヒトトランスジェニック、特に哺乳類モデルのための方法、及び組成物を含む。具体的には、本発明は、Aβペプチドを過剰発現する非ヒトトランスジェニック動物モデルを含む。
グリア、及びAβ(N3pGlu-42)の二重免疫蛍光染色により、tgN3E-42マウスのCA1領域内における非常に多数のグリアが明らかになった(図3)。ホモ接合性tgN3E-42マウスは、情動の変化(図8)、認知の低下(図9A、図9B、及び図9C)、及び運動欠陥によって生じる体重増加の障害(図10)と合わせて、神経変性を伴う進行性の表現型を示す。これらの知見は、Aβ(N3pGlu-42)の形成と組織病態との関係を明らかに示している。最も重要なことに、本明細書に記述したモデルは、数週間の年齢にて、ニューロンの喪失を伴うAβ(N3pGlu-42)の有意な蓄積をもつ最初のものである。これらの独特の結果は、トランスジェニックマウスにおいて適用されるトランスジェニックストラテジーを、異なるアミロイドペプチドの神経毒性の特性を調査するための優れたアプローチへと昇格する。従って、本明細書に記述したプレプロストラテジーは、ADan、Abri、又はまたAβに関連したペプチドのようなその他のペプチド様に対して適用されるかもしれない。

本発明は、Aβペプチドに関連した疾患の研究のためのトランスジェニック動物モデルの作成方法、及び組成物、並びにトランスジェニック非ヒト動物それ自体を含む。本発明は、特に、Aβペプチドを過剰発現するトランスジェニック動物モデルを作成する方法、及び組成物、並びにトランスジェニック非ヒト動物それ自体を含む。本発明は、Aβペプチド阻害剤を試験するための方法、及び組成物、並びにAβペプチド阻害剤により予防/治療の方法、及び医薬組成物を更に含む。
「導入遺伝子」という用語は、宿主ゲノムに組み込まれ、又は宿主細胞において自律増殖ができ、かつ1つ以上の細胞産物の発現を生じさせることができるDNAのセグメントを意味する。例示的な導入遺伝子は、対応する非形質転換細胞、又は動物に関して新規表現型もつ、宿主細胞、又はそこから発生した動物を提供するであろう。
「作動可能に連結された」という用語は、適切な分子(例えば、転写活性化タンパク質)が制御配列(群)に結合されるときに、DNA配列、及び制御配列(群)が遺伝子形質発現を可能にするような方法で連結されていることを意味する。
本発明の導入遺伝子を含むAβペプチドポリヌクレオチドには、AβペプチドcDNAを含み、かつ修飾されたAβペプチドcDNAも含むものとする。本明細書に使用される、核酸の「修飾」には、参照配列に関して1つ、又はいくつかのヌクレオチド付加、欠失、又は置換を含み得る。核酸の修飾には、遺伝暗号の縮重のためにコードされたアミノ酸配列を変化しないか、又は保存的置換を生じる置換を含み得る。このような修飾は、ポリA尾部の付加などの、故意に作製された変異、又は核酸複製の間に突然変異として生じる変異に対応することができる。

また、胚性細胞を種々の発生時期に使用して、トランスジェニック動物の産生のための導入遺伝子を導入することができる。胚性細胞の発生の段階に応じて、異なる方法が使用される。このようなトランスフェクトされた胚性幹(ES)細胞は、その後に、胚にコロニー形成し、続いて胚盤胞-段階胚の胞胚腔にこれらを導入し、生じるキメラ動物の生殖系列に寄与する(Jaenischの文献、Science 240:1468-1474(1988)において概説される)。導入遺伝子がこのような選択のために手段を提供する場合、トランスフェクトされたES細胞の胞胚腔への導入の前に、トランスフェクトされたES細胞を種々の選択プロトコルに供して、導入遺伝子が組み込まれたES細胞の比率を濃縮することができる。或いは、PCRを使用して導入遺伝子を組み込んだES細胞をスクリーニングすることができる。

活性部位以外の部位(アロステリックな結合部位)に結合し、触媒活性を減少し、又は止める酵素の高次構造上の変化を生じさせる。
阻害剤と酵素との間の平衡がゆっくりと達成される競合阻害剤であり、(konが、緩徐である)おそらく、酵素、又は阻害剤において生じなければならない高次構造上の変化に起因し、たいてい遷移状態類似体であり、酵素濃度と同様の濃度にて有効であり(nM以下のKD値)、koff値が非常に低いため、この種の阻害剤は、「ほぼ」不可逆的である。
酵素で触媒される反応の遷移状態を模倣する競合阻害剤である。従って、酵素触媒作用は、遷移状態のエネルギーの低下によって生じ、従って、基質結合よりも遷移状態結合が選択される。
2つ以上の基質を含む反応については、2つ以上の基質に似ている構造的特徴を含む競合阻害剤、又は遷移状態類似体をデザインすることができる。
(活性部位定方向(directed)不可逆阻害剤)(競合的不可逆阻害剤)は、酵素(可逆的な、特異的結合)によって、続いて共有結合形成によって認識され、かつ、
基質、遷移状態、又は産物に構造的に類似し、薬物と標的酵素との間で特異的に相互作用することができ、
反応性官能基(例えば、求核試薬、-COCH2Br)を含み、共有結合を形成することができる。

競合的可逆的酵素阻害剤は、最も好ましい。
本明細書に使用される「EC活性」という用語は、QCによるピログルタミン酸(pGlu*)へのN末端グルタミン酸残基の分子内環化として定義される。従って、スキーム3を参照されたい。
QC阻害との相関を考慮すると、好ましい実施態様において、本方法、及び医療用途では、10μM以下の、より好ましくは1μM以下の、更により好ましくは0.1μM以下、若しくは0.01μM以下の、又は最も好ましくは0.001μM以下のQC阻害に対するKi、をもつ薬剤を利用する。実際に、マイクロモル、好ましくはナノモル、及び更により好ましくはピコモルの範囲以下のKi値をもつ阻害剤が、想定される。従って、便宜のために、「QC阻害剤」として活性薬剤が本明細書に記述されているが、このような命名法は、本発明の主題を特定の作用機序に限定することは意図されないことが理解されるであろう。
一般に、本方法、又は医療用途のQC阻害剤は、例えば1000g/モル以下、500g/モル以下、好ましくは400g/モル以下、及び更により好ましくは350g/モル以下、及び更に300g/モル以下の分子量をもつ小分子であるだろう。
本明細書に使用される「Aβペプチドに関連した疾患」という用語は、Aβペプチドによって特徴づけられ、及び/又は媒介される全ての疾患、障害、又は状態をいう。
本発明の方法、及び組成物は、Aβペプチドのエフェクターの評価に、並びに軽度認知障害、アルツハイマー病、ダウンにおける神経変性症候群、家族性デンマーク人痴呆症、及び家族性英国人痴呆症などのアミロイドに関連した疾患の治療、及び予防のための薬物、並びに治療薬の開発のために、特に有用である。
この実施例により、トランスジェニックストラテジーがAD病態における異なるペプチド、例えばAβ(N3pGlu)の役割を調査するための優れた機会を提供することがわかる。従って、導入遺伝子を適合させることにより、その他のN、及びC末端Aβ種をニューロン特異的に発現すること、並びにこれらの神経毒性の可能性を調査することもできる。
最後に、pGlu-Aβ(3-42)の蓄積は、ニューロンの喪失、長期増強、及び認識における機能障害を伴うので、tgN3E-42、及びtgN3Q-42マウスは、pGlu修飾されたAβペプチドの神経毒性を証明する。これらの特色の組み合わせは、これらの動物モデルに独特であり、従って、アルツハイマー型のような病態を持つ齧歯類モデルにおける大幅な前進ということになる。
該データは、tgN3E-42マウスの遺伝子改変により、ベースラインシナプス伝達、及び従って、正常ニューロン機能の両方、更にはシナプスの可塑性(LTP)が障害されたことを示す。LTP破壊は、学習と記憶の細胞相関がマウス脳のその領域において妨げられるため、海馬依存的な記憶機能の障害に寄与する。EPSP振幅の減少は、シナプスの喪失を反映し、これは、tgN3E-42ホモ接合性マウスにおいて観察されるニューロン変性と明らかに一致する。
更に好ましい交雑マウス系統は、QCのアイソザイムを発現する。
本発明に従って、配列番号:13〜15、22、及び23で示される、そのアイソフォーム、及びスプライスフォームを含むヒトQPCTL類;ラット(配列番号:19)、及びマウス(配列番号:20)からなる群から選択されるQPCTL類を発現する交雑マウス系統が好ましい。
この点に関して、QPCTL-アイソザイムの具体的なさらなる開示について、US 60/846,244に対して具体的に参照がなされる。この出願は、参照により本明細書に組み込まれる。
また、導入遺伝子がAβN3E-42(配列番号:1)、AβN3Q-42(配列番号:2)、AβN3E-40(配列番号:3)、及びAβN3Q-40(配列番号:4)から選択される少なくとも1つAβペプチドをコードする非ヒトトランスジェニック動物が提供される。
(a)本発明に従ったトランスジェニックマウス系統に検査薬剤を投与すること、

Aβペプチドに関連した疾患は、好ましくはアルツハイマー病、又はダウン症候群における神経変性である。
本発明の一つの実施態様において、Aβペプチドの効果の阻害剤が好ましい。
本発明の化合物は、酸付加塩、特に医薬として許容し得る酸付加塩に変換することができる。

更に、哺乳類に対してエフェクターを投与することにより、骨髄球性前駆細胞の増殖を抑制することが可能であろう。
製剤は、例えば溶媒、及び/又は担体と共に活性成分を伸ばすことによって、任意に乳化剤、及び/又は分散剤を用いて、都合よく調製してもよく、例えば、水が希釈剤として使用される場合、有機溶媒を任意に補助的な溶媒として使用することができる。
上記の開示は、一般に本発明を記述する。より完全な理解は、以下の実施例を参照することによって得ることができる。これらの実施例は、単に例証のみのために記述してあり、本発明の範囲を限定することは意図されない。具体的用語を本明細書に使用したが、このような用語は、説明的な意味で、かつ限定のためではないことが意図される。
(宿主株、及び培地)
ヒトQCの発現のために使用したピチア・パストリス(Pichia pastoris)株X33(AOX1、AOX2)は、製造業者の説明書(Invitrogen)に従って、培養し、形質転換し、及び解析した。P.パストリス(P. pastoris)のために必要とされる培地、すなわち緩衝したグリセロール(BMGY)複合培地、又はメタノール(BMMY)複合培地、及び発酵基礎塩培地は、製造業者の推奨に従って調製した。
全てのクローニング手順は、標準的な分子生物学技術を適用して行った。酵母における発現のためには、ベクターpPICZαB(Invitrogen)を使用した。pQE-31ベクター(Qiagen)を使用して大腸菌(E. coli)においてヒトQCを発現した。コドン38で開始する成熟QCのcDNAを6×ヒスチジンタグをコードするプラスミドとインフレームで融合した。プライマーpQCyc-1、及びpQCyc-2(WO2004/098625)を利用する増幅、及びサブクローニングの後、断片をSphI、及びHindIIIの制限部位を使用して発現ベクターに挿入した。
プラスミドDNAを大腸菌(E. coli)JM109において増幅して、製造業者(Qiagen)の推奨に従って精製した。使用した発現プラスミド(pPICZαB)では、3つの制限部位が直線化のために提供される。SacI、及びBstXIは、QC内のcDNAを切断したので、PmeIを直線化のために選択した。20〜30μgプラスミドDNAをPmeIで直線化して、エタノールによって沈殿して、無菌の脱イオン水に溶解した。次いで、10μgのDNAを、製造業者の説明書(Biorad)に従って、電気穿孔法によってコンピテントなP.パストリス(P. pastoris)細胞の形質転換のために適用した。選択は、150μg/mlゼオシン(Zeocin)を含むプレートを使用して行った。直線化プラスミドを使用する1回の形質転換により、数百の形質転換体を得た。
本質的に「ピチア属(Pichia)発酵過程指針」(Invitrogen)に記載されているように、QCの発現は、5lの反応器(Biostat B, B. Braun biotech)において行った。簡潔には、細胞を微量塩と、及び唯一の炭素源(pH 5.5)としてのグリセロールとを補った発酵基礎塩培地において培養した。約24時間の初期バッチ相、及びその後の約5時間の供給バッチ相の間に、大量の細胞が蓄積した。一旦200g/lの細胞湿重量が達成されたら、メタノールを使用して、およそ60時間の全ての発酵時間の間に3工程フィードプロフィールを適用して、QC発現の誘導を行った。その後、細胞を6000×g、4℃にて15分間の遠心分離によってQCを含む上清から除去した。NaOHの添加によってpHを6.8に調整し、生じる濁った溶液を37000×g、4℃にて40分間、再び遠心分離した。濁りが続く場合には、セルロース膜(孔幅0.45 μm)を使用して、さらなる濾過工程を適用した。
Hisタグを付けたQCは、最初に固定された金属アフィニティークロマトグラフィー(IMAC)によって精製した。典型的な精製では、1000mlの培養上清を、750mM NaClを含む50mMリン酸緩衝液pH 6.8で平衡化したNi2+を充填したChelating Sepharose FFカラム(1.6×20cm、Pharmacia)に、5ml/分の流速で、適用した。10カラム容積の平衡緩衝液、及び5カラム容積の5mMヒスチジンを含む平衡緩衝液で洗浄後、結合したタンパク質を、150mM NaCl、及び100mMヒスチジンを含む50mMリン酸緩衝液、pH 6.8に交換することによって溶出させた。生じる溶出液を20mM Bi-Tris/HCl(pH 6.8)に対して4℃にて一晩透析した。その後、QCを、透析緩衝液で平衡化した陰イオン交換クロマトグラフィーMono Q6カラム(BioRad)によって更に精製した。QCを含む画分を、4ml/分の流速を使用してカラムに充填した。次いで、カラムを、100mM NaClを含む平衡緩衝液で洗浄した。溶出は、それぞれ30、又は5カラム容積の240mM、及び360mM NaClを含む平衡緩衝液を生じる2つの勾配によって行った。6mlの画分を収集して、純度をSDS-PAGEによって解析した。均質なQCを含む画分を限外濾過によってプールして、濃縮した。長期貯蔵(-20℃)のために、グリセロールを50%の終濃度に添加した。タンパク質は、Bradford、又はGill、及びvon Hippelの方法に従って定量化した(Bradford、M. M. 1976 Anal Biochem 72、248-254;Gill、S.C. 及びvon Hippel、P.H. 1989 Anal Biochem 182、319-326)。
QCをコードする構築物をM15細胞(Qiagen)に形質転換して、37℃にて選択的LB寒天プレートを培養した。タンパク質発現は、1%のグルコース、及び1%のエタノールを含むLB培地において室温にて実施した。培養がおよそ0.8のOD600に達したときに、発現を0,1 mMのIPTGで一晩誘導した。凍結解凍の1サイクル後、細胞を、300mM NaCl、及び2mMヒスチジンを含む50mMリン酸緩衝液、pH 8.0中の2.5mg/mlのリゾチームの添加によって、4℃にておよそ30分間溶解した。溶液を37000×g、4℃にて30分間遠心分離によって浄化し、続いてガラスフリット(DNA分離)を適用する濾過、並びに粗製、及び微細な沈殿物に対してセルロースフィルターを適用する2回のさらなる濾過工程を行った。上清(約500ml)を1ml/分の流速でNi2+-親和性カラム(1.6×20cm)に適用した。QCの溶出は、150mM NaCl、及び100mMヒスチジンを含む50mMリン酸緩衝液で実施した。QCを含む画分を、限外濾過によって濃縮した。
パパイアラテックスからのQCは、BioCAD 700E(Perseptive Biosystems、Wiesbaden、Germany)を使用して、以前に報告された方法(Zerhouni, S.らの文献、1989 Biochim Biophys Acta 138, 275-290)の変更バージョンで調製した。50gのラテックスを、その中に記述されたように水に溶解して、遠心分離した。プロテアーゼの不活性化は、S-メチルメタンチオスルホナートで行い、生じる粗製抽出物を透析した。透析後、全上清を(21×2.5 cm i.d.)SP Sepharose Fast Flowカラム(100mM酢酸ナトリウム緩衝液、pH 5.0で平衡化した(流速3ml/分))に充填した。溶出は、2ml/分の流速で、酢酸ナトリウム緩衝液濃度を増加することによって、3工程で行った。最初の工程は、0.5のカラム床体積の0.1〜0.5M 酢酸緩衝液の直線勾配であった。第2の工程は、4カラム床体積の0.5〜0.68Mの緩衝液濃度で直線的に増大した。最後の溶出工程の間に、1カラム床体積の0.85M緩衝液を適用した。最も高い酵素活性を含む画分(6ml)をプールした。濃縮、及び0.02M Tris/HCl、pH 8.0への緩衝液交換を、限外濾過を介して行った(Amicon;膜10kDaの分子量分離)。
(蛍光定量的アッセイ)
全ての測定は、30℃にてマイクロプレート(BMG Labtechnologies)のためのNovoStarリーダーで行った。QC活性は、H-Gln-βNAを使用して蛍光定量的に評価した。試料は、250μlの最終的容積で、0.2mM蛍光発生基質、0.05M Tris/HCl、pH 8.0中の0.25Uピログルタミルアミノペプチダーゼ(Qiagen、Hilden、Germany)、及びQCの適切に希釈された一定分量からなった。励起波長/放出波長は、320/410nmであった。アッセイ反応は、グルタミニルシクラーゼの添加によって開始した。QC活性は、アッセイ条件下でのβ-ナフチルアミンの標準曲線から決定した。1単位は、記述された条件下で1分あたりにH-Gln-βNAから1μモルpGlu-βNAの形成を触媒するQCの量として定義される。
この新規アッセイを使用して、大部分のQC基質についての動力学的パラメーターを決定した。QC活性は、補助的酵素としてグルタミン酸脱水素酵素を利用して、以前の不連続アッセイ(Bateman, R. C. J.の文献、1989 J Neurosci Methods 30, 23-28)を使用して、分光光度的に解析した。試料は、250μlの最終的容積で、それぞれのQC基質、0.3mM NADH、14mM α-オキソグルタル酸、及び30U/mlグルタミン酸脱水素酵素からなった。反応をQCの添加によって開始して、340nmにて吸光度の低減のモニタリングによって8〜15分間追跡した。
阻害剤試験については、試料組成は、推定上の阻害化合物を添加したこと以外、上記と同じであった。QC阻害の迅速試験については、試料には、4mMのそれぞれの阻害剤、及び1KMの基質濃度を含んだ。阻害の詳細な研究、及びKi値の決定については、補助的酵素に対する阻害剤の影響を最初に調査した。全ての場合において、検出した酵素のいずれに対しても影響はなく、従って、QC阻害の信頼できる決定が可能であった。阻害定数は、GraFitソフトウェアを使用して、競合阻害についての一般的な方程式に対してプログレス曲線のセットをフィットすることによって評価した。
マトリックス支援レーザ脱離/イオン化質量分析は、リニアの飛行時間分析計を有するHewlett-Packard G2025 LD-TOFシステムを使用することにより実施した。機器には、337nm窒素レーザ、電位加速源(5kV)、及び1.0mの飛行管を備えた。検出器の操作は、陽イオンモードで行い、シグナルは、パーソナルコンピュータに連結したLeCroy 9350Mデジタルストレージオシロスコープを使用してを記録して、フィルターをかけた。試料(5μl)は、等量のマトリックス溶液と混合した。マトリックス溶液については、我々は、30mgの2',6'-ジヒドロキシアセトフェノン(Aldrich)、及び44mgのクエン酸水素二アンモニウム(Fluka)を1mlのアセトニトリル/0.1% TFA(1/1、v/v)水溶液に溶解することによって調製したDHAP/DAHCを使用した。少量のマトリックス-分析物-混合物(〜1μl)をプローブチップ(先端部)へ移して、直ちに真空チャンバ(Hewlett-Packard G2024A試料調製アクセサリ)において蒸発させ、迅速かつ均一な試料の結晶化を確実にした
(マウスQCのクローン化)
mQCのオープンリーディングフレームの単離のためのプライマーは、推定上のmQCをコードするPubMedヌクレオチドエントリーAK017598を用いてデザインした。プライマー配列は、以下の通りであった:センス
総RNA は、RNeasy Miniキット(Qiagen)を使用してマウス膵島細胞腺腫株化細胞β-TC 3細胞から単離し、SuperScriptII(Invitrogen)によって逆転写した。その後、mQCのcDNAを、Herculase Enhanced DNAポリメラーゼ(Stratagene)での50μlの反応において、産生された産物の1:12.5の希釈で増幅し、PCR Script CAMクローニングベクター(Stratagene)に挿入して、シーケンシングによって検証した。成熟したmQCをコードするcDNA断片を、プライマー
1-2μgのプラスミドDNAは、製造者の説明書(BioRad)に従って、電気穿孔法によってコンピテントなP.パストリス細胞の形質転換のために適用した。選択は、100μg/ml Zeocinを含むプレートで行った。mQC発現により組換え酵母クローンを試験するために、2mlのBMGYを含む10mlのコニカルチューブにおいて組換え体を24時間増殖させた。その後、酵母を遠心分離して、0.5% メタノールを含む2mlのBMMYに再懸濁した。約72時間、24時間毎にメタノールを添加することによってこの濃度を維持した。その後、上清におけるQC活性を決定した。最高の活性を示したクローンをその後の実験、及び発酵のために選択した。
mQCの発現は、5Lの反応器(Biostad B, B. Braun biotech, Melsungen, Germany)において行った。発酵は、pH 5.5にて微量の塩を補った基礎塩培地において実施した。最初に、バイオマスをバッチ相、及び約28時間唯一の炭素源としてグリセロールを有する供給バッチ相に蓄積させた。mQCの発現は、およそ65時間の全発酵時間の間、Invitrogenによって推奨される3工程プロフィールに従って、メタノール供給によって開始した。その後、細胞、及び濁度を、、6000×g、及び38000×gで、それぞれ15分、及び4時間の2段階の遠心分離工程によってmQCを含む上清から除去した。精製については、発酵ブロスを、約5ms/cmの伝導率に水で希釈して、0.05M リン酸緩衝液、pH 6.4で平衡化したStreamline SP XLカラム(2.5x100cm)に逆の流れの向き(15mL/分)に加えた。2カラム体積に対して、逆の流れ方向の平衡緩衝液での洗浄工程の後、タンパク質を、1.5M NaClを含む0.15M Tris緩衝液、pH 7.6を使用して、8mL/分の流速で前進方向で溶出した。QCを含む画分をプールし、1Mの終濃度に硫酸アンモニウムを添加した。生じる溶液を、4mL/分の流速で、ブチルセファロースFFカラム(1.6x13cm)に加えた。結合したmQCを、0.75M 硫酸アンモニウムを含む0.05M リン酸緩衝液、pH 6.8で5カラム体積に対して洗浄し、0.05M リン酸緩衝液、pH 6.8で、逆の流れの向きに溶出した。mQCを含む画分をプールして、0.025M Tris、pH 7.5に対する透析によって一晩脱塩した。その後、NaOHの添加によってpHを8.0に調整し、0.02M Tris、pH 8.1で平衡化したUno Qカラム(Bio Rad)に加えた(4.0mL/分)。平衡緩衝液を使用する洗浄工程の後、mQCを、0.18M NaClを含む同じ緩衝液を使用して溶出した。QC活性を示す画分をプールし、1M ビス‐Tris緩衝液、pH 6.0の添加によって、pHを7.4に調整した。mQCは、4℃にて1箇月まで安定であった。−20℃での長期貯蔵のためには、50% グリセロールを添加した。
(実施例1:トランスジェニックマウスの作成)
(トランスジェニックマウス)
マウス甲状腺刺激ホルモン放出ホルモン-Aβ融合タンパク質mTRH-Aβ(N3E-42)、及びmTRH(N3Q-42)の産生は、本質的に他に記述されたとおりである(Cynis, H.,らの文献、グルタミニルシクラーゼの阻害が、哺乳動物細胞、におけるピログルタミン酸形成を変化させる(Inhibition of glutaminyl cyclase alters pyroglutamate formation in mammalian cells), Biochim Biophys Acta 1764、1618-1625(2006))。それぞれのcDNAを、標準的な分子生物学技術を適用して、マウスThy-1配列を含むベクターpUC18に挿入した。マウスQCは、膵島細胞腺腫株化細胞β-TC 3から単離した。mQC cDNAをベクターpTG-CAGにクローン化した。全ての構築物は、シーケンシングによって検証した。トランスジェニックマウスは、受精したC57Bl6卵母細胞(genOway、Lyon、Franceによって産生されたPNI)の雄の前核注射によって産生した。次いで、注射された卵母細胞を満期発生のために養母に移植した。生じる子孫(それぞれの系統の3つのファウンダー)を、PCR解析によって導入遺伝子組込みについて、およびC57Bl6野生型マウスに対する交雑の後に、RT-PCRによって導入遺伝子発現について更に特徴づけた(それぞれの系統n=3-5)。最高の導入遺伝子mRNAを発現する系統を、更に繁殖させるため、及び交雑実験のために選択した(tgN3E-42 X QCマウスと命名)。該tgN3E-42×QCトランスジェニックは、健康に見え、神経学的異常に関する証拠を示さなかった。tgN3Q-42 PNIの3つのファウンダーのうちの1つのみが、子孫を出産した(tgN3Q-42マウスモデル)。しかし、不妊性ファウンダーの1つは、海馬CA1層において、生後7月齢にてニューロン喪失、及びAβ(N3pGlu)に対するニューロン免疫反応性を示し、これは、その他のファウンダーの子孫でも観察された。このファウンダー、及び他のファウンダーの子供において同一の表現型が観察されたことは、観察された表現型が組込み効果のためではないことを明らかに示唆する。全ての動物は、動物保護のためのドイツ指針に従って取り扱った。
cDNA構築物の品質は、アガロースゲル上に一定分量を泳動することによって検証、及び確認し、微量の分解物は、観察されなかった。最後に、診断用BglI、EcoRI、及びHindIII制限酵素を使用した切断解析により、予想される制限プロフィールを得た。TBA-tgプラスミドをEcoRIで消化し、導入遺伝子を含む7170-bp断片をベクターバックボーンから単離した。7.1kbのトランスジェニック構築物断片を更に精製して、希釈した後、受精した卵母細胞へ注射した。単離した導入遺伝子の純度、及び濃度は、アガロースゲル電気泳動によって検証した。
導入遺伝子のランダムな組込みの検出についてのスクリーニングは、PCR増幅によって達成した。2種類のPCRをデザインした(図7を参照されたい)。
・PCR1は、導入遺伝子のランダムな組込みイベントを効率的に検出するようにデザインされる。選択されたプライマー対は、導入遺伝子配列内の短いDNA配列の増幅を可能にし、特異的な505-bpのPCR産物を産生する。
・PCR2は、導入遺伝子の組込みイベントの完全性を評価するようにデザインされる。選択されたプライマー対は、プロモーターの5'領域、及びThy1遺伝子の3'領域に延長しているDNA配列の増幅を可能にし、特異的な6279-bp PCR産物を産生する。Thy1プロモーターカセットは、マウスゲノム配列のに由来するため、導入遺伝子の完全性を調査するために使用したPCRスクリーンも、また内因性Thy1遺伝子からの7413-bp産物の増幅を生じる。
マウスを麻酔して、氷冷リン酸塩緩衝食塩水(PBS)、続いて4%パラホルムアルデヒドによって経心的に灌流した。脳試料を慎重に解剖し、4℃にて4%リン酸緩衝ホルマリンにおいて後固定した。免疫組織化学は、4μmパラフィン切片上で行った。以下の抗体を使用した:4G8(Aβ17-24, Signet)、GFAP(Chemicon)、Iba1(Waco)、ユビキチン(Dako)、及び位置3にピログルタミン酸をもつAβに対するもの(Saido, T.C., らの文献、老人斑における異なるβ-アミロイドペプチド種であるAβ N3(pE)の優勢、及び差動的沈着(Dominant and differential deposition of distinct beta-amyloid peptide species, Abeta N3(pE), in senile plaques, Neuron 14, 457-466 (1995))。ビオチン化二次抗ウサギ、及び抗マウス抗体(1:200)は、DAKOから購入した。染色は、ベクタステインキット(Vector Laboratories)、及び色素原としてのジアミノベンジジンと共にABC法を使用して視覚化した。対比染色は、ヘマトキシリンで実施した。蛍光染色法については、AlexaFluor488-、及びAlexaFluor594抱合抗体(Molecular probes)を使用した。
tgN3E-42マウスの解析については、免疫組織化学的特性付けのために更に以下の抗体を使用した:Aβ/4G8(Acris)、Aβ/6E10(Calbiochem)、Aβ-N3pGlu-42(clone6)、GFAP(Dako)、NeuN(Chemicon)。ビオチン化二次抗ウサギ、及び抗マウスの抗体(1:250)は、Vector Laboratoriesから購入した。染色は、ベクタステインキット(Vector Laboratories)、及び色素原としてのジアミノベンジジンと共に、ABC法を使用して視覚化した。蛍光染色法については、Cy2、及びCy3抱合二次抗体(Dianova)を使用した。DAPI核酸染色(Molecular probes)は、蛍光免疫染色で行った。
脳を凍結した状態で重さを量り、完全プロテアーゼ阻害剤(Roche)を含む2.5mlの2% SDSにおいて、Dounceホモジェナイザで直接ホモジナイズした。ホモジネートを30秒間超音波処理(sonified)し、その後4℃にて1分間80.000gにて遠心した。上清を採取し、直接-80℃にて凍結した。生じるペレットを0.5mlの70%ギ酸(FA)に再懸濁して、30秒間超音波処理した。ギ酸は、9.5ml 1Mで中和し、一定分量を直接-80℃にて凍結した。Aβ(x-42)、及びAβ(N3pGlu)の両方のELISA測定のためには、SDS可溶化液を10倍希釈で使用した。Aβ(x-42)測定については、ギ酸可溶化液を10倍希釈で使用した。Aβ(N3pGlu-42)測定については、希釈されていないFA可溶化液を適用した。ELISA測定は、製造業者(IBL Co., Ltd. Japan)のプロトコルに従って行った。統計分析については、SDS-、及びFA-抽出から生じるAβ(x-42)、及びAβ(N3pGlu)濃度を集積した。
N末端が修飾されたAβペプチドの電気泳動分離は、他に記述されたように、15%尿素-PAGEゲルを適用して行った(Klafki, H. W., Wiltfang, J. 及びStaufenbiel, M.(1996)Anal. Biochem. 237, 24-29)。
タンパク質を、半乾燥条件下でニトロセルロース膜(Roth)へ移した。その後、膜をTBS-T(20mM Tris/HCl;pH 7.5;500mM NaCl, 0.05%(v/v)Tween20)中の3%(w/v)粉ミルクを使用してブロッキングした。Aβペプチドを、モノクローナル抗Aβ(1-16)抗体6E10を使用して検出した(Chemicon)。可視化については、ブロット膜を西洋ワサビペルオキシダーゼに抱合した二次抗マウス抗体(Cell Signaling)と共に、5%(w/v)粉ミルクを含むTBS-Tにおいてインキュベートし、その後、製造業者のプロトコルに従って、SuperSignal West Pico System(Pierce)を使用して現像した。
グループ間の相違は、一元配置分散分析法(ANOVA)、続いて不対t検定で試験した。全てのデータは、平均±標準誤差として与えた。不対t検定の有意水準は、以下の通りに与えられた:***P<0.001;**P<0.01;*P<0.05。生存率は、ログランク検定によって算出した。全ての算出は、Windows版のGraphPad Prism version 4.03(GraphPad Software, San Diego, CA, USA)を使用して行った。
一次スクリーン:一次スクリーンは、標準的な方法を利用して、観察評価による行動的、及び機能的なプロフィールを提供する(Rogers D.C.らの文献、1997. マウス表現型の行動的および機能的な解析:SHIRPA、包括的な表現型評価のための提唱されたプロトコル。(Behavioral and functional analysis of mouse phenotype: SHIRPA, a proposed protocol for comprehensive phenotype assessment.)Mamm Genome, 8:711-713)。改変したSHIRPAプロトコルを使用して、さらなる行動試験を妨げ得る一般的な健康、及び特定機能(筋肉、及び低下した運動ニューロン機能、脊髄小脳、感覚、神経精神医学的、及び自律神経機能)を高スループットスクリーンにおいて調べた。
評価は、ホームケージにおける社会的行動(「ホームケージ観察」)、及び90秒間の透明なプレキシガラス活動領域における単一の動物の乱されていない行動(「個々の観察」)を観察することで開始した。マウス行動のこのモニタリングに続いて、さらなる特徴づけのための一連の短い試験を行った:聴覚驚愕反射、吊り下げ(hanging)行動、視覚置き直し(visual placing)、落下行動、正向反射、姿勢反射、ネガティブ重力走性、吊り下げワイヤー、耳単収縮(ear twitch)、頬髭単収縮(whiskers twitch)、及び眼の瞬き。最後に、動物の首をつかんで、異常形態学的な異常について調べた。評価を完了するために、動物の体重を計り、次いでそのホームケージに動物を戻した。
マウスにおける状況的(contextual)、及びきっかけを与えられた恐怖反射を研究するために、市販のコンピュータ制御の「恐怖条件付けシステム(FCS)」(TSE System, Bad Homburg, Germany)を使用した。
実験設定は、Oliver Stiedlのプロトコルに従って選択した(Stiedl O.らの文献、2004マウスにおけるコンテクスチュアル恐怖条件付けの間の行動および自律神経動態(Behavioral and autonomic dynamics during contextual fear conditioning in mice.) Auton Neurosci. Basic and Clinical 115(1-2):15-27)。FCSにおける調査は、2日連続で行い、3相に分けた。
第1相(トレーニング/条件付け試行):トレーニングは、常に照射された(〜300lx)恐怖条件付け箱内の透明なアクリルコンパートメントにおいて行った。拡声器により、一定の、ホワイトバックグラウンドノイズ(68dB SPL)を提供した。270秒の休止の後、マウスに30秒間、聴覚キュー(10kHz、75dB SPL)を与えた(条件刺激)。2秒のフットショック(0,7mA、定電流;無条件刺激)を、音の最後の2秒間に施した。マウスは、ショック終了の30秒後に、これらのホームケージに返した。
第3相(音依存的保持):条件刺激(音)についての記憶は、新規状況(同じような大きさの黒いアクリル箱、黒色のため光強度が減少され、ショックグリッドの代わりに平らな底板)において、第2相の1時間後に試験した。新規状況において自由に探索した270秒後に、第1相と同じ音を180秒間適用し、行動をFCSによって自動的に記録した。
明<->暗探索モデルにおける探索活動の自動評価(Crawley J. N.の文献(2007)私のマウスでどうかしましたか:不安症に関連した行動。(What's Wrong With My Mouse: Anxiety-Related Behaviors.) Wiley, 第2版、 240-241)は、PhenoMaste装置(TSE System, Bad Homburg, Germany)用の明暗箱インサートモジュールで行った。プレキシガラスチャンバーから成るインサートモジュールは、小さな通路を含む黒いプレキシガラスの箱によって、2つのコンパートメントに不均等に分けられた(チャンバの1/3〜2/3)。大きなコンパートメントは、開放的で、明るく照射され、一方で、小さなコンパートメントは、閉鎖的で、暗かった。動物を、明るく照らされたコンパートメントに個々に置き、2つのコンパートメントを10分間自由に探索させた。フォトセルのグリッドが自動的にマウスの活動(2つのコンパートメント間の移動数、それぞれのコンパートメント中で費やした時間、それぞれのコンパートメントにおいて移動する距離)を検出した。
(Y-迷路:)自発的交替率(spontaneous alternation rates)は、30cm×8cmのアームサイズをもつ黒いプラスチック材料から構築される三角形のY形の迷路を使用して評価した。20分の試験セッションの間に、それぞれのマウスをランダムに一つのアームに置き、迷路を介して自由に移動させた(Frenois, F., Cador, M., Caille, S., Stinus, L.及びLe Moine, C.の文献(2002)ナロキソンで沈殿するモルヒネ禁断の動機づけ、および体細胞成分におけるで神経相関する(Neural correlates of the motivational and somatic components of naloxone-precipitated morphine withdrawal.) Eur J Neurosci., 16, 1377-1389)。交替(alternation)は、重複する3回のセットにおいて3つのアームへ連続して侵入するものとして定義した。侵入は、マウスが4つの足で全てアームに入るとすぐに連続するものと定義した。交替率は、可能な交替数に対するに実際の比率として算出した。匂いを嗅ぐきっかけを減少させるために、迷路は、30%エタノール、60%水、及び10%無臭の石鹸を含む溶液で清潔にした。
十字迷路は、黒いプラスチック材料からなる(アームサイズ:30.0cmの長さ、8.0cmの幅、15.0cmの壁の高さ)。隣接するアームは、90°の位置にある。4つのアームは、8.0cmの四方と測定される中心空間から伸張する。従って、動物は、中心空間を介してアームを訪問する。20.0分の試験セッションの間に、それぞれのマウスを、最初にランダムに一つのアームに置き、迷路をとおって自由に横断させた。個々のアームを1〜4と記した。交替は、重複する4回のセットでの連続侵入で4つの異なったアームに侵入することとして定義する(例えば、2,3,4,1、又は4,2,3,1であり、1,2,3,2ではない)。侵入は、マウスが4つの足で全てアームに入るとすぐに連続するものと定義した。交替率は、観察の期間の間に、全体で行われた交替に対する実際のものの比率として算出される。匂いを嗅ぐきっかけを減少させるために、迷路は、それぞれの試行の後、30%エタノール、60%水、及び10%無臭の石鹸を含む溶液で清潔にした。試験は、適度の白光条件下で行われる。より短い試験のための時間枠(すなわち、10分)mお可能である。
T迷路は、Gerlaiによって提供される手段によって使用した(Gerlai, R.の文献(1998)T-迷路における新たな連続的交替課題は、マウスにおける海馬の機能障害を検出する。系統比較および病変研究。(A new continuous alternation task in T-maze detects hippocampal dysfunction in mice. A strain comparison and lesion study.) Behav Brain Res., 95, 91-101)。該装置は、黒い床、及びギロチンドアを有する黒いプラスチック材料でできていた。マウスの試験は、一回のセッションからなり、これは、1回の強制選択試行で始まり、14回の自由選択試行が続く。(i)強制選択試行:最初の試行において、2つの目標アームの1つは、ギロチンドアを降ろすことによって遮断される。マウスがスタートアームから開放された後、それは迷路を探索して、開放アームに入り、開始位置に戻るであろう。マウスがスタートアームに戻るとすぐにギロチンドアを降ろし、動物を5秒間閉じこめた。(ii)自由選択試行:スタートアームのドアを開いた後、動物は、全てのギロチンドアが開放されているので、両方の目標アームのどちらかを自由に選ぶことができる。一旦マウスが目標アームに入ると、別の目標アームは閉じられる。マウスがスタートアームに戻ったとき、次の自由選択試行は、スタートアームにおいて5秒拘束した後に開始した。試験セッションは、30分後、又は14回の自由選択試行が行われた後に終了した。動物は、課題の間に決して手で触られず、実験者は、試験動物の遺伝子型を知らされなかった。交替比率は、交替選択の数を全ての選択数で割ることによって、それぞれの動物について算出した。所与の時間枠おいて8回未満の選択を行った動物は、解析から除外した。
(クラスピング(clasping)試験:)クラスピング行動を試験するために、マウスを30秒間尾部によって吊り下げて、後肢を握っている時間を記録した。0秒のクラスピング期間には、0のスコアを、1〜10秒は、スコア1、10〜20秒はスコア2、20秒以上のクラスピングは、スコア3を与えた(Nguyen, T., Hamby, A 及びMassa, S. 装置:高架プラス迷路は、Lister(1987)の記述に従って敷設した。これは、5×5cmの中心の四角のプラットフォームをもつ黒いプレキシガラスの床を有し、そこから、透明なプレキシガラスでできた0.25cmの高さの縁をもつ2つの45×5cmの開放アーム、及び40cmの高さの壁をもつ2つの45×5cmの閉じたアームが放射状に広がる。移動を測定するために、4つのアームのそれぞれに沿って、白線を半分描いた。装置は、プラス形の合板基礎の上に、床の上に45cm上げた。装置は、60ワットの赤い電球で照射された2×5 mの研究室に置いた。
(手順:)マウスは、これらのホームケージ中で試験室に運んだ。マウスは、常にこれらの尾部の根本によって扱った。マウスは、一度に一匹ずつ、開放アームに面しているプラス迷路の中心の四角に置いた。次いで、マウスを、5分間装置を探索させた。装置から約1mにて静かに座っている観察者は、迷路上の動物の行動を記録した。迷路の中心より150cm上に置かれたビデオカメラも、行動を記録した。行動は、ライムライトを使用して記録した。5分後、マウスをこれらの尾部の根本によって迷路から取り出して、これらのホームケージに返した。次いで、迷路を70%エチルアルコールの溶液で清潔にし、検査の間に乾燥させた。

4. 閉じたアーム期間:動物が閉じたアームにおいて費やした時間の長さ。
5. 中心の四角への侵入:動物が全ての4つの足で中心の四角に侵入した頻度。
6. 中心の四角期間:動物が中心の四角において費やした時間の長さ。
7. 頭部を下げる:動物が開放アームの側面から床の方へ頭部を下げた頻度。
8. 伸ばす留まるの姿勢:動物が頭部、及び肩部を前に伸ばし、続いて元の位置に戻す頻度。
9. 立ち上がり:動物が後肢で立っているか、又は前足で迷路の壁にもたれる頻度。
10. 探索無し行動:グルーミング、又はマウスが移動していない任意の時間。
11. 排尿:尿のたまり、又は線の数。
12. 排便:生じた糞便(fecal boli)の数。
13. 移動:動物が開放、及び閉じたアームに描かれた線を横切った回数。
Claims (37)
- AβペプチドをコードするDNA導入遺伝子を含む細胞を含む、Aβペプチドを過剰発現するためのトランスジェニック非ヒト動物。
- 前記動物が前記導入遺伝子に対してヘテロ接合性である、請求項1記載のトランスジェニック非ヒト動物。
- 前記記動物が前記導入遺伝子に対してホモ接合性である、請求項1記載のトランスジェニック非ヒト動物。
- 前記動物がマウスである、請求項1〜3のいずれかに記載のトランスジェニック非ヒト動物。
- 前記導入遺伝子がマウス起源である、請求項1〜4のいずれかに記載のトランスジェニック非ヒト動物。
- 前記導入遺伝子がヒト起源である、請求項1〜4のいずれかに記載のトランスジェニック非ヒト動物。
- 前記導入遺伝子が組換遺伝子である、請求項1〜6のいずれかに記載のトランスジェニック非ヒト動物。
- 前記組換え導入遺伝子がキメラ、又はヒト化されたポリペプチドをコードする、請求項7記載のトランスジェニック非ヒト動物。
- 前記導入遺伝子がAβN3E-42(配列番号:1)、AβN3Q-42(配列番号:2)、AβN3E-40(配列番号:3)、及びAβN3Q-40(配列番号:4)の少なくとも1つをコードする、請求項1〜8のいずれかに記載のトランスジェニック非ヒト動物。
- 前記導入遺伝子がAβN3E-42(配列番号:1)、及びAβN3Q-42、(配列番号:2)の少なくとも1つをコードする、請求項1〜8のいずれかに記載のトランスジェニック非ヒト動物。
- 前記導入遺伝子が組織特異的プロモーターに作動可能に連結されている、請求項1〜10記載のいずれかに記載のトランスジェニック非ヒト動物。
- 加えて、グルタミニルシクラーゼ、又はグルタミニル-ペプチドシクロトランスフェラーゼ様タンパク質をコードするDNA導入遺伝子を含む細胞を含む、請求項1〜11記載のいずれか1項記載のトランスジェニック非ヒト動物。
- 前記グルタミニルシクラーゼ、又はグルタミニル-ペプチドシクロトランスフェラーゼ様タンパク質が、ヒトisoQC(配列番号:14、又は15)、マウス(配列番号:20)、カニクイザル(配列番号:16)、アカゲザル(配列番号:17)、ネコ(配列番号:18)、ラット(配列番号:19)、ウシ(配列番号:21)、又は少なくとも50%/75%の配列同一性/類似性を有するその類似体から選択される、請求項12記載のトランスジェニック非ヒト動物。
- 前記グルタミニルシクラーゼ、又はグルタミニル-ペプチドシクロトランスフェラーゼ様タンパク質が配列番号:13〜15、22、又は23に記載のヒトQC、若しくはイソQCのアイソフォーム、又はスプライスフォーム;ラットQC(配列番号:19)、又はマウス(配列番号:20)である、請求項12、又は13記載のトランスジェニック非ヒト動物。
- 前記グルタミニルシクラーゼ、又はグルタミニル-ペプチドシクロトランスフェラーゼ様タンパク質が配列番号:13、又は配列番号:20の1つである、請求項12〜14のいずれか1項記載のトランスジェニック非ヒト動物。
- インビボでAβペプチド効果を阻害し、又は促進する生物学的に活性な薬剤をスクリーニングする方法であって:
a)請求項1〜15のいずれかに記載のトランスジェニック非ヒト動物に試験薬剤を投与すること、及び、
b)産生されたAβペプチドの効果に対する薬剤の効果を決定すること、
を含む、前記方法。 - 前記導入遺伝子がマウス起源である、請求項16〜19のいずれかに記載の方法。
- 前記導入遺伝子がヒト起源である、請求項16〜19のいずれかに記載の方法。
- 前記導入遺伝子が組換遺伝子である、請求項16〜21のいずれかに記載の方法。
- 前記組換え導入遺伝子がキメラ、又はヒト化されたポリペプチドをコードする、請求項22記載の方法。
- 前記導入遺伝子がAβN3E-42(配列番号:1)、AβN3Q-42(配列番号:2)、AβN3E-40(配列番号:3)、及び/又はAβN3Q-40(配列番号:4)をコードする、請求項16〜23のいずれかに記載の方法。
- 前記導入遺伝子がAβN3E-42(配列番号:1)、及び/又はAβN3Q-42(配列番号:2)をコードする、請求項16〜24のいずれかに記載の方法。
- 請求項1〜15のいずれかに記載のトランスジェニック非ヒト動物に由来する細胞、又は株化細胞。
- プロモーターに作動可能に連結されたAβペプチドをコードするトランスジェニックヌクレオチド配列を、マウスのゲノムに組み込んで含むトランスジェニックマウスであって、Aβペプチド効果のAβペプチド阻害剤で逆転させ、又は寛解させることができる表現型を示す、前記マウス。
- 前記マウスがAβペプチドを過剰発現する、請求項27記載のマウス。
- 前記マウスがAβペプチドに対してヘテロ接合性である、請求項27、又は28記載のマウス。
- 前記マウスがAβペプチドに対してホモ接合性である、請求項27、又は28のマウス。
