Description
本発明は、活性化合物、具体的にはPARPの活性を阻害する活性を有する、形態Lとして化合物Aを提供する。
活性化合物または活性化合物を含む医薬組成物は、全身投与/末梢投与であるか所望の作用部位とは無関係に、簡便な投与経路によって被験者に投与することができ、それには経口(例:経口摂取により);局所(例えば経皮、経鼻、眼球、口腔および舌下など);肺(例:例えばエアロゾルを用いた、例えば口もしくは鼻を介した吸入または通気療法);直腸;膣;例えば皮下、皮内、筋肉、静脈、動脈、心臓内、硬膜内、脊髄内、嚢内、被膜下、眼窩内、腹腔内、気管内、表皮下、関節内、クモ膜下および胸骨内などの注射による非経口;例えば皮下または筋肉でのデポー剤埋込物によるものなどがあるが、これらに限定されるものではない。
活性化合物を単独で投与することは可能であるが、それを1以上の製薬上許容される担体、補助剤、賦形剤、希釈剤、充填剤、緩衝剤、安定剤、保存剤、潤滑剤その他の当業者には公知の材料ならびに適宜に他の治療薬もしくは予防薬とともに、上記で定義の少なくとも1種類の活性化合物を含む医薬組成物(例えば、製剤)として提供することが好ましい。
活性化合物および活性化合物を含む組成物の適切な用量は患者ごとに変動し得ることは明らかであろう。至適用量の決定には通常、本発明の治療のリスクまたは有害な副作用に対する治療効果レベルのバランスを取る作業が関与する。選択される用量レベルは、これらに限定されるものではない、特定の化合物の活性、投与経路、投与時刻、化合物の排泄速度、治療期間、併用される他の薬剤、化合物および/または材料、ならびに患者の年齢、性別、体重、状態、全身の健康および病歴などの多様な要素によって決まる。化合物の量および投与経路は最終的には、医師の裁量に委ねられる。ただし一般に用量は、実質的に有害または有毒な副作用を起こすことなく、所望の効果を達成する作用部位での局所濃度を与えるようなものとする。
粉末X線回折
粉末X線回折は、Bruker D5000回折計により記録した(X線波長1.5418Å Cu線源、電圧40kV、フィラメント放出40mA)。試料は、2〜40°(2θ)の範囲で、0.02°のステップ幅および1秒のタイムカウントでスキャンした。
DSCは、TSO801ROロボットシステムを備えたMettler DSC820Eを使用して記録した。通常、5mg未満の物質(穴の開いた蓋が取り付けられている40μLアルミニウム皿に入れたもの)を毎分10℃の一定の加熱速度で25℃〜325℃の温度範囲で加熱した。窒素パージガスを100ml/分の流速で使用した。
1H NMRスペクトル は、Bruker DPX400光度計を用いてそれぞれ400 MHzにて記録した。化学シフトは内部標準のテトラメチルシランを参照したδスケールにおけるppmで読み取った。別段の記載が無い限り、全てのサンプルはDMSO-d6中に溶解した。
質量スペクトルは、構造の確認にタンデム型質量分析(MS/MS)を使用して、Agilent XCTイオントラップ質量分析計に記録した。この装置は、陽イオンエレクトロスプレーモードで操作した。
2-フルオロ-5-[(4-オキソ-3,4-ジヒドロフタラジン-1-イル)メチル]安息香酸(D)(15.23g、51.07mmol)をアセトニトリル(96ml)中に窒素下で撹拌しながら懸濁した。ジイソプロピルエチルアミン(19.6ml、112.3mmol)を加えた後、1-シクロプロピルカルボニルピペラジン(I)(9.45g、61.28mmol)およびアセトニトリル(1ml)を加えた。反応混合物を18℃に冷却した。

4-(3-{[4-(シクロプロピルカルボニル)ピペラジン-1-イル]カルボニル}-4-フルオロベンジル)フタラジン-1(2H)-オン(化合物A)(20.00g、44.66mmol)を、水(50ml)とエタノール(150ml)の混合物に懸濁した。この懸濁液を撹拌しながら加熱還流した。次に得られた溶液を70℃まで冷却し、濾過した。濾過パッドを水(8ml)とエタノール(22ml)の混合物で洗浄した。
23 mgの形態Aとしての化合物 Aをバイアルに量り入れた。この固体に、8.5 mLのメタノールおよび1.5 mLの水 (すなわち15%水v/v)からなる溶液0.25mLを加えた。得られたスラリーを70℃まで加熱し、濾過して、キャップしたバイアル中にて周囲温度になるまでゆっくりと冷ました。結晶化によって固体を生じ、この固体を真空オーブンによって乾燥した。生成物は形態Lとしての化合物 Aである。
およそ50mgの形態Aとしての化合物 Aを反応チューブに加え、そしておよそ1mlのエタノール/水 (80:20 v/v)を加えた。この混合物を40℃にて3週間、スラリー化した。サンプルを濾過し、フィルター上で10分間乾燥した後、ベンチで一晩乾燥した。生成物は形態Lとしての化合物 Aである。
実施例2と同様の方法で、形態Aとしての化合物 Aをエタノール/水 (80:20 v/v)混合物中にて3週間、60℃にてスラリー化して形態Lとして化合物 Aを得た。
およそ30 mgの形態Aとしての化合物 Aを反応チューブに加え、約1mlのエタノール/水 (80:20 v/v)を加えて形態Aの形態A飽和懸濁液を作製した。この飽和懸濁液に、30 mgの形態Lとしての化合物 Aを加えた。混合物を25℃にて3日間スラリー化した。サンプルを濾過し、フィルター上で10分間乾燥した後、ベンチで一晩乾燥した。生成物は形態Aを含まない、形態Lとしての化合物 Aである。
阻害作用
本活性化合物の阻害作用を評価するため、以下のアッセイを用いてIC50値を求めた。
阻害率(%)=100−[100×(未知のcpm−平均負cpm)/(平均正cpm−平均負cpm)]
IC50値(酵素活性の50%が阻害される濃度)を計算した。IC50値は異なる濃度の範囲、通常は10μM〜0.001μMにわたって求められる。かかるIC50値は、化合物の効力が増加したことを確認するための比較値として用いる。
活性化合物の増強ファクター(PF50)は、対照細胞増殖のIC50を細胞増殖+PARP阻害剤のIC50で割った比として計算される。対照と化合物で処理された細胞の両方の増殖阻害曲線は、アルキル化剤メタンスルホン酸メチル(MMS)の存在下のものである。試験化合物は0.2マイクロモルの固定濃度で使用した。MMSの濃度は0〜10μg/mlであった。
細胞増殖をスルホローダミンB(SRB)アッセイによって評価した(Skehan, P.ら, (1990) New colorimetric cytotoxicity assay for anticancer-drug screening. J. Natl. Cancer Inst. 82, 1107-1112)。2,000個のHeLa細胞を平底96ウェルマイクロタイタープレートの各ウェルに100μlの容量で播種し、37℃で6時間インキュベートした。細胞を培地単独で置きかえるか、最終濃度が0.5、1もしくは5μMのPARP阻害剤含有培地で置き換えた。細胞をさらに1時間増殖させ、その後、一連の濃度(一般に0、1、2、3、5、7および10μg/ml)のMMSを未処理細胞またはPARP阻害剤処理細胞に添加した。PARP阻害剤のみで処理した細胞を使用して、PARP阻害剤による増殖阻害を評価した。
Claims (19)
- 結晶形態Lとしての4-[3-(4-シクロプロパンカルボニル-ピペラジン-1-カルボニル)-4-フルオロ-ベンジル]-2H-フタラジン-1-オン。
- 示差走査熱量測定(DSC)において毎分10℃で25℃〜325℃まで加熱した時、198.5℃±1℃で融解し始める、請求項1〜3のいずれか1項に記載の化合物。
- 結晶形態Aの4-[3-(4-シクロプロパンカルボニル-ピペラジン-1-カルボニル)-4-フルオロ-ベンジル]-2H-フタラジン-1-オンから結晶形態Lの4-[3-(4-シクロプロパンカルボニル-ピペラジン-1-カルボニル)-4-フルオロ-ベンジル]-2H-フタラジン-1-オンを得る方法であって、形態Aまたは形態A およびLの混合物として4-[3-(4-シクロプロパンカルボニル-ピペラジン-1-カルボニル)-4-フルオロ-ベンジル]-2H-フタラジン-1-オンを必要に応じて最大で 30% 水v/vを含む有機溶媒中にスラリー化することを含む、上記方法。
- 請求項1〜4のいずれか1項に記載の化合物と製薬上許容可能な担体または希釈剤を含む、医薬組成物。
- ヒトまたは動物の身体の治療方法に使用するための請求項1〜4のいずれか1項に記載の化合物。
- ヒトまたは動物の身体の治療におけるPARPの阻害方法に使用するための請求項1〜4のいずれか1項に記載の化合物。
- PARP活性の阻害剤の製造における請求項1〜4のいずれか1項に記載の化合物の使用。
- 血管疾患;敗血症性ショック;虚血性障害;神経毒性;出血性(haemorraghic)ショック;ウィルス感染;またはPARPの活性を阻害することより寛解される疾患の治療用薬剤の製造における、請求項1〜4のいずれか1項に記載の化合物の使用。
- 癌治療において補助剤(adjunct)として使用するための、あるいは電離放射線または化学療法剤による治療のための腫瘍細胞を増強するための、薬剤製造における、請求項1〜4のいずれか1項に記載の化合物の使用。
- 個体における、相同組換え(HR)依存的DNA DSB修復経路に欠損のある癌の治療に使用するための薬剤の製造における、請求項1〜4のいずれか1項に記載の化合物の使用。
- 前記癌が、HRによるDNA DSB修復能力が正常細胞に比べて低下または抑制された1個または複数の癌細胞を含む、請求項12に記載の使用。
- 前記癌細胞がBRCA1またはBRCA2欠損表現型を有する、請求項13に記載の使用。
- 前記癌細胞がBRCA1またはBRCA2に欠損がある、請求項14に記載の使用。
- 前記個体が、HR依存的DNA DSB修復経路の成分をコードする遺伝子中の突然変異に関してヘテロ接合である、請求項12〜15のいずれか1項に記載の使用。
- 前記個体がBRCA1および/またはBRCA2中の突然変異に関してヘテロ接合である、請求項16に記載の使用。
- 前記癌が乳癌、卵巣癌、膵臓癌または前立腺癌である、請求項12〜17のいずれか1項に記載の使用。
- 前記治療が電離放射線または化学療法剤の投与をさらに含む、請求項12〜18のいずれか1項に記載の使用。
