Description
ヘリコバクターピロリのゲノムDNAはQIAamp DNA Stool Mini Kit(Qiagen)を使用して抗原陽性患者から単離された。
E.coli BL21コンピテント細胞が挿入物を含んだpET28aプラスミドで変質あるいはガン化され、カナマイシン(30μg/ml)の存在下、37℃にて10mlのLB培地内において培養された。
ヘリコバクターピロリは、ピルビン酸塩の酸化脱炭酸作用からエネルギーを得るための基本的反応に関与するため、フラボドキシンはヘリコバクターピロリの生存に必須である。
その後、それらはIPTGで誘導され、3時間の誘導(光密度約0.8)後に−80℃で保存された。
本発明の阻害物質のヘリコバクターピロリのフラボドキシンに対する特異結合は等温適定熱量計(ITC)により確認された。この技術は標的タンパク質に対するそれぞれの化合物の親和性を測定するだけではなく、結合のエンタルピー成分およびエントロピー成分の貢献度をも測定する。
Claims (9)
- nは1から3であり、R1はCXmH3−mの基であり、このXはFまたはClであり、R2はNO2、COOHまたはSO3H基であることを特徴とする請求項1記載の化合物。
- nは2であり、mは3であり、R2はNO2であることを特徴とする請求項1または2記載の化合物。
- XはFであることを特徴とする請求項1から3のいずれかに記載の化合物。
- 感染症の治療に利用される請求項1から5のいずれかに記載の化合物を含んでいることを特徴とする薬剤組成物。
- 感染症はヘリコバクターによって引き起こされることを特徴とする請求項6記載の薬剤組成物。
- 感染症はヘリコバクターピロリによって引き起こされることを特徴とする請求項7記載の薬剤組成物。
- 感染症は胃炎、胃十二指腸潰瘍、リンパ腫または胃ガンであることを特徴とする請求項6から8のいずれかに記載の薬剤組成物。
