Description
一態様においては、本発明は、イヌRNAポリメラーゼI調節配列を含む単離された核酸を提供する。他の態様においては、本発明は、そのような核酸を含む発現ベクターおよび細胞ならびに感染性インフルエンザウイルスなどのインフルエンザウイルスを作製するためのそのような核酸の使用方法を提供する。
インフルエンザの大流行は、インフルエンザ疾患に起因する罹患率および死亡率の劇的な広範囲の増加により定義される。いくつかの因子が組み合わさって、集団における低い程度の免疫およびウイルスがヒトの間で伝染し得る効率などの流行の重篤度および程度を調節する。後者は一般的には、ウイルス自体だけでなく、集団の密度ならびに地域への、および地域からの移動の容易性によっても影響される。大流行の原因となるウイルスは、一般的には、集団の大多数が以前に経験しておらず、従って、それに対する免疫をほとんど有さないか、または全く有さない、最近出現した抗原変異体である。さらに、効率的なヒトからヒトへの伝染は、ヒト集団に人畜共通伝染性の動物ウイルスを導入する場合、急速な拡散のための必要条件であり、このウイルスはヒトにおける複製に適合し、効率的に伝染することができなければならない。

インフルエンザウイルスのプラスミドレスキューは、一般的には、好適な宿主細胞中で、ウイルスタンパク質を発現させ、ウイルスゲノムRNAを転写させるための発現ベクターの導入を含む。一般的には、RNAポリメラーゼI酵素は、ウイルスゲノムとしての使用にとって好適な末端を有する転写物を産生するため、ウイルスゲノムRNAの転写を、これらの酵素を用いて実施する。かくして、RNA pol Iプロモーターおよび他の調節エレメントを用いて、プラスミドレスキューの間にゲノムRNAの転写を開始させる。不幸なことに、RNA pol Iプロモーターは、高度に種特異的である。すなわち、1つの種に由来するRNA pol Iは、非関連の種に由来するRNA pol Iプロモーターに効率的に結合することができるか、またはできない。従って、RNA pol Iプロモーターの利用可能性は、プラスミドレスキューを実施することができる細胞を制限する。本発明より以前には、イヌ細胞中でのプラスミドレスキューは不可能であった。初めて、以下のような本発明の開示に基づいて、イヌ細胞中でのプラスミドレスキューが可能である。

別の態様においては、本発明は、細胞培養物からウイルスを組換え的にレスキューするのに有用な発現ベクターなどの、本発明の核酸を含むベクターを提供する。一般的には、前記発現ベクターは、その生活環の間に規定の末端を有するRNAの産生を必要とする当業者には公知の任意のウイルスをレスキューするのに有用である。例えば、上記で考察されたように、インフルエンザウイルスゲノムRNAは、効率的に複製され、組換え系中にパッケージングされる規定の5'および3'末端を有するべきである。参照により本明細書に組み入れられるものとする、Neumannら(2002), 83:2635-2662中の概説も参照されたい。以下の考察は、インフルエンザと共に使用するのに好適な発現ベクターに焦点を当てるものである;しかしながら、本発明のベクターを用いて、他のウイルスをレスキューすることもできることに留意すべきである。

本発明の発現ベクターを用いて、ウイルスゲノムに対して異種である配列を発現するキメラウイルスを作製することもできる。vRNA(複数も可)または対応するcRNA(複数も可)の発現を指令する発現ベクターを、新規な感染性組換えマイナス鎖RNAウイルスもしくはキメラウイルスを生成するウイルスタンパク質の発現を指令する発現ベクターと共に宿主細胞中に導入する。例えば、米国特許出願公開第US20040002061号を参照されたい。これらのウイルス中で操作することができる異種配列としては、アンチセンス核酸およびリボザイムなどの核酸が挙げられる。あるいは、ペプチドまたはポリペプチドを発現する異種配列を、これらのウイルス中に操作することができる。これらのウイルス中に操作することができる以下のペプチドまたはポリペプチドをコードする異種配列は、1)病原体に特徴的である抗原;2)自己免疫疾患に特徴的である抗原;3)アレルゲンに特徴的である抗原;および4)腫瘍に特徴的である抗原を含む。例えば、本発明のキメラウイルス中に操作することができる異種遺伝子配列としては、限定されるものではないが、ほんの数例を挙げれば、gp160などのヒト免疫不全ウイルス(HIV)のエピトープ;B型肝炎ウイルス表面抗原(HBsAg);ヘルペスウイルスの糖タンパク質(例えば、gD、gE);ポリオウイルスのVP1;ならびに細菌および寄生虫などの非ウイルス病原体の抗原決定基が挙げられる。
本発明は、ヘルパーウイルスの非存在下で、タンパク質発現ベクターおよび本発明の発現ベクターを発現するvRNAまたは対応するcRNAを宿主細胞中に導入することにより、感染性組換えマイナス鎖RNAウイルスを作製する方法を提供する。好ましくは、前記宿主細胞は、イヌ細胞、例えば、MDCK細胞である。本発明はまた、ヘルパーウイルスの存在下で、タンパク質発現ベクターおよび本発明の発現ベクターを発現するvRNAまたは対応するcRNAを宿主細胞中に導入することにより、感染性組換えマイナス鎖RNAウイルスを作製する方法を提供する。前記宿主細胞は、例えば、イヌ細胞(MDCK細胞など)、Vero細胞、Per.C6細胞、BHK細胞、PCK細胞、MDCK細胞、MDBK細胞、293細胞(例えば、293T細胞)、およびCOS細胞であってよい。

インフルエンザゲノム断片を含むベクターを、例えば、リン酸カルシウム共沈降法、エレクトロポレーション、マイクロインジェクション、リポフェクション、およびポリアミントランスフェクション試薬を用いるトランスフェクションなどの、異種核酸を真核細胞中に導入するための当業界で公知の方法(例えば、米国特許出願公開第US20050266026号および第20050158342号)に従って宿主細胞中に導入することができる。例えば、ベクター、例えば、プラスミドを、製造業者の説明書に従って、ポリアミントランスフェクション試薬TransIT-LT1 (Mirus)を用いて、例えば、MDCK細胞、COS細胞、293T細胞、またはその組合せなどの宿主細胞中にトランスフェクトすることができる。宿主細胞の集団に導入しようとする約1μgの各ベクターを、総量200μl中、160μmの培地、好ましくは無血清培地中に希釈された約2μlのTransIT-LT1と混合することができる。DNA:トランスフェクション試薬混合物を、室温で45分間インキュベートした後、800μlの培地を添加することができる。次いで、トランスフェクション混合物を宿主細胞に添加し、細胞を上記のように培養する。従って、細胞培養物中での組換えまたは再集合ウイルスの産生のために、それぞれ8種のゲノム断片(PB2、PB1、PA、NP、M、NS、HAおよびNA)を含むベクターを、約20μlのTransIT-LT1と混合し、宿主細胞中にトランスフェクトする。必要に応じて、血清を含む培地を交換した後、無血清培地、例えば、Opti-MEM 1を用いてトランスフェクトし、4〜6時間インキュベートする。
典型的には、前記ウイルスの増殖を、一般的には、宿主細胞を培養する培地成分中で達成する。インフルエンザウイルスの複製にとって好適な宿主細胞としては、例えば、Vero細胞、Per.C6細胞、BHK細胞、MDCK細胞、293細胞およびCOS細胞、例えば、293T細胞、COS7細胞が挙げられる。本発明の文脈においては、MDCK細胞が好ましい。宿主細胞としての非腫瘍原性MDCK細胞の使用も、本発明の実施形態である。上記細胞系のうちの2種、例えば、MDCK細胞および293TまたはCOS細胞を含む同時培養物を、例えば、1:1の比率で用いて、複製効率を改善することもできる。例えば、20050158342を参照されたい。典型的には、細胞を、血清(例えば、10%ウシ胎仔血清)を補給したダルベッコの改変イーグル培地などの標準的な市販の培養培地中、または無血清培地中、中性に緩衝化されたpH(例えば、7.0〜7.2のpH)を維持するのに好適な、制御された湿度およびCO2濃度下で培養する。必要に応じて、前記培地は、細菌の増殖を防止するための抗生物質、例えば、ペニシリン、ストレプトマイシンなど、および/またはL-グルタミン、ピルビン酸ナトリウム、非必須アミノ酸などの追加の栄養素、好ましい増殖特性を促進する追加の補給物質、例えば、トリプシン、β-メルカプトエタノールなどを含む。
典型的には、ウイルスを、感染させた(トランスフェクトした)細胞を増殖させた培養培地から回収する。典型的には、粗培地を清澄化した後、インフルエンザウイルスを濃縮する。一般的な方法としては、濾過、限外濾過、硫酸バリウム上への吸着および溶出、ならびに遠心分離が挙げられる。例えば、感染した培養物に由来する粗培地を、最初に、例えば、1000〜2000 x gで、細胞破片および他の大きい粒子状物質を除去するのに十分な時間、例えば、10〜30分間遠心分離することにより清澄化することができる。あるいは、前記培地を、0.8μmの酢酸セルロースフィルターを介して濾過して、無傷の細胞および他の大きい粒子状物質を除去する。次いで、必要に応じて、清澄化された培地上清を遠心分離して、例えば、15,000 x gで約3〜5時間、インフルエンザウイルスを沈降させる。ウイルスペレットを、STE(0.01 M Tris-HCl; 0.15 M NaCl; 0.0001 M EDTA)またはリン酸緩衝生理食塩水(PBS)、pH 7.4などの好適なバッファー中に再懸濁した後、ウイルスをスクロース(60%〜12%)または酒石酸カリウム(50%〜10%)上での密度勾配遠心により濃縮する。連続勾配または段階勾配、例えば、4種の12%段階中、12%〜60%のスクロース勾配が好適である。この勾配を、ウイルスを回収するための可視的バンドに濃縮するのに十分な速度、および時間で遠心分離する。あるいは、および最大規模の商業用途のために、連続モードで運転するゾーン遠心分離ローターを用いて密度勾配からウイルスを浄化する。組織培養物からのインフルエンザウイルスの調製を介して当業者を誘導するのに十分なさらなる詳細は、例えば、Furminger.「ワクチン製造(Vaccine Production)」、Nicholsonら(編) Textbook of Influenza pp. 324-332; Mertenら(1996)「ワクチン調製のための細胞培養物中でのインフルエンザウイルスの製造(Production of influenza virus in cell cultures for vaccine preparation)」、Cohen & Shafferman (編) Novel Strategies in Design and Production of Vaccines pp. 141-151、ならびに米国特許第5,690,937号、米国特許出願公開第20040265987号、第20050266026号および第20050158342号(参照により本明細書に組み入れられるものとする)に提供されている。必要に応じて、回収されたウイルスを、安定化剤としてのスクロース-リン酸-グルタミン酸(SPG)の存在下、-80℃で保存することができる。
インフルエンザウイルスのゲノムは、免疫原性ヘマグルチニン(HA)およびノイラミニダーゼ(NA)タンパク質ならびに6個の内部核ポリペプチド:ヌクレオキャプシド核タンパク質(NP);マトリックスタンパク質(M);非構造タンパク質(NS);および3種のRNAポリメラーゼ(PA、PB1、PB2)タンパク質をコードする、8個の断片の線状マイナス鎖リボ核酸(RNA)から構成される。複製の間に、ゲノムウイルスRNAは、宿主細胞の核中でプラス鎖メッセンジャーRNAおよびマイナス鎖ゲノムcRNAに転写される。8個のゲノム断片の各々は、RNAに加えて、NPおよびポリメラーゼ複合体(PB1、PB2およびPA)を含むリボ核タンパク質複合体中にパッケージングされる。
歴史的には、インフルエンザウイルスワクチンは、関連する株の経験的予測に基づいて選択されたウイルス株を用いて孵化鶏卵中で製造されてきた。より最近では、認可された弱毒化、温度感受性マスター株の文脈において選択されたヘマグルチニンおよびノイラミニダーゼ抗原を含む再集合ウイルスが製造されてきた。鶏卵中での複数回の継代を介するウイルスの培養後、インフルエンザウイルスを回収し、必要に応じて、例えば、ホルムアルデヒドおよび/またはβ-プロピオラクトンを用いて不活化する。しかしながら、この様式でのインフルエンザワクチンの製造は、いくつかの有意な欠点を有する。鶏卵から残存する夾雑物は高度に抗原性、発熱源性であり、投与の際に有意な副作用をもたらすことが多い。より重要なことに、製造のために指定された株を、典型的には、次の流行期の数ヶ月前に選択および分布して、インフルエンザワクチンの製造および不活化のための時間を可能にしなければならない。ワクチン製造にとって認可された株のいずれかが標準的な細胞培養条件下で効率的に増殖することができないことによって、細胞培養物中で組換えおよび再集合ワクチンを製造する試みが妨害された。
本発明の組換えおよび再集合ウイルスを、好適な担体または賦形剤中で予防的に投与して、1種以上のインフルエンザウイルス株に特異的な免疫応答を刺激することができる。典型的には、前記担体または賦形剤は、滅菌水、水性塩水溶液、水性緩衝塩水溶液、水性デキストロース溶液、水性グリセロール溶液、エタノール、未感染鶏卵に由来する尿膜腔液(すなわち、正常な尿膜腔液「NAF」)またはその組合せなどの製薬上許容し得る担体または賦形剤である。無菌性、pH、等張性、および安定性を保証するそのような溶液の調製を、当業界で確立されたプロトコルに従って行う。一般的には、担体または賦形剤を、アレルギー作用および他の望ましくない作用を最小化するように、ならびに投与の特定の経路、例えば、皮下、筋肉内、鼻内などに適合するように選択する。
本発明のベクターおよびベクター系の使用を容易にするために、前記ベクターのいずれか、例えば、共通インフルエンザウイルスプラスミド、変異体インフルエンザポリペプチドプラスミド、インフルエンザポリペプチドライブラリープラスミドなど、ならびに実験目的または治療目的のためのインフルエンザウイルスのパッケージングおよび感染にとって有用な、バッファー、細胞、培養培地などのさらなる成分を、キットの形態で包装することができる。典型的には、前記キットは、上記成分に加えて、例えば、本発明の方法を実施するための説明書、包装材料、および容器などを含んでもよいさらなる材料を含む。
本発明の文脈においては、イヌRNA pol I調節配列を含む核酸もしくは本発明の他の核酸、発現ベクター、インフルエンザウイルス核酸および/またはタンパク質などを、よく知られた分子生物学技術に従って操作する。増幅、クローニング、突然変異誘発、形質転換などの多くのそのような手順に関する詳細なプロトコルは、例えば、Ausubelら、Current Protocols in Molecular Biology (supplemented through 2000) John Wiley & Sons, New York (「Ausubel」); Sambrookら、Molecular Cloning - A Laboratory Manual (第2版), Vol. 1-3, Cold Spring Harbor Laboratory, Cold Spring Harbor, New York, 1989 (「Sambrook」)、ならびにBergerおよびKimmel、Guide to Molecular Cloning Techniques, Methods in Enzymology volume 152 Academic Press, Inc., San Diego, CA (「Berger」)に記載されている。
本発明の核酸、ベクター、および方法を、他の組換えウイルスの発現および精製に用いることもできる。以下の考察は、他のそのようなウイルスと共に使用するためのベクターを適合させるのに重要な考慮のための指針を提供する。
1. イヌRNAポリメラーゼI調節配列を含む単離された核酸。
以下の実施例は、単に本発明を例示するのに役立つものであり、いかなる方法においても本発明を限定することを意図するものではない。
本実施例は、インフルエンザを培養するためのいくつかの細胞系の特性評価を記載する。MRC-5、WI-38、FRhL-2、PerC6、293、NIH3T3、CEF、CEK、DF-1、Vero、およびMDCKなどの、いくつかの異なる細胞系および一次細胞を、野生型(wt)および実験室に適合させた、例えば、低温適合性(ca)のA型およびB型インフルエンザ株から誘導された遺伝子再集合体の両方の産生について評価した。多くの細胞型が、限定された程度で、いくつかの低温適合性インフルエンザ株の複製を支援したが、MDCKのみが、一貫して高力価のA型およびB型の両方のウイルスを産生した。例えば、PerC6細胞は、MDCK細胞中に認められるのと同様のレベルまで、特定のwtおよびcaのB型ウイルスの複製を支援することが見出されたが、増殖動力学は異なる(図1を参照)。対照的に、PerC6は、いくつかのcaのA型ウイルスの複製を支援することができなかった。図2は、wtならびにca A/シドニー(Sydney)/05/97およびA/北京(Beijing)/262/95ウイルスに関する増殖曲線を示す。両方の場合において、ca株はPerC6細胞中であまり複製しない。同様に、図3は、wtならびにca A/アンアーバー/6/60の増殖曲線を示し、これはca株がPerC6細胞中で効率的に複製せず、wt A/アンアーバー/6/60の複製はMDCK細胞中と同じほど強固ではない。PerC6細胞中でのインフルエンザウイルス複製のリアルタイムPCR分析により、caおよびwt Aインフルエンザウイルス株の両方のウイルスRNA(vRNA)が、感染後最初の24時間の間に増加するが、wt株のみが120時間まで増加し続け、ca株は増加しないことが示された。対照的に、wtおよびcaの両方のvRNAは、MDCK細胞中で増加し、3日目にプラトーに達した。図4を参照されたい。
一方は血清を含む培地中で増殖させ、他方は無血清培地中で増殖させた、MDCK細胞の2種のプレマスター細胞保存液の潜在的な腫瘍原性を、ワクチン製造に用いられると期待された後5回の細胞継代を行った段階で、無胸腺ヌードマウスモデルにおいて評価した。腫瘍原性を評価するために、10匹のマウス群に107個の細胞を皮下注入し、84日後、動物を犠牲にし、試験した。無血清培地中で継代された細胞を接種した10匹の動物のうちの6匹において、新生物が観察された。対照的に、10%ウシ胎仔血清を補給した培地中で継代された細胞を接種された動物のいずれにおいても、新生物の証拠はなかった;いくらかの線維肉腫が接種部位で観察されたが、血清中で継代された細胞は、表2に示されるように腫瘍原性ではなかった。
ATCCから入手したMDCK細胞を、γ照射されたFBSを含む培地中で継代する。次いで、これらの細胞を、細胞バンク作製を支援するために選択された無血清培地製剤中、限定回数継代する。無血清培地は、米国特許仮出願第60/638,166号および第60/641,139号、ならびに米国特許出願第11/304,589号に記載されている。これらの追加の継代を、37℃または33℃で実施することができる。血清よりもむしろ植物由来補給物質を含む3種の培地中でのMDCK細胞の継代により、FCSを含有する培地中で継代されたMDCK細胞のものと同様の核型を有する細胞が得られた(データは示さない)。
細胞を、限界希釈を介して生物学的にクローニングして、産生細胞がユニークな遺伝子群から誘導されることを確保した。クローンを、分裂時間および相対腫瘍原性、ならびにウイルス産生などの様々な表現型特性についてスクリーニングした。概念実験の最初の証拠においては、54個のMDCKクローンが、FCSを含有する培地中で得られた。これらのクローンを継代し、それぞれを低い感染多重度でca A/ニューカレドニア(New Caledonia)/20/99に感染させた。感染の数日後、上清を除去し、上清中のウイルス量をTCID50により測定した。少数のクローンは比較的高力価のウイルスを産生し、非クローン化親細胞における産生より多かった。優れた生物学的および生理学的特性を有するクローンを用いて、以下に記載のようなマスター細胞バンク(MCB)を確立する。
外来因子の証拠がないことを確保するために、MCBを幅広く試験する。例えば、利用可能なウイルス剤について、以下の表3に示されるような、1種以上のいくつかのPCRおよび/または抗体特異的試験を行う。
本実施例は、細胞培養物ならびに卵から製造されたインフルエンザ株の特性評価を記載し、前記系から製造されたウイルスを比較する。一般的には、インフルエンザウイルスは、ヒトにおけるワクチンとしての使用にとって好適であり、該ウイルスをそのような使用にとって好適にする生物学的特性を有する。本実施例においては、インフルエンザウイルスは、低温適合性(ca;低い温度で効率的に複製する能力を有する)、温度感受性(ts;より高い温度ではin vitroで複製が制限される)、および弱毒化(att;フェレットの肺組織において検出可能な複製を示さない)であり、本明細書ではcatsatt株と呼ぶ。この比較は、ウイルス産物の生化学、抗原、および遺伝的評価(配列決定);ヒト細胞中での複製後のウイルスの生物学的および生化学的特性評価;許容可能な動物モデルにおける複製;ならびに許容可能な動物モデルにおける免疫原性を含む。
A/H1N1、A/H5N1、A/H3N2およびB型のca ts att株は、MDCK細胞において比較的高い力価で複製した。さらに、MDCK細胞中でこれらのca ts att株を継代することにより、それらの遺伝子配列は変化しなかった。3種のca ts att株、ca A/シドニー/05/97、ca A/北京/262/95、およびca B/アンアーバー/1/94を、MDCK細胞中で1回また2回継代し、全部で6個の内部遺伝子のコード領域全体を配列決定し、出発材料と比較した。ヌクレオチド変化は観察されなかったが、これは、この基質を介するこの継代がこれらの株の遺伝子組成を変化させなかったことを示している。培地組成、入力用量(moi)、インキュベーションの温度および収穫の時間などの製造プロセスを模倣すると予想される条件下で、MDCK細胞中で産生された様々なワクチン株に対して、さらなる配列特性評価を行った。予備データに基づけば、MDCKにより産生されたウイルスのゲノム配列における変化はないであろうと予想される。
一実施形態においては、ヒト起源の細胞中でのMDCKおよび卵により産生されたワクチンの複製後の生化学的、生物学的、および構造的類似性を評価するために、ワクチンを、正常なヒト気管支上皮細胞(NHBE)などの関連する2倍体ヒト細胞中で1回継代することができる。この継代は、ヒト気道において1回の感染事象を模倣し、次いで、究極的には有効な免疫応答を引き出す原因となる子孫ウイルスの比較を可能にするのに役立つであろう。ワクチンのヘマグルチニン(結合および融合)ならびにノイラミニダーゼ活性の試験を、これらの材料上で測定し、ならびに電子顕微鏡、感染性粒子と全粒子の比、およびウイルスゲノム等価物などの他の生物学的および構造的試験を評価することができる。全体として、これらの比較は、細胞に由来するワクチンと、有効かつ安全な卵により産生されたワクチンとの比較可能性を証明するのに役立つであろう。分析試験の概要を、表5にまとめる。
フェレットは、弱毒化インフルエンザワクチンおよび成分ワクチン株の弱毒化および免疫原性を評価するのに用いられる強固な動物モデルである。MCBから産生された細胞由来インフルエンザ株の性能を、卵中で産生された同じ株と比較する。制御された試験におけるこれらの材料の面と向かった比較により、これらのウイルス産物の比較可能性の高レベルの保証が可能になる。
現在のインフルエンザワクチン株は、鳥細胞を、野生型ウイルスおよびA型もしくはB型MDVと同時感染させ、所望の6:2遺伝子群のための子孫を単離およびスクリーニングすることにより作製されている。このプロセスには、鳥細胞培養物および/またはSPF卵を介するウイルスの数回の継代が必要である。最近、インフルエンザウイルス調製物を製造するためにプラスミドレスキューが導入された。このプロセスにおいては、幅広く試験され、特性評価された細胞バンクから得たVero(アフリカミドリザル)細胞に、例えば、それぞれ8種のインフルエンザRNA断片の1つのcDNAコピーを含む、8種のDNAプラスミドをエレクトロポレーションする。エレクトロポレーションの数日後、これらのエレクトロポレーションされた細胞の上清は、インフルエンザウイルスを含む。次いで、この上清をSPF卵中に接種して、ワクチン株を増幅し、生物学的にクローニングする。これらの手順は両方とも、MVSを製造するためにSPF卵に接種されるワクチン株をもたらす。プラスミドレスキューは、より信頼性の高いタイミング、より遺伝的に正確な遺伝子断片および野生型単離物に由来する外来因子の夾雑の低い可能性などいくつかの利点を有したが、これらの2つの方法により作製された個々のMVSは、互いに識別不可能であり、これを用いて、バルクワクチン製造を開始することができる。本発明の方法および組成物を用いて、この方法を適合させて、プラスミドレスキューのためにVero細胞の代わりにMDCK細胞を用いる。

Claims (31)
- イヌRNAポリメラーゼI調節配列を含む単離された核酸。
- 前記調節配列がプロモーターである、請求項1に記載の核酸。
- 前記RNAポリメラーゼI調節配列が、配列番号26のヌクレオチド1〜469、その相補体、その逆相補体、またはその機能的に活性な断片を含む、請求項1に記載の核酸。
- 前記調節配列が、マイナス鎖ウイルスゲノムRNAまたは対応するcRNAをコードするcDNAに機能し得る形で連結されている、請求項1、2または3に記載の核酸。
- 前記核酸が、転写終結配列をさらに含む、請求項4に記載の核酸。
- 前記マイナス鎖ウイルスゲノムRNAがインフルエンザゲノムRNAである、請求項5に記載の核酸。
- 請求項6に記載の核酸を含む発現ベクター。
- 細胞中で請求項6に記載の核酸を転写させることによって、インフルエンザゲノムRNAを製造することを含む、インフルエンザゲノムRNAの製造方法。
- 請求項7に記載の発現ベクターならびにPB2、PB1、PA、HA、NP、NA、M1、M2、NS1、およびNS2からなる群より選択される1種以上のインフルエンザポリペプチドをコードするmRNAを発現する1個以上の発現ベクターを含むイヌ細胞を培養すること;ならびに組換えインフルエンザウイルスを単離することを含む、組換えインフルエンザウイルスの製造方法。
- (a)イヌ細胞の集団中に、該細胞中でインフルエンザウイルスの完全なゲノムvRNA断片を提供するゲノムvRNA断片を発現することができる発現ベクターであって、配列番号26のヌクレオチド1〜469、またはその機能的に活性な断片を含む前記ベクターを導入すること;(b)該細胞中に、該ウイルスの1種以上のポリペプチドをコードするmRNAを発現することができる発現ベクターを導入すること;ならびに(c)該細胞を培養することによって、インフルエンザウイルス粒子を産生させることを含む、組換えインフルエンザウイルスの製造方法。
- 48〜72時間、前記細胞を培養する際に産生されたインフルエンザウイルス粒子の力価が、少なくとも1.0 x 104PFU/mlである、請求項10に記載の方法。
- 48〜72時間、前記細胞を培養する際に産生されたインフルエンザウイルス粒子の力価が、少なくとも1.0 x 105PFU/mlである、請求項10に記載の方法。
- 産生されるインフルエンザウイルス粒子が感染性である、請求項9に記載の方法。
- 産生されるインフルエンザウイルス粒子が感染性である、請求項10に記載の方法。
- 請求項7に記載の発現ベクターを含む細胞。

- 培養されたイヌ細胞中で、4個以上のゲノムvRNA断片を有する組換え断片化されたマイナス鎖RNAウイルスを生成させる方法であって、(a)イヌ細胞の集団中に、該細胞中で該ウイルスの完全なゲノムvRNA断片を提供するゲノムvRNA断片を発現することができる第1の発現ベクターセットを導入すること;(b)該細胞中に、該ウイルスの1種以上のポリペプチドをコードするmRNAを発現することができる第2の発現ベクターセットを導入すること;ならびに(c)該細胞を培養することによって、ウイルス粒子を産生させることを含む、前記方法。
- 組換えインフルエンザウイルスを製造する方法であって、
i)イヌ細胞の集団中に、
a)該細胞中で、該ウイルスの完全なゲノムvRNA断片を提供するゲノムvRNA断片を発現することができる発現ベクターであって、1個以上の該発現ベクターが配列番号26のヌクレオチド1〜469、もしくはその機能的に活性な断片を含む、前記発現ベクター、ならびに、
b)該細胞中で、PB2、PB1、PA、HA、NP、NA、M1、M2、NS1、およびNS2からなる群より選択される1種以上のインフルエンザポリペプチドをコードするmRNAを発現することもできる発現ベクター、
を導入すること;ならびに、
ii)該細胞を培養することによって、インフルエンザウイルス粒子を産生させること、
を含む、前記方法。 - 前記細胞を48〜72時間培養する際に産生されるインフルエンザウイルス粒子の力価が、少なくとも1.0 x 104PFU/mlである、請求項19または20に記載の方法。
- 前記細胞を48〜72時間培養する際に産生されるインフルエンザウイルス粒子の力価が、少なくとも1.0 x 105PFU/mlである、請求項19または20に記載の方法。
- 請求項20に記載の方法により製造されたインフルエンザウイルス。

- 配列番号26に記載のヌクレオチド配列に対して少なくとも80%の同一性を有するポリヌクレオチドを含む単離された核酸配列であって、該核酸配列が、インフルエンザvRNAをコードするcDNAに機能し得る形で連結され、MDCK細胞中に導入された場合、該インフルエンザvRNAの発現を指令することができる、前記核酸配列。
- 配列番号1〜26からなる群より選択される核酸に、ストリンジェントなハイブリダイゼーション条件下でハイブリダイズするポリヌクレオチドを含む単離された核酸配列であって、該核酸配列が、インフルエンザvRNAをコードするcDNAに機能し得る形で連結され、MDCK細胞中に導入された場合、該インフルエンザvRNAの発現を指令することができる、前記核酸配列。
- 請求項26、27または28に記載の核酸を含む発現ベクター。
- 細胞中に請求項29に記載の発現ベクターを導入することによって、インフルエンザゲノムRNAを産生させることを含む、インフルエンザゲノムRNAの製造方法。
