Abstract
【解決手段】 mRNAをコードする遺伝子を提供し、mRNAのコード領域内の阻害/不安定配列(群)の位置決定を行い、多重点突然変異を作成することにより遺伝子内の阻害/不安定配列へ突然変異を導入する。この方法により、該mRNAをコードする遺伝子を改変して、遺伝子のコード能を変更しないようにクラスターヌクレオチド置換を施すことで、これらの阻害/不安定配列群を取り除く。

Description
本発明は転写を阻害あるいは減衰させる遺伝子のコード領域中の調節あるいは阻害/不安定配列(INS)に突然変異を導入した遺伝子により生産されるmRNAの安定性および/あるいは有用性の増加方法に関連する。本発明はまた本方法に従って突然変異を導入した遺伝子を保持する発現ベクターを含む構築物および当該構築物を含む宿主細胞に関連する。
転写制御機構の研究に多くの仕事が為されてきて、転写後修飾もまた、与えられた遺伝子により生産されたRNAの量及び有用性を変化させるということが次第に明らかになってきた。これらの転写後修飾は細胞質内のRNA分解に加えて核における転写後修飾(例えばスプライシング、ポリアデニル化、および輸送)を含む。これらの修飾はすべて特定の転写物の最終定常状態レベルに貢献する。これらの制御点は1つの過程が単独で生産するよりもより柔軟な制御体系を生み出す。例えば多量に転写されて効率よく修飾されても短命のメッセンジャーは安定したものよりも量が少ない。効率的な合成速度はメッセンジャーが細胞質に到達し翻訳されることを保証するが、急速な分解速度はmRNAが高レベルに蓄積しないことを保証する。例えば原癌遺伝子であるc-mycおよびc-fosのmRNAのような多くのRNAは、非常に低レベルで発現され、急速に衰退し、種々の条件下ですばやくそして一時的に修飾される点でこの種の制御を示すという研究がなされた。レビューとしてM. Hentze, Biochim. Biophys. Acta 1090:281ー292(1991)を参照。これらのmRNAの多くの分解速度はmRNAそのものの中にある1つあるいはそれ以上の不安定/抑制配列の存在に相関することが示された。
本発明は、遺伝子コード領域中に存在して該遺伝子の発現を邪魔する若しくは減らす調節もしくは阻害/不安定配列(INS)を変異させることによって、遺伝子により生産されるmRNAの安定性および/もしくは有用性を増大させる方法に関する。本発明はまた、発現ベクターを含めた、これらの方法に従い変異させられた遺伝子を含むコンストラクト(構築物)、およびこれらのコンストラクトを含む宿主細胞にも関する。

Virol. 64:6010-6017(1990))(斑点ボックスで示されている)をp17gagコード配列の3'側に含む。RREの後ろには、3'UTRのU5領域のnt8021から最後のヌクレオチドまでのHIV-1配列が続く。プラスミドp19およびp19Rは、それぞれ、プラスミドp17およびp17RのHIV-1 p17gagコード配列をRSV p19gagコード配列(黒ボックスで示されている)で置き換えることにより作製された。プラスミドp17M1234は、gagコード領域内に28のサイレントヌクレオチド置換(xxxで示されている)がある以外は、p17と同一である。波線はプラスミド配列を表す。プラスミドp17M1234(731-1424)およびプラスミドp37M1234は、すぐ後、およびその他の箇所において記載する。これらのベクターは、特定のヌクレオチド配列がINSをコードするかを決定するのに用いることのできるコンストラクトの例示である。この場合、インジケーター遺伝子(ここではp17gag)を含むベクターp17M1234はコントロールベクターを表し、ベクターp17M1234(731-1424)およびp37M1234は、問題となるヌクレオチド配列(ここではp24gagコード配列)がそれぞれ、インジケーター遺伝子の終止コドンの3'側のベクターに挿入されているか、若しくはインジケーター遺伝子のコード領域にインフレームで融合されているベクターを表す。(C)はgagINSを同定、およびさらに変異導入するための発現ベクターの構築法である。p17M1234をベクターとして用いて、改変p17gag遺伝子のコード領域の下流に付加的なHIV-1 gag配列が挿入された。ヌクレオチド番号により示された3つの異なる断片を、以下に記すようにベクターp17M1234に挿入した。プラスミドp17M1234(731-1081)、p17M1234(731-1424)およびp17M1234(731-2165)を作製するために、示された断片をp17M1234のp17gagコード配列の終止コドンの3'側に挿入した。発現アッセイ(データは示していない)において、p17M1234(731-1081)およびp17M1234(731-1424)が高レベルのp17gag蛋白質を発現した。対照的に、p17M1234(731-2165)はp17gag蛋白質を発現せず、このHIV-1 gagコード領域内に付加的なINSが存在することを示している。プラスミドp17M1234(731-1081)NS、p37M1234およびp55M1234を作製するために、改変p17gag遺伝子の末端の終止コドンおよびp17M1234の全てのリンカー配列をオリゴヌクレオチド部位特異的変異導入により取り除き、得られたプラスミドは、HIV-1と同様のgagオープンリーディングフレームを回復した。発現アッセイ(データは示していない)において、p37M1234は、ウェスタンブロットおよびELISAアッセイにより決定したところ、高いレベルの蛋白雪を発現したが、p55M1234は検出可能なgag蛋白質を発現しなかった。以上のことから、p24領域の3'側の配列の付加は蛋白質発現の排除をもたらし、ヌクレオチド配列1424-2165にはINSが含まれることが示された。本実験は、p37M1234が付加的なINSを解析するのに適したベクターであることを示した。

上記の一般的な解説および下記の詳細な解説は共に、単なる例示および説明であって、請求の範囲にあるもののみに本発明を制限するものではないことは、理解されるべきである。本明細書の一部に取り入れられ、かつその一部をなす付属の図面は、本発明の態様を例示し、解説と合わせて、本発明の原理を説明するのに役立っている。
1. INSを含むmRNAの同定
特定の蛋白質が作られる割合は、それをコードするmRNAの細胞質のレベルに比例するのが普通である。従って、阻害/不安定配列を含むmRNAをコードする候補は、そのmRNAもしくは蛋白質が検出できるほどには発現していないか、または同じ、若しくは同様の強さのプロモーターの制御下にある対照mRNAもしくは蛋白質の発現レベルと比較して僅かしか発現しないものである。同じ若しくは同様の強さのプロモーター制御下にある別の遺伝子からの定常状態レベルと比較した時の、見掛けの転写速度(例えば、核ラン-オンアッセイにより決定される)における変化では説明できない、特定のmRNAの定常状態レベル(例えばノザンブロッティングにより決定される)における相違は、その遺伝子が不安定mRNAをコードする候補であることを示す。不安定に加えて、若しくは不安定とは別に、細胞質で働く様々な阻害機構により細胞質mRNAがあまり利用されないこともある。これらの効果は、本明細書で「阻害配列」と名付けられている特定のmRNA配列により調節されている可能性がある。
不安定な、若しくはあまり利用されないmRNAが同定されると、次の工程は、それを担う(cis作用性)RNA配列エレメントを調べることである。cis作用性阻害/不安定領域の位置決定に関する詳細な方法は、上記“従来の技術”の章に記載された参考事項の各々に示されており、以下にも考察される。本発明の例示されたコンストラクトもINSの位置決定に利用することができる(下記参照)。特定のmRNAターンオーバーを担うcis作用性配列は、mRNA安定性の変化した自然に生じた変異体が時たま同定されることによる以外に、欠失および点変異導入によって同定することができる。
阻害/不安定配列がmRNAのコード領域内に位置決定されると、その遺伝子を改変してこれらの阻害/不安定配列を、該遺伝子のコード能を変えることなく排除する。また、遺伝子を改変して阻害/不安定配列を除くと同時に、保存的もしくは非保存的なアミノ酸置換をコードするように遺伝子のコード能を変える。
4. 変異mRNAの安定性の決定
得られた変異mRNAの定常状態レベルおよび/もしくは安定性を、阻害/不安定領域を含む非改変mRNAの定常状態レベルおよび/もしくは安定性を試験するのと同様にして(上記第1章に考察されているように、例えばノザンブロッティングにより)、試験することができる。阻害/不安定領域(群)を含む非改変mRNAおよび望みによっては非改変インジケーターmRNAと共に(従ってこれらと比較して)、変異mRNAを解析することができる。例示されるように、トランスフェクション実験において、ノザンブロット分析によって以降のmRNA解析を行うことにより、HIV-1 p17gag変異体を非変異HIV-1 p17gagと比較する。これらのmRNAにより生産される蛋白質は、ELISAなど、当該分野で知られた免疫ブロッティングおよび他の方法によって測定する。下記参照。
本発明の方法により変異導入が可能な遺伝子には、そのmRNAがその中にINS領域を含むことが知られている、若しくは含むと思われるものが含まれている。これらの遺伝子には、例えば、増殖因子、インターフェロン、インターロイキン、fos原癌遺伝子蛋白質、並びに、本明細書中に例示されたものに加え、他のウイルスmRNA以外に、HIV-1 gag、envおよびpolをコードする遺伝子が含まれる。本発明の方法により変異導入される遺伝子は、天然の宿主細胞もしくは生物、または異なる細胞若しくは生物において発現させることができる。変異遺伝子は、プラスミド、コスミド、ファージ、ウイルスもしくはミニ染色体などのベクターに導入することができ、当該分野でよく知られた方法により宿主細胞もしくは生物に挿入することができる。一般的に、変異遺伝子もしくはこれらの変異遺伝子を含むコンストラクトは、哺乳動物細胞(例、ヒト(例、HeLa)、サル(例、Cos)、ウサギ(例、ウサギ赤血球)、ラット、ハムスター(例、CHOおよびベビーハムスター腎細胞)もしくはマウス細胞(例、L細胞))、植物細胞、酵母細胞、昆虫細胞または細菌細胞(例、大腸菌)を含め、真核細胞または原核細胞のいずれの細胞にも利用することができる。これらの変異遺伝子のクローン化および/もしくは発現に利用することのできるベクターは、該変異遺伝子が複製され、そして/もしくは発現されて欲しい宿主細胞において、変異遺伝子を複製および/もしくは発現させる能力を持つベクターである。様々なタイプの宿主細胞に適したベクターの例としては、例えば、F.アウスベル(Ausbel)ら、Current Protocols in Molecular Biology, Greene Publishing Associates and Wiley-Interscience(1992)およびサムブルックら(1989)を参照していただきたい。そのような遺伝子の本来のプロモーターを、該遺伝子が挿入される宿主と両立し得る強力なプロモーターに置き換えることができる。これらのプロモーターは誘導性であってもよい。これらの変異遺伝子を含む宿主細胞を用いて、酵素調製品、医薬品、診断薬、ワクチンおよび治療学に有用な蛋白質を大量に発現させることができる。
HIV-1 GAG 遺伝子
ヒト免疫不全症候群ウイルス1型(HIV-1)のRev制御タンパク質と、Rev-応答エレメント(RRE)と命名されたそのRNA標的との相互作用は、ウイルス構造タンパク質の発現に必要である(レビューとしてG.パブラキスとB.フェルバー(G. Pavlakis and B. Felber)New Biol. 2:20-31(1990);B.キュレンとW.グリーン(B. Cullen and W. Greene)Cell 58:423-426(1989);C.ローゼンとG.パブラキス(C. Rosen and G. Pavlakis)AIDS J. 4:499-509(1990)を参照)。RevはRREを含むmRNAの核からの移行を促進し、その安定性を増加させることによって作用する。最近の結果ではREVがこれらのmRNAのポリソームへの効率的な会合に役割を果たすことも示されている(S.アリゴとI.チェン(S. Arrigo and I. Chen)Gene Dev. 5:808-819(1991), D.ダゴスティーノら(D, D’Agostino et al.)Mol. Cell Biol. 12:1375-1386(1992))。RREを含んだHIV-1のmRNAは、Revの非存在下ではタンパク質を効率良く産生できないことから、これらのmRNAには欠損があり、INS、CRS、またはIRと様々な表記がされている阻害/不安定配列が含まれていると予想されていた(M.エマーマンら(M. Emerman et al.)Cell 57:1155-1165(1989);S.シュワルツら(S. Schwartz et al.)J. Virol. 66:150-159(1992);C.ローゼンら(C. Rosen et al.)Proc. Natl. Acad. Sci. USA 85:2071-2075(1988);M.ハッツポロークラダラスら(M. Hadzopoulou-Cladaras et al.) J. Virol. 63:1265-1274(1989);F.マルダレリら(F. Maldarelli et al.)J. Virol. 65:5732-5743(1989);A. W.コクランら(A. W. Cochrane et al.)J. Virol. 65:5305-5313(1991))。これらの阻害/不安定配列の性質と機能に関して、詳細な記述はなされていない。不要のスプライス部位がRevの機能に必要ではないかと予想されており(D.チャンとP.シャープ(D. Chang and P. Sharp)Cell 59:789-795(1989))、こうしたスプライス部位の存在が、HIV-1 mRNAをRev依存性としているのかもしれない。
p17gag中の阻害/不安定性エレメントをさらに研究するため、p17gag発現プラスミド(p17図1)を構築した。p17gag配列はコーディング配列の直後に翻訳終止コドンを含むように設計し、そうしてp17gagだけを産生するようにした(このプラスミドの構築は後に記載する)。HIV-1上のgag AUGの上流にある主要な5’スプライス部位は、このベクターから欠失してある(B.フェルバーら(B. Felber et al.)Proc. Natl. Acad. Sci. USA86:1495-1499(1989))。p17プラスミドがRevとRREの非存在下でp17gagを産生できるかどうかを調べるため、p17をHLtat細胞に導入した(S.シュワルツら(S. Schwartz et al.)J. Virol. 64:2519-2529(1990))(以下参照)。これらの細胞はHIV-1 LTRプロモーターのtransの活性化に必要な、HIV-1 Tatタンパク質を構成的に産生する。p17プラスミドをRevの非存在下または存在下で導入し、p17gagの産生をウェスタン免疫ブロッティングにより解析した。その結果、非常に低いレベルのp17gagタンパク質が産生されていることがわかった(図2A)。gag mRNAにはRREが含まれないことから予想されるとおり、Revの存在はgagの発現を増加させなかった。次にp17gagコーディング配列とRREの両者を含むプラスミド(p17R 図1)を構築した。p17と同様にこのプラスミドは、Revの非存在下で非常に低いレベルのp17gagを産生した。だが高レベルのp17gagが、Revの存在下でのみ産生された(図2A)。これらの実験は、阻害/不安定性エレメントがp17gagコーディング配列の中に位置することを示唆した。
HIV-1 gag中の阻害配列の正確な性質を調べるため、図4に示すように4種の異なるオリゴヌクレオチドを用いて、p17gagのコーディング配列に部位特異的変異導入を行った。それぞれのオリゴヌクレオチドは、19から22ヌクレオチドの範囲に数箇所の点突然変異を導入した。これらの変異は沈黙コドン変化(silent codon changes)を導入するので、p17gagタンパク質のアミノ酸配列には影響しなかった。最初に4種のオリゴヌクレオチドを同時に用いて、記載のとおり一本鎖DNAを鋳型とした変異導入を行った(T.クンケル(T. Kunkel)Proc. Natl. Acad. Sci. USA 82:488-492(1985);S.シュワルツら(S. Schwartz et al.)Mol. Cell. Biol. 12:207-219(1992))。この結果p17ベクター中の270ヌクレオチドの広い領域にわたって、多数の点変異を同時に導入することができた。4種のオリゴヌクレオチド全ての変異を含んだ変異体を単離し、p17M1234と命名した。p17と比較した場合、このプラスミドは高いAU含有率の領域に特に集中する、総数28個の点変異を含んでいる。HLtat細胞に形質転換させた後、p17gagの発現を免疫ブロッティングで解析することにより変異の表現型を評価した。興味深いことにp17M1234は高レベルのp17gagタンパク質を産生し、そのレベルはRevの存在下でp17Rが産生する場合よりも高かった(図2A)。この結果はプラスミドp17M1234ではp17gagmRNAの阻害/不安定性シグナルが不活化されているがことを示している。予想されたとおり、Revタンパク質の存在はp17M1234からの発現を増加させず、gagの発現にわずかに阻害効果を及ぼした。このようにp17M1234変異からのp17gagの発現は、RSVのp19gagと同じ一般的特性、すなわちRevに依存しない高い構成的レベルのgagの発現を示した。ノザンブロット解析の結果、p17M1234から産生されるmRNAのレベルは、p17から産生される場合に比べて増大していることが明らかとなった(図3A)。
p17gagコーディング領域内のINS(p17gagコーディング領域は図1(B)上の記載に記されているとおりヌクレオチド336-731に広がり、図1(A)と(B)に示すとおりgagコーディング領域の3つの部分(すなわちp17、p24、p15)のうち最初の部分を含む)の除去が完全なgag遺伝子の発現に与える影響を調べるため、ベクターp17M1234上の、変異により変化させたp17gagコーディング領域の3’側すなわち終止コドンの下流にgag遺伝子の付加的な配列を挿入した発現ベクターを構築した。gagの配列を徐々に伸ばした長さで含んだベクター、すなわち図1(C)に示したp17M1234(731-1081)、p17M1234(731-1424)、p17M1234(731-2165)の解析を行った。p17gagの発現レベルを測定した結果、gagタンパク質の2番目の部分をコードするmRNA領域(すなわち731-1424ヌクレオチドに広がるp24gagタンパク質をコードする部分)が弱いINSのみを含むことが、p17M1234により発現されるp17gagタンパク質量がp17M1234(731-1424)により発現されるp17gagタンパク質量と比較して少し減少していることから決定されたのに対し、gagタンパク質の3番目の部分をコードするmRNA領域(すなわちヌクレオチド1425-2165に広がるp15gagタンパク質をコードする部分)が強いINSを含むことが、p17M1234(731-2165)により発現されるgagタンパク質量がp17M1234とp17M1234(731-1424)により発現されるタンパク質量と比較し大きく減少していることから決定された。
上記の解析により、高レベルのp37gag前駆タンパク質(p17gagとp24gagタンパク質領域の両方を含む)を発現するp37M1234ベクターの構築が可能となった。ベクターp37M1234の構築は、変化させたp17gagタンパク質をコードする遺伝子の末端の停止コドンを除去し、p24gagタンパク質をコードするヌクレオチド配列をオリゴヌクレオチド変異導入で正しい読み枠に融合して行った。これによりヌクレオチド配列が回復され、HIV-1にコードされていた時と同様に、融合したp17gagとp24gagタンパク質(すなわちp37gagタンパク質)をコードするようになった。p37gagまたはp24gagタンパク質の存在は、商業的に入手可能なELISAキットにより容易に定量可能なので、ベクターp37M1234を用いればINSを含むと疑われる付加的な配列を挿入し検定することができる。そうした用法の実施例を以下に示す。
p37M1234の構築と同様の方法で構築された他のベクターは、p17M1234(731-1081)NSとp55BM1234(図1(C))である。これら3種のベクターはいずれもp17コーディング領域の下流の領域(3’側)の翻訳を可能とするが、それぞれからのgagの発現レベルは、終止コドンから3’側のヌクレオチド配列を含んだベクター(すなわち上に記載したp17M1234(731-1081)、p17M1234(731-1424)、p17M1234(731-2165))それぞれからのgagの発現レベルと同じであった。これらの結果はまたgag遺伝子内のINS領域は、翻訳またはINS領域でのその欠失による影響を受けないことを示している。これらの結果はHIV-1 gagコーディング領域内(すなわちHIV-1 gagの1424-2165をコードする領域の中)の付加的なINS配列を検出するためのp17M1234の用法を示す。したがってこれらの結果はまた、一種またはそれ以上の阻害/不安定性領域を含む遺伝子に変異を導入して、一つの阻害/不安定性領域を除去した後、まだその遺伝子内に存在するかもしれない付加的な阻害/不安定性領域の位置を決定するにはどうすればよいかを示している。
上に記載したように、いくつかの実験からHIV-1のp17gag領域に同定され除去されているINSに加えて、HIV-1のp24とp15領域の中にINSの存在が示されている。これはシュワルツら(Schwartz et al.)のJ. Virol. 66:7176-7182(1992)の7180ページの図6に模式的に描かれている。この図においてcgagM1234はp55BM1234と同一である。
プラスミドp17RはpNL17Rと記載されてきた(S.シュワルツら(S. Schwartz et al.)J. Virol. 66:150-159(1992))。プラスミドp17はp17Rを制限酵素Asp718で消化し、再度ライゲーションすることにより作製した。この操作により3’LTRの上流にあたるヌクレオチド8021-8561に広がるRREとHIV-1配列が欠失した。p17gagの変異体を作製するため、p17gagコーディング配列を修飾したpBLUESCRIPTベクター(Stratagene社)にサブクローニングし、ウラシルを含んだ一本鎖DNAを産生した。部位特異的変異導入を記載されたとおり行った(T.クンケル(T. Kunkel)Proc. Natl. Acad. Sci. USA 82:488-492(1985);S.シュワルツら(S. Schwartz et al.)Mol. Cell. Biol. 12:207-219(1992))。適当な変異を含んだクローンは、二重鎖DNAの塩基配列決定により選択した。p19Rを作製するには、まずプラスミドp17RをBssHIIとEcoRIで消化することによりp17gagのコーディング配列全体、すなわちp17gagAUGの6ヌクレオチド上流からp17gagの終止コドンの3’側のリンカー配列9ヌクレオチドまでを除いた。p17Rのp17gagコーディング配列を、PCR増幅したRSV p19gagコーディング配列を含んだDNA断片で置換した(R.ワイスら(R. Weiss et al.)RNA Tumor Viruses. Molecular Biology of Tumor Viruses(Cold Spring Harbor Laboratory, Cold Spring Harbor, New York, 1985))。この断片はRSV gagのAUGの上流8ヌクレオチドとp19gagコーディング配列およびそれにすぐに続く翻訳終止コドンを含んでいる。RSV gag断片は感染性RSVプロウイルスクローンS-RA(R.ワイスら(R. Weiss et al.)RNA Tumor Viruses. Molecular Biology of Tumor Viruses(Cold Spring Harbor Laboratory, Cold Spring Harbor, New York, 1985))に由来する。p19はp19Rからヌクレオチド8021-8561に広がるRREと3’HIV-1配列を含むようなAsp718断片を削除することにより作製した。
HLtat細胞(S.シュワルツら(S. Schwartz et al.)J. Virol. 64:2519-2529(1990))の形質転換は、カルシウム共沈法(F.グラハムらとA.ファンデルエブ(F. Graham et al. and A. Van der Eb)Virology 52:456-460(1973))を用いて記載(B.フェルバーら(B. Felber et al.)Proc. Natl. Acad. Sci. USA86:1495-1499(1989))のとおり行い、5μgのp17、p17R、p17M1234、p19またはp19Rを2μgのRev発現プラスミドpL3crev(B.フェルバーら(B. Felber et al.)Proc. Natl. Acad. Sci. USA 86:1495-1499(1989))の非存在下(-)または存在下(+)で用いた。形質転換時のDNA総量は、pUC19キャリアDNAを用いて60mmプレートあたり0.5ml中に17μgの沈澱物となるよう調整した。形質転換の20時間後細胞を集め、細胞抽出物を12.5%変性ポリアクリルアミドゲルで電気泳動にかけ、ヒトHIV-1患者血清(Scripps社)またはウサギ抗p19gag血清を用いた免疫ブロッティングにより解析した。pRSV-ルシフェラーゼ(J.デウェットら(J. de Wet et al.)Mol. Cell. Biol. 7:725-737(1987))はホタルのルシフェラーゼ遺伝子をRSV LTRプロモーターにつないだものを含んでいるが、これを形質転換の効率の対照実験の内部標準として用い、記載(L.ソロミンら(L. Solomin et al.)J. Virol. 64:6010-6017(1990))のとおり定量した。結果は図2に示した。
HLtat細胞を上述の記載どおり形質転換し、形質転換の20時間後に集めた。総RNAをヘパリン/DNase法(Z.クラウチックとC.ウ(Z. Krawczyk and C. Wu)Anal. Biochem. 165:20-27(1987))により調製し、総RNAの20μgについて記載されたとおり(M.ハッツポロークラダラスら(M. Hadzopoulou-Cladaras et al.)J. Viro1. 63:1265-1274(1989))ノザンブロット解析を行った。フィルターは、HIV-13’LTRのヌクレオチド8304-9008に広がるDNA断片をPCR増幅し、ニック翻訳したものに対しハイブリダイゼーションさせた。結果は図3に示した。
HIV-1ENV遺伝子
env遺伝子断片をベクターp19あるいはp37M1234に挿入し、得られたプラスミドの発現をHLtat細胞に形質転換することによって分析した。いくつかの断片はたんぱく質発現を阻害することが見いだされた。同定された強力なINSのうち1つはヌクレオチド8206-8561(”断片[8206-8561]”)を含んでいる断片中にある。このINSを除去するために以下のオリゴヌクレオチドが合成され、上述に特定したような突然変異導入実験に使用した。断片はHXB2とほとんど同一の分子クローンpNL43由来である。本明細書中で使用されている番号付の体系は一貫して分子クローンHXB2の番号付けに従う。合成オリゴヌクレオチドはpNL43配列に従う。
1.1 HIVのenvmRNA発現の正及び負の決定因子
envを発現するcDNAの同定及び特徴付けのこれまでの実験によりEnvはエキソン4AE、4BE、あるいは5Eを含むmRNAより生産されることが実証された(Schwartzら., J. Virol. 64:5448-5456(1990);Schwartzら., Mol. Cell. Biol. 12:207-219(1992))。envの発現を決定するために作成したすべての構築物はpNL15E由来である。このプラスミドはHIV-1 LTRプロモーター、全長のenv cDNA15Eおよびポリアデニル化シグナルのあるHIV3’LTRを含む(Schwartz,ら. J. Virol. 64:5448-5456(1990))(図7)。pNL15Eは分子クローンpNL4-3由来で(pNL4-3は本明細書中のpNL43と同一である)(Adachiら., J. Virol. 59:284-291(1986)、tev mRNAを生じるのに使用されるエキソン6Dのスプライス受容部位を欠いている(Benkoら., J. Virol. 64:2505-2518(1990))。Env発現プラスミドをRev発現プラスミドpL3crev(Felberら., J. Virol. 64:3734-3741(1991))の存在下あるいは非存在下にて構成的にTatを発現している(Tat1エキソン)HLtat細胞(Schwartzら., J. Virol. 64:2519-2529(1990))に形質転換した。1日後、細胞をRNAおよびタンパク質を分析するために集菌した。全RNAを抽出し、ノーザンブロット分析した。タンパク質産物は細胞と結合したEnvを検出するためにウエスタンブロットによって測定した。Revが存在しないと、NL15E mRNAは効率的にスプライスされて、Nefを産生し;Revが存在すると、ほとんどのRNAはスプライスされないままで、前駆体の分泌部分であるgp120およびgp41に加工されるEnv前駆体gp160を生産する。
env発現におけるスプライシングの影響を研究するために、nt5592のスプライス供与体を部位特異的突然変異導入によって(GCAGTAをGaAtTcに変化させ、EcoRI部位を導入した)除去し、その結果プラスミド15ESDを作成した(図7)。この構築物からのmRNAは効率的にスプライスされて、Nefをコードする小さなmRNAを生産した(図8)。配列分析によってスプライスされたmRNAはnt5605(TACATgtaatg)に位置する別のスプライス供与体およびnt7925の共通のスプライス受容位の利用によって生じたということが明らかになった。発表された仕事とは反して(Luら., Proc. Natl. Acad. Sci. USA87:7598-7602(1990))、この変異体からのEnv発現はRevに依存した。次に、スプライス受容位をnt7925に突然変異を導入した。先のcDNAクローニングによりnt7925のスプライス受容位に加えて、nt7897およびnt7901(Schwartzら., J. Virol. 64:2519-2529(1990))の2つのさらなるスプライス受容位があることが明らかにされたので、nt7884からnt7926に広がる43bpの領域を除去した。これでp15EDSSが生じた(図7)。この構築物で形質転換したHLtat細胞からのmRNAをノーザンブロット分析したところ、15EDSSmRNAはスプライスされないことが確証された(図8B)。すべての機能的なスプライス部位をp15EDSSから除去したにもかかわらず、RevはEnv産物にいまだ必要である(図8A)。gag発現の研究により得られたデータと併せると、これらの結果は非効率的に使用されるスプライス部位の存在はRev依存的Env発現の第一決定因子ではないことが示唆される。少なくとも2つの使用されないスプライス部位がこのmRNA中に存在することが知られている(nt5605の選択的スプライス供与体およびnt6269エキソン6Dのスプライス受容位)。それゆえ、スプライシングを遂行しない初期のスプライソソーム形成がおこりうる可能性は除外できない。これはmRNAが核の中にとどまるのに十分で、スプライシングが起こらないためmRNAの分解を導く可能性がある。あるいは、gag/pol領域内の同定されたもの(INS)と同様のスプライス部位非依存性RNA要素がRev依存性に重要である可能性がある(Schwartzら., J. Virol. 66:7176-7182(1992);Schwartzら., J. Virol. 66:150-159(1992))。
これらの可能性を区別するために、このようなINS要素の位置を決定できるように様々な構築物を設計した。まず始めにNruIおよびMluIの制限酵素部位を伴う停止コドンをプラスミドNL15EDSS上のnt7301の細胞外gp120および膜貫通タンパク質gp41のあいだの切断部位に導入し、p120DSS(図7)を作成した。p120DSSで形質転換した細胞の培地からのgp120の免疫沈降によりRev存在下でのみgp120が高レベルで生産されることを確証した(図9B)。細胞に付着して残っている量はごくわずかに検出できる程度なので、gp120の放出はとても効率的である(データは示さない)。この発見により、env cDNAのgp41部分の翻訳はenv発現の失陥に関与する可能性が否定される。次に、gp120の3’領域の終止コドン(RREおよび3’LTRを含むgp41を含む)をSV40ポリアデニル化シグナルで(図7)置き換えた。この構築物p120pAは、Rev非存在下でgp120をとても低いレベルで生産した(図9B)。Envの背景のレベルはRREを含むgp41の5’部分(MluIからnt8200のHpaI)(図9B)を除去することによってpBS120DSSから作成したp120DR(図7)より測定した。これらの結果はgp120部位内に主要なINS様配列が存在することを示す。このmRNAに対するRevの効果を研究するために、異なるRRE(RRE330、RRE270、およびRRED345(Solominら., J. Viril. 64:6010-6017(1990))をp120pAのgp120終止コドンの下流に挿入してそれぞれp120R330、p120R270、およびp120RD345を作成した(図7)。免疫沈降により、トランスにRevおよびシスにRREが存在するとgp120を発現するプラスミドの欠損を補うことができることが示された。高レベルのgp120がRev存在下でp120R330(データは示さない)、p120R270、およびp120RD345(図9B)より生産された。
in vivoで抑制制御効果を持つ要素を同定するために、env cDNA断片を2つの異なる試験発現ベクターp19およびp37M1-10Dに挿入した。これらのベクターは、遺伝子産物の迅速な検出のための強力なプロモーター(例えば、Tat存在下のHIV-1LTR)および高レベルで発現して、測定が容易である指標遺伝子(例えば、どちらも既知のINS様要素を含んでいないRSVのp19gagあるいはHIV-1(p37M1-10D)の突然変異の入ったp37gag遺伝子)を含む。発現ベクターp19はHIV-1LTRプロモーター、RSVp19gag基質遺伝子、および完全な3’LTRを含むKpnI(nt8561)で始まるHIV-1配列(Schwartz,ら., J. Virol. 66:7176-7182(1992))を含む。HLtat細胞に形質転換すると高レベルのp19gagが構成的に生産され、ウエスタンブロットで視覚化された。発現ベクターp37M1-10DはHIV-1LTRプロモーター、突然変異の入ったp37gag(M1-10)、およびKpnI(nt8561)で始まるウイルスの3’部分を含む。HLtat細胞に形質転換するとこのプラスミドはHIV-1p24gag抗原捕捉測定によって定量できるp37gagを構成的に生産する。
gp41およびgp120部分内のINS要素が同定された。この末端にベクターp19が使用され、以下の断片(図10)が挿入された:(A)nt7684から7959;(B)nt7684から7884およびnt7927から7959;これは断片Aと同様であるがスプライス受容部位7A、7Bおよび7の領域が欠失している;(C)nt7595から7884およびnt7927から7959でBのようにスプライス部位が欠失している;(D)nt7939から8066;(E)nt7939から8416;(F)nt8200から8561(HpaI-KpnI);(G)もとのままのRREを含むnt7266から7595;(H)スプライス供与部位SD5が欠失しているnt5523から6190
断片A、B、およびDはGag発現に影響をおよぼさなかったが、断片G(RRE)はgag発現が約5倍減少した。断片C、E、およびHはGag発現を約10-20倍下げ、INS要素の存在を示唆した。
Envのアミノ酸組成に影響を与えずにこの330nt領域内に103の点突然変異を導入した6つの合成オリゴヌクレオチド(表3)を作成した。INS要素が破壊されたことを証明するために、変異を導入した断片Fをp19で試験した。オリゴ#1内への変異の導入はp19gag発現に影響を与えるには不十分だった一方、すべてのオリゴ(#1から#6)が存在すると断片FのINS効果を完全に不活性化した。これは、INS効果を除去するためにはINS要素内の1つ以上の領域を突然変異を導入する必要があるという別の例である。
envコード領域を異なる連続した断片に分離した。これらの断片およびそれらの組合せを図11に示したようなオリゴを用いてPCRで増幅し、p37M1-10D中の突然変異の導入されたp37gag遺伝子の下流に挿入した。プラスミドをHLtat細胞に形質転換し、翌日に集菌してp24gag発現を解析した。図11は、1+2+3の組合せおよび断片2、3、5の存在は実質的にgag発現を低下させることを示す。Envのアミノ酸組成を変化させずにヌクレオチドを変化させることによってenvコード領域内に位置する3つのAATAAA要素を含むATリッチなドメインを変化させる異なるオリゴ(表4)を合成した。第一のアプローチで、これらのオリゴ1-19を停止点とともにプラスミドp120R270中に導入し、あるいはRev非依存的な方法でgp120を生産する。それからオリゴ20-26のようなオリゴヌクレオチドを2つのenv部位が結合したgp41部位に導入し、完全なgp160はRev非依存的な方法で発現する。
原癌遺伝子C-FOS
fos遺伝子断片をp19ベクターに挿入して、作成したプラスミドの発現をHLtat細胞中に形質転換して解析した。いくつかの断片がタンパク質発現を阻害することが見いだされた。強力なINSがヌクレオチド3328-3450(”断片[3328-3450]”)を含む断片中に同定された(fos遺伝子のヌクレオチドはGenebankシーケンスエントリーHUMCFOT、ACCESSION#V01512に従って数えられている)。さらに、少し弱い要素がコード領域中に同定された。
HIV-1 POL遺伝子
AW Cochraneら.,”ヒト免疫不全ウイルス構造遺伝子発現を抑制する遺伝子内配列の同定および特徴付け”, J. Virol. 65:5305-5313(1991)によって特徴付けられたHIV-1のpol遺伝子から阻害/不安定配列を除去するためにベクターp37M1234が使用された。これらの研究者はCRSと名付けたpol中の領域(HIVヌクレオチド3792-4052)が阻害に重要であるということを示唆した。ヌクレオチド3700-4194を含むこの領域よりも大きい断片をベクターp37M1234中に挿入し、作成したプラスミド(プラスミドp37M1234RCRS)(図12参照)からのp37gag発現の効果をHLtat細胞に形質転換した後に分析した。
情報の配列はHIVHXB2R中に見いだされる野生型HIV-1であり、下方は突然変異の入ったオリゴヌクレオチド配列である。配列の一は括弧内に示した。

免疫予防措置あるいは免疫治療におけるP37M1-10DあるいはP55M1-13POの使用
出生後の遺伝子治療において、身体から標的細胞を除去して、それらに新しい遺伝情報を持ったウイルスのベクターを感染し、それからそれらを身体中に再び移植するといった間接的な方法で;あるいはリポソーム中に処方したDNAをカプセル化する;ウイルス外殻受容タンパク質を含むプロテオリポソームにDNAを封入する;DNAをリン酸カルシウム共沈する;およびポリリジン糖タンパク質担体複合体にDNAを結合させるといった直接的な方法で新しい遺伝情報が組織中に導入されている。さらに、リン酸カルシウム共沈の形成いかんにかかわらず、直接的な肝臓内注入後のクローン化したDNA配列のin vivoにおける感染性についても記述されている。安定性を増強する要素を含むmRNAもまた正に荷電した脂肪小胞の使用でアフリカツメガエル(Xenopus laevis)胚中で効率的に転写されることが示された。例えば、J. ベクターp37M1-10Dあるいはp55M1-1PO(図6)中の突然変異を導入したgagゲノム配列をCMVあるいはRSVのような強力な構成的プロモーターあるいはHIV-1のような誘導可能なプロモーターを用いた発現ベクター中に挿入する。
トランス優性(TD)-TD-GAG-TDまたは
TD GAG-PRO-TD REV遺伝子を用いたHIV-1発現の阻害
DNAの直接注入あるいはレトロウイルスベクター以外のベクターの使用は細胞中でトランスに優性なgag(TDgag)を構成的に高レベルに保つ。さらに、B. K. Felberら., Science 239:184-187(1988)によって採用されたアプローチによって例えばレトロウイルスベクターによるヒト細胞への感染を干渉しないHIV-1TDgagをコードしたマウス由来のレトロウイルスベクターのようなレトロウイルスベクターの作成が可能になった。Felberら.,上述、のアプローチにおいて、プロモーターおよびポリAシグナルの一部を含むHIV-1LTR断片はマウスレトロウイルスベクター内で有害な効果無しに組み込まれ、転写されないままでいることが示された。Tatタンパク質の存在によりHIV-1LTRからの転写が刺激され、HIV-1LTRに結合した遺伝子の高レベルでの発現が生じた。
例えばTronoら.,上述、中に記載されたようなトランス優性として作用するgag突然変異タンパク質は、トランス優性gagタンパク質を高度に構成的レベルで生産する修飾したベクターp37M1-10Dあるいはp55M1-13POによって作成する。

- 担体が哺乳動物に対して投与するための医薬的に許容可能な担体であり;そして哺乳動物体内で抗体を誘導するために有効な量のHIV Envタンパク質を発現することができる量で核酸構築物が存在する;哺乳動物体内でHIV Envタンパク質と反応する抗体を誘導するために有用な、請求項7に記載の組成物。
- 担体が医薬的に許容可能な担体であり;そして、哺乳動物体内において細胞傷害性Tリンパ球を誘導するために有効な量のHIV Envタンパク質を発現することができる量で核酸構築物が存在する;哺乳動物体内において細胞傷害性Tリンパ球を誘導するために有用な、請求項7に記載の組成物。
- (a)そのコード配列が多重点変異を含む変異HIV-1 env遺伝子を含む核酸構築物でトランスフェクトした宿主細胞を提供し;
(b)阻害/不安定性配列を変異させて、それにより対応する天然のHIV-1 env遺伝子の核酸配列中に存在する阻害/不安定性配列の作用を低下させ;
(c)この宿主細胞を培養して、このポリペプチドの発現を生じさせ;そして
(d)このポリペプチドを回収する;

を含む、請求項8に記載の核酸構築物。 - HIV Revタンパク質の非存在下にてHIV Polタンパク質を生成することができる核酸配列を含む核酸構築物であって、この核酸配列が、HIV-1分子クローンpHXB2のヌクレオチド配列の番号付けを使用して、以下の配列:

- 請求項10〜12のいずれか1項に記載の核酸構築物を含む宿主細胞。
- 請求項10〜12のいずれか1項に記載の核酸構築物および担体を含む組成物。
- 担体が医薬的に許容可能な担体であり;そして哺乳動物体内において抗体を誘導するために有効な量のHIV Polタンパク質を発現することができる量で核酸構築物が存在する、哺乳動物体内においてHIV Polタンパク質と反応する抗体を誘導するために有用な、請求項15に記載の組成物。
- 担体が医薬的に許容可能な担体であり;そして哺乳動物体内において細胞傷害性Tリンパ球を誘導するために有効な量のHIV Polタンパク質を発現することができる量で核酸構築物が存在する、哺乳動物体内において細胞傷害性Tリンパ球を誘導するために有用な、請求項15に記載の組成物。
- (a)そのコード配列が多重点変異を含む変異HIV-1 pol遺伝子を含む核酸構築物でトランスフェクトした宿主細胞を提供し;
