Description
1)界面活性剤により保護されたコアまたはコア/シェル構造の疎水性無機物ナノ粒子に、炭素数8〜20個の炭化水素鎖によりチオール基と親水基が結合された有機配位子を1〜30当量添加して界面活性剤を部分的に置換し、ナノ粒子の表面に金属チオラート(M−S)結合を形成することによって、一部分だけが親水性に表面改質され、無極性有機溶媒で個別分散性を維持する疎水性ナノ粒子を製造する段階;
2)前記段階1)で製造されたナノ粒子の表面上に導入された親水基に機能性分子を結合させて個別分散性を維持しながら、ナノ粒子の表面に官能性及び不規則な表面構造を付与する段階;及び
3)前記段階2)で製造されたナノ粒子の表面上に残存する残りの界面活性剤を少なくとも2個の親水基が炭素数1〜7個の炭化水素鎖により結合された有機配位子で置換することによって前記ナノ粒子を親水性ナノ粒子に変換させる段階を含む、バイオイメージングナノ粒子の製造方法を提供する。
従来のバイオイメージングナノ粒子製造のための表面改質方法は、親水性有機配位子を表面に結合することによって製造された親水性ナノ粒子が水溶液内でそれ自体として、又はバイオ分子との結合反応を行う過程で表面に導入された親水基の均一な構造により誘発される水素結合の引力による凝集及び沈澱現象によりその収率が顕著に減少した。
1)界面活性剤により保護されたコアまたはコア/シェル構造の疎水性無機物ナノ粒子に、炭素数8〜20個の炭化水素鎖によりチオール基と親水基が結合された有機配位子を1〜30当量添加して界面活性剤を部分的に置換し、ナノ粒子の表面に金属チオラート(M−S)結合を形成することによって、一部分だけが親水性に表面改質され、無極性有機溶媒で個別分散性を維持する疎水性ナノ粒子を製造する段階;
2)前記段階1)で製造されたナノ粒子の表面上に導入された親水基に機能性分子を結合させて個別分散性を維持しながら、ナノ粒子の表面に官能性及び不規則な表面構造を付与する段階;及び
3)前記段階2)で製造されたナノ粒子の表面上に残存する残りの界面活性剤を、少なくとも2個の親水基が炭素数1〜7個の炭化水素鎖により結合された有機配位子で置換することによって前記ナノ粒子を親水性ナノ粒子に変換させる段階を含む。
疎水性CdSe/CdS−ODA量子点溶液(8×10−5M)5mlを真空蒸発させて溶媒を除去してから20mlのクロロホルムに分散させ、1000当量のデシルアミン(decylamine、DA)を加えて暗い不活性雰囲気下で2日間攪拌した。その結果、得られた溶液にアセトンを添加してからその生成沈殿物を遠心分離し、クロロホルムに分散させて20mlのCdSe/CdS−DA溶液(2×10−5M)を製造した。得られたCdSe/CdS−DA試料は赤外線分光光度計及び透過電子顕微鏡(tranmission electron microscope,TEM)で分析し、その結果を図3のbと図4のbに各々示した。TEMイメージからCdSe/CdS−DA量子点間の間隔がCdSe/CdS−ODAより短くなることから配位子の交換が発生したことを確認した。
実施例1で製造した17mlのCdSe/CdS−DA溶液に5当量のメルカプトウンデカン酸(mercaptoundecanoic acid,MUA)を加えて暗い不活性雰囲気下で19時間攪拌した。前記溶液を濃縮させた後、アセトンを添加して生成された沈殿物を遠心分離し、クロロホルムに分散させて17mlのCdSe/CdS(−DA)ex(−MUA)5溶液(2×10−5M)を製造した。得られた試料は赤外線分光光度計及びTEMで分析し、その結果を図3のcと図4のcに各々示した。MUAの部分置換により図2の段階3のように均一な構造の破壊に起因して自己集合(self−assembly)構造がこれ以上発生しないということをTEMのイメージから確認した。
実施例2で製造した2mlのCdSe/CdS(−DA)ex(−MUA)5溶液にクロロホルムを添加して最終容量が10mlになるように希釈した。該溶液に5当量のジシクロヘキシルカルボジイミド(dicyclohexylcarbodiimide,DCC)を加えて暗い不活性雰囲気下で3時間攪拌した後、下記のように製造されたen−FAを50当量加え、さらに2時間攪拌した。得られた溶液を濃縮させた後、アセトンを添加して生成された沈殿物を遠心分離してクロロホルムに分散させて10mlのCdSe/CdS(−DA)ex(−MUA−en−FA)5溶液(4×10−6M)を製造した。en−FAがMUAに結合されたことを赤外線分光光度計及びTEMで分析し、その結果を図3のdと図4のdに各々示した。
441mg(1 mmol)の葉酸を乾留した10mlのトルエンに注入してから1時間攪拌した後、真空蒸発によりトルエン及び水分を除去した。その結果、得られた残留物をジメチルホルムアミド(dimethylformamide,DMF)に溶解させた後、当該フラスコを氷浴に浸して10分間攪拌した。前記混合物に226mg(1.1mmol)のDCCを加え、さらに18時間攪拌した。2.5mlのエチレンジアミン(ethylenediamine,en)を20mlのDMFに溶解させ、これに前記混合物を加えてから3時間攪拌した。その結果、得られた前記溶液に水とアセトニトリルを添加して沈殿物を形成し、同一の溶媒で再結晶してen−FA224mgを得た。
実施例2で製造した0.5mlのCdSe/CdS(−DA)ex(−MUA)5溶液にクロロホルムを添加して最終容量が2.5mlになるように希釈した。前記溶液に6当量のDCCを加えて暗い不活性雰囲気下で3時間攪拌した後、O,O’−ビス(2−アミノエチル)ポリエチレングリコール(O,O’−Bis(2−aminoethyl)polyethylene glycol,aPEGa,分子量1,000)を7当量加え、さらに16時間攪拌した。得られた溶液を濃縮した後、アセトンの添加により生成された沈殿物を遠心分離し、クロロホルムに分散させて10mlのCdSe/CdS(−DA)ex(−MUA−aPEGa)5溶液(1×10−6M)を製造した。
実施例2で製造した2mlのCdSe/CdS(−DA)ex(−MUA)5溶液にクルロロホルムを添加して最終容量が10mlになるように希釈した。前記溶液に6当量のDCCを加えて暗い不活性雰囲気下で3時間攪拌した後、下記のように製造されたaPEGa−FAを15当量加え、さらに16時間攪拌した。得られた溶液を濃縮させた後、アセトン及びメタノールを添加して生成された沈殿物を遠心分離し、クロロホルムに分散させて4mlのCdSe/CdS(−DA)ex(−MUA−aPEGa−FA)5溶液(1×10−5M)を製造した。
441mg(1mmol)のFAを乾留した10mlのトルエンに注入してから1時間攪拌した後、真空蒸発によりトルエン及び水分を除去した。得られた残留物を14mlのジメチルホルムアミドに溶解させた後、当該フラスコを氷浴に浸して10分間攪拌した。前記混合物に226mg(1.1mmol)のDCCを加え、さらに18時間攪拌した後、1mmolのaPEGaを加え、さらに3時間攪拌した。得られた溶液にジメチルエーテルを加えて沈殿物を形成し、同一の溶媒で再結晶してaPEGa−FA179mgを得た。
実施例1で製造した0.5mlのCdSe/CdS−DA溶液にクロロホルムを添加して最終容量10mlになるように希釈した3つの希釈剤を準備した。前記各希釈剤に5、10及び30当量のメルカプトウンデカン酸(mercaptoundecanoicacid,MUA)それぞれを加えて暗い不活性雰囲気下で19時間攪拌した。得られた溶液を濃縮させた後、メタノールを添加して生成された沈殿物を遠心分離し、クロロホルムに分散させて2.5mlのCdSe/CdS(−DA)ex(−MUA)n(n=5、10または30)溶液(4×10−6M)を各々製造した。
粒度8nmの疎水性SPION−OA磁性ナノ粒子溶液(3×10−6M)2mlをエタノールで2回洗浄し、20mlのトルエンに分散させてから得られた溶液を100℃に昇温した。前記溶液に10当量のメルカプトヘキサデカン酸(mercaptohexadecanoic acid,MHA)を加えて1時間還流して冷却させた。得られた溶液の2mlを濃縮した後、小量のエタノールを添加してから磁石を用いて部分的に表面改質された磁性ナノ粒子SPION(−OA)ex(−MHA)10を分離した。前記分離された部分的に表面改質された磁性ナノ粒子を赤外線分光光度計及びTEMで分析して図8のbと図9のbに各々示した。TEMのイメージにおいて、SPION(−OA)ex(−MHA)10の量子点間の間隔がSPION−OAと類似する理由は、OAの一部を炭素鎖の長さがOAより短い2つのMHAで、図2の段階Cに示したように、散発的に置換したからである。さらに、赤外線分光光度計でMHAの置換により3300cm−1の周りのO−Hピークが増加したことを確認した。
実施例7で製造した18mlのSPION(−OA)ex(−MHA)10溶液(3×10−7M)にDCC30当量を加えて3時間攪拌した後、下記のように製造されたaPEGa−MTX30当量を小量のDMFに溶解させて添加してから16時間攪拌した。得られた溶液を濃縮してエタノールを添加した後、磁石を用いてSPION(−OA)ex(−MHA−aPEGa−MTX)10を分離し、9mlのクロロホルムに分散させた。前記分散液の1mlを赤外線分光光度計及びTEMで分析し、その結果を図8のcと図9のcに各々示した。TEMのイメージにおいて、磁性ナノ粒子が自己集合によって得られる六角形の配列を破壊し、ばらばらに散在していることからaPEGa−MTXにより不規則な構造が導入されたことが分かる。また、赤外線分光光度計で1100及び1630cm−1の回りにPEG及びMTXの特性ピークが現れたことを確認した。
454mg(1 mmol)のMTX(methotrexate,USP標準参照)を乾留した14mlのジメチルホルムアミドに溶解させ、当該フラスコを氷浴に浸してから10分間攪拌した。前記溶液に226mg(1.1 mmol)のDCCを加えてから18時間攪拌した後、1 mmolのaPEGaを加え、さらに3時間攪拌した。得られた溶液にジエチルエーテルを加えて沈殿物を形成して同一の溶媒で再結晶してaPEGa−MTX138mgを得た。
実施例8で製造した8mlのSPION(−OA)ex(−MHA−aPEGa−MTX)10溶液(6×10−7M)に0.05M MPAと0.05M NaOHを溶解させたメタノール溶液を150当量添加してから2時間攪拌した。得られた溶液にメタノールを添加し、遠心分離により得られた個体SPION(−MPA)ex(−MHA−aPEGa−MTX)10をpH7.4である16mlのPBS緩衝溶液に分散させた。前記分散液1mlを赤外線分光光度計及びTEMで分析し、その結果を図8のdと図9のdに各々示した。赤外線分光光度計において、OAがMPAで置換されることによって炭素鎖の長さが減少するので、C−Hピークの強度が顕著に減少されたことが分かる。さらに、TEMのイメージにおいて、試料製造の際、水溶液の使用により表面張力に起因した重なり効果(overlap effect)があるにもかかわらず、粒子間の距離が確保されたことを確認した。
実施例7及び8で製造した6mlのSPION(−OA)ex(−MHA−aPEGa−MTX)10溶液(6×10−7M)に臭化テトラオクチルアンモニウム(tetraoctylammonium bromide,TOAB)1000当量を加えてから16時間攪拌した。前記混合物に6mlの0.1Mリジン(lysine,Lys)水溶液を加えてから19時間攪拌した。得られた溶液にメタノールを添加し、磁石を用いてSPION(−Lys)ex(−MHA−aPEGa−MTX)10ナノ粒子を分離した後、各々5mlのトルエンとエタノールで洗浄し、pH7.4であるPBS緩衝溶液に分散させた。前記分散液1mlを赤外線分光光度計及びTEMで分析し、その結果を図8のeと図9のeに各々示した。赤外線分光光度計において、OAがリジンで置換されることによって炭素鎖の長さが減少するので、C−Hピークの強度が顕著に減少されたことが分かる。さらに、TEMのイメージにおいて、試料製造の際、水溶液の使用により表面張力に起因した重なり効果があるにもかかわらず、粒子間の距離が確保されたことを確認した。
粒度11nmの疎水性SPION−OA磁性ナノ粒子溶液(3×10−6M)2mlをエタノールで2回洗浄し、20mlのトルエンに分散させた溶液を100℃に昇温した。前記混合物に5当量のメルカプトヘキサデカン酸(mercaptohexadecanoic acid,MHA)を加えてから1時間還流して冷却した。得られた溶液に15当量のDCCを加えて暗い不活性雰囲気下で3時間攪拌した後、実施例3で製造したen−FA15当量を小量のDMFに溶解させて加えてから16時間攪拌した。得られた溶液を濃縮させてエタノールを添加した後、磁石を用いてSPION(−OA)ex(−MHA−en−FA)5を分離して10mlのクロロホルムに分散させた。前記ナノ粒子の赤外線スペクトルを図8のfに示し、1630cm−1の周りにFAの特性ピークを確認した。

Claims (16)
- 1)界面活性剤により保護されたコアまたはコア/シェル構造の疎水性無機物ナノ粒子に、炭素数8〜20個の炭化水素鎖によりチオール基と親水基とが結合された有機配位子を1〜30当量添加して界面活性剤を部分的に置換し、ナノ粒子の表面に金属チオラート(M−S)結合を形成することによって、一部分だけが親水性に表面改質され、無極性有機溶媒で個別分散性を維持する疎水性ナノ粒子を製造する段階;
2)前記段階1)で製造された疎水性ナノ粒子の表面上に導入された親水基に機能性分子を結合させて個別分散性を維持しながら、ナノ粒子の表面に官能性及び不規則な表面構造を付与する段階;及び
3)前記段階2)で製造されたナノ粒子の表面上に残存する残りの界面活性剤を少なくとも2個の親水基が炭素数1〜7個の炭化水素鎖により結合された有機配位子で置換することによって前記ナノ粒子を親水性ナノ粒子に変換させる段階を含む、バイオイメージングナノ粒子の製造方法。 - 前記段階1)における疎水性無機物ナノ粒子が周期律表上のII族元素である亜鉛、カドミウム及び鉛からなる群から選ばれるいずれか一つの元素と、周期律表上のVI族元素である硫黄、セレニウム及びテルリウムからなる群から選ばれるいずれか一つの元素からなる半導体ナノ粒子、貴金属ナノ粒子または酸化鉄ナノ粒子であることを特徴とする請求項1に記載のバイオイメージングナノ粒子の製造方法。
- 前記疎水性無機物ナノ粒子がCdSe、ZnS、CdSe/CdS、CdSe/ZnS、Au、Ag、Fe2O3及びFe3O4からなる群から選択されることを特徴とする請求項2に記載のバイオイメージングナノ粒子の製造方法。
- 前記段階1)における有機配位子が分子内に少なくとも1個のチオール基と少なくとも1個の親水基とを含み、前記親水基はアミノ基、カルボキシル基、ヒドロキシル基及びチオール基からなる群から選択されることを特徴とする請求項1に記載のバイオイメージングナノ粒子の製造方法。
- 前記有機配位子がメルカプトウンデカン酸(mercaptoundecanoic acid)、メルカプトドデカン酸(mercaptododecanoic acid)及びメルカプトヘキサデカン酸(mercaptohexadecanoic acid)からなる群から選択されることを特徴とする請求項4に記載のバイオイメージングナノ粒子の製造方法。
- 前記段階1)におけるナノ粒子の表面に金属チオラート(M−S)共有結合によって化学的に結合することができる有機配位子の数が1〜30個であることを特徴とする請求項1に記載のバイオイメージングナノ粒子の製造方法。
- 前記段階2)における機能性分子が少なくとも一方の末端にアミノ基、アルデヒド基、カルボキシル基、ヒドロキシル基及びチオール基からなる群から選択される親水基が結合された生体適合性分子、標的志向性分子、これらの錯体または混合物であることを特徴とする請求項1に記載のバイオイメージングナノ粒子の製造方法。
- 前記段階3)における有機配位子が分子内に少なくとも2個の親水基を含み、前記親水基はアミノ基、カルボキシル基、ヒドロキシル基及びチオール基からなる群から選択されることを特徴とする請求項1に記載のバイオイメージングナノ粒子の製造方法。
- 前記有機配位子がメルカプトヘキサン酸(mercaptohexanoic acid)、メルカプト酢酸(mercaptoacetic acid)、メルカプトプロピオン酸(mercaptopropionic acid)、ジメルカプトコハク酸(dimercaptosuccinic acid)、2−メルカプトエタノール(2−mercaptoethanol)、2−アミノエタンチオール(2−aminoethanethiol)、リジン(Lysine)、アルギニン(arginine)及びアミノ吉草酸(aminovaleric acid)からなる群から選択されることを特徴とする請求項8に記載のバイオイメージングナノ粒子の製造方法。
- 前記生体適合性分子が両側の末端にアミノ基、アルデヒド基及びカルボキシル基のうち選ばれた親水基を有するか、一方の末端にはアミノ基、アルデヒド基及びカルボキシル基のうち一つを有し、他方の末端には炭素数1〜7個のアルコキシ基またはヒドロキシル基を有することを特徴とする請求項7に記載のバイオイメージングナノ粒子の製造方法。
- 前記生体適合性分子がポリエチレングリコール(PEG)、デキストラン、ポリ(L−ラクチド)(PLLA)、ポリ(DL−ラクチド)(PDLLA)、ポリ−DL−ラクチド/グリコリド共重合体(PLGA)、キトサン、アルギン酸、ヒアルロン酸、コラーゲン、ヘパリン及びポリ(ε−カプロラクトン)からなる群から選択されることを特徴とする請求項10に記載のバイオイメージングナノ粒子の製造方法。
- 前記標的志向性分子が生体内で特異的に認知されることができ、アミノ基、アルデヒド基、カルボキシル基、ヒドロキシル基及びチオール基からなる群から選択されるいずれかの基を有しているので、生体適合性分子とアミド結合、エステル結合及びチオエステル結合のうちいずれか一つによって結合されることができることを特徴とする請求項7に記載のバイオイメージングナノ粒子の製造方法。
- 前記標的志向性分子が葉酸(folic acid)、メトトレキサート(methotrexate、MTX)、特定細胞に選択的なペプチド、または特異抗原に選択的に反応する抗体であることを特徴とする請求項12に記載のバイオイメージングナノ粒子の製造方法。
- 請求項1に記載の製造方法の段階1)によって製造されるナノ粒子であって、界面活性剤により保護されたコア及びコア/シェル構造の疎水性無機物ナノ粒子の表面の一部分に、炭素数8〜20個の炭化水素鎖によってチオール基と親水基が結合された有機配位子を含み、その部分は親水性を示すが、全体的には疎水性を示し、無極性有機溶媒内で個別分散性を維持するバイオイメージングナノ粒子。
- 請求項1に記載の製造方法の段階2)によって製造されるナノ粒子であって、請求項14に記載のナノ粒子の表面に導入された親水基に機能性分子が結合されることにより官能性及び不規則な表面構造を有し、前記機能性分子が結合された部分は親水性を示すが、全体的には疎水性を示すバイオ−イメージングナノ粒子。
- 請求項1に記載の製造方法の段階3)によって製造されるナノ粒子であって、請求項15に記載のナノ粒子の表面に残存する残りの界面活性剤が、炭素数1〜7個の炭化水素鎖により少なくとも2個の親水基が結合された有機配位子で置換することによって親水性に変換され、水溶液内で個別分散性を維持するバイオイメージングナノ粒子。
