Abstract
Description
Claims (33)
- 血液試料中の白血球の体積当たりの計数(volumetric enumeration)を行うための試料獲得デバイスにおいて、
血液試料を受け取るための計測空洞であって、固定された所定の厚さを有する計測空洞と、
該計測空洞を画定する表面上に乾燥した形態で配置された試薬であって、該血液試料中の赤血球を溶解させる溶血剤、及び該血液試料中の白血球を選択的に染色する染色剤を含む試薬と
を含む試料獲得デバイス。 - 前記計測空洞を画定する2つの平面を有する胴体部材を含む、請求項1に記載の試料獲得デバイス。
- 前記平面が、互いから所定の距離のところに配置されて、光学計測のための試料厚さが決定されている、請求項2に記載の試料獲得デバイス。
- 前記計測空洞が50〜170マイクロメートルの均一な厚さを有する、請求項1から3までのいずれか一項に記載の試料獲得デバイス。
- 前記計測空洞が少なくとも100マイクロメートルの均一な厚さを有する、請求項4に記載の試料獲得デバイス。
- 前記計測空洞が150マイクロメートル以下の均一な厚さを有する、請求項4又は5に記載の試料獲得デバイス。
- 揮発性の液体に溶解させた前記試薬が前記表面に供給され、前記揮発性液体が蒸発して、前記試薬が乾燥した形態で残っている、請求項1から6までのいずれか一項に記載の試料獲得デバイス。
- 前記染色剤が、白血球の核を選択的に染色するように構成された、請求項1から7までのいずれか一項に記載の試料獲得デバイス。
- 前記染色剤が、ヘマトキシリン、メチレンブルー、メチレングリーン、メチレンアズール、酢酸クレシルバイオレット、トルイジンブルー、ゲンチアナバイオレット、スダン類似体、ガロシアニン及びフクシン類似体の群のうちのいずれか1つ、又はこれらの任意の組合せである、請求項1から8までのいずれか一項に記載の試料獲得デバイス。
- 前記溶血剤が、第四級アンモニウム塩、サポニン、胆汁酸、ジギトキシン、ヘビ毒、グルコピラノシド又はトリトン(Triton)型の非イオン性界面活性剤である、請求項1から9までのいずれか一項に記載の試料獲得デバイス。
- 前記計測空洞を試料獲得デバイスの外部に連絡する試料入口をさらに含み、前記入口が、血液試料を獲得するように構成された、請求項1から10までのいずれか一項に記載の試料獲得デバイス。
- 使い捨てである、請求項1から11までのいずれか一項に記載の試料獲得デバイス。
- 血液試料中の白血球の体積当たりの計数を行うための方法において、
試料獲得デバイスの計測空洞の中に血液試料を獲得すること
を含み、該計測空洞が、溶血剤、及び白血球が染色されるように該試料と反応する染色剤を含む試薬を保持し、さらに、
染色された該白血球を含む該試料に照射し、
該計測空洞の中の照射された該試料の拡大像のディジタル画像を獲得すること
を含み、該白血球が、該染色剤の選択的な染色によって区別され、さらに、
該ディジタル画像をディジタル方式で解析して、該白血球を識別し、該試料中の白血球数を決定すること
を含む方法。 - 前記血液試料が、前記計測空洞内の前記試薬と混合される、請求項13に記載の方法。
- 前記計測空洞が50〜170マイクロメートルの厚さを有し、前記ディジタル画像が、少なくとも該計測空洞の厚さに対応する被写界深度で獲得される、請求項13又は14に記載の方法。
- 分析される試料の体積が、前記計測空洞の厚さ及び画像化される前記試料の面積によって明確に画定される、請求項15に記載の方法。
- 前記試料が、前記染色剤の吸光度のピークに対応する波長の光によって照射される、請求項13から16までのいずれか一項に記載の方法。
- 前記照射がレーザ源によって実行される、請求項13から17までのいずれか一項に記載の方法。
- 前記照射が発光ダイオードによって実行される、請求項13から17までのいずれか一項に記載の方法。
- 前記ディジタル画像が、3〜200倍、より好ましくは3〜10倍の倍率を使用して獲得される、請求項13から19までのいずれか一項に記載の方法。
- 前記解析が、前記ディジタル画像内の高吸光度の領域を識別することを含む、請求項13から20までのいずれか一項に記載の方法。
- 前記解析が、前記ディジタル画像内の黒いドットを識別することを含む、請求項21に記載の方法。
- 前記解析が、前記獲得されたディジタル画像を電子的に拡大することを含む、請求項13から22までのいずれか一項に記載の方法。
- 血液試料中の白血球の体積当たりの計数を行うためのシステムにおいて、
請求項1から12までのいずれか一項に記載の試料獲得デバイスと、
計測装置と
を含み、該計測装置が、
計測空洞の中に血液試料を保持する該試料獲得デバイスを受け取るように構成された試料獲得デバイスホルダと、
該血液試料に照射するように構成された光源と、
拡大システム、及び該計測空洞の中の照射された前記試料の拡大像のディジタル画像を獲得するためのディジタル画像獲得手段を含む画像化システムであって、染色剤の選択的な染色によって、ディジタル画像内の白血球が区別される画像化システムと、
獲得されたディジタル画像を解析して、血液試料中の白血球を識別し、白血球数を決定するように構成された画像解析機と
を含む、システム。 - 前記拡大システムが、少なくとも試料獲得デバイスの計測空洞の厚さの被写界深度を有するように構成された、請求項24に記載のシステム。
- 分析される試料の体積が、前記計測空洞の厚さ及び画像化される前記試料の面積によって明確に画定される、請求項24又は25に記載のシステム。
- 前記光源が、前記染色剤の吸光度のピークに対応する波長の光を照射するように構成された、請求項24から26までのいずれか一項に記載のシステム。

- 前記光源が発光ダイオードを含む、請求項24から27までのいずれか一項に記載のシステム。
