Abstract
Description
飽和脂肪酸、モノ不飽和脂肪酸、及び非共役ポリ不飽和脂肪酸を含む第一の脂肪酸混合物をエタノールの存在下で処理して、(i)固体画分、及び(ii)飽和脂肪酸及びモノ不飽和脂肪酸の総量に対するポリ不飽和脂肪酸の総量のモル比が第一の脂肪酸混合物よりも高い、第二の脂肪酸混合物を含む液体画分を形成する工程、
前記固体画分と前記液体画分とを分離する工程、並びに
前記第二の脂肪酸混合物又はその誘導体若しくは反応生成物に溶媒の存在下で塩基を使用する処理を施して、少なくとも幾つかのポリ不飽和脂肪酸の共役異性体を形成する工程
を含む、共役ポリ不飽和脂肪酸を含む物質の製造方法を提供する。
27.3gのNaOHを200mlの95% エタノールに溶解した。100gのベニバナ油を添加して、その溶液を2時間還流した。結果として得られた混合物を硫酸で分解(split)し、次いで900mlのヘキサンで3回抽出して、ヘキサン層を合わせて、蒸留水で約pH7まで洗浄し、次いでロータリーエバポレーターを使用して有機相を蒸発させた。270gの尿素を1500mlの95%エタノールに30℃で溶解して、脂肪酸を添加し、次いでその混合物を60℃で1時間加熱して、20℃まで3時間冷却した。その物質を濾過して、尿素結晶を除去し、次いでその濾過液を蒸発させた。16.8g KOHをフラスコ中の55mlのプロピレングリコールに溶解し、次いで蒸発後に濾過液から得られた50gの脂肪酸混合物(90%より多くのリノール酸を含有する)を添加した。前記フラスコに150度で15時間、窒素を勢い良く流した。共役後に、その反応混合物を熱した10%硫酸溶液に入れて、その下層を廃棄し、上層を熱した水でpH7まで洗浄して、最後に窒素で乾燥させた。共役リノール酸は、GC、HPLCで分析した。結果を表1に提示する。
尿素との反応を95%エタノールの代わりにメタノールを使用して実施した以外は同一の条件を正確に使用して、実施例1の手順を繰り返した。反応後に、共役リノール酸を分析した。結果を表1に示す。
WO 97/18320に記載の方法を使用して、実施例1のトリグリセリドを含有するスプレッドを以下のように配合して良い。
WO 97/18320に記載の方法を使用して、実施例1の生産物のグリセリドを含有するドレッシングを以下のように配合して良い。
本実施例は、パイロットプラント規模における濃縮ベニバナ油の共役に関する。
攪拌槽型反応器において窒素ブランケット下で、7700kgの濃縮ベニバナ油を9780kgのプロピレングリコールに添加した。これに対し、5120kgの水酸化カリウム(50重量%溶液、KOH)を添加して、窒素下で110℃まで加熱した。水を除去した後に、温度を139℃まで増大させて、FAME分析におけるC18:2シス含有量が1%未満になるまで、この温度で攪拌した。総反応時間は42時間であった。
Claims (22)
- 飽和脂肪酸、モノ不飽和脂肪酸、及びポリ不飽和脂肪酸を含む第一の脂肪酸混合物をエタノールの存在下で処理して、(i)固体画分、及び(ii)飽和脂肪酸及びモノ不飽和脂肪酸の総量に対するポリ不飽和脂肪酸の総量のモル比が前記第一の脂肪酸混合物よりも高い、第二の脂肪酸混合物を含む液体画分を形成する工程、
前記固体画分と前記液体画分とを分離する工程、並びに
前記第二の脂肪酸混合物又はその誘導体若しくは反応生成物に、溶媒の存在下において塩基を使用する処理を施して、少なくとも幾つかのポリ不飽和脂肪酸の共役異性体を形成する工程
を含む、ポリ不飽和脂肪酸の共役異性体を含む物質を製造するための方法。 - 前記物質が、少なくとも90重量%のポリ不飽和脂肪酸の共役異性体を含む、請求項1に記載の方法。
- 前記ポリ不飽和脂肪酸が、12から24の炭素原子を含有する、請求項1又は2に記載の方法。
- 前記飽和脂肪酸、前記モノ不飽和脂肪酸、及び前記ポリ不飽和脂肪酸が、18の炭素原子を含有する、請求項1から3のいずれか一項に記載の方法。
- 前記共役異性体がリノール酸の共役異性体である、請求項1から4のいずれか一項に記載の方法。
- 前記第一の脂肪酸画分が尿素で処理されて、前記固体画分が尿素を含む、請求項1から5のいずれか一項に記載の方法。
- 前記第一の脂肪酸混合物を、少なくとも80重量%のエタノールを含む溶媒の存在下で処理する、請求項1から6のいずれか一項に記載の方法。
- 前記第二の脂肪酸混合物を、前記塩基を使用する処理の前に更に別の処理工程にかけて、その誘導体又は反応生成物を形成する、請求項1から7のいずれか一項に記載の方法。
- 前記更に別の処理工程が、精製、溶媒の除去、脂肪酸のC1からC6アルキルエステルを形成するエステル化、及びそれらの組み合わせから選択される、請求項9に記載の方法。
- 前記第一の脂肪酸混合物が、脂肪酸の総量に対して50重量%から85重量%の18の炭素原子を含有する脂肪酸を含む油から得られる、請求項1から9のいずれか一項に記載の方法。
- 前記第一の脂肪酸混合物が、溶媒であるエタノールの存在下において塩基を使用する植物油の加水分解を含む方法によって得られる、請求項1から10のいずれか一項に記載の方法。
- 前記植物油が、ベニバナ油、ヒマワリ油、菜種油、綿実油、大豆油、及び亜麻仁油からなる群から選択される、請求項11に記載の方法。
- 前記溶媒が、3つの炭素原子及び少なくとも2つのヒドロキシル基を有するアルコールを含む、請求項1から12のいずれか一項に記載の方法。
- 前記溶媒がプロピレングリコールである、請求項1から13のいずれか一項に記載の方法。
- 塩基を使用する処理後に、前記第二の脂肪酸混合物を酸性化する工程を含む、請求項1から14のいずれか一項に記載の方法。
- 酸性化によって形成される塩の少なくとも一部を除去し、続いて、結果として得られる混合物を精製する、請求項15に記載の方法。
- ポリ不飽和脂肪酸の共役異性体のモノ、ジ、又はトリグリセリドを形成する工程を更に含む、請求項1から16のいずれか一項に記載の方法。
- 飽和脂肪酸及びモノ不飽和脂肪酸と共にポリ不飽和脂肪酸を含有する混合物から、尿素を使用する結晶化によってポリ不飽和脂肪酸を分離するための溶媒としての、エタノールの使用。
- 請求項1から17のいずれか一項に記載の方法によって得られる生産物。
- 少なくとも90重量%の共役リノール酸、少なくとも43重量%の共役リノール酸のシス-9,トランス-11異性体及び少なくとも43重量%の共役リノール酸のトランス-10,シス-12異性体、又はそれらのモノ、ジ、若しくはトリグリセリドを含み、前記シス-9,トランス-11異性体:前記トランス-10,シス-12異性体の重量比が1.1:1から1:1.1の範囲である、請求項19に記載の生産物。
- 1.5重量%未満の共役リノール酸のトランス,トランス異性体を含む、請求項20に記載の生産物。
- 請求項19から21のいずれか一項に記載の生産物を含む、食品、栄養補助食品、又は医薬品。
