Abstract
Description
本発明は、血管新生プロセス関連分子の機能的活性領域を認識し、ブロックするポリペプチドと、プロテイナーゼ感受性領域によって分離された、オリゴマー化ドメインおよび機能的に活性な血管新生プロセスインヒビターまたはモジュレーター領域を含んでなるポリペプチドとを含んでなる多機能性多価血管新生阻害薬として有用な融合タンパク質に関する。本発明はまた、前記融合タンパク質の生産に有用な遺伝子およびベクター構築物にも関する。
発明の背景
本発明は、血管新生によって起こる病変、例えば、乾癬、関節リウマチ、網膜症、癌などを予防および/または治療するための治療薬として潜在的に有用な、抗血管新生活性を有する化合物を提供するという問題に向けられる。
一態様において、本発明は、作動可能に結合された、少なくとも:
a)血管新生プロセス関連分子の機能的活性領域を認識し、ブロックするポリペプチドをコードするヌクレオチド配列を含んでなる、第1の核酸配列(A);並びに
b)(i)オリゴマー化ドメイン、(ii)血管新生プロセスにおける機能的活性領域、および(iii)前記オリゴマー化ドメインと前記血管新生プロセスにおける機能的活性領域の間のプロテイナーゼ感受性領域、を含んでなるポリペプチドをコードするヌクレオチド配列を含んでなる、第2の核酸配列(B)
を含んでなり、
前記第1の核酸配列(A)の3’末端が、前記第2の核酸配列(B)の5’末端に結合されている、
遺伝子構築物(以下、本発明の遺伝子構築物)に関する。
(a)血管新生プロセス関連分子の機能的活性領域を認識し、ブロックする領域を含んでなるポリペプチド(A’);並びに
(b)(i)オリゴマー化ドメイン、(ii)血管新生プロセスにおける機能的活性領域、および(iii)前記オリゴマー化ドメインと血管新生プロセスにおける前記機能的活性領域の間のプロテイナーゼ感受性領域、を含んでなるポリペプチド(B’)
を含んでなる融合タンパク質を提供する。
(i)全長mAb L36、二機能性型のmAb L36、およびフレキシブルペプチドを介して、mAb L36の軽鎖可変領域(VL)と融合されたmAb L36の重鎖可変領域(VH)を含む組換えscFvからなる群から選択されるポリペプチド;並びに
(ii)哺乳類コラーゲンXVIIIのNC1ドメインを含んでなるポリペプチド(B’)
を含んでなる融合タンパク質を提供する。
次の実施例は、本発明を説明する手段となり得るが、その範囲を限定するものではない。
設計および発現、ならびに抗体フラグメントおよびコラーゲンXVIIIオリゴマーで構成される血管新生阻害薬
次の産物を発現することができるいくつかの遺伝子構築物(図3A)を作製した:
抗ラミニンモノクローナル抗体L36の単鎖Fvフラグメント(scFv)(L36 scFv)、
マウスコラーゲンXVIIIのNC1ドメインに存在する三量化ドメイン(残基10〜60)にフレキシブル連結ペプチド(リンカー)を介して融合されたL36 scFvで構成される融合タンパク質(L36 scFv−NC1ES−と表示される)、
マウスコラーゲンXVIIIのNC1ドメイン(残基10〜325)に前記リンカーを介して融合されたL36 scFvで構成される融合タンパク質(L36 scFv−NC1ES+と表示される)、
マウスコラーゲンXVIIIのNC1ドメイン(残基10〜315)(NC1)、および
マウスコラーゲンXVIIIのNC1ドメイン(残基130〜315)由来のエンドスタチン(ES)(このESはタンパク質分解により三量体形で放出され、続いてそれがおよそ20kDaの単量体ESに変換される(図2Aおよび図2B))。
細胞および培養条件
HEK−293細胞(ヒト腎臓上皮;ATCC CRL−1573)、HT−1080細胞(ヒト繊維肉腫;ATCC CCL−121)およびB16−F10細胞(マウス黒色腫;ATCC CRL−6475)は、10%のウシ胎児血清(FBS)(Life Technologies, Gaithersburg, MD, U.S.A.)で強化されたダルベッコ変法イーグル培地(DMEM)で培養した。
連結ペプチド(connector peptide)、三量化ドメイン、ヒンジペプチド、およびESドメインをコードする配列を含むNC1ドメインの配列(図2B)を、B. Olsen博士(Harvard Medical School, Boston, MA, U.S.A.)により提供されたマウス由来α1クローンmc3b(コラーゲンXVIII)[Oh et al. Genomics. 1994 Feb; 19(3):494-9]から、ポリメラーゼ連鎖反応(PCR)によって、プライマー対1および2(表1参照、ベクターの構築、その後のベクター配列の確認に使用される異なるプライマーの配列を含む)を用いて増幅した。
HEK−293細胞、HT−1080細胞およびB16−F10細胞を、リポフェクトアミン(Life Technologies)に基づく系を用いて、好適な発現ベクター(pCR3.1、pCR3.1−L36、pCR3.1−L36−三量体、pCR3.1−ES、pCR3.1−NC1またはpCR3.1−L36−リンカー−NC1)でトランスフェクトした。安定な細胞株を作製するために、トランスフェクトしたHEK−293細胞、HT−1080細胞およびB16−F10細胞を0.5mg/ml、0.6mg/mlまたは3mg/mlのG−418(Life Technologies)を含有するDMEMで選抜した。種々のベクターによって形質導入された親腫瘍細胞の増殖を、3−(4,5−ジメチルチアゾール−2−イル)−2,5−ジフェニルテトラゾイルブロミド(MTT,Promega)を用いたアッセイにより連日4日間毎日解析した。SDS−PAGE[Sanz L et al. Gene Therapy (2002) 15:1049]、ELISA[Sanz L. et al Gene Therapy (2002) 15:1049]および抗myc(Life Technologies)または抗エンドスタチン(Upstate Biotechnology, Lake Placid, NY, U.S.A.)mAb(モノクローナル抗体)を用いたウェスタンブロット(免疫ブロット法)[Blanco et al. J. Immunol (2003) 171:1070]によりタンパク質発現を確認するために、一時的な細胞集団と安定な細胞集団の上清を解析した。
トランスフェクトした、安定なHEK−293細胞を用いて、血清を含まない馴化培地で培養した。その培地にプロテイナーゼ(アプロチニン、ベスタチン、ロイペプチン、ペプスタチンAおよびE−64)阻害薬の組合せ(「カクテル」)(Sigma Biosciences)を添加して、タンパク質分解を減少させた。その培地(およそ1L)を10,000 MWCO ビバフロー50フィルター(Vivascience AG, Germany)で濃縮し(×10)、PBS(リン酸緩衝生理食塩水)、pH7.4で透析し、1mL HisTrap HPカラム(Amersham Biosciences, Uppsala, Sweden)にかけた。ESを含むタンパク質を1mL HiTrap ヘパリンHPカラム(Amersham Biosciences)でさらに精製した。0.1〜2.0NaClの直線勾配により溶出を行った。精製されたタンパク質をPBSで透析し、−20℃で保存した。ES Fc−ドメイン(Fc−ES)融合タンパク質[Cuo CJ et al. J Cell Biol. 2001 152:1233]は、K. Javaherian博士(Boston Children's Hospital, Boston, MA, U.S.A.)により提供されたものであった。前記Fc−ES融合タンパク質は、(ESドメインと免疫グロブリンのFcフラグメントとの)二量体の融合タンパク質であり、毛細血管の形態形成をブロックする。
遠心分離試験は、エポンチャコール(Epon charcoal)中央部に3mmダブルセクターを有するAn50Tiローターを用いる紫外線可視光学系を備えたOptima社製XL−A分析用超遠心機(Beckman-Coulter Inc.)により20℃で行った。低速での沈降天秤をショートカラム(23μl)において、一連の3速度(5,000rpm、6,000rpmおよび11,000rpm)にてアッセイし、天秤の画像を(20時間後)波長280nmにて取得した。試験中、水溶性のタンパク質塊の保存に留意した。沈降速度対照試験では、45時間間隔において凝集は示さなかった。高速遠心分離後(42,000rpmにて5時間)参照シグナルを測定した。EQASSOCプログラムを用いて、全細胞の見かけの重量により平均分子量を得た[Minton, A. P. (1994) in Modern Analytical Ultracentrifugation (Schuster, T., and Laue, T., eds), pp. 81-93, Birkhauser Boston, Inc., Cambridge, MA]。沈降速度試験を42,000rpmにて行い、吸光度イメージを280nmにて取得した。SEDFITプログラムで実行されるように、連続分布c(s) Lamm方程式モデルを用いて沈降係数を算出した[P. Schuck. Size-Distribution Analysis of Macromolecules by Sedimentation Velocity Ultracentrifugation and Lamm Equation Modeling. Biophysical Journal 78 (2000), pp. 1606-1619]。これらの沈降試験値を、SEDNTERPプログラムを用いて常用条件に対して補正して、対応するs20,W値を得た[T.M. Laue, B.D. Shah, T.M. Ridgeway and S.L. Pelletier, Computer-aided interpretation of analytical sedimentation data for proteins. In: S. E. Harding, A. J. Rowe and J. C. Horton, Editors, Analytical Ultracentrifugation in Biochemistry and Polymer Science, Royal Society of Chemistry, Cambridge, UK (1992), pp. 90-125]。c(M)分布を得るためのSEDFITプログラムを用いたさらなる流体力学解析(すなわち、摩擦係数比の算出)[P. Schuck. Size-Distribution Analysis of Macromolecules by Sedimentation Velocity Ultracentrifugation and Lamm Equation Modeling. Biophysical Journal 78 (2000), pp. 1606-1619]。
これらの試験は、AKTA FPLCシステムでSuperdex 200 10/300GLカラムを用いて、室温にて行った。PBS中の異なる組換えタンパク質(L36 scFv、L36 scFv−NC1ES−、L36 scFv−NC1ES+およびNC1)の0.1mLサンプルを濃度0.5〜1.0mg/mLで注入し、流速0.5ml/分で分離した。そのカラムを高分子量マーカーおよび低分子量マーカー(Amersham)で較正した。排除容量に対する溶出容量の比率とマーカーの分子量を指数曲線に調整し、その指数曲線を用いて、サンプルの分子量を算出した。
ヒトMMP−1およびMMP−9マトリックスメタロプロテイナーゼ(MMP)は、Oncogene Research Products(San Diego, CA, U.S.A.)から入手し、一方、ヒトMMP類MMP−3、MMP−8およびMMP−14(MTl−MMP)はCalbiochem(San Diego, CA, U.S.A.)から入手した。ヒトカテプシンLおよびブタ膵エラスターゼも同じくCalbiochemから入手した。
組換えタンパク質L36 scFv−NC1ES−、L36 scFv−NC1ES+およびNC1を濃度0.6μMで、以下とともに37℃にて10分間インキュベートした:
50mM酢酸ナトリウム、pH5.5、ジチオスレイトール(DTT)2mM、5mMエチレンジアミン四酢酸(EDTA)中、10nMヒトカテプシンL(ヒトカテプシンLで処置する場合);
50mM酢酸ナトリウム、pH6.0中、25nMブタエラスターゼ(ブタエラスターゼで処置する場合);および
50mM Tris−HCl、pH7.5、10mM CaCl2、150mM NaCl、0.05%のBrij−35、50μM ZnSO4中、25nMメタロプロテイナーゼ(使用するメタロプロテイナーゼで処置する場合)。
毛細血管様構造(CLS)形成アッセイでは、予備冷凍した72ウェルTerasakiプレート(Nunc, Roskilde, Denmark)にマトリゲルに基づく膜マトリックス(Becton Dickinson, Bedford, MA, U.S.A.)を2μl/ウェルで調製し、その後、それを37℃にて30分間凝固させた。3×103EC(HMEC−1またはHUVEC)を、ゲル化したマトリックスに0.5%FBSを強化した好適な培地10μlで播種した。ウェル内の培地の全容量を25μlに調整する一方で、同時に細胞を調製し、5%CO2を含む湿潤雰囲気中、37℃にて16時間インキュベートした。アッセイした組換えタンパク質(L36 scFv、L36 scFv−NC1ES−、L36 scFv−NC1ES+、NC1およびES)の管形成阻害効果は、使用した細胞株(HMEC−IまたはHUVEC)に応じて好適な培地で異なる濃度に希釈した(図5D)精製組換えタンパク質15μlを、細胞をプレーティングした時点で、マトリゲルでコーティングしたプレートに添加することにより確認した。それらの試験を3回行った。
各ウェルのデジタル画像は、Axiovert 100顕微鏡(Carl Zeiss, Germany)によりSPOTカメラ(Diagnostics Instruments, Inc.)を使用して得、328×256ピクセル、8ビットグレースケールのBMP画像として保存した。それらの画像をMatlab 5.3に基づく新ADソフトウェアを用いて処理した[Sanz, L. et al. Microvasc Res. 2002 May; 63(3):335]。
EC移動の研究では、孔径3.0μmおよび直径6.5mmのポリエチレンテレフタレートフィルターインサート(Becton Dickinson)を、無血清培地中濃度1.25μg/mlのマトリゲルにより、4℃にて一晩コーティングした。HUVEC細胞(1.5×105)を、無血清培地において、5%CO2湿潤雰囲気中、20μg/mlの精製タンパク質(L36 scFv、L36 scFv−NC1ES−、ES、NC1およびL36 scFv−NC1ES+)の存在下で37℃にて30分間プレインキュベートした後、それらを上部コンパートメントに移した。血清を含む培地600μlを下部コンパートメントに加えた。16時間後、それらの細胞をPBS中1%グルタルアルデヒドで固定し、0.1%バイオレットクリスタルで染色し、顕微鏡下で調べた。
内皮管形成アッセイにおけるエンドスタチンと抗ラミニン抗体(L36)のscFvの複合効果
オリゴマーESが上皮細胞(EC)の細胞外マトリックス(ECM)依存的な形態形成を調整することが示されている[Cuo CJ et al. J Cell Biol. 2001 152:1233]。細胞をマトリゲルにプレーティングした時点で、Fc−ES形態の二量体ESで処置すると、管状構造への構築が有意に阻害され、ECは分散したまま、プラスチック上の細胞のものと類似した形態を示した。これらの形態変化は、EC培養物をL36 scFvで処置した場合に認められるものと類似していた(図1)。
ESはコラーゲンXVIIIのNC1ドメインに由来し、このESはタンパク質分解により三量体形で放出され、続いてそれがおよそ20kDaの単量体ESに変換される(図2Aおよび2B)。異なるタンパク質を発現する異なるキメラ遺伝子を構築した。
様々な融合タンパク質のオリゴマー化状態をゲル濾過による分析的クロマトグラフィーを用いて評価し、同一サンプルでの分析的遠心分離試験を用いて評価した。IMAC(固定化金属アフィニティークロマトグラフィー)によって精製された融合タンパク質L36 scFv−NC1ES−は、ゲル濾過カラムから分子量109kDaに相当する量で本質的に単一ピーク(総面積の90%)として溶出し、このことはそのタンパク質が三量体であることを示した。沈降速度でもまた、質量が112kDaである主要な単一ピークを示し、沈降平衡法による結果、三量体種には適合させ得るが、単量体や二量体には適合させ得ない質量分布を得た(図4A)。概して、これらの結果は総て、融合タンパク質L36 scFv−NC1ES−の三量体性を示し、ゲル濾過試験および沈降速度試験で示した条件(データは示さず)と同じ条件下では、L36 scFvは単量体であることから、この性質はNC1三量化ドメインから与えられる特質であることが分かった。三量体L36 scFv(L36 scFv−NC1ES−)は、プラスチック上に固定化されているラミニンに対してより強い結合親和性を示し、マトリゲル基質でのEC分化のブロックにおいては単量体よりもはるかに効果があった(図4Bおよび図4C)。
排除クロマトグラフィーでは、精製融合タンパク質L36 scFv−NC1ES+が、三量体と一致する、概算分子量が210kDaである主要なピークと、ESの部分を含まないフラグメントに相当し得る、見かけの分子量が117kDaであるもう一つのピークとして溶出することを示した(図5A)。沈降速度でもまた、重量が181kDaおよび83kDaである二つの種を確認した。
単量体ESおよび多量体scFvの生成
いくつかのプロテイナーゼを用いてESの生成を研究するために、融合タンパク質L36 scFv−NC1ES+をインキュベートし、反応混合物を抗ES mAb(Upstate Biotechnology, Lake Placid, NY 12946, USA)を用いたウェスタンブロットにより解析した。この融合タンパク質を、アッセイする5種類のMMP(MMP−1、MMP−3、MMP−8、MMP−9およびMMP−14)で処理し、前記MMP類間でタンパク質L36 scFv−NC1ES+のプロセシングの有効性における有意差を観察したところ、MMP−3およびMMP−14で最も有効であった。融合タンパク質L36 scFv−NC1ES+の完全なプロセシングは4時間で観察された。その主要な蓄積産物は、分子量が20kDa〜25kDa間であるポリペプチドであった(図6A)。MMP−3、MMP−8、MMP−9およびMMP−14によりESフラグメントが生じ、それらのESフラグメントが4時間後に蓄積した。このことは、前記MMP類がESフラグメントを分解することができないということを示唆している。図6Aで示されるように、エラスターゼおよびカテプシンLは、融合タンパク質L36 scFv−NC1ES+からESタイプのフラグメントを生じさせるだけでなく、それらを迅速に分解もし、4時間のインキュベーション後にはESはわずかな量しか残存していない。このタンパク質分解プロセシングパターンは、未修飾の天然ネズミNC1タンパク質で観察されたパターンとほぼ同一であった(図6B)。それらの結果は、NC1ドメインにおいて、エピトープを認識するための部位を含む抗体フラグメント(scFvなど)をN末端に付加しても、プロテイナーゼによるその融合タンパク質のタンパク質分解プロセシングもES生成も妨げないことを示した。

- 請求項13〜21のいずれか一項に記載の融合タンパク質を得るための方法であって、請求項12に記載の細胞を、前記融合タンパク質の産生を可能にする条件下で増殖させること、および、必要に応じて、前記融合タンパク質を単離することおよび精製すること、を含んでなる、方法。
- 請求項13〜21のいずれか一項に記載の融合タンパク質を、少なくとも1種の薬学上許容される賦形剤とともに含んでなる、医薬組成物。
- 請求項1〜9のいずれか一項に記載の遺伝子構築物、または請求項10に記載の発現カセットを含んでなるベクターを、少なくとも1種の薬学上許容される賦形剤とともに含んでなる、医薬組成物。
- 血管新生の進行を予防し、治療し、阻害し、または最小限に抑える医薬組成物の製造における、請求項13〜21のいずれか一項に記載の融合タンパク質、または請求項1〜9のいずれか一項に記載の遺伝子構築物、または請求項10に記載の発現カセット、または請求項11に記載のベクター、の使用。
- 血管新生によって起こる病変を治療または予防するための医薬組成物の製造における、請求項13〜21のいずれか一項に記載の融合タンパク質、または請求項1〜9のいずれか一項に記載の遺伝子構築物、または請求項10に記載の発現カセット、または請求項11に記載のベクター、の使用。
- 血管新生によって起こる病変が、癌、血管腫、関節リウマチ、乾癬、バルトネラ症、移植臓器拒絶反応、出血、眼血管新生、網膜症、黄斑変性、血管新生緑内障、網膜静脈閉塞症、網膜動脈閉塞症、翼状片、ルベオーシス、または角膜血管新生を含む、請求項26に記載の使用。
