Abstract
Description
7日間熟成発酵培地を作製するためにB japonicumを振盪フラスコの栄養培地中で振盪培養器によって28℃で培養した。4種の処理剤(表2参照)を250mlの振盪フラスコに準備した。1種の処理剤を2つずつ準備したので、250mlの振盪フラスコが合計8個となった。50mlの7日間熟成発酵培地をそれぞれの振盪フラスコに加えた。すべてのフラスコの成分は、さらに、振盪培養器によって28℃で7日間平均化した。平均化させた後に、それぞれの処理液のひとつのフラスコを28℃の静止培養に移した。それぞれの処理液のもうひとつのフラスコを35℃の静止培養に移した。
フラスコを準備するための手順は、実施例1と同じであった。この実施例のための処理を表4に示す。
細菌培地を作製するためにSerratia proteomaculans(「S proteomaculans」)を標準の微生物培地(半分に薄められたトリプシンの大豆培地−「TSB」)中で22℃、24時間培養した。表5に記載された処理の1つに対応する各フラスコを1セット準備した。50mlの細菌培養液をそれぞれのフラスコに加えた。すべてのフラスコをさらに振盪培養器によって3日間、22℃で平衡化した。そして、フラスコを28℃での静止培養に移した。定期的に、サンプルを採取し、連続的に希釈し、半分に薄められたトリプシンの大豆寒天培地−「TSA」にプレーティングすることによって、細菌の数を算定した。
Claims (31)
- 部分的に乾燥した接種材料製品の製造方法であって、
実質的に静止期まで増殖させた細菌の液体接種材料を供給すること;及び、
前記液体接種材料に乾燥剤を含む乾燥処理剤を添加し、前記部分的に乾燥した接種材料製品を形成することを含む方法。 - 前記細菌が、Rhizobium属、Pseudomonas属、Serratia属、Bacillus属、Pasteuria属、Azotobacter属、Enterobacter属、Azospirillum属、Methylobacterium属及びCyanobacteria属の1以上である請求項1に記載の方法。
- 前記部分的に乾燥した接種材料製品の水分活性が、0.990未満である請求項1に記載の方法。
- 前記部分的に乾燥した接種材料製品の水分活性が、0.980未満である請求項1に記載の方法。
- 細菌を液体栄養培地に導入して培養細菌を作製すること;及び、
前記培養細菌を培養し、前記細菌を実質的に静止期まで増殖させ、液体接種材料を作製することによって、前記液体接種材料を供給する請求項1に記載の方法。 - 前記細菌の前記培養が2〜7日間行われる請求項5に記載の方法。
- 前記乾燥剤が、1以上の非還元糖及び糖アルコールである請求項1に記載の方法。
- 前記乾燥剤が、2以上のトレハロース、スクロース、グリセロール、トリエチレングリコール及びマンニトールの混合物を含む請求項1に記載の方法。
- 前記乾燥剤が、1以上のトレハロース、スクロース、グリセロール、トリエチレングリコール及びマンニトールである請求項1に記載の方法。
- 前記乾燥剤が、前記部分的に乾燥した接種材料製品の5%〜50%(重量又は体積)である請求項9に記載の方法。
- 前記乾燥剤が、トレハロースを含む請求項9に記載の方法。
- 前記トレハロースが、前記部分的に乾燥した接種材料製品の10%〜40%(重量又は体積)である請求項11に記載の方法。
- 前記トレハロースが、前記部分的に乾燥した接種材料製品の20%〜30%(重量又は体積)である請求項12に記載の方法。
- 前記乾燥剤が、トレハロースとグリセロールとの混合物を含む請求項8に記載の方法。
- 前記トレハロースが、前記部分的に乾燥した接種材料製品の5%〜40%(重量又は体積)であり、且つ、前記グリセロールが、前記部分的に乾燥した接種材料製品の1%〜10%(重量又は体積)である請求項14に記載の方法。
- 前記トレハロースが前記部分的に乾燥した接種材料製品の20%(重量又は体積)であり、且つ、前記グリセロールが前記部分的に乾燥した接種材料製品の5%(重量又は体積)である請求項15に記載の方法。
- さらに、前記部分的に乾燥した接種材料製品を包装することを含む請求項1に記載の方法。
- さらに、前記部分的に乾燥した接種材料製品を保存することを含む請求項17に記載の方法。
- さらに、前記部分的に乾燥した接種材料製品を種子に施すことを含む請求項1に記載の方法。
- 前記細菌が、Rhizobium属、Pseudomonas属、Serratia属、Bacillus属、Pasteuria属、Azotobacter属、Enterobacter属、Azospirillum属、及びCyanobacteria属の1以上である請求項19に記載の方法。
- 前記乾燥剤が、1以上の非還元糖及び糖アルコールである請求項19に記載の方法。
- 前記乾燥剤が、1以上のトレハロース、スクロース、グリセロール、トリエチレングリコール及びマンニトールである請求項19に記載の方法。
- 前記乾燥剤が、前記部分的に乾燥した接種材料製品の5%〜50%(重量又は体積)である請求項22に記載の方法。
- 前記乾燥剤が、トレハロースを含む請求項22に記載の方法。
- 前記トレハロースが、前記部分的に乾燥した接種材料製品の10%〜40%(重量又は体積)である請求項24に記載の方法。
- 前記トレハロースが、前記部分的に乾燥した接種材料製品の20%〜30%(重量又は体積)である請求項25に記載の方法。
- 前記種子が豆科作物の種子を含む請求項19に記載の方法。
- 10週間後の前記種子上の細菌数が1×105を超える請求項19に記載の方法。
- さらに、前記部分的に乾燥した接種材料製品を施した後に、種子に増量剤を施すことを含む請求項19に記載の方法。
- 乾いた担体に前記部分的に乾燥した接種材料製品を添加し、乾いた流動性のある剤形を形成する請求項1に記載の方法。
- 前記乾いた担体が泥炭である請求項30に記載の方法。
