Abstract
Description
本発明は、組換えタンパク粒様会合体(recombinant protein bodies-like assemblies、RPBLA)内に蓄積された組換えタンパク質を精製するための方法を提供する。さらに詳しくは、本発明は、所望の組換えタンパク質が濃縮され、他の細胞成分から分離され、容易に回収され得るような密度の差によって他の宿主細胞細胞小器官からの単離を可能にする組換えタンパク粒様会合体内の組換え融合タンパク質の単離を提供する。
タンパク粒(PB)は、タンパク質の蓄積に特化した細胞内小器官(すなわち、膜で囲まれた直径約1〜3ミクロンの大型小胞)である。それらの細胞内小器官は、種子のような一部の特定植物組織で自然形成され、発芽および苗成長のための主要なアミノ酸源として働く。
本発明は、形質転換した宿主から組換えタンパク質を回収するための効率的で一般的な手順または方法を提供する。本明細書において示す解決法は、密度に基づく技術によって、特に、密度クッションまたは密度勾配遠心分離技術によって、思いがけなく高収率で、他の宿主細胞細胞小器官タンパク質からの組換えタンパク粒様会合体(RPBLA)の単離を行うことができるという発見に基づく。
本発明は、一般に、形質転換した生物または細胞培養物から目的の組換えタンパク質およびペプチドを単離および精製するための下流プロセスに関する。さらに詳しくは、本発明は、宿主細胞において組換えタンパク粒様会合体(RPBLA)として発現され、蓄積する組換え融合タンパク質を精製するための方法を意図する。このRPBLAは、細胞内に高密度蓄積物を形成する貯蔵タンパク質ドメインにより誘導される組換え融合タンパク質会合体である。これらの高密度蓄積物はサイトゾル、エンドメンブレン系細胞小器官(endomenbrane system organelles)、ミトコンドリア、プラスチドに蓄積するかまたは分泌される。意図される方法によれば、融合タンパク質をRPBLAとして発現させる形質転換宿主細胞の水性ホモジネートを提供する。そのホモジネートは使用するために浄化することが好ましい。そのホモジネートにおいて異なる密度の領域を形成して、比較的高濃度のRPBLAを含む領域と比較的低濃度のRPBLAを含む領域を提供する。そのRPBLA低下領域を比較的高濃度のRPBLAの領域から分離し、その結果として前記融合タンパク質を精製する。その後、比較的高濃度のRPBLAの領域を集めるか、またはRPBLAもしくはその中の融合タンパク質の単離の前に1以上の試薬で処理するか、もしくは1以上の手順に供することができる。

動物細胞
成熟カルシトニン配列(Ct、WO2004003207)およびEGF配列に対応する合成遺伝子、ならびにhGHをコードするcDNAを、RX3 N末端γ−ゼインコード配列(特許WO2004003207)と融合し、それらをベクターpUC18に導入した。pUC18由来プラスミドpUC18RX3Ct、pUC18RX3EGFおよびpUC18RX3hGH(対応する融合タンパク質 RX3−Ct配列、RX3−EGF配列およびRX3−hGH配列を含む)のSalI−BamHI制限断片を、Xho I−Bam HIで制限したベクターpcDNA3.1−(Invitrogen)に導入した。得られた構築物をp3.1RX3CT、p3.1RX3EGFおよびp3.1RX3hGHと名づけ、それらの構築物では融合タンパク質配列はCMVプロモーターとターミネーターpA BGH下に存在した。
上記のpUC18由来プラスミドのSalI(平滑末端)−BamHI制限断片(対応する融合タンパク質 RX3−EGF配列およびRX3−hGH配列を含む)を、EcoRI(平滑末端)−Bam HIで制限したベクター pYX243(R&D Systems)に導入した。得られた構築物をそれぞれ、c117およびc118と名づけ、それらの構築物では融合タンパク質配列は誘導GALプロモーター下に存在した。
酵母
サッカロミセス・セレビシエ(Saccharomyces cerevisiae)株leu2)は、LiAc法(Ito et al. 1983, J. Bacteriol. 153:163-168)によりプラスミド構築物 c117およびc118で形質転換し、Leu−プレート上で形質転換体を選択した。発現解析は、それらの形質転換体をガラクトース含有培地で増殖させることにより行った。
タバコ(Nicotiana tabacum品種 Wisconsin)植物は、in vitro生育箱内で24〜26℃にて明期16時間で生育させた。成体植物は、温室内で18〜28℃間にて、湿度を55〜65%間に維持し、平均明期16時間で生育させた。
バイナリーベクターをA.ツメファシエンスのLBA4404株に導入した。タバコ(Nicotiana tobaccum, W38)葉のディスクを、Draper and Hamil 1988(Plant Genetic Transformation and Gene Expression. A Laboratory Manual (Eds. Draper, J., Scott, R., Armitage, P. and Walden, R.), Blackwell Scientific Publications)に記載されているとおりに形質転換した。再生した植物を200mg/Lカナマイシン含有培地で選抜し、温室に移した。導入遺伝子産物レベルが最も高いトランスジェニックタバコ植物を栽培して、T1およびT2世代を得た。
アグロインフィルトレーション方法に用いる苗は、種子からin vitro生育箱内で24〜26℃にて明期16時間で4〜6週間生育させた。
構築物 p3.1RX3.EGFおよびp3.1RX3.hGHを、リポフェクタミンに基づくトランスフェクション法(Invitrogen)により293T、Cos1またはCHO哺乳類培養細胞に導入した。高活性シアン蛍光修飾GFP(an enhanced cyan fluorescent modified GFP)の遺伝子配列を含むプラスミド pECFP−N1(Clontech)でトランスフェクトした細胞を対照として使用した。
新鮮植物材料
植物材料(湿潤または乾燥タバコ葉または苗)を液体窒素中で摩砕し、Tris−HCl 50mM pH8、200mMジチオトレイトール(DTT)およびプロテアーゼ阻害薬[10μMアプロチニン、1μMペプスタチン、100μMロイペプチン、100μMフッ化フェニルメチルスルホニル(PMSF)および100μM E64(Sigma Chemical)]を含有する抽出バッファーTでホモジナイズした。そのホモジネートを4℃にて10000×gで30分間遠心分離して、不溶性材料を除去した。総可溶性タンパク質(TSP)を、ブラッドフォードタンパク質アッセイ(BioRad)を用いて定量した。
成体トランスジェニックタバコ葉および野生型タバコ葉を濾紙中で37℃の部屋内で2週間乾燥させた。2週間後、葉を細かく切断し、室温にて5ヶ月間保存した。乾燥材料の総可溶性タンパク質を、新鮮材料について記載したように抽出し、ウエスタンブロット法により解析した。
ホモジナイゼーション
新鮮および乾燥トランスジェニックタバコ葉およびアグロインフィルトレートした(agroinfiltrated)タバコ苗(一過性形質転換)を、乳鉢と乳棒で0℃にて、10%スクロースおよびプロテアーゼ阻害薬(PMSF、ロイペプチン、アプロチニン、E−64)を添加したTris 100mM pH8、KCl 50mM、MgCl2 6mM、EDTA10mMを含有するPBP抽出バッファー中で摩砕した。そのホモジネートを、小型ローター(7.5mm径)を取り付けたポリトロン(IKA T25ベーシック、24,000rpm)を用いてさらに氷中で約10回、3〜4秒間摩砕した。未破壊組織および細胞を除去するために、固体材料を4層のミラクロス(22〜24μm)(Calbiochem)で濾過することにより除去した。
トランスフェクト細胞を培養プレートからかき取って回収し、それらをホモジナイゼーションB培地(10mM Tris−HCl pH8.0、0.9%NaCl、5mM EDTA、プロテアーゼ阻害薬)に懸濁した。その懸濁液を、およそ30回、23ゲージ針が取り付けられた5mlシリンジに入れ、それを排出した。位相差顕微鏡により細胞破壊をモニタリングした。そのホモジネートを段階スクロース勾配上に載せ、タバコ葉ホモジネートについて記載したように遠心分離した。
ホモジネートを4℃にて50×gで5分間遠心分離して、浄化済抽出物を得た。不連続スクロース勾配のために、この浄化済ホモジネート(上清)をバッファーPBP中の20%−30%−42%−56%(w/w)または42%−49%−56%−65%(w/w)スクロース2.5mlからなる段階勾配上に層にし、それを水平ローター(SW41Ti)により80,000×gで4℃にて120分間ブレーキなしで遠心分離した(Beckman社製Coulter Optima(商標)XL−100K超遠心機)。
上記のように調製したホモジネートを、42%(w/w)スクロースクッション8ml(1.18g/cm3)上で24,000gで4℃にて120分間遠心分離した。上清画分、界面画分およびペレット画分を回収した。RPBLAはクッションの下部に沈殿した。タンパク質解析では、これらの画分の等価アリコートを15%TCAで沈殿させ、サンプルを15%SDS−PAGEで分離し、銀染色により解析した。PB画分に存在する組換えタンパク質を、γ−ゼイン抗体を用いて免疫ブロット法により検出した。
段階スクロース勾配の42%〜56%(w/w)界面から単離したまたは42%(w/w)スクロースの密度クッションにより単離したRPBLAを、PBPバッファーで洗浄し、16000×gで5分間の短時間の遠心分離より回収した。RPBLA内に蓄積した組換えタンパク質を、ホウ酸ナトリウム12.5mM pH8、0.1%SDSおよび2%2−メルカプトエタノールを含有するSBバッファー1容量で可溶化した。その溶液を37℃にて一晩(約18時間)インキュベートした。一アリコートを室温にて16000×gで10分遠心分離し、その上清およびペレットをSDS−PAGEおよびウエスタンブロットにより解析して、タンパク質の完全可溶化を評価した。
組換え融合タンパク質を発現しているS.セレビシエをペレット化した。解析するために、各インキュベーション培地のアリコートを沈殿させ、−20℃にて保存した。細胞ペレットも冷凍し、解凍後、ガラスビーズと培地Y(50mM HCl−Tris pH8.0、150mM NaCl、5mM EDTA、200mM DTTおよびプロテアーゼ阻害薬)を用いる標準的な方法により細胞を破壊した。細胞および培地のいずれもを等量、SDS−PAGEおよび組換え発現タンパク質に対する特異的抗体を用いることによる免疫ブロット法により解析した。
実施例A:密度勾配によるトランスジェニック植物栄養組織からのRPBLAの単離(精製)
RX3−EGF γ−ゼイン由来融合タンパク質およびRX3−T20 γ−ゼイン由来融合タンパク質をコードする遺伝子をアグロバクテリウム・ツメファシエンスを介してタバコ植物に導入した。形質転換した植物を免疫ブロット法により解析して、組換えタンパク質の発現がより高い植物を決定した。図2Aは、RX3EGFタンパク質およびRX3T20タンパク質の両方のパターンを示している。注目すべきは、両方の組換えタンパク質が総てのトランスジェニック系に正確に蓄積しているように見えることである。主要な下方のバンドはモノマー形態の融合タンパク質に相当し、上方のバンドはそのダイマーに相当する。融合タンパク質は、通常マルチマーとして蓄積し、免疫ブロット法により検出されるモノマーおよびオリゴマーの量は、ジスルフィド結合の減少レベルに依存したものである。
分子農業において重要な点は、植物バイオマスを保存するための簡単な手段が存在するということである。これに関連して、乾燥させることにより、貯蔵量を減らし、産物を保存するための便宜な方法を提供することができる。それにもかかわらず、乾燥が目的のタンパク質の分解を促進することもよくある。工業的用途では、組換えタンパク質を含むRPBLAを単離するために乾燥植物を使用することは大変興味深いことであろう。
一過性発現系は、短期間で組換えタンパク質の蓄積挙動を検証するための便宜なツールになり得る。例えば、組換えタンパク質RX3−EGFおよびRX3−T20は、アグロインフィルトレーションにより一過性に形質転換したタバコ苗においても発現され、蓄積した。形質転換苗からのタンパク質抽出物は、免疫ブロット法による解析(図3A)で、安定的に形質転換した植物で観察される特徴的な複雑な電気泳動パターンを示し(図3A、レーン4および図2A、レーン4を比較)、融合タンパク質がこの形質転換方法を用いても正確に会合することが分かる。
組換えタンパク質を高密度組換えタンパク粒様会合体を介して精製するのに用いる手順を簡易化するために、2つのさらなる代替法を実施した:i)浄化ホモジネートを単一高密度スクロースクッションを用いて遠心分離し(図4A、B)、ii)浄化ホモジネートを低速遠心分離(すなわち、1000〜2500×g、10分間)で簡単に遠心分離した。前述の結果に一致して、1.1868g/cm3スクロースクッションを用いた遠心分離後に得られたペレットでは、RX3−EGFタンパク質およびRX3−T20タンパク質の両方が(90%を上回る)高収率で回収された(図4A、レーン4およびレーン6)。さらに、RX3−EGFタンパク質の精製は、対応するペレットで汚染タバコ内因性タンパク質がほとんど検出されなかった図4Bの銀染色ゲルより分かるように、極めて高率であった(レーン4)。
貯蔵タンパク質由来の融合タンパク質がトランスフェクト動物細胞においても高密度組換えPB様会合体の形成を誘導するかどうかを確認するために研究に着手した。ホモジナイズしたトランスフェクト哺乳類細胞の細胞小器官の細胞内分布を、段階密度勾配を用いることにより解析した。3つの異なる細胞培養タイプ、293T(ヒト由来)、Cos1(サル由来)およびCHO(ハムスター由来)を、3つの異なる融合タンパク質、RX3−Ct、RX3−EGFおよびRX3−hGHをコードするcDNAを用いることによりトランスフェクトした。pECFP−N1(Clontech)でトランスフェクトしたCos1細胞を対照として使用した。勾配画分を上記のように集め、免疫ブロット法により解析した(図5A)。
形質転換酵母における融合タンパク質を含む高密度構造の形成についても段階密度勾配により解析した。RX3−EGF融合タンパク質およびRX3−hGH融合タンパク質をコードするcDNAを、標準的な手順を用いて酵母形質転換ベクターを介してサッカロミセス・セレビシエに導入した。細胞小器官を単離するために適用される破壊方法は、方法において記載したとおり、スフェロプラストの穏やかな溶解に基づくものであった。溶解物を段階スクロース勾配上に載せ、哺乳類細胞またはタバコ葉ホモジネートについて記載したように遠心分離した。画分はSDS−PAGEおよび免疫ブロット法により解析した。
γ−ゼイン由来の融合タンパク質のそれらの密度特性を介した精製は、他の貯蔵タンパク質由来の融合タンパク質まで拡張可能である。ここで、本発明者らは、13kDのイネプロラミン(rP13)由来の融合タンパク質および22kDのα−ゼイン(22aZt)由来の融合タンパク質が段階スクロース勾配においてRPBLAに対応する高密度画分にどのように蓄積するかを示す(図7)。13kDのイネプロラミン(rP13)および22kDのα−ゼイン[全長型(22aZ)またはN末端ドメイン(22aZt)]の選択は、それらの間でもRX3ドメインに関しても相同性がないことに基づいたものであった。思いがけなく、どちらの貯蔵タンパク質からも高密度のRPBLAが生成され、そのRPBLAは段階密度勾配に供した場合にはより密度の高い界面で回収された(図7A)。
高密度の組換えPB様会合体を単離することが、トランスジェニック生物から組換えタンパク質を高収率かつ高精製レベルで回収するための有利な方法であることを証明してきた。ここでは、これらの組換えタンパク質を貯蔵細胞小器官から抽出することができることを示す。組換えPBLAは、いくつかの用途(すなわち、経口ワクチン製剤)に直接使用することができるが、他のいくつかの場合では、精製した組換えタンパク質を処理することが必要になることがある。
Claims (20)
- 宿主細胞において組換えタンパク粒様会合体(RPBLA)として発現された組換え融合タンパク質を精製するための方法であって、
(a)融合タンパク質を、所定の密度を有する組換えタンパク粒様会合体として発現させる形質転換宿主細胞の水性ホモジネートを提供する工程、
(b)そのホモジネートにおいて異なる密度の領域を形成して、比較的高濃度のRPBLAを含む領域と比較的低濃度のRPBLAを含む領域とを提供する工程、および
(c)そのRPBLA低下領域を比較的高濃度のRPBLAの領域から分離し、その結果として前記融合タンパク質を精製する工程
を含む、方法。 - RPBLAの所定の密度が、ホモジネート中に存在する内因性宿主細胞タンパク質の実質的に総てのものより高い、請求項1に記載の方法。
- 融合タンパク質が、互いに連結している2つの配列を含み、1つの配列がタンパク粒誘導配列であり、もう1つの配列が目的産物の配列である、請求項1〜2に記載の方法。
- ホモジネートにおける異なる密度の領域が、密度差を与える溶質の存在下でそのホモジネートを遠心分離することにより提供される、請求項1〜3に記載の方法。
- 異なる密度の領域が、密度勾配によって提供される、請求項1〜4に記載の方法。
- 異なる密度の領域が、前記RPBLAよりも低密度の密度クッションによって提供される、請求項1〜5に記載の方法。
- 異なる密度の領域が、ホモジネートの低速遠心分離後に得られる上清とペレットによって提供される、請求項1〜3に記載の方法。
- 宿主細胞が、高等植物細胞である、請求項1〜7に記載の方法。
- 宿主細胞が、真菌細胞である、請求項1〜7に記載の方法。
- 真菌宿主細胞が、酵母細胞である、請求項9に記載の方法。
- 宿主細胞が、藻類細胞である、請求項1〜7に記載の方法。
- 宿主細胞が、動物細胞である、請求項1〜7に記載の方法。
- 動物宿主細胞が、哺乳類細胞である、請求項12に記載の方法。
- 融合タンパク質が、タンパク粒誘導配列と目的産物の配列との間にリンカー配列をさらに含む、請求項1〜13に記載の方法。
- タンパク粒誘導配列が、プロラミンまたは修飾プロラミンを含んでなる、請求項1〜14に記載の方法。
- プロラミン配列が、γ−ゼイン、α−ゼイン、またはイネプロラミンである、請求項15に記載の方法。
- RPBLAの密度が、約1.1〜約1.35g/mlである、請求項1〜16に記載の方法。
- ホモジネートが、新鮮バイオマスから調製される、請求項1〜17に記載の方法。
- ホモジネートが、乾燥バイオマスから調製される、請求項1〜17に記載の方法。
- RPBLAを回収する工程をさらに含む、請求項1〜19に記載の方法。
