Abstract
Description
本発明の一実施形態によれば、神経伝達物質系は、神経伝達物質としてニューロキニンサブスタンスP、NKAおよびNKBを含むサブスタンスP(「SP」)系である。SPはポリペプチドであり、痛覚に対する神経伝達物質および媒介物質として作用することが知られている。SPは、同様のC末端と、SP様活性に応じて変化するN末端とを有するポリペプチドの集合であるタヒキニン群の一つである。SP受容体は、NK−1、NK−2、およびNK−3受容体を含む。SPは、NK−1受容体に選択的に結合し、NKAはNK−2受容体に選択的に結合し、NKBはNK−3受容体に選択的に結合する。
本発明の別の実施形態によれば、神経伝達物質系は、神経伝達物質としてミューおよび/またはデルタオピエート受容体に選択的に結合するエンドルフィンを含む内因性エンドルフィン系である。エンドルフィンは、オピエート受容体の結合に対するそれらの影響を通じて作用する内因性のオピエート様化合物である。ミューおよびデルタ-オピエート受容体は調和して作用し、オピエートおよびオピエート様化合物によって刺激される。ミュー受容体は、主として疼痛を修飾するが、気分も修飾する。デルタ受容体は、反対の焦点を有し、主として気分を修飾するが、疼痛も修飾する。

本発明の別の実施形態によれば、神経伝達物質系は、神経伝達物質としてセロトニンを含むセロトニン系である。セロトニンはモノアミン神経伝達物質である。セロトニンレベルが低いことは、うつ病に関連している。対抗適応は、セロトニン系のアップレギュレーションを引き起こす。
セロトニンニューロンが刺激されて興奮すると、セロトニンがシナプスへ放出される。一部のセロトニン分子はシナプスを横断して、シナプス後受容体と結合し、その後、シナプス後セロトニンニューロンの興奮を引き起こす。セロトニンがシナプス後セロトニンニューロンへ結合することにより、そのニューロンを活性化させる。そのような活性化は、一般によい気分に関連する一連の神経事象を生じる。

Claims (195)
- 患者において対抗適応を誘発することにより神経伝達物質系を調節する方法であって、前記神経伝達物質系は望ましくない精神状態または神経学的状態に関係している一種の受容体を含み、前記方法は、
前記一種の受容体に対するリガンドを患者に反復投与する工程であって、各投与は投与半減期を有し、各投与に関連する第1期間において前記リガンドをその種の受容体に結合させ、それにより対抗適応を誘発する工程を備え、
前記対抗適応は前記神経伝達物質系の調節を引き起こし、
投与間の期間に対する前記投与半減期の比率は1/2以下である方法。 - 患者において神経伝達物質系の調節を誘発する方法であって、前記神経伝達物質系は、望ましくない精神状態または神経学的状態に関係している一種の受容体を含み、前記方法は、
前記一種の受容体に対するリガンドを患者に投与することにより、対抗適応を誘発する工程と、
次いで、前記一種の受容体に対するリガンドを患者に反復投与する工程であって、各投与は投与半減期を有し、各投与に関連する第1期間において前記リガンドをその種の受容体に結合させ、それにより対抗適応を維持または向上する工程とを備え、
前記対抗適応は前記神経伝達物質系の調節を引き起こし、
投与間の期間に対する前記投与半減期の比率は1/2以下である方法。 - 前記神経伝達物質系の調節は、前記望ましくない精神状態または神経学的状態に関して治療効果をもたらす請求項1または2に記載の方法。
- 前記望ましくない精神状態または神経学的状態は、前記一種の受容体に肯定的に関係しており、前記リガンドは受容体アゴニストであり、前記調節は前記神経伝達物質系のダウンレギュレーションである請求項1乃至3のいずれか1項に記載の方法。
- 前記対抗適応は、
前記一種の受容体のうちの受容体に結合する神経伝達物質の生合成または放出の減少、
前記一種の受容体のうちの受容体に結合する神経伝達物質の再取込みの増加、
前記一種の受容体の数および前記一種の受容体のうちの受容体上における結合部位の数の少なくともいずれかの減少、並びに、
天然神経伝達物質および受容体アゴニストの少なくともいずれかによる結合に対する前記一種の受容体のうちの受容体の感受性の低下
のうちの少なくとも1つである請求項4に記載の方法。 - 前記一種の受容体に対するアンタゴニストは、各投与に関連する第1期間には投与されない請求項4乃至5のいずれか1項に記載の方法。
- 各投与はその投与に関連する第2期間を有し、第2期間はその投与に関連する第1期間に後続し、前記一種の受容体に対するアンタゴニストは、それら第2期間のうちの1つ以上において投与される請求項4乃至6のいずれか1項に記載の方法。
- 前記望ましくない精神状態または神経学的状態が前記一種の受容体に否定的に関係している場合、前記リガンドはアンタゴニストであり、前記調節は前記神経伝達物質系のアップレギュレーションである請求項1乃至3のいずれか1項に記載の方法。
- 前記対抗適応は、
前記一種の受容体のうちの受容体に結合する神経伝達物質の生合成または放出の増加、
前記一種の受容体のうちの受容体に結合する神経伝達物質の再取込みの減少、
前記一種の受容体の数および前記一種の受容体のうちの受容体上における結合部位の数の少なくともいずれかの増加、並びに、
リガンドおよび受容体アゴニストの少なくともいずれかによる結合に対する前記受容体の感受性の増大
のうちの少なくとも1つである請求項8に記載の方法。 - 前記一種の受容体に対するアゴニストは、各投与に関連する第1期間には投与されない請求項8乃至9のいずれか1項に記載の方法。
- 各投与はその投与に関連する第2期間を有し、第2期間はその投与に関連する第1期間に後続し、前記一種の受容体に対するアゴニストは、それら第2期間のうちの1つ以上において投与される請求項8乃至10のいずれか1項に記載の方法。

- 前記ノルエピネフリンシナプス後アドレナリン受容体アンタゴニストと組み合わせて、ノルエピネフリンシナプス前アルファ−2アドレナリン受容体アゴニストを投与する工程をさらに備える請求項178乃至183のいずれか1項に記載の方法。

前記一種の受容体に対するリガンドを患者に反復投与する工程であって、各投与は投与半減期を有し、各投与に関連する第1期間において前記リガンドをその種の受容体の相当数に結合させ、それにより対抗適応を誘発する工程を備え、
