Abstract
Description
本発明に従い、「抗体」又は「免疫グロブリン」は同じ意味を有し、そして本発明において等しく使用される。自然抗体中、2つの重鎖はジスルフィド結合により互いに連結し、そして各重鎖はジスルフィド結合により軽鎖と連結する。ラムダ(λ)とカッパ(κ)の2つの種類の軽鎖が存在する。抗体分子の機能活性を決定する5つの主な重鎖クラス(又はイソタイプ)が存在する:IgM、IgD、IgG、IgA、及びIgE。好ましくは、上記抗体は、IgG、例えば、IgG1、IgG2、IgG3、又はIgG4である。各鎖は異なる配列ドメインを有する。軽鎖は、可変ドメイン(VL)と定常ドメイン(CL)の2つのドメインである。重鎖は、可変ドメイン(VH)、と3つの定常ドメイン(CH1、CH2、及びCH3、これらはまとめてCHを意味する)の4つのドメインを含む。軽鎖(VL)及び重鎖(VH)の可変領域は、抗原に対する結合認識及び特異性を決定する。軽鎖(CL)及び重鎖(CH)の定常領域は、重要な生物特性、例えば、抗体鎖の会合、分泌、経胎盤移動、相補結合、及びFcレセプターに対する結合性を与える。Fvフラグメントは、1つの軽鎖及び1つの重鎖の可変部から成る免疫グロブリンのFabフラグメントのN末端部分である。抗体の特異性は、抗体結合部位と抗原決定基の構造的相補性に帰する。抗体結合部位は、主に高頻度可変性又は相補性決定領域(CDR)由来の残基から作成される。時折、非高頻度可変性由来の残基又はフレームワーク領域(FR)が、全ドメイン構造及び結合領域に影響する。CDRは、未変性免疫グロブリン結合部位の自然Fv領域の結合親和性及び特異性を一緒に規定するアミノ酸配列を意味する。免疫グロブリンの軽鎖及び重鎖は、それぞれ、L−CDR1、L−CDR2、L−CDR3、及びH−CDR1、H−CDR2、H−CDR3と命名される各3つのCDRを有する。このように、抗原結合部位は、各重鎖及び軽鎖のV領域由来のCDRセットを含んで成る、6つのCDRを含む。FRは、CDR間に入り込むアミノ酸配列を意味する。
本発明は、Her−2/neu腫瘍関連抗原を模倣するための能力により特徴づけられる、ヒト抗イディオタイプ抗体フラグメントを供する。
本発明の他の態様において、上述の抗イディオタイプ抗体フラグメントの多量体、特には二量体又は三量体が供される。
本発明は、更に、Her−2/neuを過剰発現する腫瘍の予防又は治療のために有用な医薬組成物を調製することを包含する。医薬製剤の例は、以下に供する。
腫瘍を有する患者は、本発明の抗イディオタイプ抗体フラグメントでの免疫化により治療的に処置することができ、一方腫瘍の処置を有する患者は、このような免疫化により免疫予防的に処置することができる。当該抗イディオタイプ抗体フラグメントは、免疫学的応答を増強するために適当なアジュバントとともに形成することができる。これらのアジュバントは制限することなく、ミネラルゲル、例えば、水酸化アルミニウム;界面活性剤、例えば、リゾレシチン、プロトン性ポリオール;ポリアニオン;ペプチド;油乳濁液;及び潜在的に有用なヒトアジュバント、例えば、BCG(カルメット・グラン桿菌)、及びコリネバクテリウムを含んでよい。他の改善されたアジュバント系、例えば、Me Cluskie and Weeratan,2001においてレビューされるものも使用することができる。
本発明の他の態様において、Her−2/neu特異性保護抗腫瘍免疫を誘発するための方法であって、当該方法が:
a)抗原提示細胞(APC)を上述の抗イディオタイプ抗体フラグメントと生体外において接触させる工程;
b)このように処理したAPCを、このような治療が必要な患者に投与する工程、
を含んで成る方法、を供する。
材料
Her−2/neuを過剰発現する、ヒト卵巣SK−OV−3細胞株、及びハムスター卵巣CHO細胞株は、American Type Culture Collection (Manassas, VA)から得られた。全ての生体内における実験は、実験動物研究のためのフランスのガイドラインを遵守して行った(協定番号A34220)。7週齢の雌のBALB/cマウスは Iffa Credo(L'arbresle, France)から得た。scFvフォーマットにおけるETH−2合成ヒト抗体ファージライブラリーはスイスのチューリッヒのETHから得た。当該ライブラリーは、5×108以上の異なるクローンを含む。ETH−2ライブラリーは、Pini et alの合成抗体ライブラリー(Pini et al. 1998)の改良バージョンである。抗−HER−2/neuモノクローナル抗体FRP5は、スイスのバーゼルのFMIにより供された。トラスツズマブは、Roche 社から購入した(F. Hoffmann-La Roche Ltd, Bazel, Switzerland)。コントロールとして使用したヒトIgG1は、Mabgene(Ales, France)より供された。ヒトFcフラグメントにより連結されるレセプターの2つの細胞外ドメインから構成されるHER−2/neu ECD−Fc融合タンパク質は、スイスのローザンヌ大学の生化学研究所により供された。ヒトIgG1コントロール及びトラスツズマブ F(ab’)2フラグメントは、0.2Mの酢酸ナトリウムバッファー(pH4.0)中の3%(wt/wt)比のペプシン/IgGにおける、37℃で4時間のペプシン消化により得られた(Sigma, St. Louis, MO)。当該抗体フラグメントは、セファクリル100カラム(Amersham Biosciences, Buckinghamshire, UK)でのゲルろ過クロマトグラフィーにより精製した。ドデシル硫酸ナトリウムポリアクリルアミドゲル電気泳動(SDS−PAGE)(8.5%)は、沈着したタンパク質の95%以上の純度を明らかにした。
ファージディスプレイパニングのプロトコル、及び分析は、明確に記載されている(Pini et al. 1998)。当該ファージディスプレイライブラリーは、イムノチューブ上で3ラウンドのパニングにおいて、トラスツズマブ F(ab’)2フラグメントでパニングした。チューブは、3ラウンド全てのために、PBS中、それぞれ100、50、及び10μg/mlの濃度において、トラスツズマブ F(ab’)2フラグメントでコーティングした。端的には、5×1012のファージをトラスツズマブ F(ab’)2フラグメントに添加し、30分間、持続回転下でインキュベートし、続いて室温で更に1.5時間、垂直に設置した。チューブをよく洗浄し、結合ファージを、1mlの100mMトリエチルアミンの添加により溶出させ、それから大腸菌(E.coli)TG1菌株を再感染するために使用した。一昼夜での成長後、ポリエチレングリコール沈殿によりファージを単離し、そして上記サイクルを繰り返した。ファージプールをELISA法により評価した。66ウェルマイクロタイタープレートを、PBS中のトラスツズマブ又はヒトIgG1F(ab’)2フラグメント、又は抗−fdファージIg(Sigma)のいずれかで、0.5μgのタンパク質/ウェルと成るように4℃で一昼夜コーティングした。それから当該プレートを、PBS/2%(w/v)脱脂粉乳で、37℃で2時間ブロックした。PBS中の100μlのポリクローナルファージ溶液の連続希釈を各ウェルに添加した。プレートを室温で1.5時間インキュベートした。結合ファージ−scFvは、室温で1時間30分間、西洋わさびペルオキシダーゼ(HRP)−結合抗M13mAb(Amersham Biosciences)と反応させることにより検出した。当該プレートは、業者の指示(Sigma)に従いo−フェニレンジアミン基質(OPD)を使用して展開した。
3ラウンドの選択後、ファージ結合抗体よりも可溶性抗体を産生するために、大腸菌(E.coli)HB2151細胞をファージで感染させた。単一のアンピシリン耐性感染大腸菌(E.coli) HB2151のコロニーを96ウェル組織培養プレートに採取し、そして2×TY/100μg/mlのアンピシリン/0.1%(w/v)グルコース中において37℃で3時間成長させた。scFv発現は、イソプロピルβ−D−チオガラクトピラノシド(IPTG)で、1mMの最終濃度に30℃で16時間誘発させた。細菌の上清をELISA法により、トラスツズマブ、又はヒトIgG1 F(ab’)2フラグメントとの反応性を評価した。当該ELISA法は、大腸菌(E.coli) HB2151上清のインキュベーション後に、HRP結合抗FLAG−タグM2 Ab(Sigma)を使用して結合可溶性scFvを1.5時間検出したことを除き、上述のとおり行った。
抗Id scFvを特徴づけるために、可溶性scFvのトラスツズマブF(ab’)2フラグメントに対する結合を阻害するためのHer−2/neu ECD−Fc融合タンパク質、又はHer−2/neu ECD−Fc融合タンパク質のAb1に対する結合をブロックするための精製scFvのいずれかを使用して、競合的なELISA実験を行った。各可溶性scFvは、ELISAにおいて1〜1.5のA490を与える希釈において使用した。T1と名づけられた、無関係の可溶性scFvを、ETH−2ライブラリーから単離し、そしてこれはトラスツズマブF(ab’)2フラグメントと結合せず、コントロールとして使用された(データは示していない)。最初のフォーマットにおいて、50〜0μg/mlの濃度範囲におけるHER−2/neu ECD−Fc融合タンパク質の連続希釈を阻害溶液として使用し;結合可溶性scFvは前述の通り検出した。組換えヒト癌胎児性抗原(rhCEA)は、無関係な阻害剤として使用した。阻害の当該フォーマットはまた、HER−2/neu ECD−Fc融合タンパク質によるトラスツズマブF(ab’)2フラグメント上の個々のペリプラズム画分の結合の阻害をチェックするために3ラウンドの選択後直ぐに使用した。第二フォーマットにおいて、250〜0μg/mlの範囲の溶液の多様な濃度の精製した可溶性scFv、及びELISAにおいて1〜1.5のA490を与える希釈におけるHER−2/neu ECD−Fc融合タンパク質を使用して競合を行った。結合HER−2/neu ECD−Fc融合タンパク質は、続いて、HRP−結合抗HER−2/neu FRP5 mAbで検出した。
2つの免疫化スケジュールを行った。最初に、グループあたり5匹の6〜8週齢の雌のBALB/cマウスを、3ヶ月間、月々、完全フロイントアジュバント(CFA)で乳化後、100μlのPBS又は溶出バッファー(ネガティブコントロールグループ)中の50μgの精製した可溶性抗Id scFv40又は69、又は20μgのHER−2/neu ECD−Fc融合タンパク質のいずれかにより腹腔内において免疫化した。続いて不完全フロイントアジュバント(IFA)で腹腔内において免疫化を行った(プロトコル1)。各免疫化前に血清を尾静脈から採血し、そしてアッセイのために−20℃で保管した。Magliani et al., 1998の研究により着想された二番目のものにおいて、6〜8週齢の雌のBALB/cマウスを4回免疫化した。先ず、CFAでの乳化後、1014TU/mlの力価における50μlの新鮮に調製したファージscFv40又は69、20μgのHER−2/neu ECD−Fc融合タンパク質、又はPBSのいずれかの皮下注射を行った。当該注射の2週間後、同じ用量のIFAで二回目の皮下投与を行った。2回の更なる注射は、腹腔内における最初の免疫化(プロトコル2)の後21日目及び35日目に腹腔内にて与えられた。血清抗体測定のために、マウスは、開始免疫化後21日及び42日目に採血され、そして血清を−20℃で保管した。当該血清は、上述のとおり、HER−2/neu ECD−Fc融合抗原においてフローサイトメトリー及びELISAにより抗抗Id(Ab3)応答のために確認した。得られた結果の分析はまた、Magliani et al. , 1998により記載された方法に従い行った。HER−2/neu ECD−Fc融合タンパク質で免疫化されたマウス由来のプールされた血清から成るポジティブコントロールを各測定に含んだ。結果は、1000倍にしたポジティブコントロール血清の70%の吸光度に対する検査血清の吸光度の割合として任意の単位において表した。ポジティブコントロール血清吸光度から減算された30%が抗Fc応答によるものであった(結果のセクションを参照のこと)。
96ウェルプレートを2μg/mlのHER−2/neu ECD−Fc融合タンパク質で、4℃で一昼夜コーティングすること、その後PBS/1%(w/v)BSAでブロックすることにより、間接ELISA法を行った。100μlのマウス由来の希釈血清[PBS/0.5%(w/v)BSA中、1:100]を各ウェルに添加し、そして室温で1.5時間インキュベートした。それから、100μlのヤギ抗マウスIgG(全分子)−HRP接合体、ヤギ抗マウスIgG(γ鎖特異性)−HRP接合体、又はヤギ抗マウスIgM(μ鎖特異性)−HRP接合体のいずれかを添加し、1.5時間インキュベートし、そして上述のとおり検出した。
能動免疫療法のために標的抗原としてHer−2/neu腫瘍性タンパク質を使用する場合の主な問題は、自己抗原に対する免疫耐性である。Ab1療法、並びにペプチド又はタンパク質型ワクチンが報告されている(Disis et al. 2002-Pegram et al. 1998)。全ての実験は、有効な治療方法の研究を進めるための必要性を指摘してきた。
免疫化トラスツズマブ F(ab’)2フラグメント(Ab1)における合成ETH−2 scFvライブラリーにおける3ラウンドのパニングにより行われた、抗Id scFvの選択は、組換えポリクローナルファージ上清を使用してELISAにより確認される場合、トラスツズマブ又はヒトIgG1 F(ab’)2フラグメント(コントロールとして使用した)におけるライブラリーの実質的な親和性の向上をもたらした。また、1〜3ラウンドの選択において、2×104倍に増加したファージ力価が観察された。クローニング後、3ラウンドの個々のクローンが誘発された96ウェルプレート由来の抗体上清は、それから可溶性scFv抗体を産生するために、大腸菌(E.coli)HB2151を感染させるように使用した。ELISAにより、これらのヒトIgG又はトラスツズマブ F(ab’)2フラグメントとの反応性のために、96のペリプラズム画分を試験した。ポジティブなペリプラズム画分のウェスタンブロット分析は、我々を、約30kDaの適当な分子量を伴う単一の単量体から成るscFv抗体の選択、及び低分子量を示すこれらのものを除外することに導いた(図1)。選択されたペリプラズム画分は、HER−2/neu ECD−Fc融合タンパク質又はrhCEA(無関係な阻害剤として使用される組換えヒトCEA)により、トラスツズマブ F(ab’)2フラグメントにおけるこれらの結合の阻害のために、ELISAにより確認した。顕著な阻害(>50%)を示すクローンのみを更に分析した。上記抗体のVH及びVL領域のDNA配列決定後、本発明者は、Ab1の結合部位を認識するが、VH及びVLドメインのCDR3中に5つ又は6つのアミノ酸の異なるランダムループを示すscFv39、40、及び69と称される3つのユニークなscFvフラグメントを選択した(95位、表1を参照のこと)。
抗Id scFvの結合特性を特徴づけるために、本発明者は、先ず、これらのscFvが、HER−2/neuエピトープのために、トラスツズマブ F(ab’)2フラグメント(Ab1)と競合的な結合性を示すか否かを確認した。HER−2/neu ECD−Fc融合タンパク質が阻害因子として使用される場合、トラスツズマブ F(ab’)2フラグメントにおける抗Id scFv40及び69の結合はともに、約90%阻害される(図3A)。抗Id scFv39の結合は23%のみが阻害され;従って抗Id scFv39は続く実験には使用しなかった。rhCEA(185kDa)を使用して同様の競合実験を行った(図3B)。阻害曲線は、高濃度のrhCEAでさえ、当該抗原(この分子量はHER−2/neu ECD−Fc融合タンパク質とほぼ同じである)がトラスツズマブ F(ab’)2フラグメントにおける結合のための抗Id scFvと競合できないことを示した。抗Id scFv40及び69は、トラスツズマブ F(ab’)2フラグメントにおける2つの異なる濃度においてHER−2/neu ECD−Fc融合タンパク質の結合を阻害するために引き続き使用した。当該フォーマットにおいて、観察された最大阻害は、250μg/mlのId scFv69では40%近くであった(図4)。Id scFv40でも同様の結果が得られた。
6〜8週齢の雌のBALB/cマウスを、腹腔内において可溶性の精製された抗Id scFv40及び69(プロトコル1)、又は皮下においてファージ上で示されたscFv(プロトコル2)のいずれかで免疫化した。各グループの5匹のマウスの血清を、3〜4回の注射後、Ab2免疫原(抗Id scFv40又は69)に対するこれらの反応性のためにELISAにより個々を試験した。全てのマウスは、抗Idに対して有意な免疫を生じ、そして当該力価は、両プロトコルにおける更なる免疫化により更に増加した。免疫前の血清又は希釈バッファー(PBS又は溶出バッファー)で免疫化したマウスのコントロール血清は、抗Id scFv40及び抗Id scFv69に対して陰性であることが検査された。野生型ファージで注射されたマウスの血清において、非特異的な応答が観察された。抗Id scFvに対するAb3の特異性を示すために、免疫化マウス由来の血清の連続希釈により阻害ELISAはトラスツズマブ F(ab’)2フラグメントにおける各抗Id scFvの結合を阻害することにより行った。ファージ粒子上に示された抗Id scFv69で免疫化したマウス由来の免疫血清の1:20の希釈は、Ab1[トラスツズマブ F(ab’)2フラグメント]に対して結合するAb2(抗Id scFv69)を100%近く阻害し、当該血清を含む抗体は、事実上、真の抗抗Idであったことを示唆した(図6)。抗Id scFv40も同様の結果を示した。反対に、HER−2/neu ECD−Fc融合タンパク質又はPBSで免疫化したマウス由来の免疫血清においては阻害が観察されなかった。
標的抗原と結合することができる、Ab1’と称される抗体のサブセットの検出のために、1:100に希釈したマウス血清を使用して、HER−2/neu ECD−Fc融合タンパク質においてELISAを行った。異なるHRP−結合Igの使用は、我々に、抗Id scFv及びコントロール免疫原での免疫化によりマウス血清中に誘発されるIgG、IgM、及び全Igレベルを測定することを許容した(図7)。各時間においてプロットした吸光度の値は、各グループの5匹のマウスの平均値±標準偏差(SD)に相当する。これらの結果は、(i)IgG又はIgM 抗HER−2/neu抗体の誘発パターンが、2つの免疫化プロトコルと類似したこと(図7A及びB);(ii)ポリクローナルAb3の有意な部分が、抗原と反応し(図7A及びB)、3〜4回の免疫原の注射後、Ab1’のレベルがプラトーに達したこと;(iii )図7Cに示されるとおり、IgM並びにIgG HER−2/neu ECD−Fc融合タンパク質応答の力価が、予想どおり、HER−2/neu ECD−Fc融合タンパク質で免疫化したマウスの血清において極めて高く、一方、ネガティブコントロールにおいては反応性が存在しなかったことを示した。LICRにより供される(Lausanne Branch)、他の組換えFc融合タンパク質、いわゆるMICA−Fcに対して陽性のHER−2/neu ECD−Fc融合タンパク質血清の反応性がELISAにより確認される場合に示されるとおり、HER−2/neu ECD−Fc融合タンパク質で免疫化したマウスの血清において、吸収されたタンパク質に対する極めて高い力価の部分は、抗ヒトFc応答によるものであった。得られたデータは、抗Fc応答が、HER−2/neu ECD−Fc融合タンパク質におけるELISAにより観察された全応答の約30%であるという結論に本発明者を導いた。
抗Id scFv40及び69での注射により誘発されたAb1’の性質を更に確認するために、HER−2/neu陽性SK−OV−3細胞、及びHER−2/neu陰性CHO細胞のフローサイトメトリー分析を、免疫化されたBLAB/cマウス由来の血清(1:100)で行った。トラスツズマブmAbを使用して、SK−OV−3細胞の表面におけるHER−2/neuの過剰発現、及びCHO細胞表面におけるレセプターの発現の欠如を最初に確認した(図8A及びB)。それから本発明者は、HER−2/neu ECD−Fc融合タンパク質で免疫化したマウス由来の血清中だけでなく、抗Id scFv40又は抗Id scFv69のいずれかで免疫化したマウス由来の血清中の抗体もまた、SK−OV−3細胞を特異的に染色したことを示した。CHO細胞株においては、弱い反応性のみが観察された。2つの免疫化スケジュールの最後に得られた代表的な結果は、図8A及びBにも供される。CHO細胞と比較した場合、SK−OV−3細胞上の蛍光強度の増加は、HER−2/neu ECD−Fc融合タンパク質、抗Id scFv40又は69、及びPBSで免疫化したマウス由来の血清で、それぞれ約100、25、及び2であった。希釈バッファーで注射したマウス由来の血清を使用することにより得られた細胞の染色は、免疫前の血清のものと同程度であり、従って、バックグランドシグナルと一致した。これらの実験に基づいて、本発明者は、HER−2/neuレセプターに特異的に誘導されるAbl’抗体が、抗Id scFv40又は69で免疫化したマウスの血清中に存在することを結論づけた。
