Abstract
Description
本国際出願は、それぞれその全体が参照により本明細書に援用される、2004年4月30日に出願された米国仮特許出願第60/567,069号、2005年2月14日に出願された60/653,247、そして2005年3月30日に出願された60/667,352に対する優先権を主張する。
本発明は一般に、ヒトC3タンパク質の部分のコブラ毒因子(CVF)タンパク質の対応部分による置換を有するヒト補体C3のキメラ誘導体に関する。好ましくは、C3のアルファ鎖の部分がCVFの対応する部分によって置換される。
補体の第3の成分であるC3は、補体活性化の古典的経路および代替経路の両方において中枢の役割を果たし、生理的C3活性化生成物の多くは、免疫反応および宿主防御において重要な機能を有する。代替経路において、C3の活性化型、C3bは、C3コンバターゼの構造サブユニットである。この2分子酵素は、C3bおよび因子Bの活性化型であるBbより成る。この酵素は、因子Dによって続いて開裂されるC3bの因子Bへの結合によって形成され、C3コンバターゼ、C3b、Bbの形成、および活性化ペプチドBaの放出を生じる。C3コンバターゼは分子をC3bおよびアナフィラトキシン、C3aに開裂することによってC3を活性化する。C3b分子はC3コンバターゼのごく近くの細胞または粒子に結合するであろう。最終的に結合したC3bは、C5のC5bおよびアナフィラトキシン、C5aへの活性化を可能にする。C5活性化は同じC3b、Bb酵素によって起こり、Bb酵素は追加のC3b分子に結合しているときにC5を開裂可能であり、(C3b)2,Bbより構成される3分子複合体を生成する。このC5開裂3分子酵素はC5コンバターゼと呼ばれる。C3およびC5の両方の活性化がBbサブユニットの同一の活性部位で発生するため、酵素はC3/C5コンバターゼとも呼ばれる;そして1個のECメンバのみが割り当てられている(EC 3.4.21.47)。

ヒト補体C3配列の、CVFの特異的機能の主な構造要件を表すCVF配列による置換は、CVF様機能を備えたC3誘導体の作製を可能にする。本発明の好ましい実施形態は、補体活性化が病因の一部である臨床状況で補体を枯渇させる新規な治療剤として使用するための安定なコンバターゼを形成することによって、補体を枯渇させるCVF特異的機能を示すヒトC3誘導体を提供することである。CVF特異的配列によって引き起こされたヒトC3の構造変化は最小限であるため、修飾C3分子は、免疫原性の著しい低下または非存在さえ示すであろう。
補体系は、脊椎動物免疫系の構成要素であり、宿主防御および免疫反応でのその役割を通じて健康の維持に関与している。しかしながら補体活性化も多数の疾患の病因に関与している。これらの疾患の例は、自己免疫溶血性貧血、関節リウマチおよび他の免疫複合疾患、ならびに血流が一時的に停止された後に組織損傷が発生する再潅流傷害を含む。再潅流傷害の例は、閉塞血管の再開後の組織損傷(たとえば心臓麻痺、卒中)および臓器移植後のレシピエントの血液による再潅流である。大流行している疾患を含め、多数の疾患への補体系の関与に基づいて、過去十年間には、これらの疾患状態における望ましくない補体活性化プロセスを妨害する複数の抗補体剤の開発が見られた。このような薬物の開発の試みはすべて、補体の活性化を阻害することに基づいている。
補体の第3の成分であるC3は、補体活性化の古典的経路および代替経路の両方において中枢の役割を果たし、生理的C3活性化生成物の多くは、免疫反応および宿主防御において重要な機能を有する(総説については、参考としてその全体が本明細書に援用されるMuller−Eberhard,H.J.(1988) ”Molecular Organization and Function of the Complement System,”Ann.Rev.Biochem.,57:321−347を参照)。ヒトC3は、分子量約185,000の2鎖糖タンパク質である。それは単鎖プリプロC3として合成され、β鎖とα鎖との間のアルギニン残基4個の除去による次の処理を受ける。ヒトC3の1次配列は、分子クローニングから既知である。他の哺乳類種はもちろんのこと非哺乳類種からの完全または部分配列情報が入手できる。これらとしては、マウス、ラット、モルモット、ニワトリ、コブラ、アフリカツメガエル、およびヤツメウナギが挙げられる。ヒトC3遺伝子は、長さ42kbで、エキソン41個を含み、52〜213bpの範囲の大きさである。
1.両方の酵素が、酵素活性を破壊する、各サブユニットへの自発的な崩壊解離を示す。C3b,Bb酵素は非常に短寿命であり、37℃にて1.5分間の固有半減期で崩壊するのに対して、CVF,Bb酵素は何桁もより安定であり、約7時間の固有半減期で崩壊する。
CVFは、次に枯渇につながる徹底的な活性化の機構を通じて作用する、補体インヒビタである。実際に、CVFは、他の薬物の抗補体活性を評価する標準として頻繁に使用される。CVFはこの強力な抗補体活性を示すが、その免疫原性のためにヒト用途には適していない。この理由で、CVFとC3との広範囲に渡る構造類似性を利用することによって実質的に非免疫原性CVFを調製することと、CVF分子の機能的に重要な領域をヒトC3内へ組換え手段で置換することによってCVFの所望の補体枯渇機能を備えたヒトC3誘導体を産生することが望ましい。このことは、本明細書で述べるように実施されている。

局所処置は、所望の効果に応じて、補体の枯渇または補体の活性化の結果を生じさせるために多数の方法で実施できる。1つの実施形態において、局所枯渇は、修飾C3タンパク質を臓器、組織、空洞、または皮内に局所的に投与したときに作用する。これはその範囲の補体の一時的および完全な枯渇を引き起こす。局所枯渇または活性化も、選択した部位への修飾C3タンパク質の間欠流または定常流を生じさせるインスリン型ポンプを使用して作用しうる。あるいは補体の局所活性化は、修飾C3に化学的に付着したときに、特異的組織、疾患、または罹患細胞に局在化して、その部位の補体の連続活性化を引き起こすことができる、特異的モノクローナル抗体を利用する。他の実施形態において、抗体は組換えDNA技術によって修飾C3タンパク質に付着できる。
本明細書で修飾C3タンパク質のための最善の用途を産生および解析する場合、これに限定されるわけではないが以下を含む、多数の機能および活性をそのような解析に使用できる:
因子Bに対する親和性−修飾C3タンパク質が因子Bの活性化(BaおよびBbへの開裂)を補助する効率の相違は、検出可能であり、利用できる。因子B活性化の補助を開始する、因子Bに対する単なる親和性を超えた複数の因子があるが、いったんコンバターゼが形成されると、一般に最も重要である次なる主な特性は、安定性、HおよびIによる調節、そしてC3およびC5の開裂の活性である。
ヒト補体C3/CVFハイブリッドタンパク質の生成
ヒトC3配列の、CVF特異的機能に関する重要な構造上の特徴を示すCVF配列による置換は、CVF様機能を備えたC3誘導体の生成を可能にする。それゆえ本発明の実施形態は、補体活性化が病因の一部である臨床状況で補体を枯渇させる新規な治療剤として使用するための安定なコンバターゼを形成することによって、補体を枯渇させるCVF特異的機能を示すヒトC3誘導体の産生に関する。CVF特異的配列によって引き起こされたヒトC3の構造変化は最小限であるため、修飾C3分子が免疫原性の著しい低下または非存在さえ示せることが明らかである。
修飾ヒトC3タンパク質の発現
タンパク質は、Drosophila S2細胞系において、タンパク質分泌のためのDrosophila Bipシグナル配列を使用して生成した。簡単には、プラスミドpMB/HC3−1550、pMB/HC3−1504、pMB/HC3−1496、pMB/HC3−1550/1617、およびpMB/HC3−1348をDrosophila S2細胞内に、ChenおよびOkayama(参考としてその全体が本明細書に援用される、Chen,C.,and Okayama,H.(1987) Mol.Cell.Biol.7(8),2745−2752)のリン酸カルシウム法を使用して形質移入した。S2細胞は、発現プラスミドとpCoBlastの混合物を19:1(w:w)の比で使用して形質移入した。形質移入後、ブラストサイジン(25μg/mL)を使用して、両方のプラスミドを含有する細胞を選択した。発現では、形質移入細胞の培養物1リットルを無血清培地(Hi−FiveおよびL−グルタミン)で、ブラストサイジンの非存在下で培養した。細胞が5x106細胞/mLの密度に達したとき、最終濃度25μMまでのCuSO4の添加によって、組換えタンパク質の生成を誘発した。培養物に4〜5日に渡って組換えタンパク質を発現させた。次にANX、Sephacryl H−300、およびCM−FFクロマトグラフィーの併用により、培地からハイブリッドタンパク質を精製した。
修飾ヒト補体C3タンパク質の活性測定の結果
精製した修飾ヒトC3タンパク質ハイブリッドに、以下のように多数の機能分析を受けさせた。
本アッセイは、タンパク質がヒト(または他の)血清中の補体を枯渇する能力を測定する。アッセイは2ステップで実施した。第1のステップでは、興味のあるタンパク質を通常は連続希釈によって、緩衝液中で所望の濃度まで希釈した(通例、ナノグラム/マイクロリットル未満から約320ng/マイクロリットルまたはアッセイで使用する10マイクロリットル中3.2μg)。次に希釈したタンパク質の分割量10μLを未希釈血清と混合する。混合物を37℃で3時間インキュベートして、C3コンバターゼを形成することによってタンパク質に補体を活性化させた。形成されたコンバターゼは次に血清のC3を活性化できた。次に、残っている補体活性の量を測定するために、血清を希釈して、血清中に存在するときに補体によって容易に溶解される抗体感作ヒツジ赤血球と混合した。この反応を30分間進行させて、混合物を冷緩衝液中で希釈することによって停止させた。細胞を遠心分離にかけ、放出されたヘモグロビンを測定することによって、溶解細胞を定量した。結果を図4および図9に示す。
これは、ハイブリッドタンパク質が因子Bを活性化して、C3/C5コンバターゼを形成する能力を測定するアッセイであった。コンバターゼ形成は、因子BのBbおよびBaへの開裂の機能として測定した。アッセイでは、精製したハイブリッドタンパク質は、3倍過剰の因子Bおよび因子D(すべて高度に精製)によって、37℃にてマグネシウムの存在下でインキュベートした。各種の時間にて、反応物の分割量を取り出し、マグネシウムをキレートするEDTAを添加することによって反応を停止させた。反応生成物を非還元性SDSポリアクリルアミドゲルに流して、タンパク質はクマシーブルーで染色された。変換された因子Bの量は、ゲルを専用コンピュータプログラム内にスキャンして、因子BおよびBbバンド内のタンパク質の量を測定することによって定量した。
C3コンバターゼ活性アッセイ
本アッセイは、C3aペプチドを開裂させることによって、ハイブリッドタンパク質を含有するC3/C5コンバターゼがヒトC3を活性化する活性を測定する。このアッセイを実施するために、コンバターゼを上述のように形成して、反応をEDTAの添加によって停止させた。次にコンバターゼをヒトC3と混合して、反応物を37℃にてインキュベートした。指示した時間に分割量を取り出して、SDSおよびβ−メルカプトエタノールを含有するゲル装填緩衝液と混合して反応を停止させた。SDSはタンパク質を変性させて、β−メルカプトエタノールは、タンパク質中のシステイン間のジスルフィド結合を還元する。還元条件下での電気泳動の後、ゲルをクマシーブルー染料で染色して、C3α鎖およびC3α’鎖の相対量を上述のように定量した。各反応で同量のコンバターゼを使用するように注意した。
C5変換アッセイ
C5変換活性のアッセイは本質的に、参考としてその全体が本明細書に援用されるPetrella et al.,(1987)J.Immunol.Methods 104(1−2),159−172によって述べられているように実施した。本アッセイでは、C5コンバターゼは合計3μgのタンパク質を使用して上述のように形成した。コンバターゼ形成の後、最終濃度5mMまでのEDTAの添加によって反応を停止させた。次にこの反応物5μLを、PBS中にC57μgを含有する反応物25μLに添加した。反応物を37℃で24時間インキュベートし、Laemmliゲル装填緩衝液7μLの添加と、それに続く5分間の煮沸によって反応を停止させた。反応生成物は、SDS−PAGEを還元することによって分離し、ゲルをクマシーブルー染料で染色して、C5α鎖およびC5α’鎖の相対量を上述のように定量した。現在のタンパク質のいずれも、活性C5コンバターゼを形成できなかった。
補体を局所的または全身的に枯渇させる方法
本明細書で開示した方法によって生成した修飾ヒト補体C3タンパク質を使用して、以下のように補体を局所的または全身的に枯渇させる:
局所枯渇は、修飾C3タンパク質が臓器、組織、空洞に、または経皮的に局所投与されるときに作用する。これはその範囲の補体の一時的および完全な枯渇を引き起こす。あるいは局所枯渇は、修飾C3に化学的に付着したときにそれを特定の組織、疾患部位、または感染細胞に局在化させて、その範囲の補体の連続的枯渇を引き起こす、特異的モノクローナル抗体を使用できる。
再潅流傷害の処置方法
再潅流傷害の例は、閉塞血管の再開後(たとえば心臓麻痺または虚血性卒中の後)の組織損傷、および移植臓器のレシピエントの血液による再潅流である。詰まった冠状動脈または臓器移植後の再潅流では、移植体に再潅流させる前に、または閉塞血管の開通前に多くの場合で補体を枯渇させることが望ましい。補体活性化を活性化する能力は、それが引き起こす組織損傷を回避できるため、組織損傷の唯一残存している主な源は、酸素欠乏である。通例、再潅流の間の補体活性化による組織損傷は、酸素欠乏による組織損傷の2倍大きい−すなわち組織損傷のほぼ2/3が補体活性化に起因するのに対して、1/3は酸素欠乏に起因する。それゆえ再潅流傷害は、本発明の実施形態で可能であるように、再潅流の前に補体を所望させることによって大幅に減少させることができる。この場合、最高活性コンバターゼを使用することと、高い用量を使用することが好ましい。コンバターゼの安定性は、慢性疾患に対するほど重要ではない。
遺伝子治療の有効性および/または効率を上昇させる方法
本方法は、体内での有用なウィルスの生存の延長を補助するために、補体の枯渇に依存している。補体は遺伝子治療で使用したあるウィルスベクターの本体からの除去を補助することが見出されているため、遺伝子治療ベクターの投与前に循環する補体の量を減少させることが望ましい。このことは、使用されている遺伝子治療の種類に応じて局所的にまたは全身的に実施できる。
治療剤(たとえば化学治療剤)または診断剤の送達を向上させる方法
治療薬が投与される範囲への血流を増加させるために、修飾C3タンパク質は、選択した組織への親和性を持つモノクローナル抗体に化学的に結合される。本実施例では、修飾C3は、高い活性のC3/C5コンバターゼを形成するC3である。修飾C3/抗体は、組織へ標的化されて、領域に局所補体活性化を生じさせて、補体活性化のために血管透過性を引き起こす。新しい非活性化補体が標的に到着する血液によって連続的に供給されるため、血管透過性は、活性コンバターゼ/抗体複合体が標的に結合されている限り継続する。
関節リウマチ、狼瘡、および他の自己免疫または免疫複合疾患を処置する方法
本方法は、関節リウマチの例を局所範囲における補体活性化から疼痛および炎症が発生する複数の状態の1つとして使用する。処置のために、修飾C3タンパク質が全身的または局所的に投与されて、補体を枯渇させることによって、補体反応/活性化を低下させる。これは疾患の症状および疾患の進行を低減させることができる。特に疾患症状の悪化の発現があるときには、補体系の活性の長期低下と組合せて間欠的枯渇を有することも有益でありうる。さらに本方法は、循環免疫複合体を伴う他の疾患、たとえば狼瘡および他の自己免疫疾患によって使用できる。
修飾C3タンパク質を選択する方法
各種の修飾C3タンパク質は、各種の疾患、および処置方法に有用である。それゆえ本発明の実施形態の修飾C3タンパク質の機能品質を解析して、それに従ってそれらを使用することが有用である。以下の方法を、実施例2で生成された精製修飾C3タンパク質の機能を分析するために使用する。本明細書で述べる方法は、当業者に既知の他の方法と同様に使用できる。
血漿半減期を測定する方法
これに限定されるわけではないが、免疫系によって生成される特異的抗体、血清内で循環するプロテアーゼ、非特異的免疫反応、および因子HおよびIなど特異的調節因子を含む、修飾C3タンパク質の血漿半減期に影響を及ぼす多くの因子がある。修飾C3タンパク質を活性化させて、次に補体を枯渇できるようにするために、好ましいC3タンパク質は、ヒト血漿内で少なくとも最小限の量の時間に渡って持続するであろう。それゆえ修飾C3タンパク質の血漿半減期を確認して、それらがある疾患の処置にどのように有用であるかを判定することは興味深い。
表面プラスモン共鳴を使用して測定されるような修飾ヒト補体C3タンパク質のC3コンバターゼ形成
表2は、補体タンパク質のC3、CVF、および組換えヒトC3/CVFタンパク質への相対結合を示す。すべての場合で、数字が大きいほどタンパク質相互作用が強いことを示す。タンパク質(C3bなど)をBIACORECHIPTMに結合させて、次に補体因子と接触させる。チップに結合された(それゆえ修飾C3タンパク質に結合された)補体因子それぞれの量は、表面プラスモン共鳴によって決定した。結果は:1)C5を開裂できるコンバターゼを形成できないことと一致して、C5に結合した組換えタンパク質のどちらも示さず、2)両方のタンパク質は因子Hに結合したが、CVFは結合せず、因子Hは、C3b,Bbコンバターゼ複合体を解離させて、開裂によってC3bを不活性化するために第2の補体タンパク質、因子Iを向けることができる調節タンパク質の1つであることと、3)因子Dおよびマグネシウムの存在下での因子Bへのタンパク質の親和性は、タンパク質がC3コンバターゼを形成する速度(因子B開裂を仲介するその能力によって測定された−図10を参照)にほぼ比例したことを示した。HC3−1550およびHC3−1348の両方を開裂させたが、C3bよりも低速であった。CVFはHまたはI部位を持たないため、CVFはHまたはIによって全く開裂されない。組換えCVFの2鎖分子は2または3個のI部位を有するが、因子Iによってなお開裂されないことは、興味深い。
修飾ヒト補体C3タンパク質の因子B開裂
図10の因子B開裂のグラフは、修飾ヒト補体C3bタンパク質がマグネシウムイオンおよび因子Dの存在下で因子Bの開裂を仲介する能力の時間経過を示す。これはタンパク質がC3/C5コンバターゼを形成する能力を測定するのに使用できる。本アッセイでは、それが因子Dおよびマグネシウムの存在下で因子Bを非常に効率的に結合する(実施例11のデータを参照)ことと、得られた複合体が非常に安定であることの両方のために、C3bがコンバターゼを非常に効率的に形成することは注目に値した。CVF、HC3−1496およびHC3−1504はすべてコンバターゼをほぼ同じ速度で形成したが、HC3−1550およびHC3−1550/1617は、そのコンバターゼを形成する能力がC3bとCVFとの中間である。HC3−1348は非常に低速で因子Bを結合する。これはいったん形成されたコンバターゼより長い半減期と、因子Bへのハイブリッドタンパク質のより低い親和性との組合せによって最も確からしく説明される。
修飾ヒト補体C3タンパク質のC3開裂
図11は、コンバターゼがC3を開裂する能力を示し、開裂反応の時間経過を示す。はめ込み図は、主グラフで使用したコンバターゼの20%で実施した時間経過である。結果は、CVFおよびHC3−1550がC3開裂にてほぼ等しく効率的であるコンバターゼを形成したが、HC3−1348およびHC3−1496はどちらもC3の開裂でCVFよりも約5倍効率的であるコンバターゼを形成したことを示している。HC3−1550/1617含有コンバターゼは非常に不安定であるように見えたが、約10分後にはC3開裂を補助しなかったことは興味深い。
修飾ヒト補体C3タンパク質の補体枯渇
図9の補体枯渇チャートのデータは、HC3−1550/1617を除くすべてのタンパク質が、非常に異なる効率であるが補体を枯渇できたことを示している。HC3−1550およびHC3−1504はどちらも補体の枯渇ではきわめて非効率的であったが、HC3−1496およびHC3−1348は、天然または組換えCVFと全く同様というわけではないが、補体をきわめて効率的に枯渇することができた。以前にHC3−1348を、前出願ではHC3−1325と呼んだが、置換マッピングは次に、置換が1325ではなくて1348であることを示している。
修飾C3タンパク質の変異形を生成する方法
好都合な品質を有する修飾C3タンパク質の変異形が生成される。好都合なとは、変異形がこれに限定されるわけではないが:C3コンバターゼ活性、血清補体除去、因子B結合、C3の開裂、Bbの結合およびC3の結合を含む、タンパク質の1つまたは複数の活性を向上させることを意味する。変異形は各領域に1つまたは複数の突然変異体を含むことが可能であり、1つまたは複数のアミノ酸を含むことができる。一部の特異的変異形を以下に述べる:
C末端がC3コンバターゼの安定化に関与しているため、突然変異は任意の修飾C3タンパク質のC末端(aa1617の後)に生成される。突然変異は挿入、欠失、および置換でありうる。しかしながら突然変異は好ましくは置換である。突然変異はC3コンバターゼを安定化する。C末端における突然変異の例は、これに限定されるわけではないが:位置1633、1654、および1658における突然変異を含む。非保存的変異は保存的変異と同様に、形態に影響するアミノ酸変化に加えて、実施形態に含まれる。しかしながら好ましくは、突然変異は、非保存的アミノ酸変化を生じる。
Claims (23)
- ヒトC3タンパク質の部分の、実質的にそれに関連する配列のコブラ毒因子タンパク質の対応する部分による置換を含む、修飾ヒト補体C3タンパク質。
- CVFの置換部分がC3のアルファ鎖内である、請求項1に記載の修飾C3タンパク質。
- CVFの置換部分がC3のアルファ鎖のC末端部分である、請求項2に記載の修飾C3タンパク質。
- 置換C末端部分がヒトC3タンパク質のアミノ酸1663を含む、請求項3に記載の修飾C3タンパク質。
- 置換C末端部分がヒトC3タンパク質のC末端全体を通じて延伸しない内部部分である、請求項3に記載の修飾C3タンパク質。
- 修飾タンパク質が未修飾ヒトC3タンパク質と実質的に同数のアミノ酸残基を有する、請求項1に記載の修飾C3タンパク質。
- 置換がヒトC3タンパク質のアミノ酸位置700−1663内の任意の位置を含む、請求項1に記載の修飾C3タンパク質。
- 置換が最初の位置および最後の位置を含み、最初の位置が749、874、936、1264、1348、1496、1504、および1550から成る群より選択され、最後の位置が784、921、970 1324、1550、1617、および1663から成る群より選択される、請求項1に記載の修飾C3タンパク質。
- 置換がアミノ酸:1550−1663、1504−1663、1348−1663、1550−1617、1504−1617、1496−1663、1348−1617、1496−1617、1264−1324、749−784、874−921、994−1663、994−1550および936−970から成る群より選択される、請求項8に記載の修飾C3タンパク質。
- 修飾C3タンパク質が因子Bへの親和性を有し、活性コンバターゼの形成を補助する、請求項1に記載の修飾C3タンパク質。
- コンバターゼが37℃にて約15分間の固有半減期を有する、請求項10に記載の修飾C3タンパク質。

を含む、治療剤または診断剤の送達を向上させる方法。 - 送達ステップの前に修飾C3タンパク質を特定の組織に対する親和性を備えた抗体に化学的に結合させるステップ;
をさらに含む、請求項14〜16のいずれかに記載の方法。 - 補体を枯渇させるのに十分な量で修飾C3タンパク質を投与するステップ;

- 修飾タンパク質の機能プロフィールを形成するために、修飾C3タンパク質の少なくとも1つの特性を特徴付けるステップと;
機能プロフィールを処置される疾患または状態と結び付けるステップと;
