Description

(実施例1)
培養培地「SI」(g/lにおける組成:Na2HPO4×12H2O 7.9;KH2PO4 1.8;CoCl2×6H2O 0.02;MgSO4×7H2O 1.0;コーン抽出物2.0;pH7.2±0.2)50ml含み、グルコース50g/lおよび尿素(4〜20g/l)を含むバッフルフラスコ(250ml)をロドコッカス属ロドクロウスM33接種材料培養液の1mlにより接種した。接種材料を同様な培地中で24時間にわたりインキュベートした。培養を、振とう培養器中で、180rpm(円形動作)および30℃で、96時間にわたり行った。バイオマスの収量および細胞のニトリルヒドラターゼ活性を求めた。結果を表1に示す。
培養培地「SI」(g/lにおける組成:Na2HPO4×12H2O 7.9;KH2PO4 1.8;CoCl2×6H2O 0.02;MgSO4×7H2O 1.0;尿素16.0;pH7.2±0.2)を含み、グルコース(50g/l)およびコーン抽出物(0〜10g/l)50mlを含むバッフルフラスコ(250ml)をロドコッカス属ロドクロウスM33接種材料培養液1mlにより接種した。接種材料を同様な培地中で24時間にわたりインキュベートした。培養は、振とう培養器中で、180rpm(円形動作)および30℃で、96時間にわたり行った。バイオマスの収量および細胞のニトリルヒドラターゼ活性を求めた。結果を表2に示す。
培養培地「SI」(g/lにおける組成:Na2HPO4×12H2O 7.9;KH2PO4 1.8;CoCl2×6H2O 0.02;MgSO4×7H2O 1.0;尿素16.0;コーン抽出物4.0;pH7.2±0.2)50mlを含むバッフルフラスコ(250ml)は、20〜90g/lの濃度でグルコースを含む。このフラスコの接種および培養を実施例1におけるように行った。計量されたバイオマス収量および細胞のニトリルヒドラターゼ活性を表3に要約する。
培地「SI」(g/lにおける組成:Na2HPO4×12H2O 7.9;KH2PO4 1.8;CoCl2×6H2O 0.01;MgSO4×7H2O 1.0;尿素16g;コーン抽出物 4.0;pH7.2±0.2)1.5リットルを含み、グルコースの5g/lを含む3リットル容の実験室発酵器をロドコッカス属ロドクロウスM33接種材料培養液100mlにより接種した。接種材料を同様な組成の培地中で24時間にわたりインキュベートした。

・撹拌速度 560rpm
・通気速度 1.5vvm(体積/体積/分)
培地「SI」(g/lにおける組成:Na2HPO4×12H2O 7.9;KH2PO41.8;CoCl2×6H2O 0.01;MgSO4×7H2O 1.0;尿素16.0;コーン抽出物4.0;pH7.2±0.2)1.5リットルを含み、グルコース20g/lを含む3リットル容の実験室発酵器を実施例4におけるように接種した。また、培養条件は、実施例4における通りであった。24〜36時間の発酵時間および全てのグルコースの消費の後に、尿素6g/l、酢酸ナトリウム3g/l、およびCoCl2×6H2O 0.03g/lを計量供給した。培地のpHの上昇の後に、30%強度酢酸(仕込みバッチ)を計量供給した。これらの条件下で、72時間にわたり、315比活性U/mgを示す乾燥バイオマス19.6g/lが生成したことが分かった。総活性は、6174U/mlであった。
