a)多孔質ベッドI(104,204,304)の上流の位置に、マイクロチャネル構造の流路(101,201a,301a,a’)中の多孔質ベッドII(105,205,305)を有する形態の固相材料を提供し、
b)溶質と付随する液体とが、多孔質ベッドI(104,204,304)をとって移動する前に、多孔質ベッドII(105,205,305)を通って、1又はそれ以上の妨害物質を含む液体アリコート中に溶けた溶質Sを移動させる。
多孔質ベッドII(105,205,305)は、溶媒Sに関連したダミーであり、即ち、溶質Sは、影響されずにベッドを通過でき、一方、妨害物質は、固相材料と反応してベッド中で中和される。中和は、退化、捕獲、又は溶質Sと同時又は前に、多孔質ベッドI(104,204,304)を通過するのを防止する他の方法を意味する。溶質Sは、このように、多孔質ベッドII(105,205,305)中で中和された妨害物質の存在下で、固相材料の多孔質ベッドI(104,204,304)と影響しあうことができる。
妨害物質は、得られる結果に負の影響を与える程度に、多孔質ベッドI(104,204,304)の中又は下流で起きる1又はそれ以上の所望の相互作用の結果を妨害する。一般のプロセスでは、反対方向に影響する多くの品質のパラメータ(結果)が存在する。例えば、同一プロセス/方法の、1回の実験またはランを行うのに必要な時間の量(増加)、単位時間あたりのラン数として測定された生産性(減少)、使用される1又はそれ以上の個々の反応物の回復又は損失(それぞれ減少または増加)、1又はそれ以上の得られる生産物の量(減少)、生産量、純度等のような製造パラメータについての精度(増加)である。分析評価のプロトコルでは、1又はそれ以上の、以下に示す品質パラメータが、反対に影響する。精度(低下)、検出限界(増加)、ダイナミックレンジ(狭くなる)、アナライト感度(低下)、診断感度および特異性(低下)、回復(低下)、アナライト等の使用する反応物の望まない損失(低下)である。
a)所望の物質の測定に使用する特徴と同じか、またはそうでなければこの特徴を妨害する測定上の特徴を本来的に含むことにより、またはプロセス中に、そのような測定される特徴を含む存在になる反応にあずかることによる。
b)プロセスの所望の反応と抵触することによる。例えば、加えられた反応物(例えば、アナライトおよび/または1またはそれ以上の加えられた試薬)の中和またはそうでなければ消費による。
c)粒子の状態に変わることにより、マイクロチャネル構造(例えば、多孔質ベッド、測定ゾーン、および/または他の場所)中での堆積および/または沈殿を含む沈殿を起こすことによる。
この種類の液体は、一般に合成物である。溶媒や緩衝成分から分配された妨害物質は、このように、比較的容易に避けることができる。試薬部分は、より複雑かも知れない。例えば生物学的有機試薬のような有機試薬は、一般により複雑な材料から形成され、しばしば除去が困難な妨害物質(汚染物)を含む。このように、これらの物質が、使用される液体中に残っている危険性がある。例えば、発言するペプチド/アミノ酸、炭水化物、および/または液体構造(ステロイド構造を含む)、のような生物学的成分により得られた試薬は、このように、出発材料、複生産物等により汚染されるかも知れない。分極するために使用される試薬に加えて、分極される試薬は、活性、および/または分極の数、および/またはそれぞれの分極が取り付けられる位置について異なっている分子状の存在のスペクトラムを含んでも良い。2つの物質AおよびBの結合は、活性、Aおよび/またはBの量、Aの数とBの数との比、および/またはAがBに取り付けられる位置およびBがAに取り付けられる位置、に関連して変化する分子状の存在のスペクトラムを含んでも良い。分極により得られた分極された試薬は、活性度が変化する断辺のスペクトラムを含んでも良い。分極されていない形態の試薬は、分極汚染物を含む。例えば、それらは、生物学的材料から分極され、および/またはペプチドまたはアミノ酸、ヌクレオチド、炭化水素、および/またはステロイド構造を含む液体構造から選択される、少なくとも1つの構造を示す。分極した試薬、および分極していない形態の試薬の双方は、例えば分子量のスペクトラムを有する集塊試薬を含んでも良い。このように、試薬合成物中の分子のいくつかは、検出、および/または処理される感度の限界を、例えば、多孔質ベッドI(104,204,304)やマイクロチャネル構造の内部表面への不特定な固定のような、複製産物中で関係する他のものより高くする傾向にある。
この種類の液体は、アナライトの濃度および他の成分に関連した、重要な中間試料変化を有する試料により、一般に代表される。アナライト含有液は、一般に、複雑な混合物から形成され、本発明により実行される評価を妨害する物質を多数含む。アナライト含有試料は、このように、工業的または研究的プロセスや方法のプロセスから分配されても良い。プロセスは、非生物学的プロセス、または化学的または生物学的方法により得られる少なくともいくつかの生物学的に得られた材料を用いるという意味において生物学的プロセスであっても良い。例えば、ペプチドまたはアミノ酸、ヌクレオチド、炭化水素、および/またはホルモン構造を含む液体構造から選択される少なくとも1つの構造を示す生物学的有機物を含む。試料の重要な供給源は、血液や血漿および血清のようなその一部、リンパ液、尿、髄液(CSF)、涙、唾液、腸液、胃液、吐き出し液、汗、細胞ホモジェネート、組織上清、人口生物学的流体等のような生物学的流体(即ち、液体)である。「生物学的流体」の用語は、1又はそれ以上のそれらの液体に起因する、この明細書で示される生物学的構造を一般に示す生物学的有機合成物を含む部分も意図する。人口的な生物学的流体は、ここで記載された種類の、1又はそれ以上の生物学的有機分子が加えられた液体を意図する。
A)上述のような粒子材料。血液に起因する試料にとって、粒子材料の用語は、血小板のような血球、赤血球やリンパ細胞のような血液細胞、それらの血球の断片、フィブリンのような凝固生産物、凝血およびその断片、他の血液沈殿物質を含む。一般に生物学的流体では、粒子材料が、細胞関連であり、細胞とその断片、組織断片、分極されていない形態の中性および/またはイオン性の液体等であっても良い。
B)上述のような沈殿、堆積物、またはデポジットを形成する物質。
C)プロセスで使用される試薬と望まない方法で反応し、測定される特徴から分けることができない測定上の特徴を含む物が形成される物質。試薬は、固定化した試薬R、又は分類されたおよび/または結合した形態の試薬のような溶解した形態の試薬でも良い。異好抗体は、この文脈で重要であり、即ち、内生的な抗体は、抗体決定要因を示す抗体試薬と反応し、抗体決定要因は、試料の内生的な抗体を固定することができる。アナライトのための内生的キャリアタンパクは重要であり、特に、もしアナライトが低分子量(例えば、5000ダルトン以下のような10000ダルトン以下)の場合、および/またはもしその自由形態が限定される場合に重要である。他の例は、使用される分類と同じ測定上の特徴を有する内生的な構成である。例えば、もし、試薬で使用される分類が、内生的な酵素と同じ部室を使用すれば、内生的な酵素は測定を妨害する。
D)アルブミン、特にIgGおよびIgAを含むガンマ−グロブリン、アンチトリプシン、およびハプトグロブリンを含むバルクタンパクは妨害し、C)によってキャリアタンパクとして働かない場合でさえも、除去するべきである。
多孔質ベッドI(104,204,304)および多孔質ベッドII(105,205,305)の固相材料は、同様または異なる種類でも良い。このように、下流のベッド(104,204,304)は、多孔質の一体的なプラグであり、上流のベッド(105,205,305)は、粒子又は他の周囲の方法で密集させたベッドである。両方のベッドは、多孔質の一体物または粒子を密集させた多孔質ベッドでも良い。2つのベッドの固相材料は、基本材料、粒子サイズ(および粒子サイズの分布)、極性、被覆性、親水性/疎水性、膨潤性、弾力性、剛性等の1又はそれ以上について異なってもよい。
本発明にかかる一般的なマイクロチャネル構造は、1、2、3、またはそれ以上の流路(101;201a,b;301a,a’,b)を含み、かかる流路のそれぞれは、流路の全てに対して共通の部分を有する。この共通部分(108,208)において、多孔質ベッドI(104,204,304)を含む反応マイクロキャビティI(102,202,302)があり、この多孔質ベッドIは、流路(101;201a,b;301a,a’,b)の全てに共通である。それぞれの流路は、多孔質ベッドI(104,204,304)を通って液体を移動させるために使用される。図1〜3参照。共通部分の上流では、少なくとの1つの流路(101;201a;301a,a’)中に多孔質ベッドII(105,205,305)を含む上流の反応マイクロキャビティ(102,202,302)を有する。
マイクロ流体装置は、1又はそれ以上の、マイクロリットル(μl)範囲の液体アリコート、一般にはナノリットル(nl)範囲で、例えば、アナライトおよび試薬、製造物、試料、バッファ、および/またはその他のような多くの種類の反応物がその中で処理されるマイクロチャネル構造を含む。マイクロリットル(μl)範囲の液体アリコートは、例えば100μl以下、または10μl以下のような、1000μl以下の体積を有し、多くの場合は、例えば500nl以下または100nl以下のような、1000nl以下の体積である、上限が5000nlであるナノリットル(nl)範囲を含む。ナノリットル(nl)範囲は、ピコリットル(pl)の範囲を含む。マイクロチャネル構造は、103以下、好ましくは102以下のような5×102以下の断面直径を有する、1またはそれ以上のキャビティおよび/または導管を含む。
本発明の第2の形態は、本発明の第1の形態で定義したようなマイクロチャネル構造の流路中で、マイクロ流体処理を行う方法である。このマイクロ流体処理は、一般に、例えば、本発明の方法に必要とされる型の多孔質ベッドを含みまた含まない追加の流路のような、マイクロチャネル構造中に追加の機能/部分が有ることを必要とする。装置の、少なくとも1、2、又はそれ以上のマイクロチャネル構造のそれぞれに対して、方法は以下の工程を含む。

b)固相材料の後での使用のための、固相材料上への溶質の固定であり、捕獲、触媒反応、固相触媒等の中で修正される。
a)酵素分類(酵素、コエンザイム、基質、補基質、補因子等)、放射性のあるイソプトペス、発色団、色原体、および/または蛍光団、バイオルミノファ、および/またはバイオルミノゲン、金属原子、およびイオン等のような信号放射分類、および、
b)親和力分類に固定される1の部分と、分析的に検出可能な他の部分とを含む第二の検出反応を必要とする親和力分類、である。
本発明の第3の形態は、1、2、またはそれ以上のマイクロチャネル構造を有するマイクロ流体装置を含む。それらのマイクロ流体装置の少なくとも1つは、1、2、またはそれ以上の流路(101;201a,a’;301a,a’,b)と、第1の形態で記載されたような直列接続された2つの多孔質ベッド(104,204,304;105,205,305)を含む少なくとも1、2、またはそれ以上の流路(101;201a,a’;301a,a’)のそれぞれとを含む。第1の形態のように、上流の多孔質ベッド(105,205,305)の上流、多孔質ベッド(104,204,304;105,205,305)の間、および下流の多孔質ベッド(104,204,304)の下流に、追加の機能ユニットを有しても良い。

Biol. 8 (1988) 3476-3486]。細胞は、Hamの、10%ウシ胎仔血清、100μg/mlのストレプトマイシン、およびグルタミンが補われたハムの媒体F−12中で成長する。ほぼ細胞の融合性の単層は、0.1mg/mlのウシ血清アルブミン(BSA)を補い、攪拌板上で60分間、100ng/mlのPDGF−BB100μmを用いて+/−刺激した、HamのF−12中で、夜通し飢えさせる。刺激しない細胞は、制御として使用される。この細胞のみがリン酸化されていないPDFGβレセプタを有するからである。レセプタは活性化され、内在化を禁じる高濃度のリガンドで飽和され、刺激中のデグラデーションに続く。刺激の後に、細胞は、氷冷されたPBSバッファ中で2回洗浄され、1mlPBS中で「ラバーポリスメン(rubber police men)」を用いて解体される。細胞懸濁液は、蓄えられ、200μlの氷冷されたリーシスバッファ(20mM 3重HCl pH7.5,150mM NaCl、10mM EDTA,0.5% トリトンX−100,0.5%,Deoxycholate,0.5mM Na3VO4 および1% Trasylol(バイエルン))に、15分間、氷上で溶解される。溶解物は13000rpmで15分間、4℃で遠心分離され、上清がアリコート中に蓄えられ、−20℃で保存される。より濃縮された溶解液を作製するために、細胞は、第1成長は75cm2の培養皿の上で、第2のランは175cm2の培養フラスコ中で行われる。全タンパクは、PIERCE Biotechnology (Boule Nordic AB, Huddinge S.)からの、BCAタンパク分析きっとマイクロプレート手続き(BCA Protein Assay Kit Microplate procedure)に従って定量される。
人間のPDGFβレセプタのカルボキシ末端のアミノ酸958−1106に対応した組み換えタンパクに向かうラビットポリクローナル抗体958、人間のPDGFβレセプタのカルボキシ末端のペプチドに向かう羊ポリクローナル抗体P−20、およびマウスモノクローナエウPY99は、Santa Cruz Biotechnologies (Santa Cruz, CA)から得られる。PDGFβレセプタ抗体P−20および658は、人間のPDGFレセプタタイプβの検出、および少ない程度において、ウエスタンブロッティング法、免疫沈降、および免疫組織化学により、マウスおよびラビットオリジン(origen)の検出に薦める。それらは、PDGFレセプタタイプαとは交差反応性を有すべきではない。この抗体は、同じ細胞培養を伴う、免疫沈降やウエスタンブロッティングの実験において、広範囲に使用される。[Pietras K., et al.PDGFレセプタ信号伝達の禁止による腫瘍間質中の化学療法の抗腫瘍効果の拡張,Cancer Research 62 (2002) 5476-5480]。
分析において、洗浄バッファおよび抗体は、共通の分配チャネルを通って分配され、細胞溶解プレパレーションは、それぞれの導入口を通って分配される。それぞれのバッチランは、3重のおよび空の試料蛛に標準を含む。分析は、小さな希釈続き(+/−PDGF−BB)および、リーシスバッファを有する多くのブランクを含む。分析中の全ての工程は、Gyrolab Workstation (Gyros AB) 中で自動的に行われる。
多孔質ベッドは、PBS−T(0.015M Na−PO4 pH7.4,0.15M NaCl、0.01% NaN3,0.01% Tween−20)で2回洗浄され、ストレプトアビジンが固定されたフェニルデキシトランで被覆されたポリスチレンベッドを再生するために、短時間回転する。回転に続いて、洗浄バッファ中で希釈された、ビオチニル化捕獲酵素が、667nMの濃度で加えられた。再生と同じ方法で、洗浄は2回繰り返された。
細胞溶解物が個々の導入口に加えられ、体積(200nl)が規定され、PDGFβレセプタ(アナライト)が捕獲される多孔質ベッドに運ばれる。2つの洗浄工程の後、短い回転を行い、蛍光検出中にベッドが液体で満たされるのを確実にする。背景蛍光検出は、LIF検出器で設定される、1%、5%、および25%の異なった3つの感度を有する。回転工程中にアナライトに接合することができる検出抗体を加える前に、超過のバッファは、短い回転により洗い流された。6つの洗浄工程は、最終的に、2つの洗浄工程が通常の洗浄バッファを用い、4つがイソプロパノール20%を含む洗浄バッファを用い、過剰の検出抗体を除去する。
予備実験から、P−20/958、P−20/PY99、および958/PY99の、3つの抗体の組みが選択された。
細胞溶解物実験は、ベッド中の信号分布(3倍)により、高いCV値が得られた。いくつかの実験では、細胞溶解物とブランク信号との間で信号の若干の違いが見られたが、殆ど重要な信号は背景信号から区別できなかった。ベッド中の濃化は殆ど不規則であり、通常のベッドパターンは見られなかった。いく地下の場合、信号が多孔質ベッドの下で進む傾向にあった。
以下の2つの組み合わせが試験された。
a)ストレプトアビジンが固定されたフェニルデキシトロンで覆われたポリスチレン粒子のベッド上の、フェニルデキシトロンで覆われたポリスチレンビーズのベッド(本質的に多孔質でない)、および
b)ストレプトアビジンが固定されたフェニルデキシトロンで覆われたポリスチレンビーズのベッド上の、Superdex(登録商標)ペプチド。
基本分析
TNFαのためのマイクロ流体サンドイッチ蛍光免疫測定が、WO 04083108 (Gyros AB)の実験の一部において、マイオグロビンが概説されている。固相および固定化技術および蛍光分類技術は、WO 04083108 (Gyros AB)に概説されている。分析は、IgGベッド、または対応する固定化されたIgGの無い対応するベッドを、捕獲アンチTNFαマウス抗体を示すベッドの上流に隣接して配置することにより、修正された。
双方とも、WO 04083108(Gyros AB)で使用されたものと同じである。
親水性抗体に富んだ3つの人間血清試料と、1つの通常の試料が、Aにおいて使用された。4つの全ての試料は、人間TNFα(200pg/ml)が加えられ、Bで使用された。
WO 04083108 (Gyros AB) で使用されたものと同種のストレプトアビジン粒子が、バッチモードで、ボビンIgGとマウスIgGとの4つの組み合わせ−ボビンIgG:マウスIgGが10:1、ボビンIgG:マウスIgGが1:1、ボビンIgG、マウスIgGで被覆された。続いて粒子が、捕獲アンチTNFαマウス抗体を示すベッドの上に導入された。双方のベッドに対する導入技術は、WO 04083108 (Gyros AB) のアンチマイオグロビン抗体ベッドと同じである。実験中の流路方向は、IgGベッドからアンチTNFαマウス抗体ベッドに向かう方向である。
アンチTNFαマウス抗体ベッドの前のIgGベッドの、A)上述の4つの人間血清試料の疎水性抗体の検出、B)上述のTNFαが加えられた人間血清試料のTNFα分析のためのTNFαの回復、およびC)標準カーブ、に対する影響。
図4参照。各グループのステープル(staple)は、個々の試料を示す。10:1のボビン:マウスIgGに基づくIgGベッドが、捕獲疎水性抗体中で最も有効であった。疎水性抗体は、普通の血清試料では検出されなかった。アンチTNFαマウス抗体ベッドの前のIgGベッドを使うことなく、後者のベッドは、アンチTNFαマウス抗体ベッドに基づくTNFα分析を妨害する、多量の親水性抗体を捕獲した。
図5a参照。各グループのステープルは、個々の試料を示し、ステープルの無いものは、普通の血清試料を示す。IgGベッドをアンチTNFαマウス抗体ベッドの前に配置した場合、TNFαの回復の変化が得られた。
図5b参照。IgGベッドの無い場合と比較して、異なったIgGベッドをアンチTNFαマウス抗体ベッドの前に配置した場合、標準カーブの変化が重要であった。
Claims (17)
- 1、2、またはそれ以上の流路(101;201a,b;301a,a’,b)を有し、これらの全てが、全ての流路に共通の多孔質ベッドI(104,204,304)を含み、多孔質ベッドがベッドを通過する溶質Sと作用することができる固定された反応物Rを有する、マイクロチャネル構造を含むマイクロ流体装置であって、
流路(101;201a,b;301a,a’,b)の少なくとも1つ(101;201a;301a,a’)が、多孔質ベッドI(104,204,304)の上流に配置され、溶質Sに対する作用ではダミーとなるが、溶質Sを伴う液体アリコート中に存在し、溶質Sと固定化された反応物Rとの間の作用の結果を妨害しうる物質DSとは作用しうる第2の多孔質ベッドII(105,205,305)を含むことを特徴とするマイクロ流体装置。 - 多孔質ベッドI(104,204,304)と多孔質ベッドII(105,205,305)が、物理的に互いに分離されたことを特徴とする請求項1に記載のマイクロ流体装置。
- 多孔質ベッドI(104,204,304)の上流端部が、多孔質ベッドII(105,205,305)の下流端部と接続されたことを特徴とする請求項1に記載のマイクロ流体装置。
- 上流端部と下流端部との間に、多孔質膜(106)を有することを特徴とする請求項3に記載のマイクロ流体装置。
- 多孔質ベッドI(104,204,304)および多孔質ベッドII(105,205,305)の少なくとも1つが、粒子を押し固めたベッドであり、残りのベッドが、もしあるのなら、多孔質の一体物のプラグであることを特徴とする請求項1〜4のいずれかに記載のマイクロ流体装置。
- 多孔質ベッドI(104,204,304)および多孔質ベッドII(105,205,305)の少なくとも1つが、サイズ排除材料からなる固相材料を含むことを特徴とする請求項1〜5のいずれかに記載のマイクロ流体装置。
- 上記妨害物質が溶質Sより小さく、上記少なくとも1つの流路の少なくとも1つの中の、少なくとも多孔質ベッドII(105,205,305)が、溶液に関連して、妨害物質が多孔質ベッドIIを通るのを遅らせる排除限界を有するサイズ排除材料である固相材料を含むことを特徴とする請求項1〜6のいずれかに記載のマイクロ流体装置。
- 上記1、2、またはそれ以上の流路(101;201a,b;301a,a’,b)の残りの、少なくとも1、2、またはそれ以上(201b;301b)が、多孔質ベッドIIを有さないことを特徴とする請求項1〜6のいずれかに記載のマイクロ流体装置。
- 上記少なくとも1、2、またはそれ以上の流路中の多孔質ベッドIIが、溶質とともに存在する妨害物質と作用することができる固定された試薬RDSを含むことを特徴とする請求項1〜7のいずれかに記載のマイクロ流体装置。
- 上記少なくとも1つの流路が、2またはそれ以上の流路であり、上記2またはそれ以上の少なくとも1つの中のRDSが、上記2つの流路の残りの、少なくとも1つの中のRDSとは異なることを特徴とする請求項1〜8のいずれかに記載のマイクロ流体装置。
- マイクロ流体装置のマイクロチャネル構造の流路(101;201a,b;301a,a’)中で行われ、上記流路(101;201a;301a,a’)中に配置され、移動中に溶質Sと作用することができる固定された反応物Rを有する多孔質ベッドI(104,204,304)を通って、溶質Sを含む液体アリコートを移動させる工程を含むマイクロ流体プロセスであって、
i)多孔質ベッドI(104,204,304)の上流にあり、溶質Sとの作用に関してはダミーであるが、妨害物質DSとは作用することができる多孔質ベッドII(105,205,305)を含む形態の上記流路(101;201a;301a,a’)を提供する工程と、
ii)多孔質ベッドIIの上流にある上記流路(101;201a;301a,a’)中に、上記溶質Sと上記妨害物質とを含む液体アリコートを提供する工程と、
iii)多孔質ベッドII(105,205,305)を通ってアリコートを移動させる工程と、そして、
iv)その後に、多孔質ベッドI(104,204,304)を通って溶質Sを移動させ、反応物Rとの作用を行わせる工程とを含むマイクロ流体プロセス - それらのそれぞれが上記流路の全てに共通な多孔質ベッドI(104,204,304)を含み、それらの少なくとも1つ(101;201a;301a,a’)が多孔質ベッドI(104,204,304)の上流にある多孔質ベッドII(105,205,305)を含む、1、2、又はそれ以上の流路(101;201a,b;301a,a’,b)を含むマイクロチャネル構造を有するマイクロ流体装置であって、
上記少なくとも1つの流路(101;201a;301a,a’)中の、多孔質ベッドI(104,204,304)および多孔質ベッドII(105,205,305)の一方または双方が、包括リガンドを含む固相材料を含むことを特徴とするマイクロ流体装置。 - 上記少なくとも1つの流路(101;201a;301a,a’)の1またはそれ以上の中の、多孔質ベッドII(105,205,305)中の包括リガンドが、多孔質ベッドI中と同じであることを特徴とする請求項12のマイクロ流体装置。
- 上記少なくとも1つの流路(101;201a;301a,a’)の1またはそれ以上の中の、多孔質ベッドII(105,205,305)中の包括リガンドが、多孔質ベッドI(104,204,304)中のリガンドに対して親和対(アンチリガンド)であることを特徴とする請求項12に記載のマイクロ流体装置。
- 上記リガンドが、ビオチンおよびアンチビオチンから選択されることを特徴とする請求項12〜13のいずれかに記載のマイクロ流体装置。
- 多孔質ベッドI(104,204,304)および多孔質ベッドII(105,205,305)の双方を含むたった1つの流路(101)を有することを特徴とする請求項12〜15のいずれかに記載のマイクロ流体装置。
- 多孔質ベッドII(105,205,305)の下流端部が、多孔質ベッドI(104,204,304)の上流端部に接続され、可能であればその端部の間に多孔質膜を有することを特徴とする請求項16に記載のマイクロ流体装置。
