Abstract
【解決手段】ラクトフェリン及び鉄−ラクトフェリンにエナメル質の脱灰抑作用を見出し、これらを有効成分として含有させることによって、歯質硬度維持剤、及び歯質硬度維持用飲食品を提供する。これらを使用又は摂取することにより、エナメル質の脱灰が抑制されるのでう蝕の予防に有効である。

Description
陽イオン交換樹脂のスルホン化キトパール(富士紡績社製)400gを充填したカラム(直径5cm×高さ30cm)を脱イオン水で十分洗浄した後、このカラムに未殺菌脱脂乳40L(pH6.7)を流速25ml/minで通液した。通液後、このカラムを脱イオン水で十分洗浄し、続いて0.7M塩化ナトリウムを含む0.02M炭酸緩衝液(pH7.0)で洗浄した後、0.98M塩化ナトリウムを含む0.02M炭酸緩衝液(pH7.0)で樹脂に吸着した画分を溶出した。そして、この溶出液を逆浸透(RO)膜により脱塩して、濃縮した後、凍結乾燥してラクトフェリン粉末11gを得た。なお、このラクトフェリン粉末には、ラクトフェリンが93重量%含まれていた。
このようにして得られたラクトフェリン10μmolと重炭酸ナトリウム1.2molとを含む溶液1LをA液として作製した。硫酸第二鉄を鉄イオンとして1.5mmol含む溶液1LをB液として作製した。A液にB液を加えた後、分子量5000カットの限外濾過膜にて脱塩、濃縮し、模擬緩衝液(pH8.9)にて鉄濃度が94mg/100mlになるように希釈して、鉄−ラクトフェリンを得た。このようにして得られた鉄−ラクトフェリンは、溶液100mlあたりラクトフェリンを0.9g及び鉄を94mg含有していた。

実施例1で製造したラクトフェリン、鉄−ラクトフェリンを用いて、歯質硬度維持作用について、人工口腔装置を用いて検証を行った。本装置は、3台の人工口腔部分、送液用ポンプ2台、恒温槽、冷却撹拌器3台、pHレコーダーから構成されている。上部からは5本のステンレスチューブを固定したシリコン栓を、下部からはドレイン用チューブと平面pH電極を逆さに固定したシリコン栓を装着して密閉した。人工口腔部分のウォータージャケットに温水を循環して装置内部を37℃に維持した。平面pH電極周囲にはテフロン製ホルダーにユーティリティワックスを用いて4個のウシエナメル歯片(3.5mm×3.5mm×1.5mm)を固定した。その上部から2.5% スクロース(電極上で1.0%)含有HI培地、細菌懸濁液を連続的に滴下した。対照としてPBSを、実験群として各種乳成分を滴下した。pHを連続的に記録しながら15〜18時間滴下を続け、人工プラーク形成に伴って対照のpHが4付近まで低下した時点で滴下を終了した。エナメル質歯片上およびpH電極上に形成された人工バイオフィルムを0.5N 水酸化ナトリウム溶液で処理して遠心分離し、沈殿物の濁度を500nmで測定して菌体量とした。その上清をフェノール硫酸法によって定量し非水溶性グルカン(WIG)量とした。エナメル質の脱灰度を実験前後のVicker’s硬度変化(ΔH)から評価した。なお、実験はS. sobrinus ATCC33478 (OD500=0.1)のバイオフィルム形成、pH低下、ウシエナメル歯片の脱灰に及ぼす乳成分の影響を調べた。
表1に示す配合で各成分を混合し、クリームを製造して容器に充填し、歯周病予防及び改善用歯磨き剤を製造した。
表2に示す配合で各成分を混合し、歯質硬度維持用うがい剤を製造した。
表3に示す配合に従い、ガムベースを溶解し、各成分を撹拌後、成形して歯質硬度改善用ガムを製造した。
砂糖と水飴を表4に示す配合で混合撹拌し、150℃まで煮詰め、115℃まで冷却した後、鉄−ラクトフェリンを0.1%重量となるように添加して撹拌し、冷却盤上で冷却し、成形して歯質硬度維持用飴を製造した。

- ラクトフェリン及び/又は鉄−ラクトフェリンを配合した歯質硬度維持用飲食品。
