Description
ゲノムを次の世代に伝達する能力により、ES細胞は、望みの遺伝的改変を有する、あるいは有しないES細胞を使用することによる家畜動物の生殖細胞操作のための潜在的な有用性を有している。さらに、家畜動物、例えば有蹄類の場合は、類似の未着床家畜胚からの核は除核した卵母細胞の満期(term)までの発生を支援する(Smithら、Biol. Reprod., 40:1027-1035(1989));および、Keeferら、Biol. Repord., 50;935-939(1994))。これは、報告によれば移入後8細胞期を超えて除核卵母細胞の発生を支援しないというマウス胚の核と対照的である(Cheongら、Bioil. Reprod., 48;958(1993))。従って、家畜動物からのES細胞は非常に望ましい。なぜならば、それらは核移植のための遺伝的あるいは他の方法で操作された全能性ドナー核の供給源の可能性を提供するかもしれないからである。
さらにSaitoら、Roux’s Arch. Dev. Biol., 201:134-141 (1992)は3代継代を生き延びた培養ウシ胚性幹細胞様細胞株を報告しているが、4回目の継代の後に失われた。Handysideら、Rou’x Arch. Dev. Biol., 196:185-190 (1987)はマウスICMから由来するマウスES細胞株の単離を可能とする条件下で免疫外科的に単離したヒツジ胚の内部細胞塊の培養を開示している。Handysideらはそのような条件下でヒツジICMは接着し、広がり、ES細胞様および内胚葉様細胞の両方の領域を発達させるが、長期間の培養後には内胚葉様細胞のみが顕著であることを報告している。
また最近Campbellら、Nature, 380:64-68(1996)は、マウスにおけるES細胞株の単離を促進する条件下で培養された第9日のヒツジ胚からの培養胚盤細胞(embryonic disc cell)(ED細胞)の核移植を受けた後に生きている子ヒツジの作製を報告した。この著者たちは、第9日のヒツジ胚からのED細胞は核移植により全能性であると結論付け、全能性は培養に際して維持されていると結論付けた。
Smithら、WO 94/24274、1994年10月27日公開、Evansら、WO 90/03432、1990年4月5日公開、およびWheelerら、WO 94/26889、1994年11月24日公開、はトランスジェニック動物を得るために使用されるかもしれないと称する動物幹細胞の単離、選抜および増殖を報告している。Evansらはまた、トランスジェニック動物の作製に有用であると主張する多分化能胚性幹細胞といわれるものをブタおよびウシ種から誘導したことを報告した。さらに、Wheelerら、WO 94/26884、1994年11月24日公開、はキメラおよびトランスジェニックな有蹄類の作製に有用と言われる胚性幹細胞を開示した。
このように、上述の面に基づき、多くのグループがES細胞株を作製する試みを行ってきており、これは例えば、クローン化またはトランスジェニック胚の作製および核移植におけるその潜在的応用性のためである。
培養胚盤細胞の核移植を受けた生きた子ヒツジの作製も報告されている(Campbellら、Nature, 380:64-68 (1996))。さらに、ウシ多分化能胚性細胞の核移植における使用およびキメラ胎仔の作製が報告されている(Sticeら、 Biol. Reprod.,54:100-110(1996);Collasら、Mol. Reprod. Dev., 38:264-267 (1994))。Collasらは顆粒膜細胞(成体細胞)が胚を作製するためのウシクローニング法に使用できることを証明した。しかしながら、初期胚段階(胚盤胞期)を過ぎて発生するという証明は無かった。また、顆粒層細胞は容易に培養できず、雌からしか得られない。Collasらは培養において顆粒層細胞を増殖させる試み、またはそれらの細胞を遺伝的に改変しようとする試みは行わなかった。
またトランスジェニック動物作製の領域でも問題がある。現在の方法では、異種DNAが初期胚または胎仔において種々の細胞タイプに分化する胚性細胞株に導入され、最終的にトランスジェニック動物へと発生する。しかしながら、沢山のの初期胚が1体のトランスジェニック動物の作製に必要であり、従って、この方法は非常に非効率的である。また、代理雌へ胚を入れる時間と費用が消費される前にトランスジェニック動物を選抜するための簡単で効率的な方法が存在しない。加えて、遺伝子ターゲッティング手法は初期胚遺伝子導入法では容易に行うことができない。
従って、文献において以前に報告されていることにも拘わらず、改良された哺乳動物クローニング法に対する必要性が存在する。
本発明のより具体的な目的は、分化した哺乳動物細胞の核を同種の除核卵母細胞へ移植することを含む哺乳動物細胞クローニングのための新規な方法を提供することである。
本発明の他の目的は、遺伝的に優れたあるいは他の望ましい特徴を有する成体哺乳動物を増殖させる方法を提供することである。
本発明の別の目的は遺伝的に操作された、またはトランスジェニック哺乳動物(すなわち、胚、胎仔、産仔)を作製する改良された方法を提供することである。
本発明の別の目的は分化細胞の核をその分化細胞と同じ種の卵母細胞へ移植することによって得られる遺伝的に操作されたまたはトランスジェニック哺乳動物(すなわち、胚、胎仔、産仔)を提供することである。
本発明の別の目的は、分化細胞の核をその分化細胞と同種の除核卵母細胞に移植することを含む、哺乳動物CICM細胞を作製する新規な方法を提供することである。
本発明の別の目的は、分化した哺乳動物細胞の核をその分化細胞と同種の除核卵母細胞中へ移植することによって作製されるCICM細胞を提供することである。
本発明の別の目的はそのようなCICM細胞を治療または診断に使用することである。
本発明のより具体的な目的は、ヒトおよび有蹄類のCICMを含むそのようなCICM細胞を、細胞、組織または器官移植が治療的にあるいは診断的に有益であるような一切の疾病の治療または診断のために使用することである。CICM細胞は同種内で、または種間で使用されてよい。
本発明の別の目的は、ヒトおよび有蹄類組織を含むNT胚、胎仔または産仔に由来する組織を組織または器官移植が治療的にあるいは診断的に有益であるような一切の疾病の治療または診断のために使用することである。組織は同種内または種にまたがって使用されてよい。
本発明のより具体的な目的は、本発明によって作製されたヒトCICMを分化したヒト細胞、組織または器官の作製に使用することである。
本発明の別の具体的な目的は、本発明によってin vitroで作られたCICM細胞を例えば細胞の分化の研究およびアッセイ目的、例えば薬剤研究のために使用することである。
本発明の別の目的は、本発明によって作られたCICM細胞から作製されたアイソジェニックまたはシンジェニック細胞、組織または器官の使用を含む、移植治療の改良された方法を提供することである。そのような治療には、例として、パーキンソン病,ハンチントン病、アルツハイマー病、ALS、脊髄損傷、多発性硬化症、筋ジストロフィー、糖尿病、肝臓疾患、心臓疾患、軟骨置換、火傷、血管疾患、尿路系疾患を含む疾患及び損傷の治療、および免疫疾患、骨髄移植、癌、その他の疾患の治療が含まれる。
本発明の別の目的は、分化した哺乳動物細胞または細胞核をNTユニットの形成に使用するに先だって、望みの遺伝子を分化哺乳動物細胞または細胞核へ挿入、除去または改変することによって作製された遺伝子操作された、またはトランスジェニックCICM細胞を提供することである。
本発明の別の目的は、本発明によって作られたトランスジェニックまたは遺伝子操作CICM細胞を遺伝子治療、特に上述した疾患及び損傷の治療および/または予防に使用することである。
本発明の別の目的は、本発明によって作られたCICM細胞または本発明によって作られたトランスジェニック若しくは遺伝子操作CICM細胞を核移植のための核ドナーとして使用することである。
(i)望みの分化した哺乳動物細胞または細胞核を、その分化した細胞または細胞核と同じ種の除核した哺乳動物卵母細胞中へ、核移植(NT)ユニットの形成に適した条件下で挿入すること;
(ii)生じた核移植ユニットを活性化すること;
(iii)前記活性化核移植ユニットを2細胞発生期より大きくなるまで培養すること;および、
(iv)前記培養NTユニットをNTユニットが胎仔に発生するような宿主哺乳動物に移すこと。
胎仔の細胞、組織および/または器官は細胞、組織および/または器官移植の分野で使用するのに有利である。
本発明はまた、遺伝子操作された、またはトランスジェニック哺乳動物のクローニング方法を含み、それにより、分化哺乳動物細胞または細胞核の除核卵母細胞への挿入に先だって、分化哺乳動物細胞または細胞核へ望みの遺伝子が挿入、除去または改変される。
また本発明により、上記の方法によって得られる哺乳動物およびそれらの哺乳動物の産仔が提供される。
本発明は有蹄類のクローニングに使用するのが好ましい。
(i)望みの分化した哺乳動物細胞または細胞核をその分化した細胞または細胞核と同じ種の除核した哺乳動物卵母細胞中へ、核移植(NT)ユニットの形成に適した条件下で挿入すること;
(ii)生じた核移植ユニットを活性化すること;
(iii)前記活性化核移植ユニットを2細胞発生期より大きくなるまで培養すること;および、
(iv)前記培養NTユニットから得られた細胞を培養してCICM細胞を得ること。
CICM細胞は、細胞、組織および器官移植の分野で使用するのに有利である。
前述した目的および他の目的とともに、本発明の利点及び特徴は以下で明らかとなるであろうし、本発明の性質は以下の本発明の好ましい実施態様の詳細な説明および添付の請求の範囲を参照することによりさらに明確に理解されるであろう。
本発明により、分化した胎仔または成体哺乳動物細胞由来の細胞核がドナー核と同種の除核哺乳動物卵母細胞へ核移植される。核はクローン胚の発生を指令すべく再プログラムされ、胎仔および産仔を作るためにレシピエント雌に移すことができ、またはCICM細胞を作製するために使用することができる。クローン化胚はまた受精胚と一緒にしてキメラ胚、胎仔および/または産仔を作り出すこともできる。
従来技術の方法はクローニング手順において胚性細胞タイプを使用してきた。これにはCampbellら(Nature, 380:64-68, 1996)およびSticeら(Biol. Reprod., 54:100-110, 1996)の研究が含まれる。これらのどちらの研究においても、胚性細胞株は妊娠10日未満の胚に由来するものであった。どちらの研究においても、細胞はクローニング方法に用いるドナー細胞の明確な分化を防ぐためにフィーダー層上で維持された。本発明は分化細胞を使用する。
従って、本発明により、有蹄類を含む、哺乳動物の優れた遺伝子型の増殖が可能である。これにより遺伝的に優れたあるいは他の望みの特徴を有する成体動物を増殖させることとができる。例えば、多くの重要な有蹄類種において進歩が加速されるであろう。本発明により、収穫することができ、かつクローニング手順に用いられる可能性のある無数の胎仔または成体細胞が存在する。このことは短期間で多くの同一の仔を生じさせ得るものである。
(i)ドナー核の供給源として使用される、望みの分化した哺乳動物細胞を得る工程;
(ii)ドナー核の供給源である細胞と同じ種の哺乳動物からの卵母細胞を得る工程;
(iii)前記卵母細胞を除核する工程;
(iv)望みの分化細胞または細胞核を除核した卵母細胞へ、例えば融合またはインジェクションによって移入し、NTユニットを形成させる工程;
(v)得られたNTユニットを活性化する工程;
(vi)前記活性化核移植ユニットを2細胞発生期より大きくなるまで培養する工程;および、
(vii)前記培養NTユニットを、NTユニットが胎仔に発生するような宿主哺乳動物に移入する工程。
また本発明により上記の方法によって得られた哺乳動物および、それらの動物の産仔が提供される。本発明は好ましくは有蹄類のクローニングに使用される。
本発明はさらに細胞、組織および器官移植の分野におけるNT胎仔および、NTおよびキメラ産仔の使用を提供する。
(i)望みの分化哺乳動物細胞または細胞核を、その分化細胞または細胞核と同じ種の除核哺乳動物卵母細胞中へ、核移植(NT)ユニットの形成に適した条件下で挿入すること;
(ii)得られた核移植ユニットを活性化すること;
(iii)前記活性化移植ユニットを2細胞発生期より大きくなるまで培養すること;および、
(iv)前記培養NTユニットから得られた細胞を培養してCICM細胞を得ること。
CICM細胞は細胞、組織および器官移植の分野、またはトランスジェニック胎仔または産仔を含む胎仔または産仔の作製において使用するのに有利である。
好ましくは、NTユニットは少なくとも2〜400細胞のサイズ、好ましくは4から128細胞、最も好ましくは少なくとも約50細胞のサイズまで培養されるであろう。
ヒト細胞を含む哺乳動物細胞はよく知られた方法で得られてもよい。本発明に有用な哺乳動物細胞には、例として、上皮細胞、神経細胞、表皮細胞、ケラチン細胞、造血細胞、メラニン形成細胞、軟骨細胞、リンパ細胞(BおよびTリンパ細胞)、赤血球、マクロファージ、単球、単核細胞、線維芽細胞、心筋細胞および他の筋細胞などが含まれる。さらに、核移植に用いられる哺乳動物細胞は種々の器官、例えば皮膚、胚、膵臓、肝臓、胃、小腸、心臓、生殖器官、膀胱、腎臓、尿道および他の泌尿器官などから得られることがある。これらは、適切なドナー細胞の例に過ぎない。適切なドナー細胞、すなわち、本発明に有用な細胞はどんな細胞あるいは身体器官から得てもよい。これには全ての体細胞または生殖細胞が含まれる。
線維芽細胞は、発生過程の胎仔および成体動物から大量に得ることができるため理想的な細胞タイプである。線維芽細胞は分化したもので、従って、以前はクローニング手順に用いるにはよくない細胞タイプと考えられていた。重要なことには、この細胞はin vitroで容易に増殖させることができ、迅速な倍加時間を有しており、遺伝子ターゲッティング法における使用のためにクローン増殖させることができる。分化細胞タイプが使用されるため、ふたたび本発明は新規である。本発明は、細胞が容易に増殖され、遺伝的に改変され、in vitroで選抜され得るため、有利である。
卵母細胞を単離する方法は技術的によく知られている。本質的には、これは哺乳動物、例えばウシの卵巣または生殖管から卵母細胞を単離することを含むであろう。ウシ卵母細胞の簡単な入手源は屠殺場材料である。
遺伝子工学、核移植およびクローニングのような技術を成功裡に使用するためには、卵母細胞は一般に核移植のレシピエント細胞として使用されるかもしれない前、および、卵母細胞が胚に発生すべく精子によって受精できるようになる前に成熟しなければならない。この過程は一般に未成熟(前期I)卵母細胞を哺乳動物卵巣、例えば屠殺場にて得られるウシ卵巣から集め、受精または除核に先だって成熟化培地中で卵母細胞を中期II期に達するまで成熟させることを必要とする(ウシ卵母細胞の場合は、吸引後の約18−24時間に起こる)。本発明の目的には、この期間は「成熟期間」として知られている。本明細書で期間の計算に用いる場合は、「吸引」は卵胞からの未成熟卵母細胞の吸引をいう。
除核および核移植時の卵母細胞の成熟段階はNT法の成功のために重要であることが報告されている(Pratherら、Differentiation, 48, 1-8, 1991を参照せよ)。一般には、哺乳動物胚クローニングの実施の成功にはレシピエント卵母細胞として中期II段階の卵母細胞が使用される。なぜなら、この段階の卵母細胞は受精精子を扱うように導入された核を扱うために「活性化」され得る、または充分に「活性化」されていると考えられているからである。家畜動物においては、特にウシでは、卵母細胞の活性化期間は通常吸引後約16〜52時間、好ましくは約28〜42時間の範囲である。
所定の成熟期間後(約10から40時間、好ましくは約16〜18時間)、卵母細胞は除核されるであろう。除核に先立ち卵母細胞は取り出され、卵丘細胞の除去前に1mg/mlのヒアルロニダーゼを含むHECM中に置かれるのが好ましい。これは非常に細い穴のピペットを通して繰り返してピペッティングすることにより、または短くヴォルテックスすることによりなされることがある。引きはがされた卵母細胞は次に極体についてスクリーニングされ、極体の存在により決定される、選抜した中期II卵母細胞が次に核移植に使用される。
除核は以下の通りである。
除核は、米国特許4,994,384号に記載されたような既知の方法によって行われてよい。この特許は本明細書に含まれるものとする。例えば、中期II卵母細胞は、直ちに除核するときは場合によっては7.5μg/mlのサイトカラシンBを含んでいてもよいHECM中に置かれるか、または、例えばCR1aaのような胚培養培地に10%発情期ウシ血清を加えた培地中に置き、その後に除核、好ましくは24時間を超えない、より好ましくは16〜18時間を超えない後に除核されてもよい。
除核は極体および隣接する細胞質を除くためにマイクロピペットを用いて顕微手術的に(microsurgically)行われることがある。次に卵母細胞は除核に成功したものを同定するためにスクリーニングされる。このスクリーニングは卵母細胞をHECM中の1μg/mlのヘキスト33342染色剤で染色し、卵母細胞を10秒未満の紫外線照射下で観察することによって行われることがある。うまく除核できた卵母細胞は次に適切な培地、例えば、CR1aa+10%血清中に置かれる。
次に、除核した卵母細胞と同じ種の単一の哺乳動物細胞がNTユニット形成に使用する除核卵母細胞の囲卵空隙(perivitelline space)中へ移入されるであろう。その哺乳動物細胞および除核卵母細胞はこの技術で知られた方法によってNTユニットを作り出すために使用されるであろう。例えば、細胞はエレクトロフュージョンによって融合されてもよい。エレクトロフュージョンは細胞膜の一過性の破損を起こすのに充分な電気パルスを供給することによって達成される。細胞膜は迅速に再形成するためこの細胞膜の破損は極めて短時間である。従って、2つの隣接する膜が破損を誘導され、脂質二重層が混合再形成されるとき、小さなチャンネルが二つの細胞間に開く。そのような小さな開口の熱力学的不安定性により、そのチャンネルは2つの細胞が1つになるまで広がる。この過程の更なる考察についてはPratherらの米国特許4,997,384号が参照される(この文献は本明細書にそっくり含まれるものとする)。例えばショ糖、マンニトール、ソルビトールおよびリン酸緩衝液を含む種々のエレクトロフュージョン培地を使用することができる。融合はセンダイウイルスを融合媒介物として使用して達成することもできる(Graham, Wister Inot. Symp. Monogr., 9, 19, 1969)。
また、ある場合には(例えば、小さなドナー核では)エレクトロフュージョンを使用するよりも寧ろ核を直接卵母細胞に注入することが好ましいかもしれない。そのような技法はCollasとBarnes, Mol. Reprod. Dev., 38:264-267 (1994)に開示されている。この文献は本明細書にそっくり含まれるものとする。
NTユニットは既知の方法で活性化されてよい。そのような方法には、例えば、NTユニットに本質的に冷却(cold)温度ショックまたは事実上冷涼(cool)温度ショックを与えることにより、NTユニットを生理的温度以下で培養することが含まれる。これはNTユニットを室温、すなわち胚が通常曝されている生理的温度条件に比較して冷たい温度で培養することによって行うのが最も簡便である。
あるいは、活性化は既知の活性化剤を適用することにより行われることもある。例えば、受精の際の精子による貫通は前融合卵母細胞を活性化し、核移植後により多くの生育可能な受胎および多数の遺伝的に同一の仔ウシを生じさせることが示されている。また、電気的および化学的ショックのような処置が融合後にNT胚を活性化するために用いられることがある。適切な卵母細胞活性化法はSusko-Parrishらの米国特許5,496,720号の主題である。この特許は本明細書に全て含まれるものとする。
(i)卵母細胞中の2価のカチオン濃度を上げること、および
(ii)卵母細胞中の細胞性タンパク質のリン酸化を低下させること。
これは一般に2価のカチオン、例えばマグネシウム、ストロンチウム、バリウム、カルシウムを、例えばイオノフォアの形で卵母細胞細胞質に導入することによって行われる。2価のカチオン濃度を増加させる他の方法には、電気ショック、エタノール処理およびケージキレート剤(caged chelator)処理の利用が含まれる。
リン酸化は既知の方法、例えば、キナーゼ阻害剤、例えば6-ジメチル-アミノプリン、スタウロスポリン、2-アミノプリン、およびスフィンゴシンのような、セリン−スレオニンキナーゼ阻害剤の添加によって低減されることがある。
または、細胞性タンパク質のリン酸化は卵母細胞中へのホスファターゼ、例えばホスファターゼ2Aおよびホスファターゼ2Bの導入によって阻害されてもよい。
活性化NTユニットは次に適切なin vitro培地中でCICM細胞の生成と細胞コロニーが生成されるまで培養される。胚の培養と成熟に適した培地は技術的によく知られている。ウシ胚培養及び維持のための既知の培地の例としては、ハム(Ham)のF−10+10%ウシ胎仔血清(FSC)、組織培養培地-199(TCM-199)+10%ウシ胎仔血清、チロード−アルブミン−乳酸塩−ピルビン酸塩(TALP)、ダルベッコのリン酸緩衝生理食塩水(PBS)、イーグルとウィッテンの(Eagle's and Whitten's)培地が含まれる。卵母細胞の収集と成熟のために最も一般的に使用される培地はTCM-199、および1から20%の、ウシ胎仔血清、新生仔血清、発情期雌ウシ血清、子ヒツジ血清または去勢ウシ血清を含む血清補充培地である。好ましい維持培地にはアール塩(Earl salts)、10%ウシ胎仔血清、0.2mM ピルビン酸ナトリウムおよび50μg/mlゲンタマイシン硫酸塩を含むTCM-99が含まれる。上述の何れも顆粒層細胞、卵管細胞、BRL細胞および子宮細胞およびSTO細胞のような種々の細胞タイプとの共培養に関わることもある。
CR1は、ヘミカルシウムL-ラクテートを、1.0mM〜10mM、好ましくは1.0mM〜5.0mMの範囲の量で含む。ヘミカルシウムL-ラクテートは、L-ラクテートに組み込まれたヘミカルシウム塩を含むL-ラクテートである。ヘミカルシウムL-ラクテートは、単一の成分が培地内で以下のような二つの主な要件を満たす点で重要である。即ち、(i) 該カルシウムは、コンパクションおよび細胞骨格配列に関して必要とされるカルシウム要求、および(ii)代謝並びに電子輸送のために必要な、ラクテート要求を満たす。ヘミカルシウムL-ラクテートは、また胚の生存性にとって必要な、該培地に対する、有益なミネラルおよびエネルギー源としても機能する。
有利には、CR1培地はウシ胎仔血清などの血清を含まず、しかも動物細胞の同時培養または他の生物学的メディウム、即ち卵管細胞などの動物細胞を含む培地の使用を必要としない。生物学的なメディウムは、これらが該胚にとって有害である可能性のある、かつ検出、特徴付けおよび除去が困難な、微生物または痕跡量の因子を含む点において、しばしば問題を生ずる可能性がある。

例えば、該活性化NTユニットを、2.0mMのDMAP(Sigma)を含有するCR1aa培地に移し、かつ周囲条件下で、例えば約38.5℃、5%CO2 にて、適当な時間、例えば約4〜5時間培養する。
その後、該培養したNTユニットを、好ましくは洗浄し、次いで好ましくは適切なコンフルエントなフィーダー層を含む、適当な培地、例えば10%FCSおよび6mg/mlを含有するCR1aa培地の入ったウエルプレートに配置する。適当なフィーダー層の例は、繊維芽細胞および上皮細胞、例えば有蹄類由来の繊維芽細胞および子宮上皮細胞、家禽繊維芽細胞、ネズミ(例えば、マウスまたはラット)繊維芽細胞、STOおよびSI-m220フィーダー細胞株、およびBRL細胞を包含する。
該NTユニットは、雌レシピエントに移植するのに適した、またはCICM細胞または細胞コロニーを得るために使用できる細胞を得るのに適したサイズに達するまで、該フィーダー層上で培養される。好ましくは、これらのNTユニットは、少なくとも約2〜400細胞、より好ましくは約4〜128細胞、および最も好ましくは少なくとも約50細胞となるまで、培養されるであろう。この培養は、適当な条件、即ち約38.5℃および5%のCO 2の存在下で行われ、成長を最適化するために、典型的には約2〜5日毎に、好ましくは約3日毎に、該培地を交換する。
哺乳動物細胞由来の所定の遺伝子を挿入、除去または変更するための、任意の公知の方法を使用して、該核ドナーとして使用する該分化細胞を変更することができる。これらの手順では、遺伝子の全部または一部を除去することができ、また該遺伝子は異種由来のものであり得る。これらは、同種組み換え技術を含み、該技術は1以上の遺伝子の、特異的サイトまたは該細胞ゲノムのサイトにおける、挿入、欠失または変更を可能とする。
所定のサイズのNTユニットを得た後に、CICM細胞および細胞株を得るためには、該細胞を該領域から機械的に取り出し、次いでこれを使用する。これは、好ましくは該NTユニットを含み、典型的には少なくとも約50個の細胞を含む細胞塊を取り出し、該細胞を洗浄し、該細胞をフィーダー層、例えば放射線照射された繊維芽細胞上に置くことにより実施する。典型的には、幹細胞または細胞コロニーを得るために使用する細胞は、培養されたNTユニットの最も内側の部分から得られ、該単位のサイズは、好ましくは少なくとも50個の細胞を含む。しかしながら、より少いまたはより多くの細胞数をもつNTユニット並びに該NTユニットの他の部分を由来とする細胞を使用して、ES細胞および細胞コロニーを得ることもできる。該細胞は、適当な増殖培地中で、例えば10%FCS、0.1mMのβ-メルカプトエタノール(Sigma)およびL-グルタミン酸を補充したα MEM中で、該フィーダー層内に維持される。この増殖培地を、必要な頻度で交換して、例えば約2〜3日毎に交換して、生育を最適化する。
これらの得られたCICM細胞および細胞株、好ましくはヒトCICM細胞および細胞株は、多数の治療並びに診断用途を有する。特に、このようなCICM細胞は、細胞移植治療の目的で使用することができる。ヒトCICM細胞は、多数の疾患状態の治療における用途を有している。ヒトNTユニット自体を、疾患状態の治療に使用することもできる。
この点に関連して、マウス胚幹(ES)細胞が、殆ど全ての細胞型、例えば造血幹細胞に分化できることは公知である。従って、本発明に従って作成されたヒトCICM細胞は、同様な分化能力を持つはずである。本発明による該CICM細胞は、公知の方法にしたがって、所定の細胞型を与えるように、分化すべく誘導される。例えば、本発明のヒトCICM細胞を、分化用培地内で、細胞分化を生ずる条件下で誘発して、造血幹細胞、筋肉細胞、心筋細胞、肝細胞、軟骨細胞、上皮細胞、尿管細胞等に分化させることができる。該CICM細胞の分化をもたらす培地および方法は、適当な培養条件であるとして、当分野で公知である。
例えば、Palacios等, Proc. Natl. Acad. Sci., USA, 1995, 92:7530-7537は、幹細胞を、誘導法にかけることにより、胚細胞株から造血幹細胞を製造することを教示している。該誘導法は、まずこのような細胞の集合体を、レチノイン酸を含まない懸濁培地で培養し、次いでレチノイン酸を含む同一の培地で培養し、次に細胞の付着を生ずる基質に細胞集合体を移す工程を含む。
更に、Bain等, Dev. Biol., 1995, 168:342-357は、胚幹細胞のインビトロ分化による、神経特性を持つ神経細胞の製造を教示している。これらの参考文献は、胚または幹細胞から分化細胞を得るための報告された方法の例である。これら文献および特に胚幹細胞を分化させる方法に関連する、該文献の開示事項は、その全体が本明細書に含まれるものとする。
かくして、公知の方法および培地を使用して、当業者は、遺伝子操作されたまたはトランスジェニックCICM細胞を包含する、本発明のCICM細胞を培養することができ、該培養により所定の分化細胞型、例えば神経細胞、筋肉細胞、造血細胞等を得ることができる。
本発明のCICM細胞は、任意の所定の分化細胞型を得るために使用することができる。このような分化したヒト細胞の治療における使用は、未曾有のものである。例えば、ヒト造血幹細胞は、骨髄移植を必要とする医療において使用できる。このような手順を利用して、多くの疾患、例えば卵巣癌および白血病等の、末期段階の癌並びにAIDS等の免疫系を弱める疾患等を治療する。造血幹細胞は、例えば癌またはAIDS患者の成人体細胞、例えば上皮細胞またはリンパ細胞と、除核卵母細胞と融合して、上記のようなCICM細胞を得、このような細胞を、造血幹細胞が得られるまで、分化にとって好ましい条件下で培養することにより得ることができる。このような造血細胞は、癌およびAIDSを包含する諸疾患の治療において使用することができる。
本発明の大きな利点は、本発明が移植に適したアイソジェニックまたはシンジェニックヒト細胞の、本質的に無限の供給源を提供することにある。従って、本発明は従来の移植法に係わる重大な問題、即ち宿主対移植片または移植片対宿主拒絶反応のために起こるものと考えられる、該移植組織の拒絶反応の問題を解消する。従来は、拒絶反応を、抗-拒絶反応剤、例えばシクロスポリン等の投与によって防止または軽減していた。しかしながら、このような薬物は、重大な副作用、例えば免疫抑制、発癌特性を有し、かつ極めて高価である。本発明は、抗-拒絶反応剤の使用を排除もしくは少なくとも大幅に軽減するはずである。
この方法は、欠陥のある遺伝子、例えば欠陥のある免疫系の遺伝子、嚢胞性繊維症遺伝子を置換するために、または治療上有用な蛋白質、例えば成長因子、リンホカイン、サイトカイン、酵素等の発現をもたらす遺伝子の導入のために使用することができる。例えば、脳を由来とする成長因子をコードする遺伝子を、ヒトCICM細胞、神経細胞に分化した細胞およびパーキンソン病患者に移植される細胞に導入して、このような疾患中の神経細胞の喪失を遅らせることができる。
このex vivo療法は、移植の32日後に45%まで、該ラットにおけるパーキンソン病-様の症状を軽減した。また、チロシンヒドロキシラーゼ遺伝子は、星状細胞中に配置され同様な結果を与えた(Lundberg等, Develop. Neurol., 1996, 139:39-53およびそこで引用されている参考文献)。
しかしながら、このようなex vivo系は、問題がある。特に、通常使用されているレトロウイルスベクターは、in vivoではダウンレギュレーションされ(down-regulated)、また該トランスジーンは一過性にのみ発現されるに過ぎない(MulliganによりScience, 1993, 260:926-932において概説されている)。また、このような研究は、一次細胞、即ち星状細胞を使用しており、該細胞は有限の寿命を有し、かつ緩慢に複製される。このような特性は、トランスフェクション速度に悪影響を与え、また安定にトランスフェクションされた細胞の選別を妨害する。その上、相同組み換え技術で使用すべき一次細胞を標的とする遺伝子を大量に増殖することは、殆ど不可能である。これとは対照的に、レトロウイルス系に関連する上記難点は、哺乳動物のCICM細胞の使用により排除されるはずである。
課題とするCICM細胞に導入されることがある遺伝子は、例えば表皮成長因子、塩基性繊維芽細胞成長因子、グリア由来の神経栄養成長因子、インシュリン-様成長因子(IおよびII)、ニューロトロフィン-3、ニューロトロフィン-4/5、毛様体神経栄養因子、AFT-1、サイトカイン遺伝子(インターロイキン、インターフェロン、コロニー刺激因子、腫瘍壊死因子(αおよびβ等)、治療酵素をコードする遺伝子等を包含する。
また、本発明のCICM細胞、好ましくはヒト細胞を、特に初期発生段階の調節に関与する遺伝子を研究するための、分化のin vitroモデルとして使用できる。また、本発明のCICM細胞を使用して、分化された細胞、組織および器官は、薬剤の研究において使用できる。
更に、本発明のCICM細胞は、他のCICM細胞および細胞コロニーを製造するための、核ドナーとして使用することも可能である。
本発明をより一層明確に説明するために、以下の実施例を与える。
ウシおよびブタの繊維芽細胞の一次培養物は、胎仔(ウシについては妊娠の45日目およびブタ胎仔については、妊娠の35日目)から得た。頭部、肝臓、心臓および消化管を、無菌的に取り出し、該胎仔器官をミンチ化し、30分間37℃にて、予備的に加温したトリプシンEDTA溶液(0.05%のトリプシン/0.02%のEDTA;NY州グランドアイランドのGIBCO社)中で、インキュベートした。繊維芽細胞を、組織培養皿に置き、10%のウシ胎仔血清(FCS)(UT州ローゲンのHyclone社)、ペニシリン(100 IU/ml)およびストレプトマイシン(50μl/ml)を補充したα-MEM培地(MD州ウォーカーズビルのBioWhittaker社)内で培養した。この繊維芽細胞は、5%CO2 を含む湿った大気雰囲気内で、37℃にて生育させかつ維持した。
以下のエレクトロポレーション手順は、胚(ウシおよびブタ)並びに成熟動物(ウシ)の繊維芽細胞両者について実施した。標準的なマイクロインジェクション手順をも、繊維芽細胞に異種DNAを導入するために使用したが、本例においては、より簡単な手順であることから、エレクトロポレーションを使用した。
増殖中の繊維芽細胞を含む培養プレートを、該細胞が単一細胞懸濁液となるまで、トリプシンEDTA溶液(0.05%のトリプシン/0.02%のEDTA;NY州グランドアイランドのGIBCO社)中で、インキュベートした。該細胞を、500×gにて遠沈し、5x106細胞/mlにてリン酸緩衝生理食塩水(PBS)で再懸濁した。
エレクトロポレーションの翌日に、付着した細胞を、該レポーター遺伝子の安定な組み込みについて選別した。G418(400μg/ml)を、15日に渡り成長培地に添加した(範囲:該培養した細胞の寿命終点まで3日)。この薬物は、該β-GEO遺伝子を持たないあらゆる細胞を殺した。というのは、該細胞が該neo耐性遺伝子を発現しないからである。この時間の終了時点において、安定なトランスジェニック細胞のコロニーが存在していた。各コロニーを、夫々独立に増殖させた。トランスジェニック繊維芽細胞をX-galで染色してβ-ガラクトシダーゼの発現を観察し、かつ該β-GEO遺伝子のPCR増幅を利用し、アガロースゲル上で泳動し、組み込みについて陽性であることを確認した。
ウシ胚の繊維芽細胞の一コロニーを由来とする細胞の一細胞株(CL-1)を、核移植(NT)手順におけるドナー核として使用した。一般的なNT手順は、上に記載した。
とさつ場で得た卵母細胞を、in vitroで成熟させた。該卵母細胞から卵丘細胞を剥ぎ取り、成熟後の経過時間(hpm)約18〜20時間の時点で、面取りしたマイクロピペットで除核した。除核は、TL-HEPES培地+Hoechst 33342 (3μg/ml; Sigma) 中で確認した。次に、個々のドナー細胞(繊維芽細胞)を、レシピエント卵母細胞の囲卵腔(perivitelline space)に配置した。該ウシ卵母細胞の細胞質および該ドナー核(NTユニット)を、電気融合技術を利用して、一緒に融合した。500μmギャップのチャンバーにおける、15マイクロ秒間の120Vからなる一融合パルスを、該NTユニットに印加した。これは24hpmにて生じた。該NTユニットを、CR1aa培地に、26〜27hpmまで置いた。
以下の表にこれら実験の結果をまとめた。
トランスジェニックNT胚由来のCICM細胞株(除核した卵母細胞に移植されたCL-1細胞)を使用して、キメラ胚および胎仔を生成した。トランスジェニックCICM細胞のコロニーを、機械的離解法と組み合わせた、1-5 mg/mlのプロナーゼまたは0.05%のトリプシン/EDTAを使用して離解し、5またはそれ以下の細胞からなる細胞塊を製造した。トリプシンまたはプロナーゼ活性は、該細胞を30〜100%のウシ胎仔血清を含む多数の洗浄液に通すことによって不活性化した。この離解された細胞を、TL-HEPES培地を含む、マイクロマニュビレーション用のプレートに入れた。受精した胚をも、これらのプレートに入れ、かつマイクロマニュピレーション装置を使用してキメラ胚を生成した。8〜10個のトランスジェニックCICM細胞を、8〜16細胞段階の受精胚に注入した。これらの胚を胚盤胞段階までin vitroで培養し、次いでレシピエント動物中に移植した。
同一のトランスジェニックCICM細胞株を使用して、NT胚を生成した。実施例1に記載した該NT手順を利用した。但し、該除核された卵母細胞と融合される該ドナー細胞として、繊維芽細胞の代わりにCICM細胞を使用した。トランスジェニックCICM細胞のコロニーを、機械的離解法と組み合わせた、1-5 mg/mlのプロナーゼまたは0.05%のトリプシン/EDTAを使用して離解し、5またはそれ以下の細胞からなる細胞塊を製造した。トリプシンまたはプロナーゼ活性は、該細胞を除核卵母細胞に移植する前に、該細胞を30〜100%のウシ胎仔血清を含む多数の洗浄液に通すことによって不活性化した。結果を表1に示した(第3群)。5つの胚盤胞期の胚が作製された。
Claims (21)
- (i)望みの分化哺乳動物細胞または分化哺乳動物細胞核を、前記分化細胞または細胞核と同じ種の除核哺乳動物卵母細胞へ、核移植(NT)ユニットの形成に適した条件下で挿入する工程;
(ii)得られた核移植ユニットを活性化する工程;
(iii)前記活性化核移植ユニットを2細胞発生期を越えるまで培養する工程;および、
(iv)前記培養NTユニットから得られる細胞を培養してCICM細胞株を得る工程、
を含む、CICM細胞株を作製する方法。 - 請求項1記載の方法によって作製されるCICM細胞株。
- 望みのDNAが分化哺乳動物細胞または分化哺乳動物細胞核において挿入、除去または改変され、それにより遺伝的に改変されたNTユニットが作られる、請求項1記載の方法。
