【解決手段】ヒト前立腺特異的膜抗原の特定な配列のポリペプチド、特定な配列のアミノ酸45〜750からなるヒト前立腺特異的膜抗原の外膜領域配列のポリペプチド、または特定な配列のアミノ酸45〜750からなるヒト前立腺特異的膜抗原の外膜領域配列における抗原性領域のポリペプチドであって、前記ポリペプチドは、前記前立腺特異的膜抗原に対して特異的な抗体を産生するために使用することができるポリペプチド。

「遺伝子」は核酸分子を意味し、該遺伝子の配列には、特定のタンパクを正常に調節された状態で製造するために必要な全ての情報(構造的コーディング配列、プロモータおよびエンハンサを含む)が含まれる。
<材料および方法>
遺伝子をクローニングするための手法には、免疫沈降による抗原を大量に精製することと、その後のポリメラーゼ連鎖反応に用いる縮退オリゴヌクレオチドプライマーの合成に使用するための幾つかの内部ペプチドのマイクロシークエンシング(microsequencing) が含まれていた(19,20)。部分的cDNAはPCR生成物として増幅され、このPCR生成物は、LNCap(前立腺のリンパ節癌腫)(Lymph Node Carcinoma of Prostate)細胞系cDNAプラスミドライブラリー(8)から完全な長さのcDNA分子をクローン化するための相同性プローブとして用いられた。PSM抗原の抗原決定基はPSMのグリコシル化部位であるために、CYT−356抗体(9)は細菌中で生産された抗原を検出できないことが初期の実験により明らかになった。このことによって、我々は一層困難で、かつ手の込んだ手法を要求された。
低張溶解に続いて、ショ糖密度勾配上での遠心分離にを行なうことより、膜蛋白質が細胞から単離された(21)。10〜20μg のLNCap,DU−145及びPC−3膜タンパクは4%スタッキングゲル(stacking gel)を用いた10% SDS-PAGE分離用ゲル中を16-18 時間、9-10ミリアンペアで電気泳動された。タンパクは、転写緩衝液(transfer buffer) [48mM Tris base, 39mM グリシン, 20% メタノール]中において、25ボルト及び4 ℃で一晩、PVDF膜(ミリポアR 社 (MilliporeR Corp.))上に電気ブロットされた。膜はTSB[0.15M NaCl, 0.01M Tris base, 5% BSA ]中において室温で30分間ブロックされ、引き続いて 10-15μg/mlのCYT−356モノクローナル抗体(サイトーゲン社)と共に2時間インキュベートされた。次いで、膜は10-15 μg/mlのウサギ抗マウス免疫グロブリン(アキュレート科学社(Accurate Scientific) )と共に室温で1 時間インキュベートされた後、1x106 cpm/mlの 125I- タンパクA(アマーシャムR (AmershamR) )と共に室温でインキュベートされた。その後、膜は洗浄され、-70 ℃で12-24 時間オートラジオグラフィーにかけられた(図1)。
アビジン- ビオチン法(avidin-biotin method)の免疫組織化学的検定を用いて、ヒト組織切片と細胞系の両方についてPSM抗原の発現を分析した(22)。クライオスタットで切断された(Cryostat-cut)前立腺組織切片(厚さ 4-6μm )を、メタノール/アセトン中で10分間固定化した。50,000細胞/100μl/スライドを用いて、スライドグラス上で細胞サイトスピン(cytospins) が作製された。全ての内因性ペルオキシダーゼ活性を除去するために、試料はPBS中1%過酸化水素で 10-15分間処理された。組織切片はPBS中で数回洗浄された後、適切な抑制血清(suppressor serum)と共に20分間インキュベートされた。抑制血清(suppressor serum)を排出し、次いで切片または細胞を、希釈されたCYT−356モノクローナル抗体と共に1時間インキュベートした。その後、試料はPBSで洗浄され、続いて二次抗体(1:200 希釈のウマもしくはヤギ免疫グロブリン、30分間)と共に、またアビジン- ビオチン複合体(1:25希釈で30分間)と共に連続的にインキュベートされた。DABが発色原として用いられ、ヘマトキシリン対比染色および顕微鏡板へのマウンティング(mounting)が引き続いて行われた。前立腺試料の凍結切片および2連の細胞サイトスピン(duplicate cell cytospins)が各実験についての対照として用いられた。陽性対照として、抗サイトケラチンモノクローナル抗体CAM 5.2が、上記で述べたのと同様の手法に従って用いられた。少なくとも細胞の5%が免疫活性を示せば、我々は組織切片がPSM抗原を発現していると見なした。我々の点数評価体系は以下の通りである: 陽性細胞が5%未満(<5%)=1; 陽性細胞が5〜19%=2; 陽性細胞が20〜75%=3; 陽性細胞が75%超(>75%)=4。3〜5のハイパワー光学顕微鏡の視野(400 x) において、陽性細胞と陰性細胞を評価し、100-500 個の細胞のうちの陽性細胞の割合を記録することによって、同質であるか異質であるかが説明された。免疫染色の強度は1+ から4+ のスケールで等級付けされた。正の対照と比較すると、1+ は穏和な染色を示し、2〜3+ は中程度の染色を示し、4+ は強い染色を示す。
100 mmペトリ皿内の集密度80%のLNCaP細胞を、メチオニンを含まないRPMI培地中で2時間、メチオニン欠乏の状態で生育させ、その後 100μCi/ml の35Sメチオニンを添加して、細胞を更に16-18 時間生育させた。その後、細胞を洗浄し、1ml 溶菌緩衝液[1%トライトンX-100, 50mM ヘペス pH 7.5, 10%グリセロール, 150mM MgCl2, 1mM PMSF 及び1mM EGTA]を添加して、4℃で20分間インキュベートすることによって溶菌された。この溶解物は、4℃で90分間、パンソルビンR (PansorbinR ) 細胞(カルバイオケムR (CalbiochemR ) )と共に混合することによってプレクリアーされた(pre-cleared) 。次いで、この細胞溶解物は、予めCYT−356抗体(サイトゲン社(Cytogen Corp.) )およびRAM抗体(アキュレート科学(Accurate Scinetific) )に結合されたタンパクAセファロースR CL−4Bビーズ(ファルマシアR (PharmaciaR ) )と共に、4℃で3〜4時間混合された。ペトリ皿あたり、ビーズ 3mgに対して12μg の抗体が用いられた。その後、このビーズはHNTG緩衝液[20mMヘペス pH 7.5, 150mM NaCl, 0.1% トライトンX-100, 10%グリセロール及び2mM オルトバナジン酸ナトリウム]で洗浄され、β- メルカプトエタノールを含む試料ローディング緩衝液中に再懸濁され、95℃で5-10分間変性され、4%スタッキングゲル(stacking gel)を用いた10% SDS-PAGEゲル上において10ミリアンペアで一晩電気泳動された。このゲルをクマシーブルー(Coomassie Blue)で染色し、酢酸/メタノールを用いて脱色し、真空乾燥器内で60℃で乾燥(dried down)した。その後、ゲルは70℃で16-24 時間、オートラジオグラィーにかけられた(図2A−D)。
免疫沈降に関して上に示された手法を、約6×107 個のLNCaP細胞を含む集密状態の8個のペトリ皿を用いて繰返した。免疫沈降産物はプールされ、10% SDS PAGEゲル上の2つのレーンにロードされ、9-10ミリアンペアで16時間電気泳動された。4℃において75ボルトで2時間、転写緩衝液中でタンパクをニトロセルロース BA-65膜(シュレイチャー及びシュエルR (Schleicher and SchuellR ) )上に電気ブロットした。タンパクを視覚化するために、膜をポンソーレッド(ponceau Red) で染色し、100KD のタンパクのバンドを切り出し、可溶化し、トリプシンでタンパク分解的に消化した。その後、消化された試料についてアプライドバイオシステム171C型(Applied Biosystems Model 171C) を用いてHPLCを行ない、主要なペプチドの明確なピークを選択し、改良ポストリキッド・アプライドバイオシステム477A型タンパク/ペプチド・マイクロシークエンサー(modified post liquid Applied Biosystems Model 477A Protein/Peptide Microsequencer) 上での改良エドマン分解により配列決定を行なった(23)。この書類には、全てのペプチドに関する配列決定データが含まれている。我々は、免疫沈降による抗原の精製およびPVDF膜(ミリポアR (MilliporeR ) )への電気ブロッティングによる転写を含む同様の方法により、PSM抗原のアミノ末端の配列決定を試みた。タンパクは、アプライドバイオシステム477A型のタンパク/ペプチド・シークエンサー(Applied Biosystems Model 477A Protein/Peptide sequencer) で分析され、アミノ末端がブロックされていることが判明した。従って、この技術では配列決定データは得られなかった。

PCR生成物を示す代表的な写真が図5に示されている。
これらPCR生成物をさらに解析するために、これら生成物は「TAクローニング」(インビトロゲンコーポレーションR (InvitrogenR Corp.))を用いて、適切なプラスミドベクター中にクローニングされた。ここで採用されたクローニングの戦略は、突出したR残基を挿入部位に有するプラスミドベクターに、PCR生成物を直接連結させるというものである。これは、TaqポリメラーゼがPCR生成物末端の突出したA残基を残したままにするという事実を活用している。このライゲーション混合物を、コンピテント大腸菌細胞(competent E. coli cells) に形質転換し、得られたクローンを増殖して、アルカリ溶菌法(24)によりプラスミドDNAを単離し、制限(酵素消化)分析によりスクリーニングした(図6A−B)。
PCR生成物のTAクローンは、その後、シーケナーゼ(Sequenase) (U.S.バイオケミカル(U.S. Biochemical))を用い、ジデオキシ法(25)により配列決定された。各プラスミドDNA 3-4μg を水酸化ナトリウムを用いて変性し、エタノール沈殿させた。製造者の推奨に従い、35S−ATPを用いて標識反応が行なわれ、また該反応は同様の手順に従って停止された。次いで、配列決定用生成物(sequencing products) は、6%ポリアクリルアミドゲル/7M尿素ゲル上において、IBI配列決定装置を用いて分析された。ゲルは120 ワットで2時間泳動された。電気泳動に続いて、ゲルは10% メタノール/10% 酢酸中で15-20 分間固定化され、ワットマン 3MM紙(Whatman 3MM paper) 上に移され、バイオラドR 真空乾燥器(BioradR Vacuum dryer)内において、80℃で2時間乾燥された(dried down)。その後、ゲルは室温で16-24 時間オートラジオグラィーにかけられた。PCR生成物が正しいクローンであるか否かを決定するために、我々は、得られたDNA分子の 5'-及び 3'-末端で得られた配列について分析し、正しいプライマー配列を探索すると共に、プライマーの設計に用いられていないペプチドの一部に対応する隣接配列(adjacent sequences)を分析した。

ここで下線を付したアミノ酸配列は、プライマーNを作成するときに用いたペプチド7の一部を示す。このプライマーよりも上流の全てのアミノ酸は、IN-20 クローン中で正しいもの(配列)であり、ペプチド7内に見いだされるアミノ酸と一致している。更なるDNA配列決定によって、陽性クローンのDNA配列内に我々の他のPSMペプチドが存在することを確認することができた。
完全な長さのPSM・cDNAのクローニング
LNCap・mRNA由来のcDNAライブラリーが、スーパースクリプトR プラスミド系(SuperscriptR plasmid system) (BRLRギブコ(BRLR Gibco))を用いて構築された。このライブラリーは、コンピテントDH5-α細胞 (competent DH5-α cells) を用いて形質転換され、100 μg/mlのカルベニシリン(Carbnicilin) を補充したLB培地の入っている100 mmペトリ皿上にプレーティングされた。該プレートを37℃で一晩増殖処理し、次いでクローンをニトロセルロースフィルター上に移した。このフィルターは、ランダムプライミング(27)により32PdCTPで放射性ラベルした我々の1.1 Kbの部分的cDNA相同性プローブを用い、グルンシュタインとホグネス(Grunstein and Hogness) (26)に従って処理され、スクリーニングされた。我々は8個の陽性クローンを得た。これらのクローンは、DNA制限酵素消化分析および配列決定分析において、PSM抗原をコードする完全な長さのcDNA分子を表わすことが証明された。図7で示されているのは、我々のライブラリーに表れたcDNA分子の大きさを示すオートラジオグラフィーであり、図8では完全な長さのクローンの幾つかについての制限酵素消化分析が示されている。図9は、図8の試料についてのプラスミドサザン分析であり、試料が1.1 Kbの部分的cDNAプローブに完全にハイブリダイズすることが示されている。
PSM遺伝子のノーザン分析(28)によって、その発現は前立腺と前立腺の癌腫に制限されていることが明らかになった。
PSM遺伝子発現を検定するために、15個のヒト前立腺試料についてPCRが行われた。正常な前立腺組織、良性の前立腺過形成組織および前立腺癌組織に由来する各5つの試料が用いられた(組織学的構造はMSKCC病理学科によって確かめられた)。
100 KDのPSM抗原をコードする遺伝子は既に同定されている。完全なcDNA配列は配列ID番号1に示されている。その核酸配列の下には、予想される翻訳アミノ酸配列が記載されている。アミノ酸の総数は 750である(配列ID番号2)。予想タンパク配列の親水性は図16に示される。図17に示されているのは、最も親和性が高い部位(point) を持つ次の3つのペプチドである: Asp-Glu-Leu-Lys-Ala-Glu(配列ID番号35); Asn-Glu-Asp-Gly-Asn-Glu(配列ID番号36);及び Lys-Ser-Pro-Asp-Glu-Gly(配列ID番号37)。
Ala-Gly-Ala-Leu-Val-Leu-Aal-Gly-Gly-Phe-Phe-Leu-Leu-Gly-Phe-Leu-Phe (配列ID番号38)。
PSM抗原の可能な用途:
1.腫瘍の検出:
顕微鏡的:
異なった抗原に対するプローブを使用することによって、曖昧さのない腫瘍の判定(designation) を達成することができる。前立腺癌に関しては、PSM抗原プローブが有用であることが証明され得る。従って、PSMは診断を目的として用いることあができるであろうし、これは出願人がクローン化したcDNAコーディング領域に由来するセンスプローブ(対照)およびアンチセンスプローブを用いて、自然位での(in-situ) ハイブリダイゼーションを行うことにより、顕微鏡レベルで達成され得るであろう。このことは、前立腺外への局所的拡大や、リンパ節、骨もしくは他の転位部位の関連性に関する評価に用いることができるであろう。骨転位は前立腺癌における主要な問題となっているので、転位の広がりを早期に検出することは、特にその段階を評価することに関して(for staging) 要望が強い。幾つかの腫瘍において、骨髄内での腫瘍細胞の検出は深刻な予後を予知させるものであり、転位を標的とした介入(interventions) が試みられるべきことを示唆する。骨髄吸引液(bone marrow aspirates) もしくは切片中におけるPSM抗原発現の検出は、そのような早期の情報を提供するかもしれない。PCR増幅もしくは自然位での(in-situ) ハイブリダイゼーションが用いられ得る。これは、転位の可能性がある何れの領域に対しても開発され得るであろう。
抗原に関するcDNAの知見はまた、抗原の特異的アミノ酸配列に対して使用するための抗体を開発する上で、有効な抗原として役立ち得る領域を同定することを可能にする。そのような配列は、PSM抗原の膜外領域もしくは膜内領域といった様々な領域に存在し得る。これら特異的抗体の開発は、抗原の免疫組織化学的同定をもたらすであろう。これらの誘導された抗体は、免疫診断について非常に大きな有用性を有しているから、特に凍結切片、並びにパラフィン固定された切片内で作用する抗体として使用するために開発され得るであろう。
制限酵素で切断された断片の長さの多形性(Restriction fragment length polymorphisms) (RFLPS)は、腫瘍の初期段階および促進段階の間に生じる遺伝的な損傷の経過を記録する上で有効であることが証明されている。これは、RFLP分析は次のことを立証するであろうということなのかもしれない。即ち、PSM配列の制限酵素消化マッピングにおける変化は、前立腺腫瘍の危険性、悪性化の可能性もしくは進行に関する質病素因の証拠を与え得るというてことである。
抗原特異的な抗体の開発に伴って、もし当該抗原または選ばれた抗原断片が血清中に出現したならば、それらは転位による疾病の存在に関する血清マーカーとなり得る。これら抗原もしくは選ばれた抗原断片は、個別的にまたは他の前立腺特異性マーカーとの組み合わせにおいて有用であり得る。
当該cDNA配列によって、当該抗原が外表面(exofacial Surface) 上の大部分のタンパクと同様に膜貫通タンパク (membrane spanning protein) の特性を有することが示唆されるので、抗体、特に当該腫瘍に特異的な露出されたペプチド断片に対するモノクローナル抗体は、前立腺摘出術または放射線照射の後に、転位性腫瘍の局所的拡大や残存腫瘍の画像化を提供し得る。コーディング領域についての知見によってモノクローナル抗体の生成が可能となり、これらは画像化の精度を最大にするために組み合わせて用いられ得る。cDNA分析によると、当該抗原はトランスフェリン受容体との相同性(約54%)を有しているので、この抗原に特異的な正規の(normal)リガンドがあるかもしれず、当該リガンドを同定することによって別の画像化方法が提供され得る。
PSM抗原は、それに結合する正規のリガンドを単離するために用いられ得る。特異性に依存するこれらリガンドは、ターゲッテシングに用いられ得るし、或いはそれらの血清レベルは疾病の状態を予測し得るものであろう。PSMの正規リガンドがキャリア分子であることが判明すれば、PSMは循環系から当該リガンドを除去することを助ける治療的な目的で(鉄キレート物質のように)、当該リガンドを結合するために用いられ得る。もし、当該リガンドが腫瘍の増殖もしくは転位を促進するならば、可溶性PSMを与えることによって、当該リガンドは前立腺への結合から排除されるであろう。PSM抗原の構造に関する知見は、リガンドに結合し且つ同様の目的に用いることができる、より小さい断片の生成に役立ち得る。
a)リガンド: PSM抗原のcDNA構造がトランスフェリン受容体との構造的相同性(核酸レベルで54%)を有しているとの知見は、トランスフェリン様もしくは非トランフフェリン様であり得る受容体に関する内因性リガンドが存在し得ることを示唆している。トランスフェリンは、トランスフェリン受容体に結合した後に、鉄を細胞内へ運搬するリガンドであると考えられている。しかし、アポトランスフェリンは、トランスフェリン受容体を発現する幾つかの細胞の増殖因子であることが報告されている(30)。トランスフェリンはこの抗原に対するリガンドであるのか否か、或いは他の幾つかのリガンドがこのリガンドに結合するのか否かは不明なままである。もし、或るリガンドが同定されれば、それは金属イオン(鉄もしくは亜鉛もしくはその他)のような特定の物質を腫瘍内へ運搬し得、従って毒性の物質(放射活性物質、或いは細胞毒性を有する化学薬品、例えばリシンのような毒物、細胞毒性アルキル化剤、細胞毒性プロドラッグ)を腫瘍に供給する手段として役立ち得る。
PSM抗原には他の用途があるかもしれない。前立腺に亜鉛が豊富であることはよく知られており、もし当該抗原がこの点に関する機能もしくは他の生物学的機能を提供するならば、PSM抗原は、良性肥大および/または前立腺炎のような他の前立腺病態の治療における有用性を提供し得る。
第二実験シリーズ
前立腺特異的膜抗原の発現
出願人は、最近、7E11−C5.3 抗前立腺モノクローナル抗体が認識する前立腺特異的膜抗原PSMをコードする、2.65kbの相補型DNAをクローニングした。PSM発現について、7Ell−C5.3 抗体を使ってLNCaP、DU145 およびPC-3前立腺癌細胞系を免疫組織化学的に分析すると、LNCaP細胞は強く染色されたが、DU145 およびPC-3細胞での発現は検出されなかった。インビトロにおける全長2.65kbのPSM・cDNAのカプリングされた転写/翻訳によって84kDaのタンパクが生産され、これは予測したPSMのポリペプチド分子量に相当した。イヌ膵臓ミクロソームによる該タンパクの翻訳後の修飾によって、予測した100 kDaのPSM抗原が作られた。真核発現ベクターの中の完全長PSM・cDNAでPC−3細胞をトランスフェクトした後に、出願人は、7Ell−C5.3 モノクローナル抗体を使ったウエスタン分析によって、PSM糖タンパクの発現を検出した。リボヌクレアーゼ保護分析によって、PSM・mRNASの発現はヒト組織では殆ど完全に前立腺特異的であることが証明された。PSM発現はホルモン欠乏状態において最も高いようであり、ステロイドによってホルモン的に調節されていて、DHTはヒト前立腺癌細胞系LNCaPにおけるPSM発現を8〜10倍下方調節し、テストステロンはPSM発現を3〜4倍下方調節し、コルチコステロイドは有意な効果を示さなかった。良性の前立腺肥厚においては、PSM発現は異質で且つ時には欠失していることは分っていたが、これに対して正常および悪性の前立腺組織は一貫して高いPSM発現を示す。ヌードマウスに正所移植および皮下移植されたLNCaP腫瘍はPSMを多く発現し、PSM発現の制御および調節を研究するための優れたイン・ビボのモデル系を提供する。
細胞と試薬:
LNCaP、DU−145、PC−3細胞系は、アメリカンタイプカルチャーコレクションから得た。これらの細胞系の確立と性質に関する詳細は、以前に報告されている(5A、7A、8A)。別に明記しないかぎり、LNCaP細胞はL−グルタミン、可欠アミノ酸および5%ウシ胎児血清(ギブコ−BRL、Gaithersburg,MD)を補充したRPMI1640 培地を用いてCO2 インキュベーター中で37℃で培養した。DU-145およびPC-3細胞は、10%ウシ胎児血清を補充した最小必須培地で培養した。全ての細胞培地はMSKCC培地製造部門(Media Preparation Facility)から得た。制限酵素および修飾酵素は、特に明記しない限りギブコ−BRLから購入した。
我々はアビジン−ビオチン検出法を使って、前立腺癌細胞系のPSM抗原の発現を分析した(9A)。スライド当り5×104 細胞/100 μlを使って、スライドグラス上に細胞サイトスピン(cell cytospins)を作成した。スライドグラスは、PBSで2回洗浄した後に適切な抑制血清と共に20分間インキュベートされた。抑制血清を捨て、希釈した7Ell−C5.3 (5g/mL)モノクローナル抗体と共に、細胞を1時間インキュベートした。その後、サンプルをPBSで洗浄し、続いて2次抗体と共に30分間、またアビジン−ビオチン複合体と共に30分間インキュベートした。ジアミノベンジヂンを色素原および発色剤として使用し、続いてヘマトキシリンで対比染色して封入した。二倍量の細胞サイトスピン(cytospin)を夫々の実験の対照として使用した。正のコントロールとして、抗サイトケラチンモノクローナル抗体CAM5.2 を上記の同じ方法に従って使用した。ヒトEJ膀胱癌細胞を負のコントロールとした。
プラスミドpSPORT1(ギブコ−BRL)中に完全長2.65kbのPSM・cDNAを含むプラスミド55Aを、イン・ビトロにおいて、プロメガTNTシステム(プロメガ社、マジソン、WI)を用いて転写した。ウサギ網状赤血球溶解物、メチオニン欠乏アミノ酸混合物、緩衝液および35S−メチオニン(アマシャム)を含む反応混合物内のcDNAに、T7・RMAポリメラーゼを加え、30℃で90分間インキュベートした。得られたタンパクの翻訳後修飾は、イヌ膵臓ミクロソームを反応混合液(プロメガ社、マジソン、WI)に加えることによって達成された。生成したタンパクを10%SDS−PAGEゲルで電気泳動分析し、続いて、該ゲルを増幅オートラジオグラフィーエンハンサー(アマシャム、アーリントンハイツ、IL)を用いて製造者の指示に従って処理し、80℃で真空乾燥機で乾燥した。このゲルを、 -70℃で一晩、ハイパーフィルムMP(アマシャム)を使ってオートラジオグラフにかけた。
完全長のPSM・cDNAを、pREP7真核発現ベクター(インビトロゲン、サンディエゴ、CA.)中にサブクローニングした。プラスミドDNAを、キアゲンマキシ−プレプ(Qiagen maxi-prep)プラスミド単離カラム(Qiagen Inc、Chatsworth、CA.)を用いて、形質転換されたDH5−アルファバクテリア(ギブコ−BRL)から精製した。精製したプラスミドDNA(6〜10g)をオプチメン(Optimem) 培地(ギブコ−BRL) 900ulで希釈し、既に 900lのオプチメン培地で希釈した30ulのリポフェクチン試薬(ギブコ−BRL)と混合した。この混合物を、オプチメン培地中で40-50 %の集密度状態にあるPC−3細胞が入ったT-75 フラスコに加えた。24-36 時間後に細胞をトリプシン処理し、10%ウシ胎児血清および1mg/mLのヒグロマイシンB(カルバイオケム(Calbiochem)、ラジョラ、CA.)を補充したRPME1640培地の入った100 mm皿に分けた。ヒグロマイシンBの用量は、経過時間/用量応答細胞毒性試験で前もって決定した。細胞は、この培地で2−3週間、培地とヒグロマイシンBを4-5 日ごとに交換しながら、分離性のコロニーが出現するまで培養した。コロニーを6mmのクローニングシリンダーを使って単離し、同じ培地で増殖させた。また、対照として、pREP7プラスミド単独でPC−3細胞をトランスフェクトした。トランスフェクトした細胞からRNAを単離し、RNアーゼ保護分析(後述)およびノザン分析の両方によってPSM・mRNA発現を検出した。
LNCaP、PC−3、およびPSMでトランスフェクトしたPC−3細胞から、前述したようにして粗タンパク溶解物を単離した(10A)。LNCaP細胞膜もまた、刊行物に発表された方法によって単離した(10A)。タンパク濃度は、ブラッドフォード法により、バイオラッドタンパク試薬キット(バイオラッド、リッチモンド、CA.)を用いて定量した。変性に続き、20gのタンパクを10%SDS−PAGEゲルを用いて25mAで4時間電気泳動させた。ゲルを4℃で一晩、イモビロン(Immmobilon)P膜(ミリポア、ベッドフォード、MA.)上に電気ブロットした。膜を5%BSAを加えた0.15M NaCl/0.01M Tris−HCl(TS)に入れ、7Ell−C5.3 モノクローナル抗体(10g/mL)と共に1時間インキュベートした。0.15M NaCl/0.01M Tris−HCl/0.05%トリトン−X 100(TS−X)でブロットを4回洗浄し、10g/mL濃度のウサギ抗マウスIgG(アキュレイトサイエンティフィック(Accurate Scientific) 、ウエストベリー,N.Y.)と共に1時間インキュベートした。
LNCaP腫瘍増殖長
0.25%トリプシンおよび0.02%EDTA溶液に一分間晒して、半集密状態の培養体からLNCaP細胞を回収した。細胞を5%ウシ胎児血清を含むRPMI1640培地中に再懸濁し、マトリゲル(Matrigel)(コラボラティブバイオメディカルプロダクツ(Collaborative Biomedical Products) ,ベッドフォード、MA.)またはカルシウム及びマグネシウムを含まないハンクス塩溶液(HBSS)中で洗浄し、希釈した。トリパンブルー排除試験により90%超の生存率を有する一つの細胞懸濁液だけを、イン・ビボでの注射に使用した。4〜6週令の雄の無胸腺症スイス(nu/nu)ヌードマウスを、メモリアルスローンケッタリング(Memorial Sloan-Kettering)癌センター動物施設から得た。皮下への腫瘍細胞の注射については、0.2 mLのマトリゲル中に再懸濁させた100 万個のLNCaP細胞を、28ゲージの針を備えた使い捨て注射器を用いて、各マウスの後ろ足に注射した。正所注射において、先ずペントバルビタールの腹腔内注射によりマウスを麻酔し、仰向けに寝かせた。腹をベタジンで消毒し、中線切開によって前立腺を露出させた。 0.1mL中の250万個のLNCaP腫瘍細胞を、28ゲージの針を備えた1mLの使い捨注射器を使って、一方の後葉に直接注射した。マトリゲル(Matrigel)と一緒に、またはマトリゲルを伴なわずに、LNCaP細胞が注射された。腹はオートクリップ(Autoclip)傷クリップ(クレイアダムス(Clay Adams),Parsippany,N.J.)を使って単一の層で閉じた。腫瘍は6−8週間で回収し、メモリアルスローンケッタリングガンセンター病理学部門の教授が組織学的に確認し、後のRNA単離のために液体窒素で凍結した。
RNAゾルB(チャイナ/バイオテク(Cinna/Biotecx) 、ヒューストン、TX.)の使用と共に、標準的な方法(11,12)によって、細胞および組織から全細胞性RNAを単離した。RNAの濃度および質は、ベックマンDU 640分光光度計でのUV分光と、ゲル分析とによって評価された。ヒト組織の全RNA試料は、カリフォルニア州パソアルト所在のクロテックラボラトリーズ社(Clotech Laboratories、Inc.,Paso Alto,CA)から購入した。
プラスミドベクターpSPORT1(ギブコ−BRL)中にPSM・cDNAの一部をサブクローニングし、隣接するT7及びSP6・RNAポリメラーゼプロモーターに対するcDNA挿入の方向を、制限分析によって証明した。挿入したPSMの上流で該プラスミドを線状化し、続いてSP6・RNAポリメラーゼで転写することにより、400 ヌクレオチドのアンチセンスRNAプローブが生産されるが、このうち 350ヌクレオチドはPSM・RNAによってRNアーゼによる分解から保護されるに違いない。該プローブは図20で使用された。プラスミドIN−20は、プラスミドpCRII(インビトロゲン)内に1kbのPSM・cDNAの一部を含んでおり、リボプローブ合成にも使用された。XmnI (ギブコ−BRL)によって線状化されたIN−20は、SP6・RNAポリメラーゼを使って転写されて 298ヌクレオチドのアンチセンスRNAプローブを生成したが、このうちの 260ヌクレオチドは、PSM・mRNAによってRNアーゼによる分解から保護されるに違いない。このプローブは、図21および図22で使用された。プローブは、SP6・RNAポリメラーゼ(ギブコ−BRL)、rNTP(ギブコ−BRL)、RNAsin(プロメガ)および32P−rCTP(NEN、ウイルミングトン、DE.)を用い、刊行物に発表されたプロトコル(13)に従って合成された。プローブをNENSORB 20精製カラム(NEN)で精製し、およそ100 万cpmの精製放射性アイソトープで標識したPSMプローブを夫々10gのRNAと混合し、45℃で一晩、RPAIIキット(アンビオン(Ambion)、オ−スチン、TX.)の緩衝液および試薬を使ってハイブリダイゼーションを行なった。試料は製造者の指示通りに処理し、5%ポリアクリルアミド/7M尿素変性ゲル上で Seq ACRYL試薬(ISS、Natick、MA.)を使って分析した。ゲルは予め55℃に加熱し、25ワットで約1〜2時間稼働した。その後,ゲルを10%メタノール/10%酢酸で30分間固定し、80℃のバイオラッド真空乾燥機内においてワットマン3MN紙上で乾燥し、ハイパーフィルムMP(アマシャム)を用いて一晩オートラジオグラフにかけた。走査レーザデンシトメータ(LKB、ピスキャタウエイ,NJ.)を使用して、PSM発現の定
量測定を行なった。
LNCaP細胞(200 万)を、5%ウシ胎児血清を補充したRPMI1640培地を充填したT-75 フラスコ内に撒き、約30-40 %の集密状態になるまで24時間培養した。その後、フラスコをリン酸緩衝生理食塩水で数回洗浄し、5%木炭抽出血清を補充したRPMI培地を加えた。次いで、細胞を更に24時間培養し、その際にジヒドロテストステロン、テストステロン、エストラジオール、プロゲステロン、デキサメタゾン(ステラロイド社(Steraloids Inc.) 、ウイルトン、NH.)を最終濃度2nMで加えた。細胞を更に24時間培養し、次いで前述したようにしてRNAを回収し、リボヌクレアーゼ保護分析で分析した。
PSMの免疫組織化学的検出
7Ell−C5.3 抗PSMモノクローナル抗体を使用することにより、LNCaP前立腺癌細胞系ではPSMの発現が明瞭に検出されたが、PC−3とDU−145細胞系では検出されなかった(図17)。これは、以前に刊行物に発表された結果と一致している(4A)。分析した正常および悪性の全ての前立腺組織はPSM発現について正に染色していた(未公開データ)。
図18で示したように、完全長2.65kbのPSM・cDNAのインビトロのカップリングされた転写/翻訳によって、84kDaのタンパク種が生産されたが、これはPSMのオープンリーディングフレームの 750アミノ酸から予測したタンパクと一致していた。続いて、イヌ膵臓ミクロソームを用いた翻訳後修飾によって、成熟した未変性のPSM抗原と一致する 100kDaのグリコシル化タンパク種が得られた。
PC−3細胞におけるPSM抗原の検出
pREP7発現ベクター中の完全長PSM・cDNAでトランスフェクトされたPC-3細胞を、PSM・mRNAの発現についてノーザン分析により分析した(データは示していない)。PSM・mRNAを高く発現しているクローンを選択し、7Ell−C5.3抗体を使ったウエスタンブロッティング分析を用いてPSM抗原分析を行った。図19において、LNCaPの細胞溶解物および膜フラクションでは、PSMでトランスフェクトしたPC-3細胞と同様に100 kDa のPSM抗原が良く発現されていたが、本来のPC-3細胞では発現されていなかった。トランスフェクトされたPC-3細胞でこの発現が検出されたことは、以前にクローニングされた2.65kbのPSM・cDNAが、7Ell−C5.3 抗前立腺モノクローナル抗体によって認識される抗原をコードしていることを証明しており、また該抗体はPSMの炭水化物を含む抗原決定基を認識するから、該抗原はPC-3細胞内で適切にグリコシル化されていることを証明している。
正常なヒト組織でのPSM・mRNA発現を、リボヌクレアーゼ保護分析を用いて分析した。PSMの組織発現は前立腺内で優勢に出現し、ヒトの脳および唾液腺では非常に低いレベルでの発現が検出される(図20)。ノーザン分析で分析したときは、PSM mRNA発現は非前立腺ヒト組織では検出されなかった(データは示していない)。我々はまた、正常なヒト小腸組織で時々PSM発現が検出されることに気付いたが、このmRNA発現は、使用した特異的リボプローブに応じて変化した(データは示していない)。分析した正常なヒト前立腺および前立腺癌の全ての試料においては、PSM発現を明瞭に検出できることが明らかになったのに対して、良性の肥厚を示している組織では、一般にPSMの発現は減少するか又は発現しなかった(図21)。ヌードマウスの正所および皮下の両方で増殖しているヒトLNCaP腫瘍において、我々は、皮下移植LNCaP細胞の増殖に必要なマトリゲル(matrigel)を使用した場合も使用しない場合にも、PSMが大量に発現することを検出した。PSM・mRNAの発現は、生理学的な用量のステロイドに明瞭に調節されている(図22)。DHTは、発現を24時間後に8-10倍ダウンレギュレーションし、テストステロンはPSMの発現を3-4 倍減少させた。エストラジオール及びプロゲステロンもまた、LNCaP細胞のPSM発現を下方調節したが、これはLNCaP細胞に存在することが知られている突然変異したアンドロゲン受容体に結合した結果であろう。全体的に、PSM発現は、ステロイド欠乏培地で培養した未処理のLNCaP細胞で最も高かったが、この我々の提案する状態は、インビボでのホルモン欠乏(去勢した)状態をシミュレーションしている。この実験は、ステロイド用量を 2-200 nM の範囲で変化させ、また時間を6時間から7日まで変化させて繰り返されたが、結果は同様であった。DHTについては、24時間、用量 2-20nM のときに、PSM・mRNAの最大の下方調節が観察された。
ヒト前立腺の生物学を、正常状態および腫瘍性状態の両方について更に良く理解するためには、この重要な内分泌腺に固有の種々のタンパクおよび他の特性を研究することによって、知識を深めることが必要である。以前の研究によって2つの重要な前立腺バイオマーカー、即ちPAPおよびPSAが提供され、その何れもが前立腺悪性疾患の診断、治療および管理に重大な影響を与えた。前立腺特異性膜抗原(PSM)の予備的な特徴付けに関する我々の現在の研究によって、これが多くの興味深い特徴をもった遺伝子であることが明らかになった。PSMは、PAPおよびPSAと同様に、略完全に前立腺特異的であり、これらと同様に、前立腺の固有の機能および挙動を更に描写することを可能にし得る。PSMタンパクの予想配列(3)と、ウエスタンブロッティング及び免疫組織化学で証明されたように、該タンパクがLNCaP細胞膜に存在することとは、該タンパクが一体の膜タンパクであることを示している。従って、PSMは、抗体指向性の(antibody-directed) 画像診断および細胞毒性のターゲッティング治療様式のための、魅力的な細胞表面エピトープを提供する(14)。インビトロでPSM抗原を合成でき、またPSM発現を高いレベルで維持する腫瘍の異種移植を製造できる能力によって、PSM発現の制御および調節を更に研究し、特徴づけるための便利で魅力的なモデル系が提供される。また、LNCaP細胞での高レベルのPSM発現によって、優れたインビトロのモデル系が提供される。PSM発現はステロイドに対してホルモン的に反応性であり、またホルモン非反応性疾患において高度に発現し得るので(15)、アンドロゲン依存性前立腺癌の進行におけるPSMの潜在的な役割を解明することが肝要である。脳、唾液腺、小腸における僅かな量のPSM・mRNAの発現を検出することによって更なる研究が保証されるが、これらの組織は7Ell−C5.3 抗体を使った免疫組織化学によるPSM抗原の発現については陰性であった(l6)。これら全ての組織、特に小腸において、3´末端近傍のPSM・cDNA領域に相当するプローブを用いたときにはmRNAの発現を検出したが、5´末端PSMプローブを使ったときには発現を検出できなかった。これらの結果は、PSM・mRNAの転写物が、異なった組織では代替的スプライシング(alternative spricing)を受けることを示しているのかもしれない。以前のタンパクの研究では、7Ell−C5.3 抗体が 100kDaのPSM抗原に加えて、別の2つの少し大きいタンパク種を実際に検出し得ることが示唆された(17)。これら別のタンパク種は、図19におけるLNCaP溶解物および膜の試料に見ることができる。これらタンパクの起源には、択一的にスプライシングされたPSM・mRNA、PSMとは異なるがこれと密接な関係のある他の遺伝子、または異なる翻訳後修飾をうけたPSMタンパクが含まれる可能性がある。我々は現在、これらの可能性を研究している。
遺伝子治療のための「キメラDNA」を確立するためには、異なるセグメントのDNAを結合し、この結合に関与している両方の前駆体DNA種の特徴をもった新しいDNAを作ることが必要である。この提案においては、結合される2つの断片はDNAの異なった機能的特徴を含んでおり、mRNAの形成のためにDNAを読み取るプロモーター領域は特異性を与え、また該mRNAをコードする前記DNA配列は治療に機能するDNAを供給するであろう。
効果的であるときは、抗腫瘍薬は大量の腫瘍を退化させる。抗腫瘍薬活性を得るための主要な要件は、確実に十分な細胞損傷を与えて細胞死を生じさせるためにために、十分に長い時間(t)、十分高い濃度(c)で(cxt)、腫瘍を毒性の薬に晒すことである。また、この薬剤は「活性」でなければならず、腫瘍に対する毒性は宿主の正常細胞に対する毒性よりのも高くなければならない(22)。この腫瘍に対する薬物の有効性は、腫瘍の血流および薬物の拡散能力に依存している。多くの正常組織への血流は、しばしば腫瘍への血流と同じか又は多いので、腫瘍への血流は十分な選択性を提供しない。化学療法に用いられる大部分の細胞毒性薬は、腫瘍組織に対するのと同様に正常細胞に対しても有毒である。分裂性細胞はしばしば非分裂性の正常細胞よりも感受性が高いが、前立腺癌のような多くのゆっくり成長する固形腫瘍の場合、この感受性は抗癌剤が特異性であるために十分なものではない(22)。
出願人の研究グループは、特異的に且つ無毒的に、膀胱癌または前立腺癌のモデルにおいて、動物の樹立された腫瘍を治療できることを証明した。前立腺癌は、特に前立腺で正所増殖したときは、前立腺癌の治療はより困難であった。
非前立腺腫瘍系:
非前立腺腫瘍においては、プロモーター特異的な活性化を用いることによって、トランスフェクトされた遺伝子の組織特異的な遺伝子の発現を選択的に導き得ることが観察された。黒色腫においては、メラニン発現の原因である酵素をコードするチロシナーゼプロモーターを使用することによって、黒色腫細胞でのプロモーター駆動性リポータ遺伝子の発現が50倍以上になったが、非黒色腫細胞ではならなかった。同様な特異的活性化が、マウスで増殖しているトランスフェクトされた黒色種細胞でもみられた。その実験において、非黒色腫またはメラニン細胞はトリプシナーゼ駆動性リポータ遺伝子生産物を発現しなかった。ウエルカム研究所の研究グループは、癌胎児性抗原(CEA)をコードしている遺伝子のプロモーター領域をクローニングし、配列決定した。CEAは結腸および結腸癌細胞で発現するが、5-フルオロシトシンを5-フルオロウラシルに変換する転移性シトシンデアミナーゼで特に発現し、CEAプロモーター駆動性結腸腫瘍細胞を選択的に殺傷する能力が非常に増加しているが、分裂していない正常肝細胞は殺傷しないことが観察された。イン・ビボでは、シトシンデアミナーゼ遺伝子でトランスフェクトされていない局外の腫瘍細胞も殺傷されること、並びに大きな腫瘍は治療によって後退するので宿主の動物に対しては毒性がないことが観察された。単純ヘルペスウィルス(HSV)チミジンキナーゼも同様に、プロドラッグであるガンシクロビル(gancyclovir) を活性化して、分裂する腫瘍細胞に対して有毒にし、またHSVチミジンキナーゼは組織特異的プロモーターによって特異的に活性化され得ることが示されている。
効果的な癌治療の治療上の鍵は、特異性を達成すること並びに患者に毒性を与えないことである。遺伝子治療は、前立腺および前立腺腫瘍のような非本質的組織が、酸性ホスファターゼ(PAP)、前立腺特異性抗原(PSA)のような組織特異的タンパクと、我々がクローニングした遺伝子、即ち前立腺特異性膜抗原(PSM)遺伝子とを生産するという点で、特異性に重要な役目を提供し得る。前立腺のような組織は、これら組織特異的mRNAのDNAプロモーター領域への結合の原因となる選択的な組織特異性転写因子を含んでいる。PSAのプロモーターは既にクローニングされており、我々は前立腺腫瘍細胞の前立腺特異的プロモーターとしての使用を研究している。通常、転移性前立腺癌の治療を受けている患者は、PSAのmRNAの発現の劇的な減少をもたらすアンドロゲン剥奪治療を受けている。一方、PSMの発現はホルモン剥奪によって増加しているが、これはPSMはホルモン治療を受けた患者でより強く発現することを意味している。ジョンアイザック博士の(Dr.John Isaacs') 研究所との共同研究における予備研究では、ヒトPSM遺伝子を含むヒトクロモゾーム領域をラット腫瘍AT-6に転移したときに、PSMが発現することが証明された。AT−6は、転移性のアンドロゲン非依存性腫瘍である。非前立腺由来の組織もしくは腫瘍に移植された同じクロモゾームは発現しないので、これら細胞はこれらの実験の動物のモデルとして使用できるであろう。PSA、PSM陽性のヒトLNCaP細胞はヌードマウスでの試験に使われるであろう。
第三実験シリーズ
ポリメラ−ゼ・チェイン反応(複製連鎖反応)における
PSAおよびPSM由来プライマ−を使用した
前立腺血液原性微小転移巣の高感度検出
我々は、前立腺癌患者において血液原性微小転移巣の高感度検出を可能にする、PCRに基づく分析法を開発した。我々は、「ネストPCR(重ねPCR法; nested PCR) )」を実施し、前立腺特異的抗原および前立腺特異的膜抗原に特有のmRNA配列を増幅して、夫々の結果を比較した。微小転移巣は、PSR由来プライマ−によるPCRでは2/30(6.7 %)の患者で検出されたのに対して、PSM由来のプライマ−では19/16(63.3%)の患者で腫瘍細胞を検出した。陰性対照8人は全て、PSAおよびPSM・PCRの両方について陰性であった。分析を繰返して結果を確認し、またPCR産物はDNA配列決定およびサザン分析によって確認された。PSM・PCRでは検出されるが、PSA・PCRでは検出されない循環している前立腺腫瘍細胞をもった患者の中には、先に前立腺全摘出術を受けており、今回の分析時には血清中PSAが測定可能なレベルにはない4人の患者が含まれていた。将来の疾病再罹患および進行に関するこれら知見の有意性について研究が行なわれる。
<材料および方法>
細胞および試薬
LNCaP細胞およびMCF-7細胞は、アメリカン・タイプ・カルチャ−・コレクション(Rockville, MD )から入手した。これらの細胞系の樹立および特徴に関する詳細は、先に刊行物中に公表されている(11,12)。細胞は、MSKCC培地製造施設から入手した、L−グルタミン、非必須アミノ酸類および5%子牛胎児血清(ギブコ-BRL、ガイテルスブルク(Gaithersburg)、MD)を補充したPRMI1640培地を用いて、二酸化炭素インキュベ−タ−中において37℃で増殖させた。細胞培地は全てMSKCC培地製造施設から入手した。常用の化学試薬は可能な限りの最高級品であり、シグマケミカル(Sigma Chemical Company, St., Louis, MO)から入手した。
この研究で使用された血液標本は全て、MSKCCのスタッフである秘尿器科医らの外来患者オフィスを訪れた患者から入手した。規則的にスケジュールを決めた血液採集時に、患者一人当たり二つの抗凝固性(紫色のキャップ)のチュ−ブを得た。標本の調達は、MSKCC研究所の再審理評議会(Review Board)の認可により行われた。サンプルは迅速に実験室に運ばれて、直ちに処理された。血清PSAおよびPAPの測定は、MSKCC臨床化学実験室によって、標準法により行なわれた。PSA測定は、タンデムPSA分析(Hybritech, San Diego, CA)を用いて行なわれた。陰性対照として使用する8つの血液標本は、通常の血清PSA値および生体組織検査で実証されたBPHを有する男性2人、健康な女性一人、健康な男性3人、膀胱癌患者一人、および急性前骨髄細胞性白血病患者一人から入手した。
抗凝固静脈血4mlを氷冷リン酸緩衝生理食塩水3mlと混合し、15mlポリスチレンチュ−ブ内の8mlフィコ−ル(Phamacia, Uppsala, Sweden )の上部に注意深く層状に加えた。チュ−ブを 200 x g、30分、4℃で遠心分離した。滅菌パスツ−ルピペットを使用して、バフィ−コ−ト層(約1ml)を注意深く除去し、50mlのポリスチレンチュ−ブ内で、氷冷リン酸緩衝生理食塩水を用いて50mlになるまで再希釈した。次いで、このチュ−ブを4℃において 200 x gで30分遠心分離した。上澄液を注意深く捨て、ペレットをドリップ乾燥させた。次に、1mlのRNazolBをペレットに添加し、製造業者の指示書(Cinna/Biotecx, Houston, TX) に従って全RNAを単離した。RNA濃度および純度は、ベックマン(Beckman)DU分光器を用いたUVスペクトロスコピ−、およびゲル分析によって測定した。
RNAは、LNCaP細胞、並びにLNCaP及びMCF-7細胞の混合物から、固定比率(例えば 1:100、1:1000等)で、RNAzolBを用いて単離された。この分析の検出限界を決定するために、下記に記載するように、PSAプライマーおよびPSMプライマ−の両者を用いてネストPCR法を遂行した。LNCaP:MCF-7(1:100,000 )cDNAを蒸留水で希釈し、1:1,000,000 および 1:10,000,000 の濃度を得た。MCF-7細胞が選択されたのは、これを予め試験したところ、PSMを発現しないことがPCRによって示されたからである。
PSA外側プライマ−は、エクソン4および5のスパン部分を用いて 486bpのPCR産物を生成し、またcDNAと可能な汚染ゲノムDNA増幅との区別を可能にした。PSA・cDNA配列中においてヌクレオシド 494で始まる上流のプライマ−配列は、5´- TACCCACTGCATCAGGAACA- 3´(配列ID番号39)であり、またヌクレオチド 960よりの下流のプライマ−は、5´- CCTTGAAGCACACCATTACA- 3´(配列ID番号40)である。PSAの内側上流プライマ−(ヌクレオチド 559で始まる)の5´- ACACAGGCCAGGTATTTCAG- 3(配列ID番号41)および下流プライマ−(ヌクレオチド 894で始まる)の5´- GTCCAGCGTCCAGCACACAG- 3´(配列ID番号42)は、355 bpのPCR産物を生成した。これらプライマ−は全てMSKCCマイクロケミストリ−・コア施設で合成された。全体積20リットル中において、ス−パ−スクリプト(Superscript )逆転写酵素(Gibco-BRL)を用い、製造業者の推薦書に従って、5gの全RNAがcDNAに逆転写された。このcDNAの1リットルを、外側プライマ−PCR反応の開始テンプレ−トとして使用した。20リットルのPCR混合物は、0.5 UのTaqポリメラ−ゼ(Promega Corp.,Madison,WI) 、プロメガ(Promega)反応緩衝液、1.5mM MgCl2 、200M dNTPsおよび各プライマ−1.0Mを含んでいた。次に、該混合物をパ−キンエルマ−9600DNA サ−マルサイクラ−(Perkin Elmer 9600 DNA thermalcycler)に移しれ、25サイクルのインキュベ−トを行なった。PCRのプロフィ−ルは次の通りである: 94℃×15秒、60℃×15秒、72℃×45秒。25サイクル後にサンプルを氷上に載置し、1リットルの該反応混合物を、内側プライマ−を用いて行なう別ラウンドのPCR用テンプレ−トとして使用した。最初の一組のチュ−ブをサ−マルサイクラ−に戻し、更に25サイクルにかけた。PSM−PCRでは、確実にcDNAが増幅され且つゲノムDNA増幅されないようにするため、イントロンにまで広がるプライマ−対を選択することが必要とされた。PSMゲノムDNAの配列は未だ決定されていなかったので、cDNA増幅時に期待される大きさのPCR産物を生成し、ゲノムDNAテンプレ−トからはバンドが生成されない(これは大きなイントロンが存在していることを示す)ような一対のプライマーが見付かるまで、別のプライマ−対を試用された。PSMの外側プライマ−は 946bpの産物を生成し、内側プライマ−は 434bpの産物を生成する。使用したPSM外側の上流プライマ−は、5´- ATGGGTGTTTGGTGGTATTGACC- 3´(配列ID番号43)(ヌクレオチド1401から開始)であり、下流プライマ−(ヌクレオチド2348より)は、5´- TGCTTGGAGCATAGATGACATGC- 3´(配列ID番号44)であった。PSM内側の上流プライマ−(ヌクレオチド1581より)は,5´- ACTCCTTCAAGAGCGTGGCG- 3´(配列ID番号45)であり、下流プライマ−(ヌクレオチド2015より)は、5´AACACCATCCCTCCTCGAACC- 3´(配列ID番号46)であった。使用したcDNAは、PSA分析用に用いたのと同様であった。50リットルのPCR混合物は、1UのTaqポリメラ−ゼ(Promega )、250M dNTPs、10mMメルカプトエタノ−ル、2mM MgCl2 及び166mM NH4 SO4 、670 mM Tris pH8.8 および2mg/mlのアセチル化BSAを含む5リットルの10×緩衝液混合物を含んでいた。PCRはパ−キン・エルマ−480DNAサ−マルサイクラ−(Perkin Elmer 480 DNA thermalcycler) を用い、下記のパラメ−タで行なわれた:94℃×4分を1サイクル、94℃×30秒、58℃×1分および72℃×1分を25サイクル、続いて72℃×10分で行われた。次いでサンプルを氷冷し、この反応混合物2リットルを内側PSMプライマ−を含有した新しい反応混合物による別の25サイクル用のテンプレ−トとして使用した。cDNAの定性は、466 bpのPCR産物を産生するアクチンから誘導されたプライマ−を用いた対照反応を行うことによって確認された。ここで使用した上流プライマ−は、5´- AGGCCAACCGCGAGAAGATGA- 3´(配列ID番号47)(エクソン3)であり、また下流プライマ−は、5´- ATGTCACACTGGGGAAGC- 3´(配列認識番号48)(エクソン4)であった。全PSA混合物および各PSM反応混合物10リットルを、1.5-2 %アガロ−スゲルにかけ、臭化エチジウムで染色し、イ−グル・アイ・ビデオ画像システム(Eagle Eye Video Imaging System)(Stratagene,Torrey Pines, CA )で撮影した。少なくとも3回繰り返して分析を行ない、結果を確認した。
TAクロ−ニングシステム(Invitrogen社)を用いて、PCR産物をpCRIIプラスミドベクタ−中にクロ−ン化した。これらのプラスミドを標準法(13)を用いてコンピテントな大腸菌細胞に形質転換し、プラスミドDNAをマジック・ミニプレプ(Magic Minipreps) (Promega 社)を用いて単離し、制限酵素分析でスクリ−ニングした。TAクロ−ンは、シーケナ−ゼ(Sequenase) (U.S.Biochemical 社)を用いてジデオキシ法(14)により配列決定された。各プラスミド 3-4gをNaOHで変性し、エタノ−ル沈殿を行なった。35S−dATP(NEN社)を使用して、メ−カ−推奨法に従って標識化反応を実施し、該反応を同プロトコ−ルに記載されているようにして終了させた。次に、配列決定された生成物を、6%ポリアクリルアミド/7M尿素ゲル上において、 120ワット2時間で分析した。該ゲルを、10%メタノ−ル/10%酢酸中で20分間固定化し、ワットマン3MM紙に移し、真空乾燥器で2時間80℃で乾燥した。ゲルは室温で18時間オ−トラジオグラフにかけられた。
PCR生成物のエチジウム染色されたアガロ−スゲルを、0.2N HCl中に15分浸漬し、その後 0.5N NaOH/1.5M NaCl 中と、0.1M Tris pH7.5 /1.5M NaCl 中に30分ずつ浸漬された。次いで、ゲルは 10x SSC(1.5M NaCl /0.15M クエン酸ナトリウム)中で10分間平衡化された。DNAは、10 × SSC中でのポジブロッタ(Posi-blotter)(Stratagene社)による圧力ブロッテイングによって、DNAはナイトラン・ナイロン膜(Schleicher and Schuell社)に移された。DNAは、UVストラタリンカ(stratalinker)(Stratagene社)を使用して、その膜に架橋された。ブロットは65℃で2時間プレハイブリダイズされ、続いて32Pでラベルされ且つランダムプライムされた変性cDNAプロ−ブ(PSMまたはPSAのいづれか)とハイブリダイズされた(9,15)。ブロットは 1 × SSPE /0.5% SDS中において42℃で二回、1 × SSPE/0.5% SDS中において50℃で二回、夫々20分間洗浄された。膜を空気乾燥し、70℃で30分から1時間、コダックX-Omat フィルムを用いてオ−トラジオグラフにかけた。
ネストされたプライマ−を用いた我々のPCR増幅技術によって、PSAまたはPSMから誘導されたプライマ−の何れを用いた場合にも、前立腺細胞の検出レベルは約1万個のMCF-7細胞につき1個の前立腺細胞から、百万個のMCF-7細胞につき1個以上の細胞にまで向上された(図26および図27)。これは、最小病変(minimal disease) を検出する能力が実質的に改善されたことを示している。16人の患者の特徴を彼等の臨床段階、処置、血清PSAおよびPAP値について分析し、その分析結果を表1に示した。全体に、PSA−PCRは 2/30(6.7 %)の患者で腫瘍を検出したのに対し、PSM−PCRでは、19/30(63.3%)の患者で細胞を検出した。腫瘍細胞についてPSMでは非陽性であるがPSAでは陽性である患者はいなかったのに対して、PSMはPSAで検出されな8人の陽性患者を検出した。表1の患者10および11は、両者ともかなり進行したホルモン無反応性症の患者であるが、PSAおよびPSMの両方で検出された。これら患者は、両者ともこれらサンプルを採取した後に死亡した。患者4、7および12(全員が臨床的局所病弊について前立腺全摘出処置をしており、術後1-2 年の血清PSA値は測定レベルに満たない)は、PSM−PCRによる場合は循環系の前立腺腫瘍細胞について陽性であったが、PSR−PCRによる場合は陰性であった。図28に、PSM−PCRについての代表的なエチジウム染色ゲルの写真を示した。レ−ンAにかけたサンプルは、外側プライマ−から生じるPCR生成物を示し、レ−ンBのサンプルは内側プライマ−からの生成物である。図29に、対応するPSMサザンブロットオ−トラジオグラフを示した。幾つかのサンプルで示されるように、サザンブロット分析の感度はエチジウム染色を凌いでいた。即ち、これら幾つかのサンプルの外側プライマ−産物は、図28では見えないが、図29で示したようにサザンブロットでは検出可能である。更に、図28および図29のサンプル3(図30の患者6)は、外側および内側プライマ−生成産物のバンド(他の患者において対応しているバンドよりも小さい)を含んでいるように見える。DNA配列決定において、これらバンドのヌクレオチド配列は、小欠損を除いて、PSMのヌクレオチド配列と一致していることが確認された。これは、該患者におけるPCRのアーティクラフト、即ち択一的にプライシングされたPSM・mRNAのPCR産物、或いはPSM突然変異の何れかを示す。我々は、ある時、別のサンプルでも同様な知見に気付いた (未公表デ−タ)。配列決定され且つサザンブロット分析で分析した全サンプルが、PSAおよびPSMについて真性陽性であることが確認された。
診断時に、前立腺癌患者を正確に段階分けする能力は、適切な治療法の選択において、また治療に対する長期的反応および治癒可能性の予測において、非常に重要であることは明らかである。現在のところ、前外科的段階は物理的検査、血清PSAおよびPAPの測定、直腸超音波断層画像、CTスキャン、放射線核種骨スキャンおよびMRIスキャンを含む多数の画像診断法からなる。しかし、現在の方法は血液原性微小転移病弊の問題、および予後に生じ得るマイナスの影響については向けられていない。以前の研究によって、循環する腫瘍細胞は、僅か数パ−セントあっても、不可避的に固形転移を形成することが示されている(16)。しかしながら、循環系に存在する腫瘍細胞負荷の検出および定量化ができれば、もっと正確に疾病段階の分類に役立つことが分かるであろう。循環系にこれらの細胞を有することがわかった患者の臨床経過を、テスト結果が陰性であった同様の段階にある同様の処置を受けている患者の経過とを比較することによって、血液原性微小転移病弊の長期にわたる影響を研究すべきである。
Claims (17)
- 配列番号:2に記載されたヒト前立腺特異的膜抗原の配列のポリペプチド、配列番号:2のアミノ酸45〜750からなるヒト前立腺特異的膜抗原の外膜領域配列のポリペプチド、または配列番号:2のアミノ酸45〜750からなるヒト前立腺特異的膜抗原の外膜領域配列における抗原性領域(ただし、該抗原性領域は配列番号:35に記載された配列からなるものではない)のポリペプチドであって、前記ポリペプチドは前記前立腺特異的膜抗原に対して特異的な抗体を産生するために使用することができるポリペプチド。
- 配列番号:2に記載されたヒト前立腺特異的膜抗原の配列のポリペプチド。
- ヒト前立腺特異性膜抗原に対するモノクローナル抗体であって、前記前記抗原の配列は配列番号:2に記載されている抗体。
- 請求項3に記載のモノクローナル抗体であって、以下からなる群より選択されるヒト前立腺特異性膜抗原の抗原性領域に結合する抗体:
(a)Asp-Glu-Leu-Lys-Ala-Glu(配列番号:35);
(b)Asn-Glu-Asp-Gly-Asn-Glu(配列番号:36);
(c)Lys-Ser-Pro-Asp-Glu-Gly(配列番号:37);
(d)前記ヒト前立腺特異性膜抗原の外膜領域 ;および、
(e)前記ヒト前立腺特異性膜抗原の親水性アミノ酸配列。 - 請求項3または4に記載のモノクローナル抗体であって、さらに前立腺癌細胞の表面のヒト前立腺特異的膜抗原に結合することができることを特徴とするモノクローナル抗体。
- 請求項3〜5の何れか1項に記載のモノクローナル抗体を含有する組成物。
- 前記抗体に結合された薬剤をさらに含む、請求項6に記載の組成物。
- 前記薬剤が画像化剤、治療剤、毒素、または細胞障害性の薬剤である、請求項6に記載の組成物。
- 薬学的に許容されるキャリアを含む、請求項6〜8のいずれか1項に記載の組成物。
- 薬学的に許容されるキャリアを含む薬学的組成物である、請求項6に記載の組成物。
- 前立腺癌の治療または診断に使用するための、請求項3〜5の何れか1項に記載のモノクローナル抗体または請求項6〜10のいずれか1項に記載の組成物。
- ヒト患者において癌を治療するための薬学的組成物を製造するために、請求項3〜5の何れか1項に記載のモノクローナル抗体を使用する方法。
- 被験者の前立腺癌を画像化するための組成物の製造のために、請求項6〜10のいずれか1項に記載の物質の組成物を使用する方法。
- 生物学的サンプル中のヒト前立腺特異的膜抗原の量を測定するための免疫検定法であって、
(a)少なくとも一つの前記生物学的サンプルを請求項3〜5のいずれか1項に記載のモノクローナル抗体または請求項7または8に記載の組成物と接触させて、前記抗体と前記サンプル中に存在するヒト前立腺特異性膜抗原との複合体を形成する工程と、
(b)前記複合体中のヒト前立腺特異性膜抗原の量を測定することにより、前記生物学的サンプル中のヒト前立腺特異性膜抗原の量を測定する工程と、
を具備した方法。 - サンプル中の細胞または組織切片のヒト前立腺特異性膜抗原の発現を検出するための診断組成物を調製するために、配列番号:1に記載の配列の少なくとも15個の連続するラベルされた核酸を使用する方法。
- 前記試料が血液、リンパ節、または骨髄である、請求項15に記載の使用する方法。
- 請求項1に記載のポリペプチドをコードする核酸。
