Description
本発明のアデノウイルスベクターは、かならずしもMGEに特異的である必要はないプロモーター、関心のある遺伝子のコード配列、分割(cleaving)及びポリアデニル化配列からできた発現カセットを含む。
ヒト由来のアデノウイルスベクターは反芻動物種の乳腺上皮細胞を効率的に感染できること、分泌上皮におけるその安定性はウイルスのゲノムから発現されるタンパク質の免疫原性の程度に大きく依存し、このタンパク質がウイルス起源のタンパク質及び生物医薬品タンパク質の両方を含むことが示されている。複製欠損アデノウイルスベクター(例えば△E1△E3)は組換えタンパク質の高いレベルの発現を促進できるが、感染細胞に対してウイルスタンパク質によって誘導された強い免疫反応のために安定性が低下している。
本発明はすべての哺乳動物に適用できる。それにもかかわらず、乳汁製造能力が高いために反芻動物が好ましい。性的成熟の初期段階の動物を使用できる。これらの動物は乳腺産生及び乳汁産生を誘導するためにホルモン治療プログラムに従わされる。一般的に、自然に乳汁産生している動物は乳汁生産のレベルが高く、従ってホルモン誘導乳汁産生動物よりもトランスジーンのレベルは高い。
アデノウイルスベクターを含む溶液を注入する前に、動物は槽中の乳汁容器を除去するために搾乳されなければならない。ついで、乳腺のマッサージによって、溶液が腺胞の全体に達し、乳腺が一杯になるまで、乳頭の経路を通して乳腺に等張溶液を注入し、搾乳によって溶液を除去する。この段階を2,3回繰り返し、乳腺の全体の洗浄を確認し、腺胞及び管路組織の拡張を確認する。等張溶液はPBS,NaCl0.9%,グルコース5%、HBS組織培養培地であってよい。
処理動物からの乳汁は手によるか、又は自動採乳の従来の方法で得られる。カゼイン及び脂肪を乳漿から分離し、その大部分を−20℃で保存するが、少量の分画を一般に知られている分析技術(例えばELISA、ウェスタンブロティング及び生物活性)によって関心のあるタンパク質を検出し、定量するために使用する。関心のあるタンパク質のかなりの量を含む乳漿は混合し、異質タンパク質の精製の活性原料として用いる。精製工程は目的のタンパク質に依存してかなり変化できる。
1.所望の遺伝子の発現カッセットを含む組換えアデノウイルスの構築。この段階は次を含む。
a)組換えアデノウイルスゲノムの構築
b)細胞系293におけるウイルス粒子の製造
c)アデノウイルスベクターの増幅と精製
2.乳腺上皮細胞の感染。この段階は次を含む。
a)自然の乳汁産生のない動物を使用する場合、乳腺産生と乳汁産生の誘導
b)ウイルス粒子を含む溶液の乳腺への注入
3.組換えタンパク質の回収。この段階は次を含む。
a)処理動物の搾乳
b)採取乳汁中の異種タンパク質の検出と定量
c)異種タンパク質の精製
本発明をよりよく理解するための例示的な実施例を次に示す。
複製が欠損したアデノウイルスベクターをAdeasy系(Tong−Chuan Heら;Proc Natl Acad Sci USA.1998,95(5):2509−14)に基づき、プラスミドpAdTrack−CMV(図1)を移入ベクターとして用いて構築した。AdEasyに基づくこの系は、組換えアデノウイルスを作るための早くて、容易な選択を構成する。最初に発現カセットが移入ベクター中にクローニングされ、ついで菌株BJ51832中の同種組換えによってウイルスゲノム中に移入される、2つの段階のプロセスを必要とする。アデノウイルスゲノムはついで、PacIエンドヌクレアーゼで消化され、細胞系293又は911中にトランスフェクションされる。発明者の場合、感染性ビリオン及び究極の増幅は移入ベクターpAdTrack−CMV中に含まれている緑の蛍光タンパク質(GFP)の同時発現でモニターされた。
ヘルパー依存性アデノウイルスベクターをプラスミドpSH−1の多数のクローニングサイト中にエリスロポエチンの遺伝子を含む発現カセットをクローニングすることによって構築した。得られたプラスミドに包装を可能にするための適切なサイズを付与する目的で、それを菌株BJ5183(図2)中での同種組換えによってpStuffer−26ベクターに移入した。同種組換えの結果、28Kbを超すプラスミドが生成した。PacIエンドヌクレアーゼで消化し、プラスミドを293−Cre細胞系にトランスフェクションし、その後、ヘルパーアデノウイルスAdInvΨL−GFPで感染させた。293−Cre細胞系におけるCreリコンビナーゼの発現はヘルパーアデノウイルス中で包装配列を援護しているLox P部位間の組換えを促進し、ヘルパーアデノウイルスは包装される能力を失うが、複製し、従ってヘルパー依存性アデノウイルスベクターのゲノムを含むウイルス粒子の形成に必要な「in trans」のウイルスタンパク質を提供する能力を維持する。
アデノウイルスベクター中に構築されたhGH及びhEPOの発現カセットの正常な機能性を調べるために、乳腺上皮細胞を感染させた。細胞をEGF(10ng/ml)及びインスリン(10μg/ml)を補給したDMEM中で培養した。80%のコンフルエンス(confluence)で、細胞あたり20粒子の比率で、細胞培養にアデノウイルスベクターを加えた。24時間後培地を同じ培地に変えたが、EGFとインスリンを補給した。FCSは補給しなかった。48時間後、培地を採取し、1ml中に含まれているタンパク質をトリクロロ酢酸で沈殿させ40μの水に再懸濁し、20μlを電気泳動と、ついでウエスタンブロティングに用いた。
アデノウイルスのMGE感染能力及び関心のあるタンパク質の乳汁中への分泌促進能力をマウスでアッセイした。各マウス5匹の3実験群を作った。妊娠17日目のB6D2F1雌マウスに実験群によって、ウイルスを含む製剤100μを注入した:I.hGH遺伝子を含むアデノウイルス(△E1△E3)2.5x107GTU/ml;II.hEPOの遺伝子を含むアデノウイルス(△E1△E3)1x109GTU/ml;III.生理食塩水。
乳腺への遺伝子の移入のための組換えアデノウイルスベクターはすべての哺乳動物の種で実際に使用できる。しかし乳汁の生産能力が高いために反芻動物が好ましい。乳腺産生及び乳汁産生がホルモンで誘導できる、性的成熟の早期の段階の動物を用いることができる。乳汁産生の自然相にある動物もまた、用いることができる。例えば用いる動物がヤギの場合、ホルモン誘導は1,3,5,7,9,11,13日目のエストラジオール(0.25mg/kg、i.m)及びプロゲステロン(0.75mg/kg、i.m)の投与によってできるが、プレドニソロン(0.4mg/kg、i.m)は5日目から開始する複合乳腺組織の毎日のマッサージを行って、14日目から16日目まで投与しなければならない。
処理中のストレスを減らすために、乳汁産生相のヤギに10mgのジアゼパムを筋肉内投与した。槽からできるだけ多くの乳汁を除くために動物を広範に搾乳した。乳腺を37℃で200mlの生理食塩水で2回リンスし、ついで搾乳した。すべての注入はカテーテルに繋いだ50mlシリンジで乳頭の経路を通して直接行った。注入は注入複合乳腺組織のマッサージを同時にしながら、ゆっくりと行った。ヤギでは、複合乳腺組織は独立の2つの部分に分かれており、この内の1つにはアデノウイルスベクターを含む溶液を注入し、もう1つには生理食塩水だけを投与し、陰性内部コントロールとして用いた。各乳腺に109GTU/mlのウイルス負荷を含む200mlの溶液を注入した。これは各乳腺に合計2x1011のウイルス粒子が投与されたことを意味する。注入後、溶液の均一な分布を容易にし、溶液がすべての管路と腺胞に達するようにするために、複合乳腺組織をマッサージした。注入した液体は翌日搾乳によって除去した。

本発明で提案した方法はタンパク質構造の生物医薬品製造の増大するニーズに対して、溶液を構成する。具体的には、本発明はその生物活性がトランスレーション後の複雑なプロセッシングと密接に結びつき、従って高級生物の生合成機械に依存しているタンパク質の大規模生産を可能にする。本明細書で提案した方法によって、そのようなタンパク質の生産を速く、簡単で、経済的に達成できることを可能にする。この意味で、本発明は市場の変化に対して、短期間に速い回答を可能にし、その適用には大きな技術的要求は含まれず、反応容量はニーズによって容易に調整できる。
Claims (4)
- アデノウイルスベクターを用いる乳腺上皮細胞(MGE)の形質転換を介する、ヒトでない哺乳動物の乳汁中での異種タンパク質の次の段階を含む製造法:
a)性的成熟の初期の段階中での哺乳動物の乳汁分泌の誘導
b)ウイルスの注入に先立つ乳腺内部の乳汁の除去と完全な洗浄
c)乳腺の容量の完結まで、関心のある異種タンパク質をコードする遺伝子を運ぶアデノウイルスベクターを含む溶液の、乳頭の経路を通しての注入
d)注入後4時間から24時間の間に乳腺を排出すること
e)注入48時間後に開始する、乳腺に含まれる乳汁の採取
f)関心のある異種タンパク質の乳汁からの精製。 - アデノウイルスベクターがアデノウイルス遺伝子の大多数を含む請求項1記載の方法。
- 前記アデノウイルスベクターがアデノウイルス遺伝子の大多数又はすべてを欠き、ウイルス粒子の形成に必要なすべての必要なタンパク質をイン・トランス(in trans)で提供するヘルパーアデノウイルスを要求する請求項1記載の方法。
- 関心のある異種タンパク質がヒト成長ホルモン、上皮成長因子、インスリン様成長因子、顆粒球マクロファージコロニー刺激因子、神経成長因子、エリスロポエチン、FVIII及びFIXのような凝固因子、抗体、インターロイキン−6又はインターロイキン−2のようなサイトカイン、α1−抗トリプシン、ヒト血清アルブミン、β−グロブリン、組織プラスミノーゲン賦活剤、p53、Cタンパク質又はインターフェロンのような腫瘍抑制タンパク質からなる群から選ばれる請求項1記載の方法。
