Abstract
【解決手段】 特に、CNSおよび/または眼の疾患を治療する医薬品を調整するため、標的の遺伝子または遺伝子産物を調節することができる化合物を有する組成物を使用することについて報告し、前記組成物は血液CNS関門と血液網膜関門の外に投与するように設計する。さらに、CNSまたは眼の疾患に関与するポリペプチドをコードする核酸分子を同定、入手する方法、前記疾患を診断する方法、及び前記に説明した方法に沿って同定された標的遺伝子の発現が欠乏した遺伝子組換え動物について提示する。さらに、CNSおよび/または眼に関連した疾患の治療に特に有用な薬物を同定し、単離する方法を提示する。
Description
(a)CNSまたは眼性疾患に関連した病的状態を刺激するストレス条件下、細胞、組織、または非ヒト動物を培養・育成する工程と;
(b)前記細胞、組織、または動物から核酸および/またはタンパク質を単離する工程と;
(c)少なくとも1つの前記核酸および/またはタンパク質の発現または活性プロフィールを、対応する非処理細胞、組織、または動物および、異なるストレス条件下で処理された細胞、組織、または動物と比較する工程と;
(d)異なって発現された少なくとも1つの核酸および/またはタンパク質を決定することで、前記の少なくとも1つの核酸の発現または活性量の変化、または少なくとも1つの前記タンパク質の活性、または前記タンパク質の局在化変化がCNSまたは眼の疾患に影響していることを示す、決定する工程とを有する。
(a)細胞からmRNAを採取する工程と;
(b)ハイブリダイゼーションの条件下で前記の核酸分子またはそのフラグメントを有するプローブを用い、そのように採取されたmRNAをインキュベートする工程と;
(c)プローブにハイブリダイズさせたmRNAの有無を検出する工程とを有する、
または
(a)前記被験者から細胞サンプルを採取する工程と;
(b)そのように採取された細胞サンプルを前記抗体と接触させる工程と;

(g)ステップ(e)の疾患患者のDNAに特異的なバンドパターンとステップ(f)の被験者のDNAを比較し、前記パターンが同一か、異なっているかを決定し、前記パターンが同一の場合は、それによって前記疾患と前記疾患への罹患しやすさを診断する工程とを有する、
または
(a)診断チップ、プライマー伸長法、一塩基多型、または配列決定の方法により、前記の核酸分子を有する被験者の核酸サンプルを分析する工程と;
(b)前記の結果を前記疾患に罹患した患者から得られたサンプルの結果と比較する工程とを有し、
前記発現プロフィールの同一性および/またはヌクレオチドの配列は、前記疾患を示す。

(f)ステップ(a)〜(e)により被験者のDNAを調整し、ゲル上で検出可能な標識バンドを生成する工程と;
(g)ステップ(e)の疾患患者のDNAに特異的なバンドパターンとステップ(f)の被験者のDNAを比較し、前記パターンが同一か、異なっているかを決定し、前記パターンが同一の場合は、それによって前記疾患または前記疾患への罹患しやすさを診断する、決定する工程とを有する。

前記化合物と細胞との接触に要する時間は、例えば、既知の分子を用いて時間を経過させ、時間の関数として細胞の変化を測定することで、経験的に決定する。本発明の方法の測定手段は、さらに化合物を処理した細胞と前記化合物を同様に処理し、同定された細胞と比較することで定義可能である。代わりに2つの細胞、つまり機能的な標的遺伝子を含む細胞と第1の細胞と同等であるが、機能的標的遺伝子がない第2の細胞を、いずれも前記の同一化合物と接触させ、2つの細胞の差を比較することができる。この技術は、これらのアッセイの雑音を確定する上で有用でもある。当該分野の平均的な技術の1つにより、これらの制御機序が、機能的な標的遺伝子または遺伝子産物の調節に反応した、細胞の変化の選択を容易にしていることが理解される。
細胞、非ヒト動物、標的遺伝子発現、ノックアウト系は先行技術に見ることができ、本発明の方法で採用されている;例えば本明細書で引用されている文献を参照のこと。
以下の例では、典型的な一次ブタRPE細胞の単離について説明し、これは滅菌条件下で行われる。ブタ、ヒト、ウシからのRPE細胞の単離は、原則的に同じである。
前記ROSの単離は滅菌条件下で行った。RPE細胞の調整(例1を参照)で単離した30個の網膜は、氷冷した15mlのホモジナイズ用緩衝液(20%(w/v)スクロース、20mMトリスアセテートpH7.2、2mM MgCl2、10mMグルコースを含む)に移した。前記懸濁液を1分間優しく振盪し、平織りの薄い綿布で3回ろ過し、組織フラグメントを除去し、20mMトリスアセテートpH7.2、10mMグルコースを含む、25〜60%w/vで連続的に濃度を勾配させたスクロース24ml上で層にする。Beckman SW−27ローターで24,000rpm、1時間、4℃で遠心分離を行った。勾配上部の白黄色のバンドを集め、同量の10mM Hepes緩衝液pH7.4、115mM NaCl、2.5mM KCl、1mM DTT、1mM MgCl2で注意しながら希釈し、Sigma 4K15遠心機、2989xg、4℃で10分間遠心分離した。前記の上澄みを慎重に取り除き、前記ROSのペレットをさらに使用するため、−20℃で保存した。1つの網膜から約1x107個のROSが単離された。
ROSを用いたブタ、ヒト及びウシのRPE細胞のインキュベーションは原則的に同様であるため、以下の例ではブタの眼から単離したRPE細胞のインキュベーションについて述べている(例2を参照)。
アプローチ1は、二波長蛍光染色SNAFL−2(Molecular Probes、オランダ、ライデン)を用いて共有結合により標識したROSを検出する方法を有する。前記の酸型(pH 5)は緑黄色に見えるが、アルカリ型(pH 9)は黄橙色に見える。SNAFL−2(1μlのジメチルホルムイミド中SNAFL−2 10μg)を加え、スクロース緩衝液100μl(20%スクロース、2mM MgCl2、10mMグルコース、20mMトリスアセテートpH 8.0)に単離したROSを加え、前記ROSを暗所でゆっくりと撹拌しながら、1時間室温で標識した。前記の標識したROSを、同量のhepes緩衝液pH 8.0で希釈し、Heraeus Biofuge pico中、5分間2000rpmで遠心沈降させ、100μlの前記hepes緩衝液pH 8.0で2回洗浄した。前記の標識したROSを細胞培地に再懸濁し、前記細胞に加えた。4〜8時間インキュベートした後、細胞を洗浄し、蛍光顕微鏡用に細胞培地pH 9.0を加えた。アプローチ2では、ROS処理細胞を1xPBS(Ca2+及びMg2+を含む)中で洗い、内部ROSの自己蛍光活性を蛍光顕微鏡で分析した。
以下の例では、異なる生物(つまりブタ、ヒト、ウシ)から単離した一次細胞または細胞株(つまりARPE−19)における、特定の標的遺伝子(成長因子)XのPTGSについて述べる。
Parish et al.(1998)(Parish CA,Hashimoto M,Nakanishi K,Dillon J,Sparrow J.:「A2E及びイソ型A2Eの単離と1工程での調整、ヒト網膜色素上皮のフルオロフォア」Proc Natl Acad Sci U S A.1998 Dec 8;95(25):14609−13)に報告されているとおり、A2−Eはすべてトランス型のレチナールとエタノールアミンから合成し、Eldred及びKatz(1988)(Eldred GE,Katz ML.:「ヒト網膜色素上皮のフルオロフォア:分離とスペクトル特性」Exp Eye Res.1988 Jul;47(1):71〜86)の一次展開系を用いたシリカゲル60薄層クロマトグラフィープレートでクロマトグラフィーにより精製した。50〜150mgのレチナールを1.5〜5.5mlのエタノールに溶解した。5〜15μlのエタノールアミンを加えて撹拌した。撹拌しながら5〜15μlの酢酸をゆっくりと加えた。前記混合物をアルミニウムホイルで包み、2日間室温で撹拌した。前記反応混合物を4つのエッペンドルフ試験管に4回分配し、speedvacで一晩濃縮乾固した。2つのエッペンドルフ反応試験管の内容物を、合計200〜600μlの「一次展開系」(14.5mlヘプタン、8.8mlヘキサン、9.8mlクロロホルム、3mlエーテル、3mlアセトン、14.8mlイソプロパノール、27mlエタノール、2.5mlメタノール、0.4ml酢酸、7,4ml H2O)に溶解した。次に、この溶液をそれぞれ20〜60μlずつ含む5〜15等分を、約2時間、前記「一次展開系」で展開したシリカクロマトグラフィープレートで展開した。366nmの光で照射した蛍光により、プレートのA2−Eを検出した。A2−Eを含むシリカをかき取り(A2−E:上のスポット;イソ型A2−E:下のスポット)、ボルテックスしながらクロロホルム/メタノール/水で2〜3回溶出した。上澄みを合わせ、数時間speedvacで乾燥した。前記の乾燥した物質を200〜600μlの「一次展開系」に入れ、再度クロマトグラフィーを行った。前記の抽出と乾燥も繰り返し行った。前記の乾燥した物質を約1mlのMe2SOまたはエタノールのA2−E保存溶液としてとり、暗所、−20℃で保存した。Me2SOまたはエタノールに希釈したすべてのA2−Eを36,900のモル吸光係数、439nmで定量した(Parish CA,Hashimoto M,Nakanishi K,Dillon J,Sparrow J.:「A2E及びイソ型A2Eの単離と1工程での調整、ヒト網膜色素上皮のフルオロフォア」Proc Natl Acad Sci U S A.1998 Dec 8;95(25):14609〜13)。
1〜100μM A2−E及び0.5% Me2SOまたはエタノール濃度まで、A2−Eを前記RPE細胞培地に希釈した。アポトーシス以下の(sub−apoptocical)A2E濃度を見出すため、ネガティブコントロールとしてA2−Eがない状態で0.5%Me2SOまたはエタノールを用い、アポトーシスのコントロールとして1〜200μMのスタウロスポリンを用い、コントロールインキュベーションを行った。5%CO2を用い、37℃、暗所で(アルミホルイルで包んで)1日1回24〜144時間インキュベーションを行った。光源の下に細胞を置き、390〜550nmで144時間まで様々な時間、光に照射することができる。培地のみで対照実験も行った。RPE集団あたりの自己蛍光の平均値を決定するため、蛍光顕微鏡(Nikon;励起波長450〜490nm、発光波長>510nm)及びSafire(Tecan;励起波長456nm、発光波長610nm)を用い、栄養補給後の様々な時間で細胞内蛍光を評価した。
以下の例では、Cell Death Detection ELISA Plus(Roche)を用いたアポトーシスの誘導分析について述べる(Wyllie AH,Kerr JF,Currie AR.:「細胞死:アポトーシスの重要性」Int Rev Cytol.1980;68:251〜306.Review)。マウスモノクローナル抗体を用いた定量的サンドイッチ酵素免疫測定法の原則に基づいたアッセイは、それぞれDNA及びヒストンに対して行った。これにより、アポトーシスにより死亡した細胞の細胞質に放出される、モノヌクレオチド及びオリゴヌクレオチドを特異的に定量することができる。前記サンプル(細胞可溶化物、血清、培養の上澄みなど)を、ストレプトアビジンでコーティングされたマイクロプレートに入れた。続いて、抗ヒストン−ビオチン及び抗DNA−PODの混合物を加え、2時間インキュベートした。インキュベーション中、前記抗ヒストン抗体はヌクレオソームのヒストン成分に結合し、同時に、ビオチン化によりストレプトアビジンでコートされたマイクロプレートに前記免疫複合体を固定する。さらに、前記抗−DNA−POD抗体はヌクレオソームのDNA成分と反応する。洗浄工程により結合していない抗体を除去した後、免疫複合体にある前記PODにより、ヌクレオソームの量を定量した。PODは基質としてABTS(2,2’−アジノ−ジ[3−エチル−ベンズ−チアゾリン−スルホナート])を用い、光度測定により決定した。
vol(正常酸素(normoxic)条件)により、飽和密度(100% confluence)(約107細胞/100mmプレート)まで、37℃、空気95%、CO25%に維持したインキュベーター(Heracell,Kendro)で、RPE細胞をインキュベートした。次に1時間〜7日間、37℃でO2レベルを0〜8%、5% CO2、95〜87% N2(N2を平衡状態)で維持した自動CO2/O2インキュベーター(B 5061 EC/O2,Kendro)に細胞を置き、細胞を低酸素状態とした。誘導されていない細胞は正常酸素条件のままとした。培地のPO2及びPCO2は血液ガス分析装置(Corningモデル178)で測定した。正常酸素値は、pH=7.2+/−0.1、PO2=39.3+/−0.6mmHg、及びPCO2=131.5+/−0.9mmHgであった。低酸素値は、pH=7.2+/−0.1、PO2=7〜35+/−1.1mmHg、PCO2=14.9+/−1.2mmHgであった(Liu et al.,1995;Palmer et al.,1998)(Liu Y,Cox SR,Morita T,Kourembanas S.:「低酸素症は内皮細胞の血管内皮成長因子の遺伝子発現を制御する。5’エンハンサーの同定」Circ Res.1995 Sep;77(3):638〜43;Palmer LA,Semenza GL,Stoler MH,Johns RA.:「低酸素症はHIF−1を介して肺の動脈内皮細胞のII型NOS遺伝子発現を誘導する」Am J Physiol.1998 Feb;274(2 Pt 1):L212〜9)。
以下の例では、必須要素を欠乏させることでRPE細胞にストレスを誘導するための、調整DMEM(Dulbecco’s modified Eagle’s medium)(高グルコース、Life Technologies,Inc.)でのRPE細胞の培養について述べる。正常なRPE細胞培地において、37℃で5% CO2を含む加湿した大気中、2%の熱により不活化したウシ胎仔血清(Roche)、100単位/mlのペニシリン、100μg/mlのストレプトマイシン、1×非必須アミノ酸、2mM L−グルタミン、1mMピルビン酸ナトリウム(すべてLife Technologies,Inc.)を補充した、調整DMEM(高グルコース、Life Technologies,Inc.)で前記細胞を培養させる。正常な細胞培地には様々な無機塩が含まれ、ビタミン、アミノ酸、グルコース、ヌクレオチド、その他多数の物質が加えられている。ウシ胎仔血清など、ほとんどの細胞が血清を培養するために必要である。前記血清は、タンパク質、ホルモン、成長因子、中間産物を補充する。これらの必須要素を制限するため、細胞を血清を含まない培地で培養した、さらに、調整DMEMを用いた。前記培地には無機塩(例えばKCl、NaCl)、アミノ酸(例えばL−グルタミン、L−プロリン)、ビタミン(例えばビオチン、リボフラビン、葉酸)または他の物質(例えばグルコース、リポ酸(lipon acid)、ヌクレオチド)を入れなかった。
以下の例では、pH値を変化させることで、培養したRPE細胞の代謝性アシドーシス(pH<7.2+/−0.1)を誘導させる方法について述べる。in vivoと同様、哺乳類細胞培養にはpH7.2〜7.4の最適条件のpHが必要である。培養した細胞は乳酸を産生するため、pH値を低下させる。培地のpH値は血液ガス分析装置(Corningモデル178)で測定した。代謝性アシドーシスを誘導するため、2つの方法、つまり培地の漏出したHCO3 −とともに(5%から0%に)CO2圧を変化させる方法(Palmer et al.,1998)(Palmer LA,Semenza GL,Stoler MH,Johns RA.:「低酸素症はHIF−1を介して肺の動脈内皮細胞のII型NOS遺伝子発現を誘導する」Am J Physiol.1998 Feb;274(2 Pt 1):L212〜9)と、20 mM Hepes緩衝液を含む調節DMEMを利用する方法を用いた。Hepesで緩衝とした培地は37℃、5%CO2を含む加湿した大気中で酸性となるため、Hepes緩衝培地は細胞のpHストレスに利用することができる。
一次RPE細胞から調整したRNAを開始物質とし、リアルタイムPCR及びDNAマイクロアレイの発現分析を行った。ストレスの提示後(低酸素培養条件、栄養、成長因子の欠乏、またはpH変化など)、RPE細胞を細胞の溶解前にPBSで1回洗浄した。約1x106個の細胞(例えば細胞培養プレート6ウェル中1ウェル)を、プレートを軽く振盪させ、RLT緩衝液(Qiagen GermanyのRneasyミニキット)を含む800μlのβ−メルカプトエタノールで溶解した。可溶化液をすぐに−80℃に凍結した。37℃で15分間解凍後、製造業者(Rneasyミニキット、Qiagen)のプロトコールに従い、可溶化物を処理して全RNAを単離した。(1x106細胞ごとの)1カラムの充填材のRNAを、適当量(10〜100μl)のRNアーゼを含まない水で溶出した。全く同様に培養した細胞のRNAをプールし、1つの培養条件から均一なRNAを生産した。
その後RNAを分析するためには、ゲノムDNAの混入を取り除くため、注意が必要である。従って、DNAポリメラーゼI(RNアーゼを含まないDNアーゼI)を用いた酵素制限により、ゲノムDNAの消化を行った。要するに、100μlの反応用量には、50μgまでのRNAと、20μlの25mM MgSO4(最終濃度5mM)、3.4μl 3M NaAc pH 4.7(最終濃度100mM)、及び20U DNアーゼI(Roche Diagnosticsなどの10U/μlストック2μl)を一緒に含めた。37℃で1時間消化を行った。その後、供給者の指針に従ってRneasyミニキット(Qiagen)を利用し、再度DNアーゼと制限したDNA断片からRNAを精製した。
リアルタイムPCR及びマイクロアレイ技術により発現分析を行うため、全RNAからcDNAを合成することが必要である。異なる培養条件に由来するmRNAの遺伝子発現を比較できるようにするため、1μlのオリゴ−dT−プライマー(500μg/ml、Qiagen−Operon、# 55000142)、ブタRPE細胞のRNA2μg、1μlの10 mM dNTP−mix(Invitrogen)、及びRNアーゼを含まない水(Qiagen)、合計12μlまでにより、同量の総RNA(ここでは、例えば異なるRNAにつき2μg)をcDNAに逆転写した。前記混合物を65℃で5分インキュベートした後、2分氷冷した。短時間遠心分離を行った後、4μl 5x First Strand緩衝液、2μl 0.1M DTT、及び1μl RNasin(40U/μl、Promega、# N2511)のキットの成分(Invitrogen、# 18064〜014)を加えた。上下にピペットで混ぜた後、前記反応液を水浴で42℃にして、2分間インキュベートした。次に、1μlの逆転写酵素(Superscript II、Invitrogenキット、上記参照)を加え、上下にピペットで混合した。前記反応液を水浴中、42℃で50分間インキュベートし、次に前記反応試験管を15分間70℃の加熱ブロックに切り替えて反応を停止した。短時間氷でインキュベートした後、新たに合成されたcDNAを遠心分離し、使用前まで−20℃で保存した。
リアルタイムPCRで鋳型として用いるcDNAの濃度は、(最初のRNAによって)100pg〜100ngと変化する。所定の遺伝子と対照遺伝子それぞれについて、(蛍光色素と失活剤分子を含む)特定のTaqMan(登録商標)プローブを設計することができる。代わりに、前記色素SYBR(登録商標)−Greenを用いることもでき、すべての二本鎖DNA分子でインターカレートし、生じたPCR産物を処理中に測定することができる。(リアルタイムPCRとマイクロアレイ分析で)PCR増幅するオリゴヌクレオチドを設計し、通常150〜600塩基対(bp)のサイズのPCR断片を達成した。所定の遺伝子に加え、βアクチン(ACTB)、グリセルアルデヒド−3−リン酸デヒドロゲナーゼ(GAPD)、またはヒポキサンチンホスホリボシルトランスフェラーゼ(HPRT1)などの対照またはハウスキーピング遺伝子とともに、PCRプローブを2回または3回設定した。典型的な25μlのPCR反応は、鋳型cDNA(100pg〜100ng)、HotStart Taq−ポリメラーゼ(Invitrogenなど)0.5 U/μl、1xポリメラーゼ反応緩衝液、0.2mMの各dNTP(Invitrogenなど)、1.5〜7mMのMgCl2(Invitrogenなど)、50〜300nMの範囲のオリゴヌクレオチド(ドイツのQiagen−Operonなど)、SYBR(r)−Green(Bio Whittaker Molecular Applicationsなど、#50512、濃度10,000x)0.1〜0.5x(ストックを希釈)などで構成された。リアルタイムPCRの典型的なPCR条件は、5〜15分の95℃での活性化段階、94℃で各30sの変性サイクル45回、30sのプライマーのアニーリング(温度はプライマーの融解温度による)、及び最長1分間の72℃でのプライマーの伸長であった。各サイクルの後、PCR増幅中の前記プローブの蛍光増加を、リアルタイムPCR装置の光学ユニットで決定した(例えばBIORADのiCycler)。対照と処置を行ったサンプルから作成したリアルタイムPCRのデータは、mRNAの発現変化について、意味深い情報を与えた。
選択したすべての標的遺伝子から、以下のプロトコールにより、(鋳型として)RPE/網膜−または肝臓特異的なcDNAから150〜600bpのPCR断片を増幅した:PCR反応は、典型的には合計50μlに設定した。この反応には通常、1xポリメラーゼ反応緩衝液(Invitrogenなど)、1.5〜4mM(最終濃度)のMgCl2(Invitrogenなど)、0.2mMの各ヌクレオチド(10mM dNTPストック、Invitrogenなど)、最高1μMの遺伝子特異的forward及びreverseプライマー(Qiagen Operon)、0.025〜1UのTaqポリメラーゼ(Invitrogenなど)、適当量の鋳型cDNA(1〜10μl、cDNAの品質による)、及びヌクレアーゼを含まない水を最終量50μlまで含めた。PCRの典型的な条件は、5分間の95℃での単回変性段階、94℃で各30sの変性サイクル30回、30sのプライマーのアニーリング(通常45〜65℃、温度はプライマーの融解温度による)、最長1分間の72℃でのプライマーの伸長であった。
製造業者の指針に従いQiaQuick精製キット(Qiagen)を利用し、PCRの精製を行った。
スポット用の緩衝液として50% DMSOを用いたGenepakスポッター(Genetix)により、2つに分かれたピン(Telechem)でCMT GAPSコートしたスライド(Corning)上のPCR産物のDNAを配列した。スポットしたスライドは6ヵ月まで保存した。
各標識反応について、RNAは5μg(少なくとも300ng/μl、OD260/280は1.8〜2.0)が望ましい。Qiagenのキット「Label Star」を用い、供給者のプロトコールに従って、(対照/非処理細胞の)対照RNAと(ストレス/処理細胞の)被験RNAをCy3及びCy5(Amersham BiosciencesまたはPerkin Elmerから供給)で直接標識した。
自動Lucidea Slideproハイブリダイゼーションステーション(Amersham Biosciences)により、洗練されたハイブリダイゼーション及び洗浄の条件下でマイクロアレイにスポットしたDNAと混合したRNAのハイブリダイゼーションを、42℃で一晩行った:ハイブリダイゼーション緩衝液は、25%ホルムアミド、5x SSC、及び0.1% SDSで構成された。
ハイブリダイゼーション後、マイクロアレイのシグナルをレーザースキャナー(ScanArray 4000,Perkin Elmer)でスキャンし、未処理の画像データを得た。上記で作成した画像からタブで区切ったテキストファイルにシグナル情報を抽出するため、ScanAlyze(Mike Eisen、カリフォルニア州スタンフォード大学、http://rana.lbl.gov/EisenSoftware.htm)などのソフトウェアツールを適用した。データの規格化は、MicrosoftのExcel及びAccessプログラム(Microsoft Officeパッケージの一部)を適応して行った。そのように調整したデータは、例えば、異なって発現された遺伝子を見つけるため、複雑なクラスター分析が可能なSilicon GeneticsのGeneSpringなどのソフトウェアを用いて分析した。
この例は、マウス動物モデルにおける標的遺伝子eGFPのdsRNAによる特異的転写後遺伝子サイレンシングについて説明しており、この間、全身適用において、転写後遺伝子サイレンシングに対する最適なdsRNA濃度が決定される(実験手順1、結果は図1を参照のこと)。前記手順には遺伝子組換えマウス(FVB.Cg−Tg(GFPU)5Nagy,The Jackson Laboratory)のin vivo処理が含まれ、標的遺伝子eGFPに対するdsRNAオリゴリボヌクレオチド分子の全身適用により、これらの体細胞中で緑色蛍光タンパク質(eGFP)の活性型を発現する。対照動物も非サイレンシングdsRNA分子を用いて全身に処理する。転写後遺伝子サイレンシングの目的で、鎮痛剤または麻酔の影響を受けていない動物に、100または200μg eGFP特異的dsRNA/kg体重(BW)、また対照群では200μgの非サイレンシングdsRNA/kg BWの尾静脈注入を連日静脈内から行う(投与1日目:0日、投与最終日:20日)。緩衝液で処理した対照群の動物(0.1mgの緩衝液を尾静脈に連日静脈内注射)も保持した。実験動物の各群は8匹とし、最大注射量/注射は0.1mlである。21日目に前記動物はCO2吸入により屠殺する。
前記マウス中の眼における緑色蛍光タンパク質の発現は免疫組織学的に検討する(自発的eGFP蛍光:蛍光顕微鏡評価、eGFP特異的免疫蛍光染色:蛍光顕微鏡評価)。
dsRNA分子の設計に対して、2ヌクレオチド長で対称的な3’オーバーハングを有する21ヌクレオチド(nt)長のセンス鎖及びアンチセンス鎖を得るため、タイプAA(N19)TT(Nはすべてのヌクレオチドを表す)の配列を標的mRNAの配列から選択した。3’オーバーハングでは、ウリジンの代わりに2’デオキシチミジンを用いた。前記dsRNA分子が標的遺伝子のみに対応していることを確認するため、前記の選択したdsRNA配列をBLAST解析でマウスゲノムに対して検討する。前記21−ntRNA分子は化学的に合成、精製する。二本鎖を形成するため、100μgの各センス及びアンチセンスオリゴリボヌクレオチドを、10mMTris/HCl、20mMNaCl(pH7.0)中で混合し、95℃に加熱、18時間以上室温に冷却する。前記dsRNA分子をエタノールで沈殿し、滅菌緩衝液(100mM酢酸カリウム、30mMHEPES−KOH、2mM酢酸マグネシウム、pH7.4)に再懸濁する。前記dsRNAの整合性及び二本鎖の特性は、ゲル電気泳動法で確認する。代わりに、前記dsRNA分子を市販の供給業者から購入してもよい。前記標的遺伝子の配列及び対応するdsRNA分子は以下の通りである:
全身に適用するため、前記動物を固定し、前記dsRNAを尾静脈中に静脈注射し(最大注射量:0.1ml)、鎮痛または麻酔は静脈注射自体より動物にストレスを与えるため控える。球後注射(最大注射量:0.005ml)を行うため、前記動物に短期間イソフルラン吸入麻酔を行い、鎮痛を行うためメタミゾールナトリウムを投与する。次に、前記動物をいつも飼育している動物ケージの周りで保定する。in vivo診断の完了後(各動物実験の最後は例1〜5にそれぞれ記載する)、前記動物をCO2吸入により屠殺し、眼球除去し、この眼は組織学的(免疫組織学的)に研究する。
除去後、前記眼を4%ホルマリン/PBS溶液で24時間固定する。標準的な方法により、次に前記固定サンプルを連続的に濃度を増加させたアルコール中で脱水し、パラフィンに包む。ミクロトームを用い、標準的には5〜12μmの連続切片を作成し、加熱した水浴中で伸展し、ポリリジンでコートしたカバーグラスに移す。次に前記切片をインキュベーター中で2時間、52℃で乾燥する。前記乾燥切片はキシロール中で脱パラフィン化し、連続的に濃度を減少させたアルコール中に移した後、Tris/HCl(pH7.4)に入れる。ブロッキング後、前記切片を一次抗eGFP抗血清(ポリクローナルヤギ抗eGFP抗血清、Santa Cruz No.sc−5384)で2時間インキュベートする。Cy2共役ウサギ抗ヤギIgG(Dianova,No.305−225−045)を用い、免疫蛍光染色の手法により検出する。前記サンプルを包み、次に20X及び40X/1.3対物レンズを装備したEclipse TE−2000−S顕微鏡(Nikon)で顕微鏡検査を行うため、マウントする。脱パラフィン化を行った、未処理切片中の自然eGFP特異的蛍光は、蛍光顕微鏡を用いて解析する。

- 中枢神経系(CNS)と眼の疾患の両方または一方を治療する医薬品を調合するため、標的遺伝子または遺伝子産物を調節することができる化合物の使用法であって、前記組成物は血液脳関門と血液網膜関門の両方または一方の外に適用されるように設計するものである。

- 請求項35〜37のいずれか1つの方法または使用法において、前記表現型は血管新生および/または新血管新生、変性性網膜障害のすべてまたはいずれかに関連するものである。
- 請求項38の方法において、前記インヒビターは請求項10〜19のいずれか1つで定義されたインヒビターであるものである。
- 請求項24〜39のいずれか1つの方法であって、さらに前記の少なくとも1つの核酸および/またばタンパク質の配列を同定し、任意に対応するコード遺伝子またはそのcDNAを同定する工程を有するものである。
- 請求項40の方法であって、さらに前記遺伝子、cDNA、またはその断片をクローニングする工程を有するものである。
- 請求項24〜41のいずれか1つの方法により得られる核酸分子。
- 血管新生および/または新血管新生、および/または網膜疾患に関与するポリペプチドをコードした、請求項42の核酸分子。
- 請求項42または43の核酸分子に特異的にハイブリダイズする核酸分子であって、請求項35〜38のいずれか1つで定義したとおり、表現型の反応変化を誘導することができなくなったタンパク質の変異体をコードする核酸分子。
- 請求項42〜44のいずれか1つの核酸分子において、この核酸分子はDNAである。
- 請求項45の核酸分子において、この核酸分子はcDNAである。
- 請求項42〜64のいずれか1つの核酸分子において、この核酸分子は哺乳類に由来するものである。
- 請求項47の核酸分子において、前記哺乳類はマウスまたはヒトである。
- 請求項42〜48のいずれか1つの核酸分子、またはその相補鎖と特異的にハイブリダイズする、少なくとも15ヌクレオチド長の核酸分子。
- 請求項42〜49のいずれか1つの核酸分子を有するベクター。
- 請求項50のベクターにおいて、前記核酸分子は原核生物または真核生物の宿主細胞の発現を可能にする制御要素と操作可能に結合するものである。
- 請求項50または51のベクターを有する宿主細胞。
- 請求項52の宿主細胞は、細菌、真菌、植物、または動物細胞である。
- 請求項53の宿主細胞は、哺乳類細胞である。
- 請求項54の宿主細胞は、RPE細胞または神経感覚網膜細胞である。
- 請求項35〜38のいずれか1つで定義されるとおり、表現型の反応変化を誘導することができるポリペプチドを生産する方法であって、ポリペプチドの発現を可能にする条件下で請求項52〜55のいずれか1つの宿主細胞を培養する工程と、前記培養で生産されたポリペプチドを回収する工程と
を有するものである。 - 請求項42〜49のいずれか1つの核酸分子でコードされているか、または請求項56による方法で得られるポリペプチド。
- 請求項57のポリペプチドを特異的に認識する抗体。
- 請求項42〜49のいずれか1つの核酸分子、またはそのような核酸分子に相補的な核酸分子、請求項50または51のベクター、請求項52〜55のいずれか1つの宿主細胞、請求項57のポリペプチド、請求項58の抗体のいずれかまたはすべてと、任意に医薬品として認められた担体を有する医薬品。
- 請求項42〜49のいずれか1つの核酸分子、請求項50または51のベクター、請求項52〜55のいずれか1つの宿主細胞、請求項57のポリペプチドおよび/または請求項58の抗体と、任意に適当な検出法とを有する診断構成。
- 請求項1〜6のいずれか1つで定義された疾患を治療する方法において、有効量の請求項59の医薬品を被験者に投与する方法を有するものである。
- 請求項1〜6のいずれか1つで定義された疾患に関与する遺伝子の発現を検出する方法であって、
(a)細胞からmRNAを採取する工程と;
(b)ハイブリダイゼーションの条件下で請求項42〜49のいずれか1つの核酸分子またはそのフラグメントを有するプローブを用い、そのように採取されたmRNAをインキュベートする工程と;
(c)プローブにハイブリダイズさせたmRNAの有無を検出する工程と、
を有するものである。 - 請求項1〜6のいずれか1つで定義された疾患に関与する遺伝子の発現を検出する方法であって、
(d)前記被験者から細胞サンプルを採取する工程と;
(e)そのように採取された細胞サンプルを請求項58の抗体と接触させる工程と;
(f)前記遺伝子でコードされたタンパク質に結合した抗体の有無を検出する工程と、
を有するものである。 - 請求項44の核酸分子によってコードされたタンパク質発現を検出するための、請求項62〜63の方法。
- 請求項1〜6のいずれか1つで定義された疾患であるか、またはそのような疾患に罹患しやすい被験者を診断する方法であって:
(a)前記疾患に罹患した患者からDNAを単離する工程と;
前記ステップ(a)で単離されたDNAを少なくとも1つの制限酵素で消化する工程と;
(b)サイジングゲル上で得られたDNAフラグメントを電気泳動により分離する工程と;
(c)検出可能なマーカーで標識した、請求項42〜49の何れか1つの核酸分子またはそのフラグメントを有するプローブとともに、前記の得られたゲルをインキュベートする工程と;
(d)前記疾患患者のDNAに特異的なバンドパターンを作ると定義されているプローブとハイブリダイズしたゲル上で、標識バンドを検出する工程と;
(e)ステップ(a)〜(e)により被験者のDNAを調整し、ゲル上で検出可能な標識バンドを生成する、調整する工程と;
(f)ステップ(e)の疾患患者のDNAに特異的なバンドパターンとステップ(f)の被験者のDNAを比較する工程であって、前記パターンが同一か、異なっているかを決定し、前記パターンが同一の場合は、それによって前記疾患または前記疾患への罹患しやすさを診断する、比較する工程と、
を有するものである。 - 請求項1〜6のいずれか1つで定義された疾患であるか、そのような疾患に罹患しやすいと、診断する方法であって:
(a)診断チップ、プライマー伸長法、一塩基多型、または配列決定の方法により、請求項42〜49の核酸分子を有する被験者の核酸サンプルを分析する工程と、
(b)前記結果を前記疾患に罹患した患者から得られたサンプルの結果と比較する工程とを有し、
前記発現プロフィールおよび/またはヌクレオチドの配列の同一性は、前記疾患を示すものである。 - 体細胞遺伝子治療により請求項1〜6のいずれか1つで定義した被験者の疾患を治療、予防、および/または遅延させる組成物を調整するための、請求項42〜49のいずれか1つの有効量の前記核酸分子、またはそのような核酸分子に相補的な核酸分子、または請求項50または51のベクターの利用法。
- 請求項1〜6のいずれか1つで定義されるとおり、被験物質が疾患に関与する核酸分子またはポリペプチドに作用するか否かを決定する方法であって:
(a)請求項57のポリペプチドを発現した細胞とスクリーニングする化合物とを接触させる工程と;
(b)前記化合物が、前記ポリペプチドの発現または活性を調節するか否かを決定する工程と、
を有するものである。 - 請求項68の方法において、前記ポリペプチドはGGTBプロモーターの制御下で発現されるものである。
- 請求項68または69の方法において、前記被験物質は化学療法剤である。
- 請求項68〜70のいずれか1つの方法において、前記被験物質は化学療法剤の混合物である。
- 請求項68〜71のいずれか1つの方法において、前記細胞は単細胞生物または多細胞生物に由来するものである。
- 請求項68〜72のいずれか1つの方法において、前記細胞は請求項28、29、35、または52〜55のいずれか1つで定義された細胞である。
- 請求項68〜73のいずれか1つの方法において、好ましくは第1のスクリーニングで前記被験物質が被験物質の集合を構成し、被験物質の集合となるものである。
- 請求項74の方法において、前記被験物質の集合は約103〜約105の多様性を持つものである。
- 請求項74または75の方法において、前記被験物質の集合の多様性は連続的に減少するものである。
- 請求項68〜76のいずれか1つの方法において、前記方法は組織または非ヒト動物を構成するものである。
- 請求項1〜6のいずれか1つで定義された疾患を治療する薬物またはプロドラッグを生産する方法であって:
(a)被験化合物または被験化合物の集合を合成する工程と;
(b)前記被験化合物または被験化合物の集合に対して、請求項68〜77のいずれか1つの方法を実施する工程と;
(c)ステップ(b)の修飾因子またはその誘導体として同定された化合物を生産する工程と、
を有するものである。 - 請求項68〜78のいずれか1つの方法によって同定、単離、生産された被験物質であって、前記被験物質が、これまで請求項1〜6のいずれか1つで定義された疾患の治療薬として知られていないものものである。
- 請求項1〜6のいずれか1つで定義された疾患を治療する組成物を調整するため、請求項68〜78のいずれか1つの方法によって同定、単離、生産された化合物の使用法。
- 請求項1〜6のいずれか1つで定義された疾患を治療する方法であって、前記疾患に罹患した被験者に、請求項80の使用法または請求項78の方法に沿って調整した組成物を投与する方法を有するものである。
- 固体支持体を有し、そこに請求項42〜49のいずれか1つの核酸分子を1つ以上結合させたチップ。
- 請求項24〜41または62〜78のいずれか1つの方法で使用するキットであって、請求項82のチップ、または請求項42〜49のいずれか1つの核酸分子、または請求項58のポリペプチドの発現および/または活性を検出する他の方法を有するものである。
- 請求項68〜78の方法で同定、単離、生産された化合物を、新薬発見及び薬物またはプロドラッグの調整のリード化合物として使用する方法。
- 請求項1〜6のいずれか1つで定義した疾患を治療するため、または請求項42〜49のいずれか1つの核酸分子またはそのコードされた遺伝子産物を有する遺伝子の調節に反応する、下流遺伝子を同定及び単離するため、被験物質、リード化合物、薬物及びプロドラッグの妥当性を確認するために、請求項42〜49のいずれか1つの核酸分子または請求項57のポリペプチドを使用する方法。
- 請求項57のポリペプチドの異常な発現または活性を示す遺伝子組換え非ヒト動物。
- 請求項86の非ヒト動物において、前記動物は請求項1〜6のいずれか1つで定義された疾患を再現するものである。
- 請求項86または87の動物において、哺乳類である。
- 請求項86〜88のいずれか1つの遺伝子組換え非ヒト動物を、請求項1〜6のいずれか1つで定義された疾患を治療するため、新薬発見のプロセスに利用する方法。
- 請求項1〜6のいずれか1つで定義された疾患を治療するために用いる医薬品であって、1つ以上の二本鎖オリゴヌクレオチド(dsRNA)を有し、請求項42〜49のいずれか1つの核酸分子1つ以上の対応するmRNAのRNA干渉と、任意に医薬品として認められた担体を介するものである。
