Abstract
【解決手段】 特定の非イオン性界面活性剤を臨界ミセル濃度付近において含有せしめると共に、ポリエチレングリコールを、かかる非イオン性界面活性剤に対して、大割合にて含有せしめるようにした。

Description
先ず、精製水に対して、所定の配合成分を、下記表1に示される各種割合において、それぞれ添加して、溶解せしめることにより、pHが6.5とされた各種試験液1〜7をそれぞれ調製した。また、それら得られた7種の試験液の浸透圧は、何れも、生理食塩液に対する浸透圧比で、約1.1となるものであった。
上記した実施例1の結果を踏まえて、ポリエチレングリコールと非イオン性界面活性剤(ポリソルベート80)の2成分を併用している試験液1と試験液3についての比較検討を行うために、眼表面への影響を調べるNi−BUT(Non-invasive Tear Break-up Time )測定試験を実施した。
実施例1において調製された試験液1、2、6及び7を用いて、それら試験液を更にホウ砂の適量を用いてpH=7.3とした後、それぞれ2mlずつ、バイアル瓶に収容せしめ、そしてその後、それぞれのバイアル瓶に、市販のコンタクトレンズ(ジョンソン・エンド・ジョンソン社製「1DAY ACUVUE」)の3枚を、それぞれ2日間浸漬した。次いで、このように浸漬処理されたコンタクトレンズを、1枚ずつ、人工涙液(リゾチーム:496μg/ml、塩化ナトリウム:0.9%、塩化カルシウム:1mM、リン酸二水素ナトリウム:4mM、pH=7.0)の5mlが入ったバイアル瓶(25℃保持)に収容して浸漬せしめ、そして5分間浸漬した後の人工涙液中のリゾチーム濃度を、高速液体クロマトグラフィーにて測定した。そして、コンタクトレンズ浸漬前の人工涙液中のリゾチーム濃度と、コンタクトレンズ浸漬後の人工涙液中のリゾチーム濃度との差から、コンタクトレンズに付着したリゾチーム量を算出した。3つの測定値(n=3)の平均値を求め、その結果を、下記表5に示した。また、比較検討を行うために、試験液1、試験液6、試験液7及び試験液2の順番に、表5の結果を、図3に、棒グラフで示した。なお、比較対照の生理食塩水処理レンズのリゾチーム付着量は、96.0μg/lensであった。
実施例1における試験液の調製に用いられた非イオン性界面活性剤であるポリソルベート80に代えて、他の非イオン性界面活性剤であるポリオキシエチレン硬化ヒマシ油、ポリオキシエチレンステアレート又はポリオキシエチレン−ポリオキシプロピレンブロックコポリマー(BASF社製)を用いて、実施例1と同様にして、調製された試験液にあっても、また、ポリエチレングリコールとして、平均分子量が4000であるもの(ナカライテスク株式会社製)を用いた場合にあっても、装用中のコンタクトレンズ表面に付着した脂質汚れを効果的に除去することが出来、また、タンパク質の付着を効果的に抑制し、更に、生体に対して安全である眼科用液剤組成物を得ることが出来た。
実施例1における試験液1の配合成分である、非イオン性界面活性剤としてのポリソルベート80の配合割合を、0.001w/w%とし、またポリエチレングリコール(PEG#6000)の配合量を0.05w/w%として、同様な試験液を調製したところ、その脂質可溶化量の絶対値においては、実施例1の試験液1には劣るものの、生理食塩液よりも高い値を示すことを認めた。また、BUT測定試験においては、非イオン性界面活性剤(ポリソルベート80)の含有量が少ないために、それを、実施例2の場合と同様に点眼しても、涙液が正常な状態に戻る時間が速く、より一層安全な眼科用液剤組成物であることを認めた。

- 水性媒体中に、ポリオキシエチレンソルビタンアルキレート、ポリオキシエチレン硬化ヒマシ油、ポリオキシエチレンステアレート及びポリオキシエチレン−ポリオキシプロピレンブロックコポリマーからなる群より選ばれた少なくとも1種以上の非イオン性界面活性剤を、0.0001〜0.04w/w%の範囲内で含有すると共に、ポリエチレングリコールを、該非イオン性界面活性剤の1重量部に対して20重量部以上の割合において含有していることを特徴とするコンタクトレンズ用液剤組成物。
- 前記ポリエチレングリコールが、1000〜100000の平均分子量を有している請求項1に記載のコンタクトレンズ用液剤組成物。
- 前記ポリオキシエチレンソルビタンアルキレートが、ポリオキシエチレンソルビタンモノオレートである請求項1又は請求項2に記載のコンタクトレンズ用液剤組成物。
- ヒドロキシエチルセルロース、ヒドロキシプロピルセルロース、ヒドロキシプロピルメチルセルロース、コンドロイチン硫酸塩及びヒアルロン酸塩からなる群より選ばれた少なくとも1種以上の増粘剤を、更に含有している請求項1乃至請求項3の何れかに記載のコンタクトレンズ用液剤組成物。
- 前記液剤組成物の浸透圧が、生理食塩液に対する浸透圧比にて、0.85〜1.55の範囲内にある請求項1乃至請求項4の何れかに記載のコンタクトレンズ用液剤組成物。
- 等張化剤、pH緩衝剤、キレート化剤、防腐剤、殺菌剤、清涼化剤、消炎剤、血管収縮剤、ビタミン類及びアミノ酸類からなる群より選ばれた少なくとも1種以上の添加剤を、更に含有している請求項1乃至請求項5の何れかに記載のコンタクトレンズ用液剤組成物。
- 前記液剤組成物が、2mEq/L〜30mEq/Lの範囲内のK+ を含有している請求項1乃至請求項6の何れかに記載のコンタクトレンズ用液剤組成物。
- コンタクトレンズ用点眼液として用いられることを特徴とする請求項1乃至請求項7の何れかに記載のコンタクトレンズ用液剤組成物。
