実施例
この実施例で使用するヒストンタンパク質は、本明細書中でヒストンH1.4と称したヒストン断片であり、ヒトリンカーヒストンH1遺伝子(GeneBank アクセッション番号第M60748号、使用するタンパク質は、本明細書では配列番号1として同定される)から調製された。遺伝子を細菌中で発現させ、変性条件下で精製した後、酸性pHのリン酸緩衝液中でリフォールディングさせた。

トランスフェクション実験は、ルシフェラーゼ遺伝子をコードするリポータープラスミドpGL3−c(Promega)2μgと、部分的リンカーヒストン1.4タンパク質の増加量(μg)とを混合することにより実施した。タンパク質−DNA複合体の様々な重量対重量比をトリス生理食塩水pH8中で調製した。HeLa細胞を培地で洗浄して、以下のいずれか中でインキュベートした:
(1)10%血清を含む培地1ml;
(2)10%血清および2mMのカルシウムを含む培地1ml;
(3)10%血清および100μMのクロロキンを含む培地1ml;または
(4)血清全くなしの培地1ml

細胞を溶解し、ルミノメーターを用いて、Promegaのルシフェラーゼアッセイキットにより、ルシフェラーゼ酵素活性を測定した。結果は、血清なしの培地中でのトランスフェクションは比較的高く、約106の相対光単位(RLU)に達した。血清を含むトランスフェクションは、非常に低い値のルシフェラーゼ発現をもたらしたが、培地にクロロキンまたはカルシウムを補充すると、トランスフェクションの増加が観察された。結果を表1に示す。
