このタンパク質を2-Dゲルから抽出し、アミノ酸配列決定に付した。それはN-末端アミノ酸配列MILTELEKALN（配列番号１）を与え、これはタンパク質データベースの検索により見出されたS100-A8タンパク質のアミノ酸配列の報告と100％同一であった。セット中における他の３つのタンパク質もS100タンパク質ファミリーに属するものと一致した。２つは４つのアミノ酸とは離れて位置する２つの翻訳開始部位をもつS100-A9と一致し、１つはモノクローナル抗体との反応性による決定および二次元タンパク質分離の発表された図との比較から、他のS100関連タンパク質であるカルギゼリンと一致した。これらのタンパク質の様々な腫瘍タイプの2-Dパターンにおける位置は図１および図２に示す。
血清および他の生物学的液体中に検出される、腫瘍により合成されるタンパク質の可能性の主な決定因要素はその分泌される性質である。タンパク質が細胞により分泌されるのか否かを決定するタンパク質の特徴はあまり理解されていないままである。分泌過程に影響を及ぼす要因は、タンパク質における翻訳後の修飾の発生、および一定のシグナル経路の活性化に依存し得る。分泌性タンパク質の同定を容易にするため、培養腫瘍細胞により分泌されるタンパク質を精製する方法が開発され、クーマシーまたは銀染色により２次元ゲルでタンパク質を可視化し、それらの構成ペプチドのＮ−末端配列決定または質量分析により、分泌されたタンパク質スポットを分析する。この方法を用いて、Ｓ１００ファミリーの３つのメンバーを、乳癌における分泌されるタンパク質であると同定した。二つの分泌タンパク質のＮ−末端配列決定により、それらは以下の配列に基づく、ＭＲＰ８およびＭＲＰ１４として同定された：
