Claims (1)
- 【特許請求の範囲】 1. 植物または植物細胞にプロトポルフィリノーゲンオキシダーゼ阻害性除草 剤に対する耐性を付与する方法であって、あるDNA断片もしくはその生物学的機 能等価物またはそのDNA断片もしくはその生物学的等価物を含有するプラスミド を植物または植物細胞に導入することを含み、該DNA断片または該生物学的機能 等価物は発現され、かつ、次の特徴を持つ: (1)該DNA断片は植物中でプロトポルフィリノーゲン活性を持つタンパク質ま たはそのタンパク質の一部をコードする; (2)該DNA断片は、配列番号1、配列番号2または配列番号3からなる群より選 択されるアミノ酸配列をコードする核酸と相同であり、配列番号1または配列番 号2または配列番号3のVal13に対応するアミノ酸が別のアミノ酸で置換されてい るタンパク質またはそのタンパク質の一部をコードし、DNA-DNAまたはDNA-RNAハ イブリダイゼーション法によって検出および単離できる; (3)該DNA断片は、植物または藻類の細胞中で発現された場合、その植物また は藻類の細胞内でプロトポルフィリノーゲンオキシダーゼ阻害性除草剤に対する 耐性を付与する能力を持つ。 2. DNA断片もしくはその生物学的機能等価物またはそのDNA断片を含むプラス ミドが、双子葉植物においてプロトポルフィリノーゲンオキシダーゼ活性を持つ タンパク質またはそのタンパク質の一部をコードする請求項1に記載の方法。 3. 双子葉植物がArabidopsis thalianaであり、DNA断片が配列番号2のVal13 が別のアミノ酸で置換されているタンパク質をコードする請求項2に記載の方法 。 4. DNA断片が単子葉植物においてプロトポルフィリノーゲンオキシダーゼ活 性を持つタンパク質またはそのタンパク質の一部をコードする請求項1に記載の 方法。 5. DNA断片がトウモロコシにおいてプロトポルフィリノーゲンオキシダーゼ 活性を持つタンパク質またはそのタンパク質の一部をコードし、DNA断片が配列 番号3のVal13が別のアミノ酸で置換されているタンパク質をコードする請求項4 に記載の方法。 6. DNA断片がChlamydomonasにおいてプロトポルフィリノーゲンオキシダーゼ 活性を持つタンパク質またはそのタンパク質の一部をコードし、該DNA断片が配 列番号1のVal13が別のアミノ酸で置換されているタンパク質をコードする請求項 1に記載の方法。 7. Val13または対応するアミノ酸がメチオニンで置換されている請求項1〜6 のいずれか一つに記載の方法。 8. 耐性を付与される植物または植物細胞が緑藻Chlamydomonasである請求項1 〜6のいずれか一つに記載の方法。 9. Val13または対応するアミノ酸がメチオニンで置換されている請求項8に記 載のプロトポルフィリノーゲン阻害性除草剤耐性付与法。 10. 請求項1〜9のいずれか一つに記載の方法によって耐性が付与された植物 または植物細胞または緑藻。 11. プロトポルフィリノーゲン阻害性除草剤に対する耐性が付与された植物 または藻類の細胞を選択する方法であって、プロトポルフィリノーゲン阻害性除 草剤に対する 耐性が請求項1〜9のいずれか一つに記載の方法によって付与された植物または藻 類の細胞の集団を、除草剤感受性プロトポルフィリノーゲンオキシダーゼだけを 発現させる該植物または藻類の細胞の生育を通常遮断する量のプロトポルフィリ ノーゲン阻害性除草剤で処理することを含む方法。 12. 請求項1〜9のいずれか一つに記載の方法によってプロトポルフィリノー ゲン阻害性除草剤に対する耐性が付与された作物用植物の栽培域においてプロト ポルフィリノーゲン阻害性除草剤に対する耐性を持たない植物を防除する方法で あって、該栽培域に少なくとも1種類のプロトポルフィリノーゲン阻害性除草剤 を該プロトポルフィリノーゲン阻害性除草剤に対する耐性を持たない植物の生育 を阻害するのに有効な量で適用することを含む方法。 13. 適用されるプロトポルフィリノーゲン阻害性除草剤が式X-Qの化合物群か ら選択され、Qがからなる群より選択され、Xが:からなる群より選択される請求項12に記載の非耐性植物防除法。 14. 適用されるプロトポルフィリノーゲン阻害性除草剤が、式:の化合物、 ラクトフェン、 [N-(4-クロロ-2-フルオロ-5-プロパルギルオキシ)フェニル-3,4,5,6-テトラ ヒドロフタルイミド、 [2-クロロ-5-(シクロヘキサ-1-エン-1,2-ジカルボキシイミド)-4-フルオロフ ェノキ シ]酢酸ペンチル、 7-フルオロ-6-[(3,4,5,6-テトラヒドロ)フタルイミド]-4-(2-プロピニル) -1,4-ベンゾオキサジン-3(2H)-オン、 6-[(3,4,5,6-テトラヒドロ)フタルイミド]-4-(2-プロピニル)-1,4-ベンゾ オキサジン-3-(2H)-オン、 2-[7-フルオロ-3-オキソ-4-(2-プロピニル)-3,4-ジヒドロ-2H-1,4-ベンゾオ キサジン-6-イル]ペルヒドロイミダゾ[1,5-a]ピリジン-1,3-ジオン、 2-[(4-クロロ-2-フルオロ-5-プロパルギルオキシ)フェニル]ペルヒドロ-1H- 1,2,4-トリアゾロ[1,2-a]ピリダジン-1,3-ジオン、 2-[7-フルオロ-3-オキソ-4-(2-プロピニル)-3,4-ジヒドロ-2H-1,4-ベンゾオ キサジン-6-イル]5,6,7,8-1,2,4-トリアゾロ[4,3-a]ピリジン-3H-オン、 2-[3-オキソ-4-(2-プロピニル)-3,4-ジヒドロ-2H-1,4-ベンゾオキサジン-6- イル]-1-メチル-6-トリフルオロメチル-2,4(1H,3H)-ピリミジンジオン、 2-[6-フルオロ-2-オキソ-3-(2-プロピニル)-2,3-ジヒドロベンズチアゾール- 5-イル]-3,4,5,6-テトラヒドロフタルイミド、および 1-アミノ-2-[3-オキソ-4-(2-プロピニル)-3,4-ジヒドロ-2H-1,4-ベンゾオキ サジン-6-イル]-6-トリフルオロメチル-2,4(1H,3H)-ピリミジンジオン からなる群より選択される請求項12に記載の非耐性植物防除法。 15. 次の特徴を持つDNA断片またはその生物学的機能等価物: (1)該DNA断片は植物においてプロトポルフィリノーゲンオキシダーゼ活性を 持 つタンパク質またはそのタンパク質の一部をコードする; (2)該DNA断片は、配列番号1、配列番号2および配列番号3からなる群より選 択されるアミノ酸配列をコードする核酸配列と相同な核酸配列とのDNA-DNAまた はDNA-RNAハイブリダイゼーションによって検出および単離されうる配列を持つ ; (3)該DNA断片は配列番号1、配列番号2または配列番号3のVal13に対応するア ミノ酸が別のアミノ酸で置換されているタンパク質をコードする; (4)該DNA断片は、植物または藻類の細胞内で発現された場合、その植物また は藻類の細胞においてプロトポルフィリノーゲン阻害性除草剤に対する耐性を付 与する能力を持つ。 16. DNA断片が双子葉植物においてプロトポルフィリノーゲンオキダーゼ活性 を持つタンパク質またはそのタンパク質の一部をコードする請求項15に記載のDN A断片またはその生物学的機能等価物。 17. 双子葉植物がArabidopsis thalianaであり、DNA断片が、配列番号2のVal 13の別のアミノ酸による置換によってもたらされるアミノ酸配列をコードする請 求項16に記載のDNA断片またはその生物学的機能等価物。 18. 植物が単子葉植物である請求項15に記載のDNA断片またはその生物学的機 能等価物。 19. 単子葉植物がトウモロコシであり、DNA断片が、配列番号3のVal13の別の アミノ酸による置換によってもたらされるアミノ酸配列をコードする請求項18に 記載のDNA断片またはその生物学的機能等価物。 20. 植物が緑藻Chlamydomonasであり、DNA断片が配列番号1のVal13の別のア ミノ酸による置換によってもたらされるアミノ酸配列をコードする請求項15に記 載のDNA断片またはその生物学的機能等価物。 21. 該別のアミノ酸がメチオニンである請求項15〜20のいずれか一つに記載 のDNA断片またはその生物学的機能等価物。 22. DNA断片がChlamydomonasのゲノムDNAから単離されうる配列を持ち、プロ トポルフィリノーゲンオキシダーゼ活性を持つタンパク質またはそのタンパク質 の一部をコードし、配列番号4の37番目にあるグアニン(G37)に対応するヌクレ オチドが別のヌクレオチドで置換されている請求項20に記載のDNA断片またはそ の生物学的機能等価物。 23. 該別のヌクレオチドがアデニンである請求項22に記載のDNA断片またはそ の生物学的機能等価物。 24. 請求項15〜23のいずれか一つに記載のDNA断片またはその生物学的機能等 価物を含むプラスミド。 25. 請求項24に記載のプラスミドを保持する微生物。 26. プロトポルフィリノーゲンオキシダーゼに対するある化合物の阻害効果 を評価する方法であって、(a)プロトポルフィリノーゲン阻害剤感受性のプロ トポルフィリノーゲンオキシダーゼ活性を持つタンパク質をコードする遺伝子を 含有する感受性微生物と、配列番号1、配列番号2または配列番号3のVal13に対応 するアミノ酸が別のアミノ酸で置換されておりプロトポルフィリノーゲン阻害剤 に耐性なプロトポルフィリノ ーゲンオキシダーゼ活性を持つタンパク質をコードする遺伝子だけが該感受性微 生物とは異なる耐性微生物とを、試験化合物の存在下に培養し、(b)プロトポ ルフィリノーゲンオキシダーゼに対するその試験化合物の阻害効果を評価するた めに、該感受性微生物と耐性微生物の両方の生育を測定することを含む方法。 27. 配列番号1、配列番号2または配列番号3のVal13が別のアミノ酸で置換さ れておりポルフィリン性除草剤に対して耐性なプロトポルフィリノーゲンオキシ ダーゼ活性を持つタンパク質をコードする遺伝子を、活性なプロトポルフィリノ ーゲンオキシダーゼを欠く微生物に導入してその微生物の生育能を回復させるこ とによって耐性微生物が得られる請求項26に記載のプロトポルフィリノーゲンオ キシダーゼ阻害効果評価法。 28. 配列番号1、配列番号2または配列番号3のVal13が別のアミノ酸で置換さ れておりプロトポルフィリノーゲンオキシダーゼ活性を持つタンパク質をコード する耐性遺伝子を、プロトポルフィリノーゲンオキシダーゼ阻害性除草剤感受性 のChlamydomonas株に導入することによって耐性微生物が得られる請求項26に記 載のプロトポルフィリノーゲンオキシダーゼ阻害効果評価法。 29. 耐性を付与できる遺伝子が請求項20または22に記載のDNA断片を含む遺伝 子である請求項26のプロトポルフィリノーゲンオキシダーゼ阻害効果評価法。 30. Val13がメチオニンで置換されているか、G37がアデニンで置換されてい る請求項26〜29のいずれか一つに記載のプロトポルフィリノーゲンオキシダーゼ 阻害効果評価法。 31. プロトポルフィリンオキシダーゼ阻害剤を同定し評価するin vivo法であ って、 (a)プロトポルフィリノーゲン阻害剤感受性のプロトポルフィリノーゲンオキ シダーゼ活性を持つタンパク質をコードする遺伝子を持つ感受性微生物と、配列 番号1、配列番号2または配列番号3のVal13に対応するアミノ酸が別のアミノ酸で 置換されておりプロトポルフィリノーゲンオキシダーゼ阻害剤に耐性なプロトポ ルフィリノーゲンオキシダーゼ活性を持つタンパク質をコードする遺伝子の存在 だけが該感受性微生物と異なる耐性微生物とを、試験化合物の存在下に培養し、 (b)特定の用量で感受性微生物の生育だけを阻害する化合物を同定することを 含む方法。 32. 配列番号1、配列番号2または配列番号3のVal13が別のアミノ酸で置換さ れておりポルフィリン性除草剤に対して耐性なプロトポルフィリノーゲンオキシ ダーゼ活性を持つタンパク質をコードする遺伝子を、活性なプロトポルフィリノ ーゲンオキシダーゼを欠く微生物に導入してその微生物の生育能を回復すること によって耐性微生物が得られる請求項31に記載のプロトポルフィリノーゲン阻害 剤選択法。 33. 配列番号1、配列番号2または配列番号3のVal13が別のアミノ酸で置換さ れておりプロトポルフィリノーゲンオキシダーゼ活性を持つタンパク質をコード する遺伝子を、プロトポルフィリノーゲンオキシダーゼ阻害性除草剤感受性のCh lamydomonas株に導入することによって耐性微生物が得られる請求項31に記載の プロトポルフィリノーゲンオキシダーゼ阻害剤選択法。 34. プロトポルフィリノーゲンオキシダーゼ阻害剤に耐性なプロトポルフィ リンオキシダーゼ活性を持つタンパク質をコードする該遺伝子が請求項20または 22に記載のDNA断片を含む遺伝子である請求項31のプロトポルフィリノーゲンオ キシダーゼ阻害 剤選択法。 35. (請求項30と同じである)請求項31〜34のいずれか一つに記載のプロト ポルフィリノーゲンオキシダーゼ阻害剤選択法。 36. プロトポルフィリノーゲンオキシダーゼ活性を阻害しない化合物を同定 するin vivo法であって、(a)プロトポルフィリノーゲンオキシダーゼ阻害剤感 受性のプロトポルフィリノーゲンオキシダーゼ活性を持つタンパク質をコードす る遺伝子を含有する感受性微生物と、配列番号1、配列番号2または配列番号3のV al13に対応するアミノ酸が別のアミノ酸で置換されておりプロトポルフィリノー ゲンオキシダーゼ阻害剤に耐性なプロトポルフィリノーゲンオキシダーゼ活性を 持つタンパク質をコードする遺伝子だけが該感受性微生物と異なる耐性微生物と を、試験化合物の存在下に培養し、(b)該感受性微生物と耐性微生物の両方の 生育を阻害する化合物を同定することを含む方法。 37. 配列番号1、配列番号2または配列番号3のVal13が別のアミノ酸で置換さ れておりポルフィリン性除草剤に対して耐性なプロトポルフィリノーゲンオキシ ダーゼ活性を持つタンパタ質をコードする遺伝子を、活性なプロトポルフィリノ ーゲンオキシダーゼ活性を欠く突然変異微生物に導入してその突然変異体の生育 能を回復することによって耐性微生物が得られる請求項36に記載のプロトポルフ ィリノーゲンオキシダーゼ活性に影響を及ぼさない化合物の同定および評価法。 38. 配列番号1、配列番号2または配列番号3のVal13が別のアミノ酸で置換さ れておりポルフィリン性除草剤に対して耐性なプロトポルフィリノーゲンオキシ ダーゼ活性を持つタンパク質をコードする遺伝子をプロトポルフィリノーゲンオ キシダーゼ阻害 性除草剤感受性のChlamydomonas株に導入することによって耐性微生物が得られ る請求項36に記載のプロトポルフィリノーゲンオキシダーゼ活性に影響を及ぼさ ない化合物の同定および評価法。 39. プロトポルフィリノーゲン阻害剤に耐性なプロトポルフィリンオキシダ ーゼ活性を持つタンパク質をコードする該遺伝子が請求項20または22に記載のDN A断片を含む遺伝子である請求項36のプロトポルフィリノーゲンオキシダーゼ活 性に影響を及ぼさない化合物の同定および評価法。 40. 配列番号、配列番号2または配列番号3のVal13がMetで置換されているか 、配列番号4、配列番号5または配列番号6のG37がアデニンで置換されておりプロ トポルフィリノーゲンオキシダーゼ活性を持つタンパク質をコードする遺伝子を 導入することによって該耐性微生物が得られる請求項36〜39のいずれか一つに記 載のプロトポルフィリノーゲンオキシダーゼ活性に影響を及ぼさない化合物の同 定および評価法。
