Abstract
Description
Claims (1)
- 【特許請求の範囲】 1.SDS-PAGEによる測定で約170キロダルトンの分子量を有する、単離された、 二量体FAPα分子であって、前記二量体FAPα分子が細胞外マトリックスタンパク 質を分解することができる分子。 2.請求項1に記載の二量体FAPα分子であって、前記二量体FAPα分子の各単量 体が配列番号2のアミノ酸配列からなる分子。 3.請求項1に記載の二量体FAPα分子であって、組換えで産生される分子。 4.請求項3に記載の二量体FAPα分子であって、真核細胞によって産生される 分子。 5.(i)FAPα触媒領域と (ii)非FAPαタンパク質の少なくとも一部 からなる単離されたタンパク質。 6.式Xaa-Proの末端ジペプチドを分子から切断する方法であって、前記分子を 第2の分子と接触させることを含み、前記の第2の分子がFAPα酵素活性を有す るものである切断方法。 7.請求項6に記載の方法であって、前記第2の分子が単離された、二量体FAP αである方法。 8.請求項6に記載の方法であって、前記第2の分子がFAPα触媒領域を含む方 法。 9.FAPα活性を有する分子と相互作用する物質を同定する方法であって、前記 分子をテストすべきサンプルと一緒にし、FAPα活性を有する分子と相互作用す る分子の指標としての、前記分子とのなんらかの相互作用を測定することを含む 方法。 10.請求項9に記載の方法であって、前記FAPα分子が二量体である方法。 11.請求項9に記載の方法であって、前記分子がFAPα触媒領域を含む方法。 12.請求項9に記載の方法であって、前記物質がFAPα活性のアンタゴニスト である方法。 13.請求項9に記載の方法であって、前記物質がFAPα活性のアゴニストであ る方法。 14.請求項9に記載の方法であって、前記物質がFAPα活性のインヒビターで ある方法。 15.請求項9に記載の方法であって、前記物質とFAPα活性を有する細胞抽出 物とを一緒にすることを含む方法。 16.請求項15に記載の方法であって、前記細胞抽出物がFAPα活性を有する 分子をコードする核酸分子でトランスフォーメーションまたはトランスフェクシ ョンされた細胞の抽出物である方法。 17.請求項16に記載の方法であって、前記細胞が原核細胞である方法。 18.請求項16に記載の方法であって、前記細胞が真核細胞である、。 19.異常なFAPα活性で特徴づけられる病的状態の患者を治療する方法であっ て、前記の患者においてFAPα活性レベルを正常化するに十分な量の、FAPα活性 を有する分子と相互作用する物質を、それを必要としている患者に投与すること を含む方法。 20.請求項19に記載の方法であって、FAPα活性のインヒビターを投与する ことを含む方法。 21.請求項20に記載の方法であって、前記インヒビターがコラーゲン誘導体 である方法。 22.請求項20に記載の方法であって、前記インヒビターが(S)-バリル-ピロ リジン-2(R)-ボロン酸である方法。 23.請求項19に記載の方法であって、前記物質がFAPα活性のアゴニストで ある方法。 24.請求項19に記載の方法であって、前記物質がFAPα活性のアンタゴニス である方法。 25.物質がFAPα活性を有する分子と相互作用するかどうかを測定する方法で あって、前記物質と前記分子をAla-Pro-AFCと一緒にし、前記分子とAla-Pro-AFC との相互作用を測定し、前記相互作用と、前記物質の非存在下における前記分子 とAla-Pro-AFCとの相互作用とを比較することを含み、その差が前記物質と前記 分子との相互作用を示すものである方法。 26.FAPα活性を維持するのに十分なFAPα分子の一部および非FAPαアミノ酸 配列を含む融合タンパク質であって、前記融合タンパク質が水溶性であるタンパ ク質。 27.請求項26に記載の融合タンパク質であって、非FAPαアミノ酸配列がCD 8タンパク質に見いだされるアミノ酸配列である融合タンパク質。 28.請求項27に記載の融合タンパク質であって、前記CD8タンパク質がマウ スのタンパク質である融合タンパク質。 29.請求項27に記載の融合タンパク質であって、前記CD8タンパク質がヒト のタンパク質である融合タンパク質。 30.請求項27に記載の融合タンパク質であって、FAPαのアミノ酸27から760 に結合したマウスCD8のアミノ酸1から189を含む融合タンパク質。
