- 【特許請求の範囲】 1.細胞、特に細菌、胎児細胞、骨髄株細胞及び循環性癌細胞の磁気免疫分離方 法において、常磁性球上に標的細胞を固定すること、及び固定された細胞、遊離 細胞及び余剰の常磁性球を含む標本に対し磁界を作用させて常磁性球を分離させ ることから成るタイプの方法であって、不均一な磁界が適用される長さよりもは るかに短かい断面をもつ管(2)の中で標本を循環させることを特徴とする方法 。 2.磁界勾配の中に置かれたらせん状に巻きつけられた管(2)の中で標本を循 環させることを特徴とする請求の範囲第1項に記載の磁気免疫分離方法。 3.管の断面が0.5〜3ミリリットルの間に含まれていることを特徴とする請 求の範囲第1項又は第2項のいずれか1項に記載の磁気免疫分離方法。 4.管の長さは10センチメートルを上回っていることを特徴とする請求の範囲 第1項〜第3項のいずれか1項に記載の磁気免疫分離方法。 5.調製物は分離管に連結されたタンクの中に導入され、かかるタンクは重力に よる標本の循環を確保するべく管に比べ高く配置されていることを特徴とする請 求の範囲第1項〜第4項のいずれか1項に記載の磁気免疫分離方法。 6.タンクの中に混合せずにかつ界面層無しで、第1の体積の標本と第 2の体積の洗浄液を導入することを特徴とする請求の範囲第5項に記載の磁気免 疫分離方法。 7.調製物は、磁気勾配の中に置かれた管の中に吸収され、この管の下流端部が ぜん動ポンプに連結されていることを特徴とする請求の範囲第1項〜第4項のい ずれか1項に記載の磁気免疫分離方法。 8.磁石が、管との関係において、好ましくは交互運動に従って可動であること を特徴とする請求の範囲第1項〜第7項のいずれか1項に記載の磁気免疫分離方 法。 9.試薬及び標本が磁気分離管の中を0.1cm/秒〜10cm/秒の速度で連続的 に流れることを特徴とする請求の範囲第1項〜第8項のいずれか1項に記載の磁 気免疫分離方法。 10.常磁性球の直径よりも大きく固定された細胞の直径よりも小さい多孔度を もつ微細孔膜フィルターを用いて、その後のろ過段階を行なうことを特徴とする 請求の範囲第9項に記載の磁気免疫分離方法。 11.常磁性球及びいくつかの余剰細胞の直径よりも大きく固定された細胞の直 径よりも小さい多孔度をもつ微細孔膜フィルターを用いてその後のろ過段階を行 なうことを特徴とする請求の範囲第10項に記載の磁気免疫分離方法。 12.常磁性球及びいくつかの余剰細胞の直径よりも大きく固定された細胞の直 径よりも小さい多孔度をもつ微細孔膜フィルターで構成され ていることを特徴とする請求の範囲第10項又は第11項のいずれか1項に記載 の磁気免疫分離方法の使用のためのろ過装置。 13.細胞特に細菌、胎児細胞、骨髄株細胞及び循環性癌細胞の磁気免疫分離用 設備において、長さよりもかなり短かい断面をもつ管及びこの管が横断する空間 の中に磁界を作り上げるように配置された少なくとも1つの磁石で構成されてい ることを特徴とする設備。 14.さらに、管の上流に配置されたタンクを含んで成ることを特徴とする請求 の範囲第13項に記載の細胞の磁気免疫分離用設備。 15.増大する断面をもつ2つの区分を呈し、第2の断面が第1の断面の少なく とも10倍であるような管を含んでいることを特徴とする請求の範囲第13項又 は第14項に記載の細胞の磁気免疫分離用設備。 16.らせんを形成する溝を呈し磁石を支持する2枚の対称なプレート(15, 15)を有することを特徴とする請求の範囲第13項に記載の細胞の磁気免疫分 離用設備。 17.細胞特に細菌、胎児細胞、骨髄株細胞及び循環性癌細胞の磁気免疫分離用 設備において、長さよりもかなり短かい断面をもつ管によって構成されており、 この管は、標本の吸込みのための第1の端部及びぜん動ポンプに連結された第2 の端部を呈し、管は磁界勾配の中に置かれていることを特徴とする設備。 18.細胞特に細菌、胎児細胞、骨髄株細胞及び循環性癌細胞の磁気免 疫分離用設備において、交番磁界がカゴ形に配置された永久磁石によって生成さ れていることを特徴とする請求の範囲第17項に記載の設備。 19.負圧供給源に気密な形で連結されているサポート及び細胞分離管の下流端 部の連結のための手段を有する補足的なカバーで構成され、互換性あるろ過膜が サポートとカバーの間に配置されていることを特徴とする請求の範囲第11項に 記載の分離方法の使用のためのろ過装置。 20.カバーが有孔プレート及び単数又は複数の細胞分離管の端部によって孔を あけることのできるゾーンを呈する成形部品によって構成されていることを特徴 とする請求の範囲第19項に記載のろ過装置。 21.細胞表面に存在するか又は遊離した補足的物質に特異的に結合することの できるリガンドを用いて常磁性微小球上に標的細胞を固定すること及び、不均質 な磁界を横断し断面よりかなり大きい長さをもつ短管を横断して標本を循環させ ることを特徴とする、母親の血液中に存在する胎児細胞を分離する方法。 22.細胞表面に存在するか又は遊離した補足的物質に特異的に結合することの できるリガンドを用いて常磁性微小球上に標的細胞を固定すること及び、不均質 な磁界を横断し断面よりかなり大きい長さをもつ短管を横断して標本を循環させ ることを特徴とする循環性又は微小転移性癌細胞の早期検出方法。 23.細胞表面に存在するか又は遊離した補足的物質に特異的に結合することの できるリガンドを用いて常磁性微小球上に標的細胞を固定すること及び、不均質 な磁界を横断し断面よりかなり大きい長さをもつ短管を横断して標本を循環させ ることを特徴とする細胞、特に化学療法又は放射線療法による抗癌治療の後に破 壊された骨髄を再度播種できるようにする骨髄株細胞の調製方法。 24.常磁性球の断面より大きく、固定された研究対象の細胞よりも小さい多孔 度を示す膜フィルターを用いて、磁界の中に置かれた短管から再度出てくる細胞 のろ過を行なうことを特徴とする請求の範囲第23項に記載の、細胞、特に化学 療法又は放射線療法による抗癌治療の後に破壊された骨髄を再度播種できるよう にする骨髄株細胞の調製方法。
