Claims (12)
1. Postupak pripreme frakcija matičnih stanica porijeklom iz masnog tkiva, obuhvaća korake:
(a) uzimanje ili dobivanje uzorka masnog tkiva koji sadrži matične stanice;
(b) ispiranje uzorka iz koraka (a) prikladnim vodenim puferom;
(c) inkubacija uzorka iz koraka (b) pomoću enzima koji može razgraditi masno tkivo i iz njega ekstrahirati matične stanice;
(d) izdvajanje vodene faze iz proizvoda iz koraka (c);
(e) pročišćavanje vodene faze dobivene u koraku (d);
(f) titracija vodene faze dobivene u koraku (e) i, ako je potrebno, razrjeđivanje radi dobivanja konačne frakcije matičnih stanica željene koncentracije i volumena
gdje se materijal koji sadrži matične stanice obrađuje unutar štrcaljke barem kroz korake: (a), (b) i (c), ta štrcaljka ima funkcije sredstva za uzimanje, separatora faza i reaktora postupka.
2. Postupak prema zahtjevu 1, gdje štrcaljka ima volumen punjenja u rasponu od 20 do 100 ml.
3. Postupak prema zahtjevima 1-2, gdje štrcaljka ima jednu ili više oznaka koje pokazuju optimalni volumen(e) punjenja i/ili ima područja za pisanje ili označavanje.
4. Postupak prema zahtjevima 1-3, gdje je masni materijal lipoaspirat.
5. Postupak prema zahtjevima 1-4, gdje se koraci (a), odnosno (b-d) provode od strane dva različita operatera na međusobno udaljenim lokacijama.
6. Postupak prema zahtjevima 1-5, gdje u koraku (b) pufer je PBS pufer s dodacima enzimskih nutrijenata.
7. Postupak prema zahtjevima 1-6, gdje koraci (b) i/ili (c) obuhvaćaju okretanje štrcaljke prema dolje (igla prema dolje) ili prema gore (igla prema gore), nakon čega slijedi izbacivanje faze koja se nalazi uz iglu.
8. Postupak prema zahtjevima 1-7, gdje u koraku (c) enzim je liberaza, i inkubacija se provodi 20 do 80 minuta, na temperaturi od 30 do 45°C.
9. Postupak prema zahtjevima 1-8, gdje se u koraku (d) inkubacijska smjesa miješa s otopinom koja sadrži albumin, zatim se lipidna faza odbacuje i dobiva vodena faza za sljedeće korake postupka.
10. Postupak prema zahtjevima 1-9, gdje se materijal s matičnim stanicama dalje održava unutar štrcaljke tijekom jednog ili više od koraka (d)-(e).
11. Postupak prema zahtjevima 1-10, gdje se pročišćavanje u koraku (e) provodi centrifugiranjem(ima) i/ili filtracijom(ama).
12. Postupak prema zahtjevima 1-11, gdje se konačna frakcija matičnih stanica oblikuje kao jedna ili više jedinica od 1-5 ml, tako da je ukupna koncentracija nukleotidnih stanica između 108 do 104 stanica/ml.
