Abstract
The invention comprises diphenylmethylpiperidines (preferably 2-diphenylmethyl piperidines) of the formula: <FORM:0863715/IV (b)/1> wherein A represents an alkylene group of 1-5 carbon atoms, Z is a nitrogen atom monoor di-substituted by alkyl radicals of 1-5 carbon atoms or disubstituted by an alkylene radical to form a ring which may be interrupted by further hetero atoms, and the phenyl groups optionally bear halogen or C1-5 alkyl or alkoxy substituents. The compounds are prepared by introducing an aminoalkanoyl radical into a diphenyl-methyl piperidine, suitably by reacting the diphenylmethyl piperidine with an alkanoic acid or functional derivative thereof, such as the chloride, ester or anhydride, which acid contains an amino substituent or a substituent convertible to or replaceable by an amino group for example a halogen atom, which may be replaced by reaction with an amine, followed by conversion or replacement of this substituent where necessary. The products are used pharmaceutically, suitably in the form of their acid addition salts such as the hydrohalides, sulphates, nitrates, phosphates, acetates, propionates, oxalates, malates, citrates, methanesulphonates, ethanesulphonates, hydroxyethanesulphonates, benzoates, salicylates, p-aminosalicylates and toluenesulphonates. Preferred products include the diethylamino-, isopropylamino-, dimethylamino-, dibutylamino-, isoamylamino-, piperidino-, pyrrolidino-, morpholino-, and 4-methylpiperazino-, acetyl-2-diphenylmethyl-piperidine, the corresponding 1-a - and b -morpholinopropionyl-and a -diethylamino-propionyl piperidines and 2-p-chlorobenzhydryl- and 3- and 4-benzhydryl- 1-diethylaminoacetyl piperidine.ALSO:Preparations useful as medicaments and local anaesthetics comprise diphenylmethyl-piperidines (preferably 2-diphenylmethyl-piperidines) of the formula <FORM:0863715/VI/1> or acid addition salts thereof, wherein A is an alkylene group of 1-5 carbon atoms, Z is a nitrogen atom mono- or di-substituted by alkyl radicals of 1-5 carbon atoms or di-substituted by an alkylene radical to form a ring, which may be interrupted by further heteroatoms, and the "phenyl" groups optionally bear halogen or C1-5 alkyl or alkoxy substituents (see Group IV (b)), in admixture with a pharmaceutical organic or inorganic, solid or liquid carrier suitable for topical administration.
