Abstract

The invention comprises acridan compounds of the formula <FORM:0933875/IV(a)/1> and acid addition salts thereof, wherein R1 and R2 are hydrogen or lower-alkyl groups and R3 and R4 are lower-alkyl groups or R4 may also be hydrogen or benzyl, or NR3R4 is a heterocyclic amino group having a saturated 5- or 6-membered ring which may also bear alkyl substituents, and processes for the preparation of the above compounds wherein an appropriate 10-unsubstituted acridan is reacted with an aminopropyl compound of the formula R3R4N,CH2,CH2,CH2,Z, where Z is the acid residue of a reactive ester, e.g. a halogen atom or an arylsulphonyloxy group, preferably in an inert solvent, in the presence of an acid-binding agent, at a temperature of 50-200 DEG C. Suitably R1 and R2 are alkyl groups of 1-3 carbon atoms such as methyl and R3 and R4 are methyl groups or together with the nitrogen atom form a pyrrolidine, piperidine, morpholine, thiomorpholine or piperazine ring. Pharmaceutical preparations having sedative, hypothermic, mydriatic, adrenolytic and in some cases anaesthetic effects comprise the above acridan compounds of the invention or salts thereof such as hydrohalides, phosphates, sulphates, acetates, tartrates, maleates, citrates, malonates, oxalates and methane or ethane sulphonates, and a pharmaceutical carrier suitable for oral or parenteral administration. The preparations suitably take the form of tablets, capsules, powders, syrups or solutions.
