Abstract
The invention comprises compounds of formula <FORM:0741667/IV(a)/1> wherein R and R1 are hydrogen or alkyl radicals and may be the same or different or may be united to form with the adjacent nitrogen atom a heterocyclic ring, and R2 is hydrogen, a hydroxy group or an alkyl radical. The compounds are prepared by heating a pyrimidine derivative of formula <FORM:0741667/IV(a)/2> (1) with chloral and hydrolysing the resulting 5 - (2:2:2 - trichloro - 1 - hydroxyethyl) derivative in alkaline medium; or (2) with chloroform in presence of an alcoholic solution of an alkali metal hydroxide. The examples describe the preparation of 4:6 - dihydroxy - 5 - formyl - 2-piperidinopyrimidine and the 2:4-dinitrophe-nylhydrazone thereof, 4 - hydroxy - 5 - formyl-6 - methyl - 2 - di - n - butylaminopyrimidine and the semicarbazide and 2:4-dinitrophenyl-hydrazone thereof, 2 - amino -, 2 - methyl-amino -, 2 - dimethylamino -, 2 - piperidino-and 2 - hexamethyleneimino - 4 - hydroxy - 5-formyl-6-methylpyrimidine. 2 - Methylamino -, 2 - dimethylamino -, 2 - di - n - butylamino -, 2 - piperidino - and 2-hexamethyleneimino - 4 - hydroxy - 5 - (2:2:2-trichloro - 1 - hydroxyethyl) - 6 - methylpyrimidine are prepared as intermediates by process (1). 4:6 - Dihydroxy - 2 - piperidinopyrimidine is prepared by refluxing a mixture of guanylpiperidine sulphate, diethyl malonate and sodium in ethanol, and precipitating an aqueous solution of the product with hydrochloric acid. 2 - Di - n - butylamino - and 2 - hexamethyleneimino - 4 - hydroxy - 6 - methylpyrimidine are prepared by refluxing 4-hydroxy-6-methyl-2 - methylthiopyrimidine in b - ethoxyethanol with di-n-butylamine and hexamethyleneimine respectively.
