Abstract
In the recovery and purification of penicillin contained in a hydrophobic organic solvent, i.e. an organic solvent for penicillin which is insoluble or substantially insoluble in water such as a hydrocarbon, chlorinated hydrocarbon, ether, ester, higher alcohol or higher ketone, the penicillin is extracted from solution with a hydrophilic solvent, which is substantially immiscible with the said hydrophobic solvent and Which is an ether alcohol or polyhydric alcoho containing not more than 3 hydroxyl groups. In place of the hydrophilic solvent an aqueous solution thereof may be used. Suitably, the extraction is carried out using 50-90 per cent aqueous ethylene glycol or diethylene glycol at a pH of 4-7 and the resulting solution of penicillin in aqueous ethylene or diethylene glycol is diluted with water to a 10-40 per cent concentration of hydrophilic solvent and is re-extracted with a hydrophobic solvent at a pH of 1-5. In examples (1) and (2) penicillin in ether is extracted with 80 per cent aqueous glycol and the glycol solution is acidified, diluted and extracted with chloroform; (3) diethylene glycol is used in place of glycol in the process of (1); and (4) an amyl acetate extract of penicillin is treated as in (1).
