Abstract

L'urokinase est purifiée par la mise en contact d'une solution contenant de l'urokinase brute avec un échangeur de cations fortement acide qui contient un groupe acide sulfonique et est réglé sur le type NH 4 **+ afin d'adsorber l'urokinase sur cet échangeur de cations, puis par l'extraction de l'urokinase à partir de l'échangeur par élution. On ajoute au préalable de l'urée à la solution contenant l'urokinase brute dans une proportion telle que la concentration finale d'urée dans la solution se situe entre 3 et 8 M. Ce procédé est propre à fournir une urokinase purifiée qui présente une activité pratiquement constante et très spécifique, à partir d'une solution contenant de l'urokinase brute, avec un rendement élevé, quel que soit le degré de pureté de l'urokinase brute. L'urokinase est un enzyme qui est capable de produire la plasmine, enzyme fibrinolytique, à partir de plasminogène, et l'on sait que cette substance est non seulement utile pour le traitement de diverses sortes de thrombose, mais qu'elle est également capable de renforcer remarquablement le pouvoir des agents carcinostatiques lorsqu'elle est utilisée en association avec ces agents.
