Claims (8)

1. Chimär plasmid med en gen, som kodar för termos-tabilt a-amylas, kännetecknad därav, att den 5 innehäller en DNA-sekvens, som innehäller α-amylasgenen frän plasmid-DNA, som erhällits ur en Bacillus stearother-mophilus-stam och DNA pä en lämplig E. coli- eller B. sub-tilis-vektor.
2. Chimär plasmid enligt patentkravet 1, k ä n - 10 netecknad därav, att DNA-sekvensen som innehäller a-amylasgener erhälles ur B. stearothermophilusstammen ATCC 31 195, 31 196, 31 197, 31 198, 31 199 eller 31 783.
3. Förfarande för konstruktion av en chimär plasmid enligt patentkravet 1 med en gen, som kodar termosta- 15 bilt α-amylas, där man a) spjälker ett i naturen förekommande DNA av mik-roorganismursprung, vilken innehäller en gen som kodar för termostabilt α-amylas, med restriktionsendonukleas Hind III för erhällande av en lineär DNA-sekvens innehällande 20 a-amylasgenen; b) spjälker en lämplig E. coli- eller B-subtilis-vektor med restriktionsendonukleas Hind III för erhällande av en annan lineär DNA-sekvens, och c) sammanbinder de i stegen a) och b) erhällna li- 25 neära DNA-sekvensserna med T4-DNA-ligas, känne- t e c k n a t därav, att det i naturen förekommande DNA:t av mikroorganismursprung är plasmid-DNA som erhällits ur en Bacillus stearothermophilus-stam.
4. Förfarande enligt patentkravet 3, känne- 30 tecknat därav, att vektorn är en plasmid, som är E. coli-plasmiden pBR322, pBR325 eller pWL625 eller B. sub-tilis plasmiden pC194.
5. Förfarande enligt patentkravet 3 eller 4, kännetecknat därav, att det i naturen förekom- 11
