Claims (18)
1. Torkat hydrogelodlingsmedium för mikrober, känneteck-nat därav, att det omfattar en hydrogel baserad pä cellu-losaetrar med kovalenta tvärbindningar, varvid hydrogel- 5 odlingsmediet kan fästas direkt pä ett för vatten ogenomträng1igt underlag utan bärare- eller adhesions-skikt.
2. Hydrogelodlingsmedium enligt patentkravet 1, kännetecknat därav, att den gelbildande beständsdelen inne- 10 halier bäde hydrofila och hydrofoba cellulosaetrar.
3. Hydrogelodlingsmedium enligt patentkravet 1 eller 2, kannetecknat därav, att väte i cellulosans OH-grupper i cellulosaetrar har substituerats med alkyl-, som till exempel metyl-, etyl- eller propylgrupper, hydroxialkyl-, 15 som till exempel hydroxietyl- eller hydroxipropylgrupper, bensyl- eller hydroxibensylgrupper eller med karboximety-lens alkalimetallsalter, varvid i var och en glukosenhet hos cellulosan kan vara 0-3 OH-grupper, som substituerats med ovannämnda grupper.
4. Hydrogelodlingsmedium enligt nägot av de föregäende patentkraven, kännetecknat därav, att cellulosaeterns mo-lekylvikt är 500 - 1 000 000, företrädesvis 1 000 - 500 000. «
5 NjN'-bismetylenakrylamid, till den erhällna tvärbundna hydrogelen tillsätts näring-sämnen samt eventuellt andra behövliga komponenter, och det erhällna hydrogelodlingsmediet utbreds pä ett för vatten ogenomträngligt underlag och torkas, varvid od-10 lingsmediet fastnar direkt pä underlaget utan bärare- eller adhesionsskikt.
5. Hydrogelodlingsmedium enligt nägot av de föregäende 25 patentkraven, kännetecknat därav, att cellulosaeterns viskositet är 100 - 100 000 mPas, heist 1 000 - 10 000 mPas (2 % lösning, 20 °C).

Hydrogelodlingsmedium enligt nägot av de föregäende 111011 patentkraven, kännetecknat därav, att det utöver den pä tvärbundna cellulosaetrar baserade hydrogelen omfattar näringsämnen samt eventuellt andra behövliga komponenter, ' säsom andra gelbildande föreningar, pH-regleringsmedel, 5 detergenter, substrat, andra reagens som hör tili bioke-' miska detektionsreaktioner, selektiva inhibitorer, anti- biotika och färgmedel.
8. Hydrogelodlingsmedium enligt nägot av de föregäende patentkraven, kännetecknat därav, att det har torkats pä 10 ett för vatten ogenomträngligt underlag tili en fuktighet av 0,01 - 20 vikt-%.

Hydrogelodlingsmedium enligt patentkravet 1, kännetecknat därav, att tili gelen kan tillsättas cellulosaetrar, kopolymerer polymeriserade av vinylmonomerer, 15 som akrolein, akrylamid, 2-akrylamido-2-metylpropansul-fonsyra, 2-akrylamido-2-metylpropansulfonsyrans salter, akrylsyra, akrylsyrans salter, akrylonitril, metakryla-mid, metakrylamidopropyltrimetylammoniumklorid, metakryl-syra, metakrylsyrans salter, 2-metakroyloxietansulfonsy-20 ra, 2-metakroyloxietansulfonsyrans salter, 2-metakrylo- xietyltrimetylammoniumklorid, 3-metakryloxi-2-hydroxipro-pyltrimetylammoniumklorid, metakroylklorid, Ν,Ν'-metylen-bisakrylamid eller blandningar därav, agar-agar, albu-miner, alginat, gelatin, guargum, karragener, xantangum-25 mi, polypeptider och andra modifierade polysackarider.
10. Förfarande för framställning av ett torkat hydroge-lodlingsunderlag för mikrober, kännetecknat därav, att cellulosaetrar tvärbinds tili varandra för bildning av hydrogel t · j · * 30 a) genom att omsätta cellulosaetrarnas OH-grupper med till exempel divinylsulfon, formaldehyd, glyoxal, urea eller monometylol- och dimetylolderivat av melamin, ureaformaldehyd- och melaminformaldehydförpolymerer, di-metyloletylenurea, epiklorhydrin, diepoxider eller diiso- 111011 cyanat, eller b) genom att besträla eller c) genom att graftpolymerisera cellulosaetrarna med fri-radikalmekanism med vinylmonomerer, säsom akrylamid och

Förfarande enligt patentkravet 10, kännetecknat dä-rav, att tili gelen tillsätts utöver näringsämnen ocksä andra komponenter, säsom gelbildande föreningar, som ab- 15 sorberar vattenlösningar, pH-regleringsmedel, detergen-ter, substrat, andra reagens som hör tili biokemiska de-tektionsreaktioner, selektiva inhibitorer, antibiotika eller färgmedel, och det erhällna hydrogelodlingsmediet utbreds pä ett för vatten ogenomträngligt underlag, tor-20 kas vid 20 - 60 °C tili en fuktighet av 0,01 - 20 vikt-% och klipps tili testremsor.

Förfarande enligt patentkravet 10, kännetecknat dä-5 rav, att hydrogeler kan bildas av cellulosaetrar genom att graftpolymerisera med friradikalmekanism med akro-lein, akrylamid, 2-akrylamido-2-metylpropansulfonsyra, 2-akrylamido-2-metylpropansulfonsyrans salter, akrylsyra, akrylsyrans salter, akrylonitril, metakrylamid, metakry-10 lamidopropyltrimetylammoniumklorid, metakrylsyra, meta-krylsyrans salter, 2-metakroyloxietansulfonsyra, 2-meta-kroyloxietansulfonsyrans salter, 2-metakryloxietyltrime-tylammoniumklorid, 3-metakryloxi-2-hydroxipropyltrimetyl-ammoniumklorid, metakroylklorid, N,N'-metylenbisakrylamid 15 eller blandningar av dessa.

Mikrobiologisk testförpackning, som omfattar ett tor-kat hydrogelodlingsmediumunderlag för mikrober, kanne- / tecknad därav, att hydrogelodlingsmediets gelbildande in- y/ grediens omfattar en hydrogel, som baserar sig pä cellu- 20 losaetrar med kovalenta tvärbindningar, varvid hydrogel-odlingsmediet kan fästas direkt pä ett för vatten ogenomträngligt underlag utan bärare- eller adhesions-skikt.
