Abstract
PROCEDIMIENTO PARA PRODUCIR UN ZIMOGENO DE UROQUINASA. CONSISTE EN CULTIVAR CELULAS DE RIÑON HUMANO EN UN MEDIO DE CULTIVO Y RECUPERARLO DEL FLUIDO DE CULTIVO POR CROMATOGRAFIA DE AFINIDAD EMPLEANDO UNA COLUMNA DE ANTICUERPO INMOVILIZADO ANTI-ZIMOGENO; DONDE EL ZIMOGENO ES UN POLIPEPTIDO QUE TIENE UNA UNICA CADENA CON UN PESO MOLECULAR DE 50.000, Y AUNQUE EL ZIMOGENO POR SI MISMO PRESENTA POCA ACTIVIDAD DE ACTIVADOR PLASMINOGENO, MANIFIESTA UNA ACTIVIDAD DE ACTIVADOR PLASMINOGENO SIGNIFICATIVA POR CANTIDADES CATALITICAS DE PLASMINA; Y DONDE EL ZIMOGENO TIENE UNA CAPACIDAD TROMBOLITICA TAN ELEVADA COMO TRES VECES LA DE LA UROQUINASA DE LA ORINA HUMANA, PUDIENDO ESTABILIZARSE POR ADICION DE ALBUMINA Y POR CONSIGUIENTE PUDIENDO CONSEGUIRSE SUS PREPARACIONES LIOFILIZADAS ESTABLES. SE UTILIZA FARMACEUTICAMENTE PARA EL TRATAMIENTO DE LOS TROMBOS HUMANOS.
