Polipéptidos de HCELL
Los inventores describen glucoproteínas de HCELL aisladas y porciones biológicamente activas de las mismas, o
20 derivados, fragmentos, análogos u homólogos de las mismas. También se describen fragmentos de polipéptidos apropiados para su uso como inmunógenos para generar anticuerpos anti-HCELL. En una realización, las glucoproteínas de HCELL nativas se pueden aislar de células o tejidos mediante un esquema de purificación adecuado usando técnicas estándar de purificación de proteínas.
25 En otra realización, las glucoproteínas de HCELL se producen por técnicas de ADN recombinante. De forma alternativa a la expresión recombinante, una glucoproteína o polipéptido de HCELL se pueden sintetizar químicamente usando técnicas de síntesis peptídica estándar.
Una proteína “aislada” o “purificada” o una porción biológicamente activa de la misma está sustancialmente libre de
30 material celular o de otras proteínas contaminantes de la fuente celular o tisular de la que la glucoproteína HCELL deriva o sustancialmente libre de precursores químicos u otros productos químicos cuando se sintetiza químicamente. La expresión “sustancialmente libre de material celular” incluye preparaciones de glucoproteína HCELL en las que la proteína está separada de los componentes celulares de las células de las que se aísla o produce de forma recombinante. Para investigar adicionalmente actividades de ligando para L-selectina de HCELL y PSGL-1 expresadas en células
hematopoyéticas humanas, se realizaron ensayos de Stamper-Woodruff de la adherencia linfocitaria mediada por L
selectina a monocapas de células hematopoyéticas fijadas con glutaraldehído. Las células KG1a poseían actividad
5 de ligando a HCELL desde >40-120 rpm, la cual fue máxima a 80 rpm, mientras que las células HL60 exhibieron
actividad de ligando a L-selectina predominantemente a 80 rpm (Figura 12C). Además, la adherencia linfocitaria a
células KG1a mediada por L-selectina fue 10 veces más alta que la de las células HL60 a 80 rpm, y no hubo
evidencia de unión de linfocitos a las células K562 y RPMI-8402 (Figura 12 C). Dado que el ligando primario para L
selectina en las células HL-60 es PSGL-1 y su expresión es equivalente en células KG1a y HL60, estos datos 10 sugieren además que, basándose en una célula, la actividad HCELL en KG1a posee una capacidad más alta para
funcionar como ligando sobre un rango más amplio de tensión de corte.
Aunque el nivel de expresión de PSGL-1 es equivalente entre células KG1a y HL60, se examinó si PSGL-1 en las
células KG1a era funcionalmente equivalente a PSGL-1 en las células HL-60. Dado que el determinante de unión N15 terminal crítico de PSGL-1 para selectina-P se solapa con el determinante (s) de unión estructural para L-selectina
(Snapp, K.R., Ding, H., Atkins, K., Warnke, R., Luscinskas, F.W y Kansas, G.S. (1998) Blood 91, 154-164; De Luca,
M., Dunlop, L.C., Andrews, R.K., Flanner y, J.V., Ettling, R., Cumming, D.A., Veldman, G.M. and Berndt, M.C. (1995)
J. Biol. Chem. 270 (45), 26734-26737), se razonó que las capacidades de KG1a y de HL60 para unirse a selectina-P se correlacionan con la eficacia de unión de PSGL-1 a L-selectina. Así, se realizaron experimentos en la cámara de 20 flujo de la actividad de ligando a selectina-P utilizando células de ovario de hámster chino transfectadas con ADNc que codifica selectina-P humana de longitud completa (CHO-P). Tanto las células HL60 como las KG1a sostuvieron un rodamiento equivalente de CHO-P mediado por PSGL-1, las células K562 y RPMI-8402 no poseían ninguna actividad (Figura 13A). La actividad de ligando a selectina-P de las células KG1a y HL60 se eliminó mediante tratamiento con mocarhagin (Figura 13B). Distinto a la diferente a la capacidad de sostener actividad de ligando para 25 L-selectina entre las células KG1a y las HL60, estos datos sugieren que la PSGL-1 nativa como se expresa en la membrana celular era similar estructural y funcionalmente en estas líneas celulares. 5 inmunotransferencia con HECA-452 de cantidades equimolares de CD44 y PSGL-1 inmunoprecipitado de proteínas de membrana de KG1a revelaron que CD44 fue distintivamente más reactivo a HECA-452 que PSGL-1 (Figura 16), lo que sugiere que HCELL contiene un número mayor de epítopos HECA-452 que PSGL-1, lo que podría ser la explicación de su mayor actividad hacia L-selectina.
10 Ejemplo 11: Purificación por inmunoafinidad de HCELL y PSGL1
Se inmunoprecipitaron HCELL y PSGL-1 de células hematopoyéticas humanas. El procedimiento de inmunoprecipitación se siguió como está descrito (Dimitroff, C.J., Lee, J.Y., Fuhlbrigge, R.C. y Sackstein, R. (2000) Proc. Natl. Acad. Sci. 97 (25), 13841-13846). Brevemente, las proteínas de membrana de células hematopoyéticas 15 humanas (Dimitroff, C.J., Lee, J.Y., Fuhlbrigge, R.C. and Sackstein, R. (2000) Proc. Natl. Acad. Sci. 97 (25) 1384113846) se solubilizaron primero en NP-40 al 2 % y preaclararon en Proteína G-agarosa (Life Technologies). Para el SDS-PAGE/transferencia Western, 25 los inmunoprecipitados se lavaron 5 veces con tampón de lisis/NP-40 al 2 %/SDS 1 %/BSA 1 % y 3 veces con tampón de lisis sin BSA, y hervidos en tampón de muestra reductor para su análisis. Para la valoración funcional de CD44 o PSGL-1 en pruebas de adherencia, los inmunoprecipitados respectivos se lavaron 5 veces con tampón de lisis/NP-40 al 2 %/SDS al 1 %/BSA al 1 % y tres veces con tampón de lisis sin NP-40, SDS o BSA, luego
