Abstract

Procedimiento para la obtención de una solución de albúmina humana con elevada capacidad de enlace de moléculas de plasma humano, que comprende: a) obtención de un filtrado de plasma de sangre, que es una solución de albúmina esencialmente libre de residuos insolubles de proteínas desnaturalizadas y otros compuestos insolubilizados por etanol, y una primera diafiltración; b) estabilización del producto de la etapa (a) con NaCl y N-acetiltriptófano sin la adición de ácidos grasos; c) pasteurización del producto de la etapa (b); d) una segunda diafiltración del producto de la etapa (c) hasta que el N-acetiltriptófano se ha eliminado ampliamente; y e) estabilización del producto de la etapa (d) con >= 0,15 M de NaCl.

Los datos de estabilidad a tiempo real de 3 lotes de la albúmina indicada, muestran que el producto se mantiene estable. En concreto, la capacidad de enlace no presenta variaciones importantes respecto al inicio del estudio, al
5 menos tras 30 meses de almacenamiento a 5 ºC ± 3ºC y a 30 ºC ± 2ºC (figura 3). De hecho las oscilaciones observadas en los resultados de capacidad de enlace a lo largo del tiempo ocurren en paralelo a 5 ºC y a 30 ºC, por lo que son atribuibles a la propia variabilidad del ensayo, realizado en diferentes días, más que a una variación real de dicha capacidad de enlace.
10 Ejemplo 4: Estabilidad de una solución de albúmina obtenida según el proceso descrito en la presente invención y estabilizada con cloruro sódico 0,15 M.
Los datos de estabilidad a tiempo real de la albúmina indicada, realizado a las temperaturas de almacenamiento de 5 ºC, 30 ºC y 40 ºC muestran que el producto se mantiene estable a la temperatura de 5 ºC. A esta temperatura, la
15 capacidad de enlace (figura 4) no presenta variaciones importantes respecto al inicio del estudio y tampoco comparada con la albúmina estabilizada con N-Acetiltiptófano. Los datos de Distribución molecular y de Turbidez (tabla 1) también apoyan la estabilidad de esta solución de albúmina.

Tal como se aprecia por lo anterior, mediante las investigaciones realizadas sobre la estabilización de la albúmina y la eliminación de los estabilizantes asociados a la misma y su implicación en la capacidad de unión de la albúmina, los inventores han comprobado la gran ventaja del uso de una albúmina humana con una elevada capacidad de
45 enlace para la terapia de detoxificación.
Esta albúmina humana usada en terapia de detoxificación se caracteriza por que su formulación final carece totalmente de estabilizantes o bien comprende agentes estabilizantes que resultan en una elevada capacidad de enlace.
50 Esta terapia de detoxificación puede ser realizada por infusión de albúmina al paciente, por plasmaféresis y reposición de albúmina al paciente o también realizarse en un sistema extracorpóreo.

55 se indican, éstos no están destinados a limitar el alcance de la misma, de manera que se deberá considerar que quedan incluidas dentro del alcance de la presente invención todas aquellas variantes que queden comprendidas en el alcance de las siguientes reivindicaciones.
