Abstract

Un método para determinación de la secuencia de bases nucleotídicas en un analito polinucleotídico caracterizado por los pasos de (1) generar una corriente de trifosfatos nucleotídicos monobásicos a partir del analito por pirofosforólisis progresiva en presencia de una enzima de pirofosforólisis; (2) producir moléculas capturadas por reacción de cada trifosfato nucleotídico monobásico en presencia de una polimerasa y una ligasa con un sistema de captura constituido por: (i) (a) un primer oligonucleótido que comprende una región bicatenaria y una región monocatenaria y (b) un segundo oligonucleótido monocatenario cuya secuencia de bases nucleotídicas es al menos parcialmente complementaria a la de la región monocatenaria del primer oligonucleótido; o (ii) un oligonucleótido simple que comprende una región nucleotídica monocatenaria cuyos extremos están unidos a dos regiones oligonucleotídicas bicatenarias diferentes; (3) amplificar al menos parte de la molécula capturada para producir una pluralidad de amplicones característicos del trifosfato nucleotídico monobásico simple, (4) marcar los amplicones con una sonda correspondiente que tiene un elemento característico detectable y (5) detectar una propiedad característica del elemento detectable.
