Se dan a conocer las secuencias de ácido nucleico y de aminoácidos de varias proteínas PRG4 o lubricina nativas y recombinantes, y la caracterización de las proteínas PRG4 y diversas isoformas, por ejemplo, en las patentes estadounidenses n.os 5.326.558; 6.433.142; 7.030.223; 7.361.738 de Turner et al., y las patentes estadounidenses
n. os 6.743.774 y 6.960.562 de Jay et al.. La publicación estadounidense n.º 20070191268 de Flannery et al. también da a conocer moléculas de PRG4 o lubricina recombinantes útiles en la presente invención.
En la técnica se conocen bien métodos para el aislamiento, la purificación y la expresión recombinante de una proteína PRG4. En determinadas realizaciones, el método comienza con la clonación y el aislamiento de ARNm y
15 ADNc que codifican para proteínas PRG4 o isoformas usando técnicas de biología molecular convencionales, tales como PCR o RT-PCR.
