Abstract

Promotor y construcciones para expresión de proteínas recombinantes en hongos filamentosos. Se describe un nuevo promotor para la expresión de proteínas recombinantes en hongos filamentosos (1-740 en SEQ ID No. 1), que es el promotor gdhA del gen de la glutamato deshidrogenasa A de Aspergillus niger var. awamori, así como una nueva secuencia de ADN que codifica la glutamato deshidrogenasa (741-2245 en SEQ ID No. 1). Para la expresión de proteínas recombinantes resultan especialmente útiles las construcciones de ADN que comprenden el nuevo promotor, seguido de una secuencia de ADN que codifica una proteína de fusión formada por: un gen que codifique la proteína a obtener (por ejemplo la taumatina), una secuencia espaciadora tipo KEX2, y un gen altamente expresado de un hongo filamentoso. Entre las ventajas de este sistema están: altas concentraciones de proteína expresada, menores tiempos de fermentación, y utilización de una fuente de nitrógeno más económica.
