Abstract

Die Erfindung betrifft Verbindungen gemäß der allgemeinen Formel (1), sowie deren Verwendung zur Herstellung eines Arzneimittels insbesondere zur Behandlung einer Erkrankung, die mit Bromodomänen in Zusammenhang steht, wie Krebserkrankungen.

Inhibitoren für Bromodomänen
B e s c h r e i b u n g
Durch posttranslationale Modifikation kontrollieren Histonacetyltransferasen (HATs) und Histondeacetylasen (HDACs) den Acetylierungszustand von Lysinen im Chromatin. Hierdurch wird die Expression von Genen positiv oder negativ reguliert. Für diesen Vorgang sind hochselektive Acetyllysin-spezifische Protein-Protein-Wechselwirkungen von Bedeutung, die durch Protein- Interaktionsmodule orchestriert werden. Zu diesen gehören die Bromodomänen (BRD), die eine tiefe hydrophobe Acetyllysin-Bindungstasche aufweisen. Proteine mit zwei BRD und einer extraterminalen Domäne kennzeichnen die Bromo- und Extra-Terminale Domänen (BET) Familie, zu denen beispielsweise BET Familien-Proteine BRD2, BRD3, BRD4 und BRDT gehören. BET Proteine sind wichtig, da sie normale Transkriptionsvorgänge regulieren, bei verschiedenen Krebsarten bestimmen sie jedoch auch die Transkription von Onkogenen wie c-myc und Bcl-2. Eine selektive Inhibition der BET Proteine ist für Krebstherapien daher von großer Bedeutung. Unter anderem konnte gezeigt werden, dass in der akuten myeloischen Leukämie (AML) und myeloproliferativen Neoplasien (MPN) BRD4 als therapeutisches Ziel in Frage kommt.
In den Jahren 2012 und 2013 berichteten Fish et al. (J. Med. Chem. 2012, 55, 9831-9837) und Picaud et al. (Cancer Res. 2013, 73, 3336-3346) über die Identifizierung eines BET Inhibitors (PFI-1), der selektiv BRD4 adressiert. Die heterocyclischen Verbindungen werden ebenfalls in der WO 2013/027168 A1 beschrieben. Seither wurden weitere strukturell sehr ähnliche Moleküle beschrieben, die ebenso mit Bromodomänen wechselwirken, bevorzugt aber Vertreter der "Plant Homeodomain Finger-containing" Proteinfamilie inhibieren. Beispielsweise beschreiben die WO 2011/137089 A1 und WO 2010/042867 A2 Aktivatoren der humanan Pyruvatkinase. Weiter beschreibt die WO 2010/141074 A2 Inhibitoren der UDP-N-Acetylglucosamin-Peptid N-acetylglucosaminyltransferase (O-GlcNAc-Transferase). Die Verbindungen sind strukturell sehr ähnlich. Neben einem heterocyclischen Fragment auf der Ostseite verbindet eine sekundäre Sulfonamidgruppe den westlichen Teil des Moleküls, an dem unterschiedlich substituierte Aryle gebunden sind. Modifizierungen der Verbindungen konzentrieren sich hierbei auf Variationen der Ost- und Westhälften der Moleküle. Es besteht Bedarf an weiteren Verbindungen, die in der Behandlung von Krebserkrankungen und der Entwicklung neuer Medikamente verwendbar sind.
Aufgabe der vorliegenden Erfindung war es daher, neue Verbindungen bereitzustellen, die als Inhibitoren für Bromodomänen und damit zur Behandlung von Krebserkrankungen geeignet sind.
Diese Aufgabe wird gelöst durch die Verbindung gemäß der allgemeinen Formel (1) wie nachstehend angegeben und/oder deren Stereoisomere, Tautomere, Solvate, Flydrate und pharmazeutisch verträgliche Salze: worin:
X ist ausgewählt aus O oder N-R1, wobei R1 ausgewählt ist aus der Gruppe umfassend Wasserstoff, Cyano, C1-6- Alkyl und/oder jeweils unsubstituiertes oder einfach oder mehrfach mit C1-6-Alkyl substituiertes C3 -7-Cycloalkyl, Aryl oder Hetaryl;
Y ist ausgewählt aus der Gruppe umfassend C1-6-Alkyl, jeweils unsubstituiertes oder einfach oder mehrfach mit C1-6-Alkyl substituiertes C3 -7-Cycloalkyl, Aryl oder Hetaryl, NH2, NHR2 und/oder NR2R3, wobei R2 und R3 ausgewählt sind aus der Gruppe umfassend C1-6- Alkyl und/oder jeweils unsubstituiertes oder einfach oder mehrfach mit C1-2-Alkyl substituiertes C3 -7-Cycloalkyl, Aryl oder Hetaryl;
Z ist ausgewählt aus Cyclohexyl Aryl oder Hetaryl, jeweils unsubstituiert oder einfach oder mehrfach substituiert mit C1-6-Alkyl, C1-5-Alkoxy oder einfach oder mehrfach mit Halogen substituiertem C1-6-Alkyl; und Me ist Methyl. Es wurde gefunden, dass die Verbindungen eine ausgeprägte inhibitorische Aktivität in verschiedenen hämatopoetischen Zelllinien zeigen. Dies ist insbesondere unerwartet, da gegenüber dem bekannten BET Inhibitor PFI-1 signifikante Strukturunterschiede der zentralen Funktionalität bestehen, und insbesondere Änderungen zentraler funktionaler Gruppen wie der Sulfonamid-Gruppe von PFI-1 üblicherweise in einem Verlust der Aktivität resultieren. Eine Variation der Substituenten an den Stickstoffatomen der Sulfondiimid- bzw. der Sulfonimidamidgruppe oder eine Variation der Alkylgruppe der Sulfoximin- bzw. Sulfondiimidgruppe erlaubt eine Optimierung und Anpassung der Selektivität. Derartige Variationen sind bei Sulfonamidstrukturen nicht möglich.
Insbesondere eröffnen die Sulfoximin-, Sulfonimidamid- und Sulfondiimidgruppe die Nutzung der Stereochemie am zentralen Schwefelatom. Sulfoximin-, Sulfonimidamid- und Sulfondiimidgruppen weisen mit diesem ein Stereozentrum auf, dessen Substituenten ihre relative Tage zueinander nicht ändern können und hierdurch sind verschiedene räumliche Anordnungen möglich. Die Verbindungen der Formel (1) können daher in Form der Racemate, Diastereomere oder Enantiomerenpaare vorliegen. Die Stereoisomere können nach klassischen Methoden, beispielsweise mittels Hochleistungsflüssigkeitschromatographie (high performance liquid chromatography, HPLC), Racematspaltung oder asymmetrischer Synthese, erhalten werden. Die stereoisomeren Verbindungen können sich in Bezug auf ihre inhibitorische Aktivität unterscheiden. So konnte beispielsweise gezeigt werden, dass das (S)-Enantiomer eines Sulfoximines eine verglichen mit PFI-1 höhere inhibitorische Wirkung auf das Wachstum und die Viabilität von Feukämie-Zelllinien zeigte.
Weiterhin können Sulfonimidamide, Verbindungen, bei welchen der Substituent Y ausgewählt ist aus NH2, NHR2 oder NR2R3, auch in Form ihrer Tautomere vorliegen.
Der Begriff "C1-6-Alkyl" umfasst, wenn nicht abweichend angegeben, geradkettige oder verzweigte Alkylgruppen mit 1 bis 6 Kohlenstoffatomen, insbesondere ausgewählt aus Methyl, Ethyl, Propyl, Isopropyl, Butyl, tert-Butyl, Isobutyl, Pentyl, Isopentyl, Neopentyl, Hexyl und Isohexyl.
C3-7-Cycloalkylgruppen sind vorzugsweise ausgewählt aus der Gruppe umfassend Cyclopropyl, Cyclopentyl und/oder Cyclohexyl. C1-5- Alkoxy gruppen sind vorzugsweise ausgewählt aus der Gruppe umfassend Methoxy, Ethoxy, lineares oder verzweigtes Propoxy und/oder Butoxy.
Unter dem Begriff "Aryl" sind aromatische Reste mit 6 bis 10 Kohlenstoff-Atomen zu verstehen. Der Begriff "Aryl" umfasst bevorzugt Carbocyclen, insbesondere Phenyl.
Unter dem Begriff "Hetaryl" sind mono- oder bi-cyclische Heteroarylgruppen umfassend ein, zwei, drei oder vier Heteroatome ausgewählt aus der Gruppe umfassend N, O und/oder S zu verstehen. Bevorzugte Heteroarylgruppen sind monocyclische Heteroarylgruppen, insbesondere C5-6- Heteroaryle. Bevorzugte monocyclische Heteroarylgruppen weisen ein Heteroatom ausgewählt aus N, O oder S auf. Bevorzugte Heteroarylgruppen sind ausgewählt aus der Gruppe umfassend Pyridyl und/oder Pyrimidyl, wobei Pyridyl bevorzugt ist.
Der Begriff "Halogen" umfasst Fluor, Chlor, Brom und Jod, wobei Fluor oder Chlor bevorzugt sind.
Unter dem Begriff "Salz" sind im Sinne der vorliegenden Erfindung Formen der Verbindung zu verstehen, in denen diese eine ionische Form annimmt bzw. geladen ist und mit Kationen oder Anionen als Gegenion vorliegt oder sich in Fösung befindet. Insbesondere versteht man unter pharmazeutisch verträglichen Salzen im Sinne dieser Erfindung mit einer physiologisch verträglichen anorganischen bzw. organischen Base gebildete Salze, die physiologisch insbesondere bei Anwendung im Menschen verträglich sind. Pharmazeutisch verträgliche Salze der erfindungsgemäßen Verbindungen sind beispielsweise ausgewählt aus der Gruppe umfassend Natrium-, Kalium- und/oder Lithium-Salze. Unter dem Begriff "Salz" sind im Sinne der vorliegenden Erfindung insbesondere auch pharmazeutisch verträgliche Additionssalze zu verstehen, insbesondere Basenadditionssalze, beispielsweise Salze der Verbindungen mit anorganischen Basen wie Alkali- oder Erdalkalihydroxiden.
Eine bevorzugte Gruppe an Verbindungen sind Sulfoximine. Bei diesen Verbindungen ist der Substituent X Sauerstoff. In Ausführungsformen der Verbindung (1) sind die Substituenten:
X Sauerstoff; Y ausgewählt aus der Gruppe umfassend C1-6- Alkyl und/oder jeweils unsubstituiertes oder einfach oder mehrfach mit C1-2-Alkyl substituiertes C3 -7-Cycloalkyl, Aryl oder Hetaryl; und
Z ausgewählt aus Cyclohexyl, Aryl oder Hetaryl, jeweils unsubstituiert oder einfach oder mehrfach substituiert mit C1-6-Alkyl, C1-5-Alkoxy oder einfach oder mehrfach mit Halogen substituiertem C1-6-Alkyl.
In bevorzugten Ausführungsformen der Verbindung (1) sind die Substituenten:
X Sauerstoff;
Y ausgewählt aus der Gruppe umfassend C1-5- Alkyl und/oder jeweils unsubstituiertes oder einfach oder mehrfach mit C1-2-Alkyl substituiertes C5-6-Cycloalkyl oder Phenyl, vorzugsweise ausgewählt aus C1-3-Alkyl, insbesondere Methyl oder Ethyl; und
Z ausgewählt aus Cyclohexyl oder Phenyl, jeweils unsubstituiert oder einfach oder mehrfach substituiert mit C1-3-Alkoxy, insbesondere Cyclohexyl oder Phenyl jeweils unsubstituiert oder einfach substituiert mit Methoxy. Die Substitution ist dabei vorzugsweise in ortho-Position.
In bevorzugten Ausführungsformen ist die Verbindung ausgewählt aus der Gruppe umfassend die Verbindungen gemäß den nachstehenden Formeln (MF1), (MF2), (MF3) oder (MF4):
Die Verbindungen MF1, MF2 und MF3 zeigten eine ausgeprägte inhibitorische Aktivität in verschiedenen hämatopoetischen Zelllinien, insbesondere Erythroleukämie -Zelllinien und Zelllinien der akuten myeloischen Leukämie (AML). Insbesondere konnte eine zu PFI-1 vergleichbar hohe Aktivität der jeweiligen racemischen Gemische der Verbindungen MF1, MF2 und der achiralen Verbindung MF3 gezeigt werden.
In bevorzugten Ausführungsformen ist die Verbindung das (S)-Enantiomer der Verbindungen der Formeln (MF1) oder (MF2). In besonders bevorzugten Ausführungsformen ist die Verbindung das (S)-Enantiomer der Verbindung der Formel (MF2). Dieses weist die nachstehende Formel (MF2-S) auf:
Das (S)-Enantiomer der Verbindung der Formel (MF2) MF2-S zeigte eine hervorragende Inhibition der Zellproliferation hämatopoetischer Zelllinien. So konnte MF2-S in einer Konzentration von 1 μM signifikant stärker die metabolische Aktivität der Erythroleukämie -Zelllinie HEL (JAK2V617F positiv) und der AML -Linie Molm-14 (FLT3-ITD positiv) reduzieren als der bekannte BET-Inhibitor PFI-1. Das (S)-Enantiomer der Verbindung der Formel (MF2) zeigte weiterhin eine bessere Wirksamkeit in der Inhibition der Zellteilung und der Induktion des Zelltods in den HEL- und AML- Zelllinien. Somit kann mit MF2-S eine Verbindung zur Verfügung gestellt werden, die im Vergleich zu PFI-1 eine verbesserte Effizienz aufweist. Von besonderem Vorteil ist ferner, dass das (S)- Enantiomer der Verbindung der Formel (MF2) eine bessere Inhibition des Zellwachstums als PFI-1 in humanen Proben von AML-Patienten zeigte.
Die Verbindung kann auch das (R)-Enantiomer der Verbindung der Formel (MF2) entsprechend der Verbindung gemäß der nachstehenden Formel (MF2-R) sein:
MF2-R zeigte eine Inhibition der Zellproliferation von Feukämie-Zelllinien, während MF2-S eine höhere Effizienz aufwies. In weiteren Ausführungsformen der Sulfoximine kann der Substituent Z, insbesondere Cyclohexyl oder Phenyl, jeweils einfach mit Methoxy substituiert sein. Die Substitution ist dabei vorzugsweise in ortho- Position. Das Sulfoximin kann eine Struktur gemäß der nachstehenden Formel (MF1) aufweisen:
Das racemische Gemisch der Verbindung (MF1) zeigte ebenfalls eine inhibitorische Aktivität in den hämatopoetischen Zelllinien, insbesondere Erythroleukämie -Zelllinien und Zelllinien der akuten myeloischen Leukämie (AML).
Vorzugsweise ist die Verbindung das (S)-Enantiomer der Verbindung der Formel (MF1). Dieses weist die nachstehende Formel (MF1-S) auf:
Das (S)-Enantiomer MF1-S, zeigte einen vergleichbaren Effekt auf die metabolische Aktivität der hämatopoetischen Zelllinien von HEL- und Molm-14-Zellen wie PFI-1.
Die Verbindung kann weiter das (R)-Enantiomer der Verbindung der Formel (MF1) sein. Dieses weist die nachstehende Formel (MF1-R) auf:
Das (R)-Enantiomer MF1-R zeigte eine geringe inhibitorische Wirkung in hämatopoetischen Zelllinien. In bevorzugten Ausführungsformen ist die Verbindung eines der Enantiomere der Verbindung der Formel (MF4) ausgewählt aus (+)-N-(3- Methyl -2-oxo- 1 ,2,3,4-tetrahydrochinazolin-6-yl)-S- cycIohexyI-5-methyIsuIfoximin und (-)-N-(3- Methyl -2-oxo- 1 ,2,3,4-tetrahydrochinazolin-6-yl)-S- cycIohexyI-5-methyIsuIfoximin.
Eine weitere bevorzugte Gruppe an Verbindungen sind Sulfondiimide. Bei diesen Verbindungen ist der Substituent X eine Gruppe N-R1. In diesen Ausführungsformen der Verbindung (1) sind die Substituenten:
X ist N-R1, wobei R1 ausgewählt ist aus der Gruppe umfassend Wasserstoff, Cyano, C1-6- Alkyl und/oder jeweils unsubstituiertes oder einfach oder mehrfach mit C1-2- Alkyl substituiertes C3 -7-Cycloalkyl, Aryl oder Hetaryl;

In bevorzugten Ausführungsformen der Verbindung (1) sind die Substituenten:

In bevorzugten Ausführungsformen ist die Verbindung ausgewählt aus der Gruppe umfassend die Verbindungen gemäß den nachstehenden Formeln (JHS1), (JHS2) oder (JHS3) oder ist eines deren Stereoisomere:
Die Verbindung (JHS1) zeigte ebenfalls eine inhibitorische Aktivität in den hämatopoetischen Zelllinien, insbesondere Erythroleukämie -Zelllinien und Zelllinien der akuten myeloischen Leukämie (AML). So konnte JHS1 die metabolische Aktivität von HEL- und Molm-14-Zellen bei Konzentrationen von 10 und 20 μM inhibieren.
In weiter bevorzugten Ausführungsformen ist die Verbindung das (S)-Enantiomer der Verbindung der Formel (JHS1). Dieses weist die nachstehende Formel (JHS1-S) auf:
Das (S)-Enantiomer JHS1-S zeigte einen vergleichbaren Effekt auf die metabolische Aktivität der hämatopoetischen Zelllinien von HEL- und Molm-14-Zellen wie PFI-1.
Auch bevorzugt ist das (R)-Enantiomer der Verbindung der Formel (JHS1). Dieses weist die nachstehende Formel (JHS1-R) auf:
Das (R)-Enantiomer JHS1-R zeigte eine nahezu gleich hohe inhibitorische Wirkung in hämatopoetischen Zelllinien wie das (5)-Enantiomer JHS1-S.
Weitere Sulfondiimide können ausgewählt sein aus den Verbindungen gemäß den Formeln (JHS2) und (JHS3). Diese Verbindungen zeigten eine geringe inhibitorische Aktivität in den untersuchten Leukämie -Zelllinien.
Eine weitere bevorzugte Gruppe an Verbindungen sind Sulfonimidamide und deren Tautomere. Bei diesen Verbindungen ist der Substituent Y eine Aminogruppe NH2, NHR2 oder NR2R3. In diesen Ausführungsformen der Verbindung (1) sind die Substituenten:

In bevorzugten Ausführungsformen der Verbindung (1) sind die Substituenten:

In bevorzugten Ausführungsformen ist die Verbindung ausgewählt aus der Gruppe umfassend die Verbindung gemäß der nachstehenden Formel (JHS4) oder eines deren Enantiomere:
Die Verbindungen sind als Inhibitoren der Bromodomäne, insbesondere der Proteinfamilie der BET- Proteine z. B. BRD2-4, die die Initiation und Elongation der Gen-Transkription und das Zellwachstum beeinflussen, zur Behandlung von Erkrankung, die mit Bromodomänen in Zusammenhang stehen, geeignet. Hierzu zählen Autoimmunerkrankungen, inflammatorische Erkrankungen und Krebserkrankungen. Eine Verwendung der Verbindungen in der Behandlung von Autoimmunerkrankungen, inflammatorischen Erkrankungen und von Krebserkrankungen, insbesondere durch BET-Proteine vermittelten krebsartigen Zellwachstums ist daher ein weiterer Aspekt der vorliegenden Erfindung. Unter dem Begriff "Krebserkrankung" werden im Sinne der vorliegenden Erfindung allgemein bösartige (maligne) Erkrankungen, die durch eine unkontrollierte Vermehrung von veränderten Zellen gekennzeichnet sind, verstanden.
Ein weiterer Gegenstand der Erfindung betrifft eine Verwendung der Verbindungen zur Herstellung eines Arzneimittels, insbesondere zur Herstellung eines Arzneimittels zur Behandlung einer Erkrankung, die mit Bromodomänen in Zusammenhang steht. Erkrankungen, die mit Bromodomänen in Zusammenhang stehen sind vorzugsweise ausgewählt aus Autoimmunerkrankungen, inflammatorischen Erkrankungen und Krebserkrankungen, insbesondere der akuten myeloischen Leukämie oder myeloproliferativen Neoplasien. Ein entsprechender Gegenstand der Erfindung betrifft eine erfindungsgemäße Verbindung zur Verwendung als Arzneimittel, insbesondere zur Verwendung in der Behandlung einer Erkrankung, die mit Bromodomänen in Zusammenhang steht, vorzugsweise von Autoimmunerkrankungen, inflammatorischen Erkrankungen und Krebserkrankungen, insbesondere der akuten myeloischen Leukämie oder myeloproliferativen Neoplasien. Krebserkrankungen sind insbesondere Knochenmarkerkrankungen bzw. Leukämien ("Blutkrebs"). Als Leukämie werden bösartige Erkrankungen bezeichnet, die zumeist im Knochenmark bzw. hämatopoetischen Stammzellen ihren Ursprung haben und die zumeist zuerst im Blut nachgewiesen werden können. Hierbei kann ebenfalls zwischen akuten und chronischen Leukämien unterschieden werden. Beispiele dieser Knochenmarkserkrankungen sind die akute lymphatische Leukämie (ALL) und chronische lymphatische Leukämie (CLL), die die lymphozytären Zellen beeinflussen, und die akute myeloische Leukämie (AML) oder das myelodysplastische Syndrom (MDS). Unter dem Begriff myeloproliferative Neoplasien (MPN) wird eine Gruppe seltener, bösartiger Erkrankungen des Knochenmarks zusammengefasst, bei denen zu viele rote Blutkörperchen, weiße Blutkörperchen und/oder Blutplättchen gebildet werden. Zu den MPN gehört die Philadelphia-Chromosom-positive chronisch myeloische Leukämie (CML) und die Philadelphia-Chromosom-negativen Polycythaemia Vera (PV), Essentielle Thrombozythämie (ET) und die primäre Myelofibrose (PMF), wobei aus der PV und der ET eine Myelofibrose entstehen kann, was das Krankheitsbild drastisch verschlechtert. Eher seltener kann sich aus einer MPN eine akute Leukämie entwickeln. In unterschiedlichen Formen der Leukämie und myeloproliferativer Erkrankungen wurden BET Proteine als mögliche Ziele einer Therapie, sowohl Einzel-Therapien als auch Kombinations-Therapien, ausgemacht.
Die Behandlung kann eine therapeutische und/oder prophylaktische Behandlung beinhalten·
Bevorzugt ist eine Behandlung einer Erkrankung, die mit Bromodomänen in einem Säuger in Zusammenhang steht.
Für die Beschreibung der Verbindungen, insbesondere gemäß der allgemeinen Formel (1), wird auf die vorstehenden Ausführungen verwiesen. Eine bevorzugte Gruppe an Verbindungen sind Sulfoximine, vorzugsweise die Verbindungen gemäß der Formel (MF2), insbesondere die (S)- Enantiomere der Formeln (MF2-S), oder der Formel (MF1) insbesondere das (S)-Enantiomer der Formel (MF1-S). Eine weitere bevorzugte Gruppe an Verbindungen sind Sulfondiimide, vorzugsweise die Verbindung der Formel (JHS1), insbesondere das (S)-Enantiomer der Formel (JHS1-S) oder das (R)-Enantiomer der Formel (JHS1-R). Eine weitere bevorzugte Gruppe an Verbindungen sind Sulfonimidamide, bei welchen der Substituent Y eine Aminogruppe NH2, NHR2 oder NR2R3 oder deren tautomere Form ist, vorzugsweise die Verbindungen gemäß der Formel (JHS4). Ein weiterer Gegenstand der Erfindung betrifft eine pharmazeutische Zusammensetzung oder ein Arzneimittel enthaltend wenigstens eine erfindungsgemäße Verbindung, insbesondere zur Verwendung in der Behandlung einer Erkrankung, die mit Bromodomänen in Zusammenhang steht, vorzugsweise von Krebserkrankungen, insbesondere der akuten myeloischen Leukämie oder myeloproliferativen Neoplasien.
Es konnte festgestellt werden, dass die Verbindungen gemäß der allgemeinen Formel (1) eine ausgeprägte inhibitorische Aktivität in verschiedenen hämatopoetischen Zelllinien zeigen. Diese Effekte erlauben es, die Verbindungen in Arzneimitteln zur Behandlung von Erkrankungen, die mit Bromodomänen in Zusammenhang steht, vorzugsweise von Autoimmunerkrankungen, inflammatorischen Erkrankungen und Krebserkrankungen, insbesondere der akuten myeloischen Leukämie oder myeloproliferativen Neoplasien, zu verwenden. Für die Beschreibung der Erkrankungen wird auf die vorstehenden Ausführungen verwiesen.
Für die Beschreibung der Verbindungen, insbesondere gemäß der allgemeinen Formel (1), wird auf die vorstehenden Ausführungen verwiesen. Vorzugsweise enthält die pharmazeutische Zusammensetzung oder das Arzneimittel ein Sulfoximin, vorzugsweise die Verbindungen gemäß der Formel (MF2), insbesondere die (S)-Enantiomere der Formeln (MF2-S), oder der Formel (MF1) insbesondere das (S)-Enantiomer der Formel (MF1-S). Weiter bevorzugt enthält die pharmazeutische Zusammensetzung oder das Arzneimittel ein Sulfondiimid, vorzugsweise die Verbindung der Formel (JHS1), insbesondere das (S)-Enantiomer der Formel (JHS1-S) oder das (R)-Enantiomer der Formel (JHS1-R). Weiter bevorzugt enthält die pharmazeutische Zusammensetzung oder das Arzneimittel ein Sulfonimidamid, bei welchen der Substituent Y eine Aminogruppe NH2, NHR2 oder NR2R3, oder deren tautomere Form ist, vorzugsweise die Verbindungen gemäß der Formel (JHS4).
Die Zusammensetzung oder das Arzneimittel können neben der Verbindung als Wirkstoff weiterhin pharmazeutisch verträgliche Hilfsstoffe und/oder Träger umfassen. Die Art der Zusatz-, Träger- und/oder Hilfsstoffe hängt von der gewünschten Verabreichungsart ab. Orale Mittel können beispielsweise als Tablette, Filmtablette oder Kapsel, auch in retardierter Form, vorliegen und können übliche Exzipienzien enthalten, wie Bindemittel, Füllstoffe, Gleitmittel, desintegrierende Mittel wie Stärke oder Netzmittel. Vorzugsweise ist eine Tablette in Wasser lösbar. Orale flüssige Präparate können in Form wässriger oder öliger Suspensionen, Lösungen, Emulsionen, Sirupen, Elixieren oder Sprays vorliegen. Als Trägersubstanzen sind beispielsweise organische oder anorganische Stoffe verwendbar, die sich für eine enterale, beispielsweise orale oder rektale, oder parenterale Applikation eignen und mit den Verbindungen nicht reagieren, beispielsweise Wasser, pflanzliche Öle, Benzylalkohole, Polyethylenglykole, Glycerintriacetat und andere Fettsäureglyceride, Gelatine, Sojalecithin, Kohlenhydrate wie Lactose oder Stärke, Magnesiumstearat, Talkum oder Cellulose.
Das Arzneimittel oder die Zusammensetzung kann als flüssige, halbfeste oder feste Arzneiform, beispielsweise in Form von Injektionslösungen, Tropfen, Säften, Sirupen, Sprays, Suspensionen, Tabletten, Kapseln oder in Form von Pellets oder Granulaten vorliegen und/oder verabreicht werden. Für eine orale Verabreichung eignen sich bevorzugt Zubereitungen in Form von Tabletten, Dragees, Kapseln oder Granulaten. Das Arzneimittel oder die Zusammensetzung kann sterilisiert sein. Die Zusammensetzung oder das Arzneimittel kann oral, rektal, pulmonal, enteral und/oder parenteral verabreichbar sein.
Wenn nicht anders ausgeführt, weisen die verwendeten technischen und wissenschaftlichen Ausdrücke die Bedeutung auf, wie sie gemeinhin von einem Durchschnittsfachmann in dem Gebiet, zu dem diese Erfindung gehört, verstanden wird.
Beispiele und Figuren, die der Veranschaulichung der vorliegenden Erfindung dienen, sind nachstehend angegeben.

Methoden:
Patientenmaterial
Peripheres Blut und Knochenmark der Patienten wurde durch die Klinik für Flämatologie, Onkologie, Flämostaseologie und Stammzelltransplantation (Med. Klinik IV) (EK 127/12) und der zentralisierten Biomaterialbank, Universitätsklinikum Aachen, zur Verfügung gestellt. Die gesunden Kontrollen wurden vom Institut für Transfusionsmedizin (EK099/149), Universitätsklinikum Aachen, zur Verfügung gestellt. Alle Donoren/Patienten haben eine schriftliche Einverständniserklärung abgegeben.
Zellkultur
Alle Arbeiten mit den genannten Zelllinien FIEL, Molm-14- und K562 (vom DSMZ bezogen: ACC- 11, ACC-10 und ACC-777), wurden unter keimfreien Bedingungen unter der Sterilbank durchgeführt. Die Inkubation erfolgte bei 37 °C im Brutschrank bei einer C02-Sättigung von 5%. Für die Passagierung der Suspensionszellen wurden die Zellen in einem Falcon mit vorgelegtem PBS (phosphate buffered saline) überführt und bei 5 min bei 1200 rpm zentrifugiert. Alle drei Zelllinien wurden in RPMI-1640 Medium (Gibco, Thermo Fischer Scientific, Darmstadt, Germany) mit 10% FCS und 1 % Penicillin-Streptomycin kultiviert.
Zellzahlbestimmung und Proliferationsassay
Die Bestimmung der Zellzahl und der Viabilität erfolgte nach Trypan-Blau Färbung. Pro mL Medium wurden die entsprechenden Konzentrationen der Inhibitoren hinzugegeben bzw. das entsprechend höchste Volumen an DMSO hinzugegeben als Kontrollgruppe.
MTT-Assay
Die Viabilität und Stoffwechselaktivität der Zellen wurde mit dem MTT-Assay gemessen. Das gelbe, wasserlösliche MTT (3-(3,5-dimethylthiazol-2-yl)-2,5-dephenyltetrazolium bromide) ist ein Tetrazoliumsalz, das in lebenden Zellen in das dunkelblaue Formazan reduziert wird. Das unlösliche Formazan wird durch Zugabe von Isopropanol-HCl freigesetzt und seine Intensität kann photometrisch bei einer Wellenlänge von 550 nm bestimmt werden. Somit wird der Anteil lebender Zellen im Vergleich zur einer bestimmten Kontrollprobe gemessen.
Die Zellen wurden gezählt, mit PBS gewaschen und 30.000 Zellen wurden pro Well in einer 96-Well- Platte in Triplikaten ausgesät. Für die Inhibitorbehandlung wurden die angegebenen Endkonzentrationen eingesetzt. Dafür wurden 10 μL der entsprechenden Inhibitor- Verdünnung in die 96-Well-Platten vorgelegt und anschließend 90 μL Zellsuspension hinzu pipettiert. Die Platten wurden für 72 h im Brutschrank (37 °C, 5% CO2) inkubiert. Nach der Inkubation wurden 10 μL MTT-Lösung (5 mg/mL) mit einer Mehrkanalpipette auf die Wehs gegeben und gründlich resuspendiert. Danach erfolgte eine Inkubation für 4 h im Dunkeln bei Raumtemperatur. Anschließend wurden die Zellen in 100 μL Isopropanol-HCl resuspendiert, lysiert und die Absorption wurde photometrisch mit dem Plate Reader (Kayto, RT-2100C) bei einer Wellenlänge von 550 nm gemessen.
Zellzyklus-Analyse
Der Einfluss der eingesetzten Inhibitoren auf den Zellzyklus wurde mittels Propidium-Iodid-Färbung der fixierten Zellen untersucht. Die Zellen wurden hierfür mit 95% Ethanol bei -20 °C fixiert, RNA wurde durch RNase (100 μg/mL) für 15 min bei 37 °C zerstört und die DNA mittels Propidium-Iodid (50 μg/mL) angefärbt. An einem Durchflusszytometer (Gallios (Beckman Coulter, Krefeld, Germany) wurden die Zellzyklusphasen analysiert und die Daten mittels FlowJo Software (Version 10) ausgewertet.
SDS-Page und Western Blot
Zelllysate wurden mit RIPA Lysepuffer hergestellt (50 mM Tris pH 7.4, 150 mM sodium Chloride, 1 mM EDTA, 1 % Triton-X, 15 % glycerol, 0.5 % sodium deoxycholate, and Protease/Phosphatase Inhibitoren). Protein-Konzentrationen wurden mit dem Bradford Protein Assay bestimmt an einem Nanodrop Spectrometer (NanoDrop 2000/2000c System, Thermo Fisher Scientific, Darmstadt, Germany) bei 595 nm. 25 μg der Proteinlysate wurde mit 4xLämmli-Probenpuffer versetzt, 5 min bei 65 °C denaturiert und in die SDS-Page eingesetzt. Die Proteine wurden im Wet-Blot-Verfahren in Towbin-Puffer (3 g Tris, 14.4 g Glycine, 5 % Methanol pro Liter ddH2O) auf eine PVDF-Membran übertragen bei 100 mA über Nacht. Die Membran wurde mit 10% BSA in TBS-T-Puffer (20 mM Tris-HCl, pH 7.6, 137 mM NaCl, 0.05 % IGEPAL) blockiert. Die primären Antikörper wurden über Nacht bei 4 °C und der HRP-gelabelte 45 min bei Raumtemperatur mit der Membran inkubiert. Die Proteine wurden mittels PCA-ECL-Lösung an einem Chemilumineszenz-Detektor (Fusion SL, PeqLab) detektiert.
CFU- Assay
Mit Hilfe des CFU Assays kann das Koloniebildungsvermögen einzelner Zellen in einem semi-soliden Methylcellulose-Medium (MethoCult H4434, StemCell Technologies, Köln, Germany) bestimmt werden. Die Anzahl der Kolonien wurde nach 7-11 Tagen bestimmt.
Unterschiedliche Zellzahlen wurden eingesetzt:
Mononukleäre Zellen des Knochenmarks aus AML-Patienten: 7x105Zellen/mL PBMCs von PV, ET bzw PMF Patienten: 4x105Zellen/mL
CD34+ Zehen (G-CSF mobilisiert von Stammzellspendern) der gesunden Donoren: 2x104Zellen/mL; PBMCs der gesunden Probanden: 4x105 Zellen/mL.
Vital sterile mononukleäre Knochenmarkszellen von AML-Patienten wurden für 24 h in 1.5 mL BIT Medium in 8.5 mL IMDM Medium (versetzt mit 5 μL FLT3 ligand, 10 μL rh SCF, 2 μL rh IL-3, 2 μL rh G-CSF, 50 μL Ciprofloxacin und 6.99 μL ß Mercaptoethanol in 1: 100 Verdünnung) kultiviert, bevor sie in den CFU-Assay eingesetzt wurden. Schmelzpunkte (Smp.)
Die Schmelzpunkte wurden in offenen Kapillaren in einem Metallheizblock mit Digitalthermometer ermittelt.
Kernspinresonanz (NMR)-Daten
NMR-Messungen erfolgten für die Kerne 1 H und 13C bei den Messfrequenzen 600 MHz oder 400 MHz (1H) bzw. 151 MHz oder 101 MHz (13C) in deuterierten Lösungsmitteln wie z. B. Chloroform, Dichlormethan oder Dimethylsulfoxid. 13C-NMR-Spektren wurden ausnahmslos 1 H- breitbandentkoppelt gemessen.
Infrarotspektroskopische (IR) Daten
Die Probenmessung erfolgte mit abgeschwächter Totalreflexion (ATR).
Massenspektrometrische (MS) Daten
Die Daten wurden durch Elektronenstoß-Ionisation (EI), chemische Ionisation (CI) oder Elektrospray- Ionisation (ESI) erhoben.
Polarimetrie
Die Messungen wurden bei Raumtemperatur mit monochromatischer Strahlung der Wellenlänge λ = 589 nm in einer Küvette der Länge d = 1 dm durchgeführt. Die spezifischen Dreh werte [α]D tragen die Einheit deg cm3 dm -1 g-1. Die Konzentration c wurde mit der Einheit g (100 mL) -1 aufgeführt.
Hochleistungsflüssigkeitschromatographie (HPLC)
Für die analytische HPLC wurden Säulen mit chiraler stationärer Phase eingesetzt, die eine Länge von 250 mm und einen Durchmesser von 4.6 mm besaßen. Enantiomerenüberschüsse (ee) wurden aus den Enantiomerenverhältnissen der HPLC-Chromatogramme berechnet.
Chemikalien
6-Brom-3-methyl-3,4-dihydrochinazolin-2( 1 H)-on wurde hergestellt wie in WO 2013/027168 A1 beschrieben. Die Sulfoximine, achiral, razemisch oder ihre jeweiligen (S)- und (R)-Enantiomere, wurden hergestellt nach Methoden wie in Angew. Chem. 2016, 128, 7319-7323 oder Chem. Commun. 2017, 53, 348-351 oder Tetrahedron: Asymmetry 1997, 8, 909-912 oder Acta Chem. Scand. 1996, 50, 305-315 beschrieben. Die Sulfondiimide wurden hergestellt nach Methoden wie in Angew. Chem. 2012, 124, 4516-4519 beschrieben. Die Sulfonimidamide wurden hergestellt nach Methoden wie in Chem. Eur. J. 2017, 23, 15189-15193 beschrieben.
Beispiel 1: Herstellung der Verbindungen
1.1 Herstellung von (3-Methyl-2-oxo-l,2,3,4-tetrahydrochinazolin-6-yl)boronsäure
Ein Dreihalskolben wurde mit [1,1'-Bis(diphenylphosphino)ferrocen]dichlorpalladium(II) (0,02 Äquiv.) und 2-Dicyclohexylphosphino-2‘,6‘-dimethoxybiphenyl (0,04 Äquiv.) befüllt. Nach Zugabe von trockenem Ethanol (c = 0.2 mol/L) wurde 20 min unter Argon bei einer Ölbadtemperatur von 95 °C gerührt. Zu dem abgekühlten Gemisch wurden im Argon-Gegenstrom 6-Brom-3-methyl-3,4- dihydrochinazolin-2( 1 H)-on, Kaliumacetat (2,9 Äquiv.) und Tetrahydroxydiboron (3,0 Äquiv.) hinzugefügt. Es wurde 5 h bei einer Ölbadtemperatur von 95 °C gerührt und anschließend über Nacht bei 50 °C. Kontrolle über Dünnschichtchromatographie (Ethylacetat/ Aceton = 1:1) zeigte unvollständigen Umsatz an, so dass weiteres Tetrahydroxydiboron (0,5 Äquiv.) hinzugegeben und erneut bei einer Ölbadtemperatur von 95 °C gerührt wurde. Das Reaktionsgemisch wurde auf Raumtemperatur abgekühlt, mit Wasser versetzt und 1 h gerührt. Der Feststoff wurde abgesaugt und NMR-spektroskopisch untersucht. Das Volumen des Filtrats wurde im Membranpumpenvakuum bei einer Wasserbadtemperatur von 40 °C reduziert, bis erneut Feststoff ausfiel, der wieder abgesaugt und NMR-spektroskopisch analysiert wurde. Diese Prozedur wurde mehrfach wiederholt. Hinreichend saubere Feststoff-Fraktionen wurden vereinigt und so das Produkt als sandfarbener Feststoff mit einem Schmelzpunkt >240 °C (Zersetzung) isoliert. Die Verbindung wurde mittels Kernspinresonanzspektroskopie- und Massenspektrometrie-Untersuchungen bestätigt.
Synthese vor Schriften und Analytik der Sulfoximine:
1.2 Herstellung von rac-N-(3-Methyl-2-oxo-1,2,3,4-tetrahydrochinazolin-6-yl)-S-(2-methoxyphenyl)- S-methylsulfoximin (MF1)
Ein Druckröhrchen wurde mit 5-(2-Methoxyphenyl)-5-methylsulfoximin (1.6 Äquiv.), 6-Brom-3- methyl-3,4-dihydrochinazolin-2(1H)-on, Cäsiumcarbonat (1.6 Äquiv.), Tris(dibenzylidenaceton)dipalladium(0) (0.13 Äquiv.), 2-Dicyclohexylphosphino-2’,4’,6’- triisopropylbiphenyl (0.22 Äquiv.) und einem Magnet-Rührfisch befüllt. Das Druckröhrchen wurde im Hochvakuum evakuiert und mit Argon belüftet. Im Argon-Gegenstrom wurde trockenes 1 ,4-Dioxan (c = 0.06 mol/L) hinzugegeben und das Reaktionsgemisch 22 h bei einer Ölbadtemperatur von 110 °C gerührt. Die abgekühlte Suspension wurde über Kieselgur abgesaugt und die Kieselgur-Schicht mit Ethylacetat gewaschen. Das Lösungsmittel des Filtrats wurde im Membranpumpenvakuum bei einer Wasserbadtemperatur von 50 °C entfernt. Das Rohprodukt wurde zweimal säulenchromatographisch (Kieselgel, Aceton bzw. Diethylether/ Aceton = 2:1) aufgearbeitet. Das Produkt wurde mit wenig Diethylether versetzt und die Suspension im Ultraschallbad behandelt. Der Diethylether wurde abpipettiert und das Produkt als weißer Feststoff mit einem Schmelzpunkt von 194-196 °C isoliert.
Die Verbindung wurde mittels Infrarotspektroskopie-, Kernspinresonanzspektroskopie- und Massenspektrometrie-Untersuchungen bestätigt.
1.3 Herstellung von (S)-N-(3- Methyl -2-oxo- 1 , 2,3, 4-tetrahydrochinazolni -6-yl )-.S-(2-methoxy phenyl)- S-methylsulfoximin (MF1-S)
Ein Reagenzglas wurde mit (S)-S-(2-Methoxyphenyl)-S-methylsulfoximin, (3-Methyl-2-oxo-1, 2,3,4- tetrahydrochinazolin-6-yl)boronsäure (1.8 Äquiv.), Kupfer(II)acetat (0.17 Äquiv.) und einem Magnet- Rührfisch befüllt. In das Reagenzglas wurde trockenes Methanol (c = 0.26 mol/L) gegeben. Das Reagenzglas wurde in einen Rundkolben gestellt, auf den ein mit Calciumchlorid gefülltes Trockenrohr gesetzt wurde. Das Reaktionsgemisch wurde bei Raumtemperatur 43 h an der Luft gerührt. Das Lösungsmittel wurde im Membranpumpenvakuum bei einer Wasserbadtemperatur von 50 °C entfernt. Der verbliebene Rückstand wurde säulenchromatographisch (Kieselgel,
Diethylether/ Aceton = 2:1) behandelt und das Produkt als hellbrauner Feststoff mit einem Schmelzpunkt >218 °C (Zersetzung) erhalten. Die Verbindung wurde mittels Kernspinresonanzspektroskopie-Untersuchungen bestätigt. Der spezifische Drehwert [α ]D betrug +279,3 (c = 0,51 in CHCl3).
1.4 Herstellung von (R)-N-(3-Methyl-2-oxo- 1 ,2,3,4-tetrahydrochinazolin-6-yl)-5-(2-methoxyphenyl)- 5-methylsulfoximin (MF1-R) Ein Reagenzglas wurde mit (R)-S-(2-Methoxyphenyl)-S-methylsulfoximin, (3-Methyl-2-oxo-1, 2,3,4- tetrahydrochinazolin-6-yl)boronsäure (1.8 Äquiv.), Kupfer(II)acetat (0.15 Äquiv.) und einem Magnet- Rührfisch befüllt. In das Reagenzglas wurde trockenes Methanol (c = 0.25 mol/L) gegeben. Das Reagenzglas wurde in einen Rundkolben gestellt, auf den ein mit Calciumchlorid gefülltes Trockenrohr gesetzt wurde. Das Reaktionsgemisch wurde bei Raumtemperatur 22 h an der Luft gerührt. Das Lösungsmittel wurde im Membranpumpenvakuum bei einer Wasserbadtemperatur von 50 °C entfernt. Der verbliebene Rückstand wurde säulenchromatographisch (Kieselgel,
Diethylether/ Aceton = 2:1) behandelt und das Produkt als hellbrauner Peststoff mit einem Schmelzpunkt >212 °C (Zersetzung) erhalten. Die Verbindung wurde mittels Kernspinresonanzspektroskopie-Untersuchungen bestätigt. Der spezifische Drehwert [α ]D betrug -270, 1 (c = 0,47 in CHCl3) .
1.5 Herstellung von rac-N-(3-Methyl-2-oxo-1,2,3,4-tetrahydrochinazolin-6-yl)-5-methyl-5- phenylsulfoximin (MF2) Ein Druckröhrchen wurde mit S-Methyl-S-phenylsulfoximin (1.7 Äquiv.), 6-Brom-3-methyl-3,4- dihydrochinazolin-2(1H)-on, Cäsiumcarbonat (2.2 Äquiv.), Tris(dibenzylidenaceton)dipalladium(0) (0.1 Äquiv.), 2-Dicyclohexylphosphino-2’,4’,6’-triisopropylbiphenyl (0.14 Äquiv.) und einem Magnet-Rührfisch befüllt. Das Druckröhrchen wurde im Hochvakuum evakuiert und mit Argon belüftet. Im Argon-Gegenstrom wurde trockenes 1 ,4-Dioxan (c = 0.2 mol/L) hinzugegeben und das Reaktionsgemisch 42 h bei einer Ölbadtemperatur von 130 °C gerührt. Die abgekühlte Suspension wurde über Kieselgur abgesaugt und die Kieselgur-Schicht mit Ethylacetat gewaschen. Das Lösungsmittel des Filtrats wurde im Membranpumpenvakuum bei einer Wasserbadtemperatur von 50 °C entfernt. Das Rohprodukt wurde zweimal säulenchromatographisch (Kieselgel,
Diethylether/ Aceton = 1:1 bzw. Diethylether/ Aceton = 2:1) behandelt. Das Produkt wurde als gelber Feststoff mit einem Schmelzpunkt von 196-197 °C isoliert. Die Verbindung wurde mittels Infrarotspektroskopie-, Kernspinresonanzspektroskopie- und Massenspektrometrie-Untersuchungen bestätigt.
1.6 Herstellung von (S)-N-(3-Methyl-2-oxo-1,2,3,4-tetrahydrochinazolin-6-yl)-S-methyl-S- phenylsulfoximin (MF2-S)
Ein Reagenzglas wurde mit (S)-S-Methyl-S-phenylsulfoximin, (3-Methyl-2-oxo-1, 2,3,4- tetrahydrochinazolin-6-yl)boronsäure (2.0 Äquiv.), Kupfer(II)acetat (0.11 Äquiv.) und einem Magnet- Rührfisch befüllt. In das Reagenzglas wurde trockenes Methanol (c = 0.3 mol/L) gegeben. Das Reagenzglas wurde in einen Rundkolben gestellt, auf den ein mit Calciumchlorid gefülltes Trockenrohr gesetzt wurde. Das Reaktionsgemisch wurde bei Raumtemperatur 40 h an der Luft gerührt. Das Lösungsmittel wurde im Membranpumpenvakuum bei einer Wasserbadtemperatur von 50 °C entfernt. Der verbliebene Rückstand wurde säulenchromatographisch (Kieselgel,
Diethylether/ Aceton = 2:1) behandelt und das Produkt als hellgelber Feststoff mit einem Schmelzpunkt von 135-137 °C erhalten. Die Verbindung wurde mittels
Kernspinresonanzspektroskopie-Untersuchungen bestätigt. Der spezifische Drehwert [α ]D betrug +136,9 (c = 0,35 in CHCl3). 1.7 Herstellung von (R)-N-(3-Methyl-2-oxo-1,2,3,4-tetrahydrochinazolin-6-yl)-S-methyl-S- phenylsulfoximin (MF2-R)
Ein Reagenzglas wurde mit (R)-S-Methyl-S-phenylsulfoximin, (3-Methyl-2-oxo-1,2,3,4- tetrahydrochinazolin-6-yl)boronsäure (2.3 Äquiv.), Kupfer(II)acetat (0.11 Äquiv.) und einem Magnet- Rührfisch befüllt. In das Reagenzglas wurde trockenes Methanol (c = 0.3 mol/L) gegeben. Das Reagenzglas wurde in einen Rundkolben gestellt, auf den ein mit Calciumchlorid gefülltes Trockenrohr gesetzt wurde. Das Reaktionsgemisch wurde bei Raumtemperatur 23 h an der Luft gerührt. Das Lösungsmittel wurde im Membranpumpenvakuum bei einer Wasserbadtemperatur von 50 °C entfernt. Der verbliebene Rückstand wurde zweimal säulenchromatographisch (Kieselgel, Diethylether/ Aceton = 2:1 bzw. Dichlormethan/ Aceton = 1:1) behandelt und das Produkt als weißer Leststoff mit einem Schmelzpunkt von 133-135 °C erhalten. Die Verbindung wurde mittels Kernspinresonanzspektroskopie-Untersuchungen bestätigt. Der spezifische Drehwert [α ]D betrug -100,7 (c = 0,29 in CHCl3).
1.8 Herstellung von N-(3-Methyl-2-oxo-1,2,3,4-tetrahydrochinazolin-6-yl)-5,5-diphenylsulfoximin (MF3)
Ein Druckröhrchen wurde mit 5,5-Diphenylsulfoximin, 6-Brom-3-methyl-3,4-dihydrochinazolin- 2(1H)-on (1.7 Äquiv.), Cäsiumcarbonat (2.1 Äquiv.), Tris(dibenzylidenaceton)dipalladium(0) (0.11 Äquiv.), 2-Dicyclohexylphosphino-2’,4’,6’-triisopropylbiphenyl (0.18 Äquiv.) und einem Magnet- Rührfisch befüllt. Das Druckröhrchen wurde im Hochvakuum evakuiert und mit Argon belüftet. Im Argon-Gegenstrom wurde trockenes 1,4-Dioxan (c = 0.1 mol/L) hinzugegeben und das Reaktionsgemisch 27 h bei einer Ölbadtemperatur von 130 °C gerührt. Die abgekühlte Suspension wurde über Kieselgur abgesaugt und die Kieselgur-Schicht mit Ethylacetat gewaschen. Das Lösungsmittel des Filtrats wurde im Membranpumpenvakuum bei einer Wasserbadtemperatur von 50 °C entfernt. Das Produkt wurde säulenchromatographisch (Kieselgel, Diethylether/ Aceton = 2:1) gereinigt. Anschließend wurde die Substanz mit wenig Diethylether versetzt und die Suspension kurz im Ultraschallbad behandelt. Der Diethylether wurde abpipettiert und das Produkt als weißer Feststoff mit einem Schmelzpunkt von 225-227 °C isoliert. Die Verbindung wurde mittels Kernspinresonanzspektroskopie-, Massenspektrometrie- und Infrarotspektroskopie-Untersuchungen bestätigt.
1.9 Herstellung von rac-N-(3- Methyl -2-oxo- 1 ,2,3,4-tetrahydrochinazolin-6-yl)-S-cyclohexyl-.S- methylsulfoximin (MF4)
Die Verbindung wurde mittels zweier Verfahren A und B synthetisiert.
Methode A: Ein Druckröhrchen wurde mit S-Cyclohexyl-S-methylsulfoximin, 6-Brom-3-methyl-3,4- dihydrochinazolin-2( 1 H)-on (1.7 Äquiv.), Cäsiumcarbonat (2.4 Äquiv.), Tris(dibenzylidenaceton)dipalladium(0) (0.1 Äquiv.), 2-Dicyclohexylphosphino-2’,4’,6’- triisopropylbiphenyl (0.15 Äquiv.) und einem Magnet-Rührfisch befüllt. Das Druckröhrchen wurde im Hochvakuum evakuiert und mit Argon belüftet. Im Argon-Gegenstrom wurde trockenes 1 ,4-Dioxan (c = 0.13 mol/L) hinzugegeben und das Reaktionsgemisch 27 h bei einer Ölbadtemperatur von 130 °C gerührt. Die abgekühlte Suspension wurde über Kieselgur abgesaugt und die Kieselgur-Schicht mit Ethylacetat gewaschen. Das Lösungsmittel des Filtrats wurde im Membranpumpenvakuum bei einer Wasserbadtemperatur von 50 °C entfernt. Der verbliebene Rückstand wurde säulenchromatographisch (Kieselgel, Diethylether/ Aceton = 2:1) behandelt. Das isolierte Produkt wurde durch eine zweite Säulenchromatographie (Kieselgel, Ethylacetat/Methanol = 10:1) gereinigt und als weißer Feststoff erhalten.
Methode B: Ein Reagenzglas wurde mit S-Cyclohexyl-S-methylsulfoximin, (3-Methyl-2-oxo-1, 2,3,4- tetrahydrochinazolin-6-yl)boronsäure (1.4 Äquiv.), Kupfer(II)acetat (0.1 Äquiv.) und einem Magnet- Rührfisch befüllt. In das Reagenzglas wurde trockenes Methanol (c = 0.25 mol/L) gegeben. Das Reagenzglas wurde in einen Rundkolben gesteht, auf den ein mit Calciumchlorid gefülltes Trockenrohr gesetzt wurde. Das Reaktionsgemisch wurde bei Raumtemperatur 45 h an der Luft gerührt. Das Lösungsmittel wurde im Membranpumpenvakuum bei einer Wasserbadtemperatur von 50 °C entfernt. Der verbliebene Rückstand wurde durch Säulenchromatographie (Kieselgel, Diethylether/ Aceton = 2:1) aufgearbeitet. Das Produkt wurde zwei weitere Male säulenchromatographisch (Kieselgel, Aceton bzw. Ethylacetat/Methanol = 10:1) gereinigt und als gelbbrauner Feststoff mit einem Schmelzpunkt von 165-167 °C isoliert. Die Verbindung wurde mittels Infrarotspektroskopie-, Kernspinresonanzspektroskopie- und Massenspektrometrie- Untersuchungen bestätigt.
Die Produkte der zwei Ansätze wurden vereinigt und die Enantiomere mittels einer präparativen HPLC-Säule unter den Bedingungen für die analytische AD-Säule voneinander getrennt.
Anschließend wurden die einzelnen Enantiomere jeweils erneut säulenchromatographisch (Kieselgel, Ethylacetat/Methanol = 10:1) aufgereinigt und als weiße Feststoffe erhalten.
Das Enantiomer (+)-N-(3-Methyl-2-oxo-1,2,3,4-tetrahydrochinazolin-6-yl)-S-cyclohexyl-S- methylsulfoximin wies einen Schmelzpunkt von 191-193 °C und einen spezifischen Drehwert [α ]D von +22,4 (c = 0,38 in CHCl3) auf. Das Enantiomer (-)-N-(3-Methyl-2-oxo-1, 2,3,4- tetrahydrochinazolin-6-yl)-5-cyclohexyl-5-methylsulfoximin wies einen Schmelzpunkt von 195—
197 °C und einen spezifischen Drehwert [α ]D von -22,2 (c = 0,37 in CHCl3) auf.
1.10 Herstellung und Analytik der Sulfondiimide und Sulfonimidamide:
Methode A:
In ein ausgeheiztes Schlenk-Rohr mit Magnet-Rührfisch und Septum wurde das Sulfondiimid oder Sulfonimidamid, 6-Bromo-3-methyl-3,4-dihydrochinazolin-2(1H)-on (1.5 Äquiv.), Cäsiumcarbonat (1.5 Äquiv.), Tris(dibenzylidenaceton)dipalladium(0) (0.15 Äquiv.), und 2-Dicyclohexylphosphino- 2',4',6'-triisopropylbiphenyl (0.3 Äquiv.) gegeben. Das Schenk-Rohr wurde im Hochvakuum evakuiert und mit Argon belüftet. Im Argon-Gegenstrom wurde trockenes 1 ,4-Dioxan hinzugefügt und das Reaktionsgemisch 12 h bei einer Ölbadtemperatur von 110 °C gerührt. Die abgekühlte Suspension wurde über eine Celite-Schicht (Celite Standard Super Cel, pH = 7-8) filtriert und mit einer Dichlormethan/Methanol-Lösung (10:1) nachgespült. Das Lösungsmittel des Filtrats wurde im Membranpumpenvakuum bei einer Wasserbadtemperatur von 40 °C entfernt und der Rückstand säulenchromatographisch an Kieselgel mit einer Dichlormethan/Methanol-Gradientenlösung (1:0 bis 30:1) aufgereinigt.
Methode B : In ein ausgeheiztes Schlenk-Rohr mit Magnet-Rührfisch und Septum wurde das Sulfondiimid oder Sulfonimidamid, (3-MethyI-2-oxo-1,2,3,4-tetrahydrochinazoIin-6-yl)boronsäure (1.5 Äquiv.) und Kupfer(II)acetat (0.2 Äquiv.) gegeben. Das Schenk-Rohr wurde im Hochvakuum evakuiert und mit Argon belüftet. Im Argon-Gegenstrom wurde trockenes Dimethylformamid oder Methanol hinzugefügt, das Septum durch ein mit Calciumchlorid gefülltes Trockenrohr ersetzt und die Lösung für 24 Stunden bei Raumtemperatur gerührt. Die Lösung wurde über eine Celite-Schicht (Celite Standard Super Cel, pH = 7-8) filtriert und mit einer Dichlormethan/Methanol -Lösung (10:1) nachgespült. Das Lösungsmittel des Filtrats wurde im Membranpumpenvakuum bei einer Wasserbadtemperatur von 40 °C entfernt und der Rückstand säulenchromatographisch an Kieselgel mit einer Dichlormethan/Methanol-Gradientenlösung (1:0 bis 30:1) aufgereinigt.
1.10.1 rcr-N-(3- Methyl -2-oxo- 1 ,2,3,4-tetrahydrochinazolin-6-yl)-N’-phenyl-S-methyl-S- phenylsulfondiimid (JHS1)
Die Verbindung wurde mittels Methode A wie Methode B in DMF erfolgreich hergestellt, mittels Kernspinresonanzspektroskopie-, Massenspektrometrie- und Infrarotspektroskopie-Untersuchungen bestätigt und wies einen Schmelzpunkt von 102-103 °C auf.
1.10.2 (S)-N-(3-Methyl-2-oxo-1,2,3,4-tetrahydrochinazolin-6-yl)-N'-phenyl-S-methyl-S- phenylsulfondiimid (JHS1-S)
Die Verbindung wurde mittels Methode A hergestellt, mittels Kernspinresonanzspektroskopie-, Massenspektrometrie- und Infrarotspektroskopie-Untersuchungen bestätigt und wies einen Schmelzpunkt von 83-84 °C und einen spezifischen Drehwert [α ]D von -612,8 (c = 1.95 in CHCI3) auf.
1.10.3 (R)-N-(3-Methyl-2-oxo-1,2,3,4-tetrahydrochinazolin-6-yl)-N'-phenyl-S-methyl-S- phenylsulfondiimid (JHS1-R) Die Verbindung wurde mittels Methode A hergestellt, mittels Kernspinresonanzspektroskopie-, Massenspektrometrie- und Infrarotspektroskopie-Untersuchungen bestätigt und wies einen Schmelzpunkt von 84-85 °C und einen spezifischen Drehwert [α ]D von 601,5 (c = 1.95 in CHCI3) auf.
1.10.4 rac-N-(3- Methyl -2-oxo- 1 ,2,3,4-tetrahydrochinazolin-6-yl)-N,-H-S-methyl-S- phenylsulfondiimid (JHS2)
Die Verbindung wurde mittels Methode B in Methanol hergestellt, mittels
Kernspinresonanzspektroskopie-, Massenspektrometrie- und Infrarotspektroskopie-Untersuchungen bestätigt und wies einen Schmelzpunkt von 119-120 °C auf. 1.10.5 rac-N-(3- Methyl -2-oxo- 1 ,2,3,4-tetrahydrochinazolin-6-yl)-N,-methyl-S-methyl-S- phenylsulfondiimid (JHS3)
Die Verbindung wurde mittels Methode B in Methanol hergestellt, mittels
Kernspinresonanzspektroskopie-, Massenspektrometrie- und Infrarotspektroskopie-Untersuchungen bestätigt und wies einen Schmelzpunkt von 92-93 °C auf.
1.10.6 rac- 1 - [ N-(3- Methyl -2-oxo- 1 ,2,3, 4-tetrahydroch i nazol i n-6-y l -S-phenl yl su lfon i m idoyl ] piperidin (JHS4)
Die Verbindung wurde mittels Mathode A und Methode B in DMF hergestellt, mittels Kernspinresonanzspektroskopie-, Massenspektrometrie- und Infrarotspektroskopie-Untersuchungen bestätigt und wies einen Schmelzpunkt von 129-130 °C auf.
Beispiel 2: Untersuchung der Effekte der Enantiomere MF1-R, MF1-S, JHS1-R und JHS1-S auf die metabolische Aktivität in Leukämie -Zelllinien
Für Zellexperimente wurden die Erythroleukämie -Zelllinie HEL (JAK2V617F-positiv), die AML- Zelllinie Molm-14 und die chronisch myeloische Leukämie (CML) -Zelllinie K562 (BCR-ABL- positiv) verwendet.
Die Verbindungen MF1-R, MF1-S, JHS1-R und JHS1-S wurden in DMSO gelöst und jeweils in Konzentrationen von 0,1 μM, 1 μM, 10 μM und 20 μM eingesetzt. Entsprechend wurde DMSO jeweils als Kontroll-Behandlung mitgeführt und deren metabolische Aktivität als 100% gesetzt. Die Zelllinien HEL, Molm-14 und K562 wurden mit den jeweiligen Verbindungen in den verschiedenen Konzentrationen behandelt und nach 72 h in Kultur wurde die metabolische Aktivität, die Auskunft über das Wachstum der Zellen gibt, mittels MTT [3-(4,5-Dimethylthiazol-2-yl)-2,5- diphenyltetrazoliumbromid] -Assay bestimmt.
Die bestimmten Absorptionen, die der metabolischen Aktivität entsprechen, sind für die HEL-, Molm- 14- und K562-Zellen jeweils in Figur 1A-C dargestellt. Wie man der Figur 1 entnimmt, zeigte das (S)- Enantiomer MF1-S einen vergleichbaren Effekt auf die metabolische Aktivität von HEL- und Molm- 14-Zellen wie PFI-1, während das (R)-Enantiomer MF1-R demgegenüber nahezu inaktiv war. Dies zeigt, dass die Chiralität der Verbindung MF1 Auswirkung auf die Aktivität hat. Ein solcher Effekt war bei den Enantiomeren JHS1-R und JHS1-S kaum zu beobachten. K562-Zellen wurden als Negativkontrollen genutzt, die kaum einen Effekt auf die Inhibition von BRD4 zeigen (Figur IC). Entsprechend sank die metabolische Aktivität der behandelten K562-Zellen durch die Behandlung mit den Verbindungen kaum.
Beispiel 3: Untersuchung der Effekte der Verbindungen JHS1, JHS2, MF2 und JHS3 auf die metabolische Aktivität in Leukämie -Zelllinien
Racemische Gemische der Verbindungen JHS1, JHS2, MF2 und JHS3 wurden in DMSO gelöst und jeweils in Konzentrationen von 10 nM, 100 nM, 1 μM, 10 μM und 20 μM eingesetzt und entsprechend DMSO jeweils als Kontroll-Behandlung mitgeführt. Die Zelllinien HEL, Molm-14 wurden wie in Beispiel 2 beschrieben mit den jeweiligen Verbindungen in den verschiedenen Konzentrationen behandelt und nach 72 h die metabolische Aktivität mittels MTT- Assay bestimmt.
Die bestimmten metabolischen Aktivitäten sind für die HEL-, Molm-14- und K562 -Zellen jeweils in Figur 2A-C dargestellt. Wie man der Figur 2 entnimmt, reduzierten die racemischen Gemische der Verbindungen MF2 und JHS1 die metabolische Aktivität von HEL und Molm-14-Zehen bei Konzentrationen von 10 und 20 μM. Demgegenüber waren die Verbindungen JHS2 und JHS3 inaktiv.
Beispiel 4: Untersuchung der Effekte der Enantiomere MF2-R und MF2-S auf die metabolische Aktivität in Leukämie-Zelllinien Im Folgenden wurden die zwei Enantiomere von MF2 jeweils individuell synthetisiert und MF2-R und MF2-S in dem MTT-Zell versuch wie unter Beispiel 2 beschrieben einzeln untersucht und mit dem racemischen Gemisch MF2 und PFI-1 verglichen. Des Weiteren wurde die mittlere inhibitorische Konzentration (IC50) berechnet.
Die bestimmten metabolischen Aktivitäten sind für die HEF-, Molm-14- und K562 -Zellen jeweils in Figur 3A-C dargestellt. Die IC50-Werte für die HEF-, Molm-14- und K562-Zellen sind jeweils in Figur 3D-F dargestellt. Wie man der Figur 3 entnimmt, zeigte sich, dass die Verbindung MF2-S signifikant stärker die metabolische Aktivität von HEF-Zellen (Figur 3A, D; 37.24% ± 6.19 vs.
72.95% ± 12.7) und Molm-14-Zellen (Abb. 3B, E; 55.19% ± 0.72 vs. 80.43% ± 1.324) reduzierte als die Vergleichs Verbindung PFI-1 in einer Konzentration von 1 μ M.
Dies zeigt, dass die Aktivität des racemischen MF2 verbessert und mit dem (S)-Enantiomer MF2-S eine Verbindung generiert wurde, die eine verbesserte Effizienz aufweist im Vergleich zu PFI-1. Demgegenüber verlor das (R)-Enantiomer MF2-R im Vergleich zu MF2 an inhibitorischer Aktivität. Bei höheren Konzentrationen der Inhibitoren (10 μ M und 20 μM) glich sich die Effizienz der Reduktion der metabolischen Aktivität wieder an. Eine Reduktion der benötigten Dosis ist jedoch von entscheidender Wichtigkeit, da hierdurch potentielle Nebenwirkungen reduziert werden könnten.
Beispiel 5 : Untersuchung der Effekte der Enantiomere MF2-R und MF2-S auf die Proliferation in Leukämie -Zelllinien
In weiteren Versuchen wurden HEL-, Molm-14- und K562-Zellen in Triplikaten mit jeweils 1 oder 5 μM der Enantiomere MF2-R und MF2-S inkubiert und die Zellzahlen der so behandelten Zellen wurden zum Startzeitpunkt sowie nach 24, 48 und 72 h nach Trypan-Blau-Färbung in der Thoma- Kammer bestimmt und dokumentiert. Das hier beschriebene Zellwachstum wird als Proliferation bezeichnet und gibt Aufschluss über die durchgeführten Zellteilungen im angegebenen Zeitraum.
Die jeweiligen durchschnittlichen Zellzahlen sind für HEL-, Molm-14- und K562-Zellen jeweils in Figur 4A-C dargestellt. Figur 4D zeigt die relative Zellzahl und Figur 4E die durchschnittliche Zellpopulation. Wie man der Figur 4 entnimmt, wurde durch das (S)-Enantiomer MF2-S im Vergleich zu PFI-1 die Zellproliferation von HEL und Molm-14 signifikant reduziert. Figur 4D zeigt die signifikante Reduktion für 5 μ M MF2-S im Vergleich zu PFI-1.
Während der Proliferation durchlaufen die Zellen unterschiedliche Stadien des Zellzyklus, in denen die genomische DNA repliziert, der Zellkern abgebaut und die Zellteilung stattfindet, wodurch zwei identische Zellen entstehen. Wie man der Figur 4E entnimmt, wurde die Synthesephase (S-Phase) durch MF2-S stärker gehemmt als durch PFI-1, wobei der Anstieg der G0/G1 -Phase typisch für einen Block des Zellzyklus ist. Die subGl -Phase gibt den prozentualen Anteil der toten Zellen an und auch hier waren signifikant höhere Anteile der HEL- und Molm-14-Zellen, wenn mit MF2-S behandelt, angereichert.
Während der Untersuchung der Zellproliferation wurde ebenfalls die Viabilität der Zelllinien anhand der Trypan-Blau-Färbung bestimmt. Trypan-Blau kann nicht in lebende Zellen eindrigen. Die Viabilität ist für die HEL-, Molm-14- und K562-Zellen jeweils in Figur 5A-C dargestellt. Figur 5D zeigt die relative Viabilität für 5 μM PFI-1 und MF2-S. Wie man der Figur 5A-D entnimmt, reduzierte vor allem MF2-S in einer Konzentration von 5 μM die Viabilität der HEL- und Molm-14-Zellen signifikant im Vergleich zu PFI-1.
Weiterhin wurde eine mögliche Induktion der Apoptose in den beschriebenen Zelllinien durch die Verbindung MF2 untersucht. Hierzu wurde die Spaltung des Proteins Poly(ADP-Ribose)-Polymerase 1 (PARP1) nach 24 und 48 h Behandlung der HEL-, Molm-14- und K562 -Zellen mit jeweils 5 μM des racemischen Gemischs MF2 sowie der (S)- und (R) -Enantiomere MF2-S und MF2-R im Vergleich zu PFI-1 untersucht. Die Spaltung des Proteins PARP1 ist ein Hinweis auf die beginnende Apoptose (Zelltod) der Zellen und entsprechend wurden mittels Western Blot die Spaltprodukte von PARP1 untersucht. Im Detail, wurden nach Inhibitor-Behandlung Zelllysate mit RIPA Lysepuffer hergestellt. Protein-Konzentrationen wurden mit dem Bradford Protein Assay bestimmt an einem Nanodrop Spectrometer bei 595 nm. 25 μg der Proteinlysate wurde mit 4xLämmli-Probenpuffer versetzt, 5 min bei 65 °C denaturiert und in die SDS-Page eingesetzt. Die Proteine wurden im Wet-Blot-Verfahren auf eine PVDF-Membran übertragen bei 100 m A über Nacht. Die Membran wurde mit 10% BSA in TBS- T-Puffer blockiert. Die primären Antikörper wurden über Nacht bei 4 °C und der HRP-gelabelte 45 min bei Raumtemperatur mit der Membran inkubiert. Die Proteine wurden mittels PCA-ECL-Lösung an einem Chemilumineszenz-Detektor (Fusion SL, PeqLab) detektiert. Mit DMSO behandelte Zellen dienten als Kontrolle. Das Protein Glycerinaldehyd-3-phosphat-Dehydrogenase (GADPH) wurde bestimmt, um eine gleichmäßige Auftragung der Proteinmenge nachzuweisen.
Figur 5E zeigt die Western Blots der HEL-, Molm-14- und K562-Zellextrakte. Wie man der Figur 5E entnimmt, war in HEL-Zellen die markierte Bande für das PARP1 -Spaltprodukt nach 48 h Behandlung mit MF2-S stärker ausgeprägter als bei PFI-1 -Behandlung, während die Bande in Molm- 14-Zellen gleich stark erschien. Jedoch war weniger ungespaltenes PARP1 nach 48 h Behandlung mit 5 μM MF2-S zu erkennen. Es wird angenommen, dass die Inhibition von BRD4 in den untersuchten Zelllinien verstärkt zur Reduktion der metabolischen Aktivität und Proliferation führte und weniger zu einer Induktion der Apoptose der Zehen. Dennoch induzierte MF2-S (5μM) stärker Apoptose als die weiteren untersuchten Verbindungen.
Beispiel 6: Untersuchung der Effekte der Enantiomere MF2-R und MF2-S auf humanes Gewebe
Um zu untersuchen, ob das Enantiomer MF2-S ebenfalls in Primärmaterial von AML (akute myeloische Leukämie) -Patienten und MPN (myeloproliferative Neoplasie)-Patienten ein verbessertes Ansprechen zeigt, wurden sogenannte colony formation assays (CFU-Assays) durchgeführt. Hierfür wurden mononukleäre Zehen (PB MC: Mononukleäre Zehen aus dem peripheren Blut) oder CD34- positive Stammzehen (CD34+ Zellen) aus dem Blut von vier gesunden Kontrollen und drei AML- bzw. MPN-Patienten isoliert und in semi-solides Medium, das mit jeweils 1 μM oder 5 μM an MFS-2 oder PFI-1 versetzt wurde, gegeben. Einzelzellen bilden im CFU-Assay nach 12 Tagen in Kultur Zellkolonien, sofern ihre Ausbildung nicht durch Inhibitoren verhindert wird.
Figur 6A zeigt die Kolonie-Anzahl der vier Kontrollen. Wie man der Figur 6A entnehmen kann, reduzierte in den Kontrollen 1 (PB MC) und 2 (CD34+ Zellen) eine Behandlung mit 5 μM PFI-1 oder MF2-S die Menge der Kolonien gleichsam. Um diesen unspezifischen Effekt auf gesunde Zellen zu reduzieren, wurde in den CD34+-Kontrollen 3 und 4 ebenfalls eine Konzentration von 1 μM MFS-2 oder PFI-1 eingesetzt. Wie der Figur 6A entnommen werden kann, wurde durch 1 μM MFS-2 oder PFI-1 die Kolonienanzahl der Kontrollen weniger stark reduziert. In der Kontrolle 4 wurde die Kolonienanzahl durch 1 μM MF2-S nicht signifikant reduziert. Figur 6B zeigt die Ergebnisse der CFU-Assays von drei AML-Patienten. Wie man der Figur 6B entnimmt, reduzierte MF2-S in den jeweiligen Konzentrationen von 1 μM und 5 μM jeweils die Kolonienanzahl signifikant stärker als PFI-1. Ferner zeigte sich hier 1 μM MF2-S ebenso effizient wie 5 μM PFI-1.
Figur 6C zeigt die Ergebnisse der CFU-Assays der drei MPN-Patienten. Myeloproliferative Neoplasien beinhalten drei klassische Philadelphia Chromosom-negative (BCR-ABF-negative) Krankheitsentitäten, welche die Polycythemia vera (PV), die essentielle Thrombozythämie (ET) und die primäre Myelofibrose (PMF) sind. Sowohl die PV als auch die ET kann in eine Myelofibrose (MF) fortschreiten, was meist eine fatale Verschlechterung des Krankheitszustands und des Behandlungserfolgs bedeutet. Alle in diesen Studien involvierten MPN-Patienten waren JAK2V617F- positiv. Wie man der Figur 6C entnehmen kann, führte die Behandlung der Zellen im CFU-Assay mit 5 μM MF2-S jeweils zu einer signifikant stärkeren Reduktion der Kolonienzahl. Zudem wurde durch 1 μM MF2-S die Kolonienzahl aus Material des PMF-Patienten sehr effizient reduziert, während dies im Vergleich zur PFI-1 beim Material des ET-Patienten nicht der Fall war.
Zudem wurde durch in silico docking Analysen und molecular dynamics (MD) Simulationen mit Hilfe der Röntgen-Struktur der von PFI-1 -gebundenen Bromodomäne des BRD4 bestätigt, dass MF2-S der beste Ligand der Bromodomäne sein sollte. Hierbei zeigte sich, dass sich im Falle von MF2-S die (S)- S-Methyl-S-phenylsulfonimidoylgruppe zu einer hydrophoben Region des BRD4 orientiert, während das (R)-S-Methyl-S-phenylsulfonimidoylfragment des MF2-R in die entgegengesetzte Richtung zeigte. Es wird angenommen, dass hierdurch die Interaktion verloren geht. Ohne auf eine bestimmte Theorie festgelegt zu sein, wird angenommen, dass dies die verbesserte Effizienz des (S)-Enantiomers MF2-S im Vergleich zum (R)-Enantiomer MF2-R erklärt.
Beispiel 7 : Untersuchung der Selektivität des Enantiomers MF2-S
Um die Selektivität der Verbindung MF2-S zu untersuchen, wurde bei der Firma Reaction Biology Corp. (PA, USA) ein Thermal Shift Assay (BromoMELT™ assay) mit der Verbindung MF2-S und als Vergleichsverbindung PFI-1 durchgeführt. Dieser Assay gibt Auskunft über die spezifische Bindung von MF2-S an 76 Bromodomänen-beinhaltende Proteine. Der Versuch wurde in Duplikaten bei einer Konzentration von 10 μM durchgeführt. Die Dissoziation der Inhibitoren (3 = PFI-1; (S)-4a = MF2-S) von den Bromodomänen-beinhaltenden Proteinen bei steigenden Temperaturen im Vergleich zu einer DMSO-Kontrolle wird durch den Wert ATm wiedergegeben.
Figur 7A zeigt die durch den Thermal Shift Assay erhaltenen ΔTm-Werte gegen DMSO-Kontrolle für die Verbindung MF2-S und die Vergleichs Verbindung PFI-1. Wie der Figur 7A entnommen werden kann, bindet MF2-S, bis auf BRDT-2, gleich gut oder besser Proteine der BET Familie als die Vergleichsverbindung PFI-1. Ebenfalls konnte eine höhere Spezifität für BRD4 festgestellt werden. Figur 7B illustriert die Spezifität sämtlicher analysierter Bromodomänen-beinhaltender Proteine für die Vergleichs Verbindung PFI-1, und Figur 7C für die Verbindung MF2-S. Flierbei sind Temper aturänderungen mit steigendem Radius für höhere Tm-Werte angegeben. Wie man den
Figuren 7B und 7C entnimmt, binden sowohl PFI-1 als auch MF2-S spezifisch die Proteine der BET Familie, wobei PFI-1 stärker BRD2 und MF2-S spezifischer BRD4 bindet.
Die Ergebnisse zeigen insgesamt, dass neue Bromodomänen-Inhibitoren zur Verfügung gestellt werden können, wobei die Verbindung MF2-S im Vergleich zum ursprünglichen Bromodomänen- Inhibitor PFI-1 bessere Ergebnisse zeigte. Diese Ergebnisse konnten in primärem Patientenmaterial von AML- und MPN-Patienten bestätigt werden.

Claims

P a t e n t a n s p r ü c h e
1. Verbindung gemäß der allgemeinen Formel (1) wie nachstehend angegeben und/oder deren Stereoisomere, Tautomere, Solvate, Hydrate und pharmazeutisch verträgliche Salze: worin:
X ist ausgewählt aus O oder N-R1, wobei R1 ausgewählt ist aus der Gruppe umfassend Wasserstoff, Cyano, C1-6- Alkyl und/oder jeweils unsubstituiertes oder einfach oder mehrfach mit C1-6-Alkyl substituiertes C3 -7-Cycloalkyl, Aryl oder Hetaryl;
Y ist ausgewählt aus der Gruppe umfassend C1-6-Alkyl, jeweils unsubstituiertes oder einfach oder mehrfach mit C1-6-Alkyl substituiertes C3 -7-Cycloalkyl, Aryl oder Hetaryl, NH2, NHR2 und/oder NR2R3, wobei R2 und R3 ausgewählt sind aus der Gruppe umfassend C1-6- Alkyl und/oder jeweils unsubstituiertes oder einfach oder mehrfach mit C1-2-Alkyl substituiertes C3 -7-Cycloalkyl, Aryl oder Hetaryl;
Z ist ausgewählt aus Cyclohexyl, Aryl oder Hetaryl, jeweils unsubstituiert oder einfach oder mehrfach substituiert mit C1-6-Alkyl, C1-5-Alkoxy oder einfach oder mehrfach mit Halogen substituiertem C1-6-Alkyl; und
Me ist Methyl.
2. Verbindung nach Anspruch 1, dadurch gekennzeichnet, dass:
X ist Sauerstoff;
Y ist ausgewählt aus der Gruppe umfassend C1 -6- Alkyl und/oder jeweils unsubstituiertes oder einfach oder mehrfach mit C1-2- Alkyl substituiertes C3 -7-Cycloalkyl, Aryl oder Hetaryl; und
Z ist ausgewählt aus Cyclohexyl, Aryl oder Hetaryl, jeweils unsubstituiert oder einfach oder mehrfach substituiert mit C1-6-Alkyl, C1-5-Alkoxy oder einfach oder mehrfach mit Halogen substituiertem C1-6-Alkyl.
3. Verbindung nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass die Verbindung ausgewählt ist aus der Gruppe umfassend die Verbindungen gemäß den nachstehenden Formeln (MF1), (MF2), (MF3) oder (MF4):
4. Verbindung nach Anspruch 3, dadurch gekennzeichnet, dass die Verbindung das (S)- Enantiomer der Verbindungen der Formeln (MF1) oder (MF2) ist.
5. Verbindung nach Anspruch 1, dadurch gekennzeichnet, dass:
X ist N-R1, wobei R1 ausgewählt ist aus der Gruppe umfassend Wasserstoff, Cyano, C1-6- Alkyl und/oder jeweils unsubstituiertes oder einfach oder mehrfach mit C1-2- Alkyl substituiertes C3 -7-Cycloalkyl, Aryl oder Hetaryl;
Y ist ausgewählt aus der Gruppe umfassend C1 -6- Alkyl und/oder jeweils unsubstituiertes oder einfach oder mehrfach mit C1-2-Alkyl substituiertes C3 -7-Cycloalkyl, Aryl oder Hetaryl; und
Z ist ausgewählt aus Cyclohexyl, Aryl oder Hetaryl, jeweils unsubstituiert oder einfach oder mehrfach substituiert mit C1-6-Alkyl, C1-5-Alkoxy oder einfach oder mehrfach mit Halogen substituiertem C1-6-Alkyl.
6. Verbindung nach Anspruch 5, dadurch gekennzeichnet, dass die Verbindung ausgewählt ist aus der Gruppe umfassend die Verbindungen gemäß den nachstehenden Formeln (JF1S1),
(JHS2) oder (JHS3):
7. Verbindung nach Anspruch 1, dadurch gekennzeichnet, dass: X ist Sauerstoff;
Y ist ausgewählt aus der Gruppe umfassend NH2, NHR2 und/oder NR2R3, wobei R2 und R3 ausgewählt sind aus der Gruppe umfassend C1 -6- Alkyl und/oder jeweils unsub- stituiertes oder einfach oder mehrfach mit C1-2-Alkyl substituiertes C3 7-Cycloalkyl, Aryl oder Hetaryl; und Z ist ausgewählt aus Cyclohexyl, Aryl oder Hetaryl, jeweils unsubstituiert oder einfach oder mehrfach substituiert mit C1-6-Alkyl, C1-5-Alkoxy oder einfach oder mehrfach mit Halogen substituiertem C1-6-Alkyl.
8. Verbindung nach Anspruch 7, dadurch gekennzeichnet, dass die Verbindung ausgewählt ist aus der Gruppe umfassend die Verbindung gemäß der nachstehenden Formel (JHS4) oder eines deren Enantiomere:

Verwendung einer Verbindung nach einem der vorherigen Ansprüche zur Herstellung eines Arzneimittels, insbesondere zur Herstellung eines Arzneimittels zur Behandlung einer Erkrankung, die mit Bromodomänen in Zusammenhang steht, vorzugsweise von Krebserkrankungen, insbesondere der akuten myeloischen Leukämie oder myeloproliferativen Neoplasien.
10. Arzneimittel enthaltend wenigstens eine Verbindung nach einem der Ansprüche 1 bis 8, insbesondere zur Verwendung in der Behandlung einer Erkrankung, die mit Bromodomänen in Zusammenhang steht, vorzugsweise von Krebserkrankungen, insbesondere der akuten myeloischen Leukämie oder myeloproliferativen Neoplasien.
