Abstract
Die vorliegende Erfindung betrifft Cyanothiophene der allgemeinen Formel (I), in der R wie in den Ansprüchen definiert sind, deren Tautomere, deren Stereoisomere, deren Gemische und deren Salze, welche wertvolle pharmakologische Eigenschaften aufweisen, insbesondere eine Glugacon-Rezeptor-antagonistische Wirkung.
Description
Cyanothiophene, deren Herstellung und deren Verwendung als Arzneimittel
Die vorliegende Erfindung betrifft substituierte Cyanothiophene, deren Herstellung und deren die Verwendung für die Herstellung eines Medikamentes zur Behandlung oder Vorbeugung von Krankheiten, bei denen Glucagon-Rezeptoren beteiligt sind.
Diabetes ist eine komplexe Krankheit, die durch Hyperglykämie aufgrund eines Mangels an Insulinbildung oder mangelnder Insulinwirkung gekennzeichnet ist. Die metabolischen Komplikationen des Diabetes - die Hyperglykämie und die Ketose - sind verknüpft mit dem relativen oder absoluten Anstieg des Verhältnisses von Glucagon zu Insulin. Folglich ist Glucagon ein hyperglykämischer Faktor, der den Anstieg des Blutzuckers hervorruft.
Daher stellen geeignete Antagonisten, die den Glucagon-Rezeptor blockieren, Mittel zur Behandlung von Diabetes dar, wobei die Glucoseproduktion in der Leber inhibiert wird und die Glucosespiegel im Patienten reduziert werden.
Verschiedene Publikationen offenbaren peptidische und nicht-peptidische Glucagon- Rezeptor Antagonisten (McCormick et al., Curr. Pharm. Des. 7, 1451 (2001) eine Übersicht). Insbesondere wurde die Inhibition der Glucagon-stimulierten Glucose Produktion in Menschen für Bay 27-9955 berichtet (Petersen et al., Diabetologia 44, 2018 (2001)).
Der vorliegenden Erfindung lag die Aufgabe zu Grunde, neue nicht-peptidische Wirkstoffe aufzuzeigen, die als hochwirksame Glucagon-Rezeptor-Antagonisten für die Behandlung von Diabetes geeignet sind.
Cyanothiophene und ihre Verwendung als Glucagon-Rezeptor-Antagonisten sind bereits bekannt. So werden beispielsweise in den US-Patentanmeldungen US 2004/0097552 und US 2004/0097557 substituierte Cyanothiophene beschrieben, die
in 2-Stellung durch eine Amid-, Alkylcarbonylamino-, Arylcarbonylamino-, Heteroarylcarbonylamino- oder Heterocyclylaminogruppe substituiert sind.
Ferner werden in den internationalen Anmeldungen WO 2004/024066 und WO 2004/024065 bicyclische Heterocyclen mit Glucagon-Rezeptor-antagonistischer Wirkung offenbart.
Überraschenderweise wurde nun gefunden, dass 3-Cyanothiophene, welche in 4- Stellung ein Halogenatom oder eine Cyano-, Nitro- oder Alkoxygruppe tragen und/oder in 2-Stellung durch eine Bicycloalkylcarbonylaminogruppe substituiert sind, hochwirksame Glucagon-Rezeptor-Antagonisten sind, welche besonders zur Her¬ stellung von Arzneimitteln geeignet sind.
Gegenstand der vorliegenden Erfindung sind somit die Verbindungen der allge- meinen Formel I
deren Tautomere, Enantiomere, Diastereomere, deren Gemische und deren Salze, insbesondere deren physiologisch verträglichen Salze mit anorganischen oder organischen Säuren oder Basen, welche wertvolle pharmakologische Eigenschaften aufweisen, insbesondere eine Hemmwirkung auf Glucagon-Rezeptoren, ihre Ver¬ wendung zur Behandlung und Vorbeugung von Krankheiten, insbesondere von Diabetes sowie ihre Herstellung.
In der obigen Formel (I) bedeuten
R1 eine Ci-4-Alkylgruppe, die endständig durch ein Fluor-, Chlor-, Brom- oder lodatom oder durch eine Aminogruppe substituiert sein kann,
wobei die Wasserstoffatome der Aminogruppe unabhängig voneinander durch eine Ci-3-Alkylgruppe oder eine gegebenenfalls durch eine Ci-3-Alkylgruppe substituierte Phenyl- oder Pyridylgruppe substituiert sein können,
oder eine Phenyl- oder Pyridylgruppe, die jeweils durch ein bis drei Ci-3-Alkylgruppen substiuiert sein können,
R2 ein Wasserstoff-, Fluor-, Chlor-, Brom- oder lodatom oder eine Cyano-, Nitro-, C1- 3-Alykl-, Trifluormethyl- oder Ci-4-Alkyloxygruppe und
R3 eine Bicylco[2.2.1]hept-2-enylgruppe oder
eine Cyclohexylgruppe, in der das Kohlenstoffatom in Position 2 mit dem Kohlen¬ stoffatom in Position 5 über eine -CH2-, -CH2-CH2- oder -CH=CH- Gruppe verbrückt sein kann.
Bevorzugt sind diejenigen Verbindungen der allgemeinen Formel (I), in denen
R1 eine Ci-4-Alkylgruppe, die endständig durch ein Fluor-, Chlor-, Brom- oder lodatom oder durch eine Aminogruppe substituiert sein kann,
wobei die Wasserstoffatome der Aminogruppe unabhängig voneinander durch eine Ci-3-Alkylgruppe oder eine gegebenenfalls durch eine d-3-Alkylgruppe substituierte Phenyl- oder Pyridylgruppe substituiert sein können,
oder eine Phenyl- oder Pyridylgruppe, die jeweils durch ein bis drei Ci-3-Alkylgruppen substiuiert sein können,
R2 ein Fluor-, Chlor-, Brom- oder lodatom oder eine Cyano-, Nitro-, Ci-3-Alykl-, Trifluormethyl- oder Ci-4-Alkyloxygruppe und
R3 eine Bicylco[2.2.1]hept-2-enylgruppe oder
eine Cyclohexylgruppe, in der das Kohlenstoffatom in Position 2 mit dem Kohlen¬ stoffatom in Position 5 über eine -CH2-, -CH2-CH2- oder -CH=CH- Gruppe verbrückt sein kann, bedeuten,
deren Tautomere, Enantiomere, Diastereomere, deren Gemische und deren Salze.
Besonders bevorzugt sind diejenigen Verbindungen der allgemeinen Formel (I), in denen
R1 eine Ci-4-Alkylgruppe, die endständig durch ein Fluor-, Chlor-, Brom- oder lodatom oder durch eine Aminogruppe substituiert sein kann,
wobei die Wasserstoffatome der Aminogruppe unabhängig voneinander durch eine Ci-3-Alkylgruppe oder eine gegebenenfalls durch eine d-3-Alkylgruppe substituierte Phenyl- oder Pyridylgruppe substituiert sein können,
oder eine Phenyl- oder Pyridylgruppe, die jeweils durch ein bis drei Ci-3-Alkylgruppen substiuiert sein können,
R2 ein Fluor-, Chlor-, Brom- oder lodatom oder eine Cyano-, Nitro- oder Ci-4-Alkyl- oxygruppe und
R3 eine Bicylco[2.2.1]hept-2-enylgruppe oder

Eine zweite bevorzugte Untergruppe betrifft diejenigen Verbindungen der allge¬ meinen Formel (I), in denen
R1 eine Ci-4-Alkylgruppe, die endständig durch ein Fluor-, Chlor-, Brom- oder lodatom oder durch eine Aminogruppe substituiert sein kann,
wobei die Wasserstoffatome der Aminogruppe unabhängig voneinander durch eine Ci-3-Alkylgruppe oder eine gegebenenfalls durch eine Ci-3-Alkylgruppe substituierte Phenyl- oder Pyridylgruppe substituiert sein können,
oder eine Phenyl- oder Pyridylgruppe, die jeweils durch ein bis drei d-3-Alkylgruppen substiuiert sein können,
R2 ein Wasserstoff-, Fluor-, Chlor-, Brom- oder lodatom oder eine Cyano-, Nitro-, Ci- 3-Alykl-, Trifluormethyl- oder Ci-4-Alkyloxygruppe und

Ganz besonders bevorzugt sind diejenigen Verbindungen der allgemeinen Formel (I), in denen
R1 eine C2-4-Alkylgruppe, die endständig durch eine N-Phenyl-N-methyl-aminogruppe substituiert ist,
oder eine Phenylgruppe, die durch ein oder zwei Methylgruppen substiuiert sein kann,
R2 ein Chlor- oder Bromatom oder eine Cyano- oder Nitrogruppe und
R3 eine Cyclohexylgruppe, in der das Kohlenstoffatom in Position 2 mit dem Kohlen- stoffatom in Position 5 über eine -CH2- oder -CH2-CH2- Gruppe verbrückt sein kann, bedeuten,
deren Tautomere, Enantiomere, Diastereomere, deren Gemische und deren Salze.
Folgende Verbindungen der allgemeinen Formel (I) sind besonders hervorzuheben:
(a) 2-[(1S,2R,4R)-Bicyclo[2.2.1]hept-2-ylcarbonylamino]-4-chlor-3-cyano-5-(2,5- dimethyl-phenyl)-thiophen,
(b) 2-[(2R)-Bicyclo[2.2.2]oct-2-ylcarbonylamino]-4-chlor-3-cyano-5-(2,5-dimethyl- phenyl)-thiophen,
(c) 4-Chlor-3-cyano-2-(cyclohexylcarbonylamino)-5-phenyl-thiophen,
(d) 4-Brom-3-cyano-2-(cyclohexylcarbonylamino)-5-phenyl-thiophen,
(e) 3,4-Dicyano-2-(cyclohexylcarbonylamino)-5-phenyl-thiophen,
(f) 3-Cyano-2-(cyclohexylcarbonylamino)-4-nitro-5-phenyl-thiophen,
(g) 2-[(1S,2R,4R)-Bicyclo[2.2.1]hept-2-ylcarbonylamino]-4-brom-3-cyano-5-(2,5- dimethyl-phenyl)-thiophen,
(h) 2-[(1 S,2R,4R)-Bicyclo[2.2.1 ]hept-2-ylcarbonylamino]-4-brom-3-cyano-5-{3- [methyl-(4-methylphenyl)-amino]-propyl}-thiophen und
(i) 2-[(1 S,2R,4R)-Bicyclo[2.2.1 ]hept-2-ylcarbonylamino]-3,4-dicyano-5-{3-[methyl-(4- methylphenyl)-amino]-propyl}-thiophen
sowie deren Salze.
Erfindungsgemäß erhält man die Verbindungen der allgemeinen Formel I nach an sich bekannten Verfahren, beispielsweise nach folgenden Verfahren:
a) Umsetzung einer Verbindung der allgemeinen Formel
in der R1 und R2 wie oben erwähnt definiert sind,
mit einem Säurechlorid der allgemeinen Formel
R3-COCI (III),
in der R3 wie oben erwähnt definiert ist.
Die Umsetzung wird zweckmäßigerweise in einem Lösungsmittel wie Methylen¬ chlorid, Chloroform, Tetrachlorkohlenstoff, Ether, Tetrahydrofuran, Dioxan, Benzol, Toluol, Acetonitril oder Sulfolan gegebenenfalls in Gegenwart einer anorganischen oder organischen Base wie z.B. Pyridin oder 4-Dimethylamino-pyridin bei Tempe- raturen zwischen -20 und 2000C, vorzugsweise jedoch bei Temperaturen zwischen -10 und 1600C, durchgeführt
b) Zur Herstellung einer Verbindung der Formel (I), in der R2 ein Fluor-, Chlor-, Brom¬ oder lodatom oder eine Nitrogruppe bedeutet: Chlorierung, Bromierung, lodierung oder Nitrierung einer Verbindung der allgemeinen Formel
in der R1 und R3 wie oben erwähnt definiert sind.
Die Chlorierung kann beispielsweise mit N-Chlorsuccinimid in Lösungsmitteln wie Eisessig, Tetrachlorkohlenstoff oder Dichlormethan bei Temperaturen zwischen 25

und 100 0C durchgeführt werden. Alternativ kann auch Chlor, insbesondere in Verbindung mit Lewis-Säuren wie etwa Aluminiumchlorid, als Reagenz verwendet werden.
Die Bromierung kann zweckmäßigerweise mit N-Bromsuccinimid in Lösungsmitteln wie Eisessig, Tetrachlorkohlenstoff oder Dichlormethan bei Temperaturen zwischen 25 und 100 0C, vorzugsweise bei 60-75 0C erfolgen. Alternativ kann auch Brom, insbesondere in Verbindung mit Lewis-Säuren wie etwa Aluminiumchlorid, als Reagenz verwendet werden.
Die lodierung kann zweckmäßigerweise mit N-Iodsuccinimid in Lösungsmitteln wie Eisessig, Tetrachlorkohlenstoff oder Dichlormethan bei Temperaturen zwischen 25 und 100 0C erfolgen. Alternativ kann auch lod, insbesondere in Verbindung mit Lewis-Säuren wie etwa Aluminiumchlorid, als Reagenz verwendet werden.
Die Nitrierung erfolgt zweckmäßigerweise mit konzentrierter Salpetersäure oder Nitriersäure in Lösungsmitteln wie beispielsweise Essigsäure oder Essigsäureanhydrid bei Temperaturen zwischen -5 0C und 40 0C, vorzugsweise bei Raumtemperatur. Alternativ kann auch Nitroniumtetrafluoroborat in Dichlormethan verwendet werden.
c) Zur Herstellung einer Verbindung der Formel (I), in der R2 eine Cyanogruppe bedeutet: Umsetzung einer Verbindung der allgemeinen Formel
in der R1 und R3 wie oben erwähnt definiert sind, mit Cyanid.

Die Umsetzung erfolgt beispielsweise mit Kupfer(l)-cyanid in Lösungsmitteln wie N, N- Dimethylformamid, Dimethylacetamid, Dioxan, Benzol, Toluol, oder Aceton itril unter Erwärmung, vorzugsweise unter Rückfluß.
d) Zur Herstellung einer Verbindung der Formel (I), in der R2 ein Fluoratom bedeutet: Umsetzung einer Verbindung der allgemeinen Formel
in der R1 und R3 wie oben erwähnt definiert sind und in der keine weiteren Fluor-, Chlor-, Brom-, lodatome oder Nitrogruppen enthalten sind, mit n-Butyllithium und anschließende Umsetzung mit N-Fluordibenzolsulfonimid.
Die Umsetzung mit n-Butyllithium erfolgt in Lösungsmitteln wie Diethylether oder Tetrahydrofuran bei Temperaturen unter -78°C, vorzugsweise in Tetrahydrofuran. Die anschließende Umsetzung mit N-Fluordibenzolsulfonimid erfolgt ebenfalls in Lösungsmitteln wie Diethylether oder Tetrahydrofuran bei Temperaturen unter -78°C, vorzugsweise in Tetrahydrofuran.
e) Zur Herstellung einer Verbindung der Formel (I), in der R1 eine eine Ci-4-Alkyl- gruppe, die endständig durch eine Aminogruppe substituiert sein kann, wobei die Wasserstoffatome der Aminogruppe unabhängig voneinander durch eine Ci-3- Alkylgruppe oder eine gegebenenfalls durch eine d-3-Alkylgruppe substituierte Phenyl- oder Pyridylgruppe substituiert sein können, bedeutet: Umsetzung einer Verbindung der allgemeinen Formel
(VI),

in der R2 und R3 wie oben erwähnt definiert sind und
X eine Abgangsgruppe wie beispielsweise ein Chlor-, Brom- oder lodatom oder eine Tosyl- oder Triflatgruppe bedeutet, mit einem entsprechenden sekundären Amin und anschließend gegebenenfalls Umwandlung der Substituenten an der so eingeführten Aminogruppe.
Die Umsetzung erfolgt zweckmäßigerweise ohne Zusatz weiterer Lösungsmittel (außer dem sekundären Amin) unter Erwärmen, z.B. durch Mikrowelleneinstrahlung.
Anschließend können gegegebenenfalls während der Umsetzung verwendete Schutzgruppen abgespalten werden und/oder
die so erhaltenen Verbindungen der allgemeinen Formel I in ihre Enantiomeren und/oder Diastereomeren aufgetrennt werden und/oder
die erhaltenen Verbindungen der Formel I in ihre Salze, insbesondere für die pharmazeutische Anwendung in ihre physiologisch verträglichen Salze mit anorganischen oder organischen Säuren, überführt werden.
Ferner können die erhaltenen Verbindungen der allgemeinen Formel I, wie bereits eingangs erwähnt wurde, in ihre Enantiomeren und/oder Diastereomeren aufgetrennt werden. So können beispielsweise cis-/trans-Gemische in ihre eis- und trans-lso- mere, und Verbindungen mit mindestens einem optisch aktiven Kohlenstoff atom in ihre Enantiomeren aufgetrennt werden.
So lassen sich beispielsweise die erhaltenen cis-/trans-Gemische durch Chromato¬ graphie in ihre eis- und trans-lsomeren, die erhaltenen Verbindungen der allgemei¬ nen Formel I, welche in Racematen auftreten, nach an sich bekannten Methoden (siehe Allinger N. L. und Eliel E. L. in "Topics in Stereochemistry", Vol. 6, Wiley Inter- science, 1971) in ihre optischen Antipoden und Verbindungen der allgemeinen For¬ mel I mit mindestens 2 asymmetrischen Kohlenstoffatomen auf Grund ihrer physi-

kalisch-chemischen Unterschiede nach an sich bekannten Methoden, z.B. durch Chromatographie und/oder fraktionierte Kristallisation, in ihre Diastereomeren auf¬ trennen, die, falls sie in racemischer Form anfallen, anschließend wie oben erwähnt in die Enantiomeren getrennt werden können.
Die Enantiomerentrennung erfolgt vorzugsweise durch Säulentrennung an chiralen Phasen oder durch Umkristallisieren aus einem optisch aktiven Lösungsmittel oder durch Umsetzen mit einer, mit der racemischen Verbindung Salze oder Derivate wie z.B. Ester oder Amide bildenden optisch aktiven Substanz, insbesondere Säuren und ihre aktivierten Derivate oder Alkohole, und Trennen des auf diese Weise erhaltenen diastereomeren Salzgemisches oder Derivates, z.B. auf Grund von verschiedenen Löslichkeiten, wobei aus den reinen diastereomeren Salzen oder Derivaten die freien Antipoden durch Einwirkung geeigneter Mittel freigesetzt werden können. Besonders gebräuchliche, optisch aktive Säuren sind z.B. die D- und L-Formen von Weinsäure oder Dibenzoylweinsäure, Di-O-p-toluoyl-weinsäure, Äpfelsäure, Mandelsäure,
Camphersulfonsäure, Glutaminsäure, Asparaginsäure oder Chinasäure. Als optisch aktiver Alkohol kommt beispielsweise (+)- oder (-)-Menthol und als optisch aktiver Acylrest in Amiden beispielsweise (+)-oder (-)-Menthyloxycarbonyl in Betracht.
Desweiteren können die erhaltenen Verbindungen der Formel I in ihre Salze, insbe¬ sondere für die pharmazeutische Anwendung in ihre physiologisch verträglichen Salze mit anorganischen oder organischen Säuren, übergeführt werden. Als Säuren kommen hierfür beispielsweise Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Methansulfonsäure, Phosphorsäure, Fumarsäure, Bernsteinsäure, Milchsäure, Zitro- nensäure, Weinsäure oder Maleinsäure in Betracht.
Außerdem lassen sich die so erhaltenen neuen Verbindungen der Formel I, falls diese eine Carboxygruppe enthalten, gewünschtenfalls anschließend in ihre Salze mit anorganischen oder organischen Basen, insbesondere für die pharmazeutische Anwendung in ihre physiologisch verträglichen Salze, überführen. Als Basen kom¬ men hierbei beispielsweise Natriumhydroxid, Kaliumhydroxid, Cyclohexylamin, Ethanolamin, Diethanolamin und Triethanolamin in Betracht.

Die als Ausgangsstoffe verwendeten Verbindungen der allgemeinen Formeln Il bis VI sind entweder literaturbekannt oder man erhält diese nach an sich literaturbekannten Verfahren (siehe Beispiele I bis VIII).
Wie bereits eingangs erwähnt, weisen die erfindungsgemäßen Verbindungen der allgemeinen Formel I und ihre physiologisch verträglichen Salze wertvolle pharma¬ kologische Eigenschaften auf, insbesondere eine Hemmwirkung auf Glucagon- Rezeptoren.
Die biologischen Eigenschaften der neuen Verbindungen wurden wie folgt geprüft:

Die Bindung der erfindungsgemäßen Verbindungen der Formel I an den Glucagon- Rezeptor wurde in einem Verdrängungs- Bindungs-Assay bestimmt, der auf der Verdrängung von radioaktiv markiertem Glucagon von einer den rekombinanten humanen Glucagon-Rezeptor enthaltenden Membran Fraktion beruht. Die für den humanen Glucagon-Rezeptor kodierende cDNA wurde in den Expressionsvektor pcDNA3.1 (Invitrogene) kloniert. BHK-21 Zellen (Baby hamster kidney C-13 Zellen, ATCC) wurden mit diesem Konstrukt transfiziert und ein stabiler Zellklon durch Behandlung mit G-418 (Gibco) selektioniert und isoliert. Eine Membranfraktion, die den rekombinanten humanen Glucagon-Rezeptor enthält, wurde von diesem Klon durch folgende Schritte hergestellt: Konfluent wachsende Zellen wurden durch eis-gekühlten PBS-Puffer (Gibco) mit 0.05% EDTA abgelöst und suspendiert. Nach einer Zentrifugation wurde das Pellet in einem Puffer (1OmM Tris/HCI, pH 7.2; 0.01 mM PMSF (Phenylmethylsulfonylfluorid)) suspendiert und für 90 Minuten bei 4°C inkubiert. Nach einer Behandlung des Lysates mit einem Homo- genisator (Dounce) wurden Zellkerne und andere zelluläre Bestandteile durch eine Zentrifugation bei 500 g für 10 Minuten abgetrennt. Der Überstand wurde anschlie¬ ßend bei 100.000 g für 35 Minuten zentrifugiert, um die Membranen zu pelletieren. Die präzipitierten Membranen wurden in Inkubationspuffer (50 mM Tris/HCI, pH 7.2;

10OmM NaCI; 5 mM MgCI2; 1 mM EDTA; 0.2% BSA (bovine serum albumin)) sus¬ pendiert, aliquotiert und bei -800C gelagert.
Die Verdrängung von Glucagon wurden gemessen, indem 20μg der Membranfrak¬ tion, 50.00 cpm 1251-Glucagon (Amersham Pharmacia) und eine Konzentration der Testsubstanz für 60 Minuten bei 200C in einem Volumen von 100μl in Inkubations¬ puffer in einer Mikrotiterplatte (Optiplate, Packard Instruments) inkubiert wurden. Der gebundene Radioligand wurde von dem freien Liganden durch Filtration und Waschung über GC/B Filter (Packard) auf einem Multiscreen-Vakuumfiltrations- system (Millipore) getrennt. Die Messung erfolgte in einem Topcount Scintillations- Zähler (Packard). Die Bindung in Gegenwart von 1μM unmarkiertem Glucagon (Wherl GmbH) wurde als unspezifisch definiert. Die Analyse der Daten wurde so durchgeführt, dass der Prozentsatz der gebundenen Aktivität in Gegenwart einer Testsubstanz bestimmt wurde. Die Ergebnisse wurden als IC5o-Werte berechnet. Die in den Beispielen 1 bis 25 aufgeführten Verbindungen ergeben IC50-Werte kleiner
Die erfindungsgemäßen Glucagon-Rezeptor-Antagonisten können oral, transdermal, inhalativ oder parenteral verabreicht werden. Die erfindungsgemäßen Verbindungen liegen hierbei als aktive Bestandteile in üblichen Darreichungsformen vor, beispiels- weise in Zusammensetzungen, die im wesentlichen aus einem inerten pharmazeu¬ tischen Träger und einer effektiven Dosis des Wirkstoffs bestehen, wie beispiels¬ weise Tabletten, Dragees, Kapseln, Oblaten, Pulver, Lösungen, Suspensionen, Emulsionen, Sirupe, Suppositorien, transdermale Systeme etc.. Eine wirksame Dosis der erfindungsgemäßen Verbindungen liegt bei einer oralen Anwendung zwischen 1 und 100, vorzugsweise zwischen 1 und 50, besonders bevorzugt zwischen 5-30 mg/Dosis, bei intravenöser oder intramuskulärer Anwendung zwischen 0,001 und 50, vorzugsweise zwischen 0,1 und 10 mg/Dosis. Für die Inhalation sind erfindungs¬ gemäß Lösungen geeignet, die 0,01 bis 1 ,0, vorzugsweise 0,1 bis 0,5 % Wirkstoff enthalten. Für die inhalative Applikation ist die Verwendung von Pulvern bevorzugt. Gleichfalls ist es möglich, die erfindungsgemäßen Verbindungen als Infusionslösung, vorzugsweise in einer physiologischen Kochsalzlösung oder Nährsalzlösung einzusetzen.

Die erfindungsgemäßen Verbindungen können allein zur Anwendung gelangen oder in Kombination mit anderen erfindungsgemäßen Wirkstoffen, gegebenenfalls auch in Kombination mit weiteren pharmakologisch aktiven Wirkstoffen aus der Gruppe bestehend aus: acarbose, beraprost, bexarotene, Captopril, denileukin, diftitox, etanercept, farglitazar, fidarestat, glibenclamide, glibornuride, gliclazide, glimepiride, glipizide, glucagon, ilomastat, Imidapril, insulin, lanreotide, linogliride, lisinopril, metformin, mexiletine, miglitol, minalrestat, mitiglinide, moxonidine, nafagrel, nateglinide, octreotide, orlistat, oxcarbazepine, pegvisomant, pioglitazone, ponalrestat, pramlintide, ramipril, repaglinide, rosiglitazone, sirolimus, sorbinil, tolrestat, troglitazone, voglibose, zenarestat und zopolrestat.
Geeignete Anwendungsformen sind beispielsweise Tabletten, Kapseln, Zäpfchen, Lösungen, Säfte, Emulsionen oder dispersible Pulver. Entsprechende Tabletten können beispielsweise durch Mischen des oder der Wirkstoffe mit bekannten Hilfsstoffen, beispielsweise inerten Verdünnungsmitteln, wie Calciumcarbonat, Calciumphosphat oder Milchzucker, Sprengmitteln, wie Maisstärke oder Alginsäure, Bindemitteln, wie Stärke oder Gelatine, Schmiermitteln, wie Magnesiumstearat oder Talk, und/oder Mitteln zur Erzielung des Depoteffektes, wie Carboxymethylcellulose, Celluloseacetatphthalat, oder Polyvinylacetat erhalten werden. Die Tabletten können auch aus mehreren Schichten bestehen.
Entsprechend können Dragees durch Überziehen von analog den Tabletten herge- stellten Kernen mit üblicherweise in Drageeüberzügen verwendeten Mitteln, bei¬ spielsweise Kollidon oder Schellack, Gummi arabicum, Talk, Titandioxid oder Zucker, hergestellt werden. Zur Erzielung eines Depoteffektes oder zur Vermeidung von Inkompatibilitäten kann der Kern auch aus mehreren Schichten bestehen. Desgleichen kann auch die Drageehülle zur Erzielung eines Depoteffektes aus mehreren Schichten bestehen wobei die oben bei den Tabletten erwähnten Hilfsstoffe verwendet werden können.

Säfte der erfindungsgemäßen Wirkstoffe beziehungsweise Wirkstoff kombinationen können zusätzlich noch ein Süßungsmittel, wie Saccharin, Cyclamat, Glycerin oder Zucker sowie ein geschmacksverbesserndes Mittel, z.B. Aromastoffe, wie Vanillin oder Orangenextrakt, enthalten. Sie können außerdem Suspendierhilfsstoffe oder Dickungsmittel, wie Natriumcarboxymethylcellulose, Netzmittel, beispielsweise
Kondensationsprodukte von Fettalkoholen mit Ethylenoxid, oder Schutzstoffe, wie p- Hydroxybenzoate, enthalten.
Injektionslösungen werden in üblicher weise, z.B. unter Zusatz von Konservierungs- mittein, wie p-Hydroxybenzoaten, oder Stabilisatoren, wie Alkalisalzen der Ethylen- diamintetraessigsäure hergestellt und in Injektionsflaschen oder Ampullen abgefüllt.
Die eine oder mehrere Wirkstoffe beziehungsweise Wirkstoffkombinationen enthal¬ tenden Kapseln können beispielsweise hergestellt werden, indem man die Wirkstoffe mit inerten Trägern, wie Milchzucker oder Sorbit, mischt und in Gelatinekapseln ein¬ kapselt.
Geeignete Zäpfchen lassen sich beispielsweise durch Vermischen mit dafür vorge¬ sehenen Trägermitteln, wie Neutralfetten oder Polyäthylenglykol beziehungsweise dessen Derivaten, herstellen.
Eine therapeutisch wirksame Tagesdosis beträgt zwischen 1 und 800 mg, bevorzugt 10 - 300 mg pro Erwachsener.

26.1 g (145 mmol) 2,5-Dimethylphenylacetonitril werden in Ethanol gelöst und mit

Anschließend erhitzt man 30 Stunden zum Rückfluss. Nach Abkühlung der Reaktionslösung wird der Eisessig abdestilliert, der Rückstand mit Wasser versetzt und mit Ether extrahiert. Die organische Phase wird mit Wasser, 10-proz. Natriumcarbonatlösung und Wasser waschen. Nach Trocknen über Magnesiumsulfat wird die organische Phase bis zur Trockene eingeengt. c. 2-Amino-3-cvano-4-trifluor-5-(2,5-dimethylphenvh-thiophen 21.3 g (99 mmol) 1 ,1 ,1-Trifluor-3-(2,5-dimethylphenyl)-propan-2-on werden in 70 ml Ethanol gelöst und mit 6.54 g (99 mmol) Malonsäuredinitril, 3.17 g (99 mmol) Schwe¬ fel und 11 ml (100 mmol) N-Methylm Orphol in versetzt. Anschließend erhitzt man in der Mikrowellenapparatur (CEM Discovery) 30 Minuten auf 1200C bei 200 Watt. Danach wird das Lösungsmittel im Vakuum entfernt und der Rückstand mit 100 ml Wasser und 100 ml Essigester versetzt. Die Phasen werden getrennt und die wässrige Phase mit Essigester extrahiert. Das Reaktionsgemisch wird 30 Stunden bei Raumtemperatur gerührt und an¬ schließend mit 50 ml Wasser und 50 ml Essigester versetzt. Die Phasen werden getrennt und die wässrige Phase mit Essigester extrahiert. Die vereinigten orga- nischen Phasen werden über Natriumsulfat getrocknet und am Rotationsverdampfer konzentriert.

(24.9 mmol) Malonsäuredinitril gelöst und mit 7.43 g (72.8 mmol) Aluminiumoxid
(basisch, Aktivitätsstufe I) versetzt und 20 Minuten bei Raumtemperatur gerührt.
Danach wird Dichlormethan zugegeben und vom Feststoff abgesaugt. Das Filtrat wird über Magnesiumsulfat getrocknet und vom Lösemittel befreit. Das so erhaltene Rohprodukt wird in 5.5 ml Ethanol gelöst und mit 750 mg (23.4 mmol) Schwefel versetzt. Bei Raumtemperatur werden 1.44 ml (13.9 mmol) Diethylamin zugetropft.

Allgemeine Herstellunαsvorschrift für die Beispiele 1 bis 8
1 mmol der entsprechenden Norboman-, Norbornen- oder Bicyclo[2.2.2]octan- carbonsäure wird in 5 ml Dichlormethan gelöst, mit 0.3 ml Oxalylchlorid versetzt und 1 Stunde bei Raumtemperatur gerührt. Danach wird bis zum Rückstand eingeengt. Das entsprechende Säurechlorid wird zu einer auf 5 0C gekühlten Lösung von 0.3 g (1 mmol) 2-Amino-3-cyano-4-trifluormethyl-5-(2,5-dimethylphenyl)-thiophen (Beispiel I) und 0.24 ml (3 mmol) Pyridin in 5 ml Dichlormethan zugetropft. Die Reaktions¬ lösung wird 12 Stunden bei Raumtemperatur gerührt, mit Dichlormethan verdünnt, und zweimal mit 1 N Salzsäure gewaschen. Die organische Phase wird über Mag¬ nesiumsulfat getrocknet und bis zur Trockene eingeengt. Der Rückstand wird an Kieselgel (Cyclohexan/Essigester = 9:1) chromatographiert.

Beispiel 15 3-Cvano-2-(cvclohexylcarbonylamino)-4-nitro-5-phenyl-thiophen
1 g (3 mmol) 3-Cyano-2-(cyclohexylcarbonylamino)-5-phenylthiophen (Beispiel 12a) werden in 80 ml Essigsäureanhydrid vorgelegt. Bei 50C tropft man langsam 4 ml 60- proz. Salpetersäure zu. Nach 10 Minuten Rühren bei dieser Temperatur wird die Lösung auf Raumtemperatur erwärmt und weitere 2 Stunden gerührt. Das Reaktions¬ gemisch wird auf 10 ml Eis gegeben und verrührt. Anschließend werden die Phasen getrennt. Die wässrige Phase wird mit Dichlormethan extrahiert. Die vereinigten organischen Phasen werden über Magnesiumsulfat getrocknet und eingeengt. Der Rückstand wird mit Methanol und Dichlormethan versetzt, das Dichlormethan ab¬ destilliert und der ausgefallene Niederschlag abgesaugt. Ausbeute: 0.1 g (9% der Theorie)

0.68 g (3.5 mmol) 2-Amino-3-cyano-4-methoxy-5-propylthiophen (Beispiel VIII) und 0.84 ml (10.4 mmol) Pyridin werden in 25 ml Dichlormethan gelöst. Innerhalb von 10 Minuten werden bei 100C 0.48 ml Cyclohexancarbonsäurechlorid in 5 ml Dichlor¬ methan zugetropft. Eine Lösung von 100 mg (0.31 mmol) 2-[(1 S,2R,4R)-Bicyclo[2.2.1]hept-2-ylcarbonyl- amino]-5-(3-chlorpropyl)-3-cyano-thiophen in 0.4 ml Dichlormethan wird mit 82 mg (0.62 mmol) wasserfreiem Aluminiumchlorid versetzt. Anschließend werden 18 μ\ (0.34 mmol) Brom zugegeben. Man erhitzt 3 Stunden zum Rückfluss. Danach wer¬ den nochmals 50 mg (0.38 mmol) Aluminiumchlorid und 10 /vi (0.19 mmol) Brom zugegeben. Man rührt 16 Stunden bei Raumtemperatur und erhitzt nochmals 2 Stun¬ den zum Rückfluss. Anschließend wird die Reaktionslösung auf 5-proz. Natriumbi- sulfitlösung gegossen. Man extrahiert mit Essigester, wäscht die organische Phase mit gesättigter Natriumchloridlösung und trocknet über Magnesiumsulfat. Das nach Entfernen des Lösungsmittels erhaltene Rohprodukt (110 mg) wird sofort weiter um¬ gesetzt.

Dest. Wasser wird auf 700C erhitzt. Hierin wird unter Rühren p-Hydroxybenzoe- säuremethylester und -propylester sowie Glycerin und Carboxymethylcellulose- Natriumsalz gelöst. Es wird auf Raumtemperatur abgekühlt und unter Rühren der
Wirkstoff zugegeben und homogen dispergiert. Nach Zugabe und Lösen des Zuckers, der Sorbitlösung und des Aromas wird die Suspension zur Entlüftung unter Rühren evakuiert.
5 ml Suspension enthalten 50 mg Wirkstoff.

Ampullen mit 10 mα Wirksubstanz
Zusammensetzung:

Die Wirksubstanz wird in der erforderlichen Menge 0,01 n HCl gelöst, mit Kochsalz isotonisch gestellt, sterilfiltriert und in 2 ml Ampullen abgefüllt.

Zusammensetzung:
Wirkstoff 50,0 mg
0,01 n Salzsäure s.q.

Herstellung:
Die Wirksubstanz wird in der erforderlichen Menge 0,01 n HCl gelöst, mit Kochsalz isotonisch gestellt, sterilfiltriert und in 10 ml Ampullen abgefüllt.
Claims
1. Verbindungen der allgemeinen Formel
(I)
in der
R1 eine Ci-4-Alkylgruppe, die endständig durch ein Fluor-, Chlor-, Brom- oder lodatom oder durch eine Aminogruppe substituiert sein kann,
wobei die Wasserstoffatome der Aminogruppe unabhängig voneinander durch eine Ci-3-Alkylgruppe oder eine gegebenenfalls durch eine Ci-3-Alkylgruppe substituierte Phenyl- oder Pyridylgruppe substituiert sein können,
oder eine Phenyl- oder Pyridylgruppe, die jeweils durch ein bis drei Ci-3-Alkylgruppen substiuiert sein können,
R2 ein Wasserstoff-, Fluor-, Chlor-, Brom- oder lodatom oder eine Cyano-, Nitro-, Ci- 3-Alykl-, Trifluormethyl- oder Ci-4-Alkyloxygruppe und
R3 eine Bicylco[2.2.1]hept-2-enylgruppe oder
eine Cyclohexylgruppe, in der das Kohlenstoffatom in Position 2 mit dem Kohlen¬ stoffatom in Position 5 über eine -CH2-, -CH2-CH2- oder -CH=CH- Gruppe verbrückt sein kann, bedeuten,
deren Tautomere, Enantiomere, Diastereomere, deren Gemische und deren Salze. 44
2. Verbindungen der allgemeinen Formel (I) gemäß Anspruch 1 , in denen
R1 eine Ci-4-Alkylgruppe, die endständig durch ein Fluor-, Chlor-, Brom- oder lodatom oder durch eine Aminogruppe substituiert sein kann,
wobei die Wasserstoffatome der Aminogruppe unabhängig voneinander durch eine Ci-3-Alkylgruppe oder eine gegebenenfalls durch eine Ci-3-Alkylgruppe substituierte Phenyl- oder Pyridylgruppe substituiert sein können,
oder eine Phenyl- oder Pyridylgruppe, die jeweils durch ein bis drei Ci.3-Alkylgruppen substiuiert sein können,
R2 ein Fluor-, Chlor-, Brom- oder lodatom oder eine Cyano-, Nitro-, C1-S-AIyW-, Trifluormethyl- oder Ci-4-Alkyloxygruppe und
R3 eine Bicylco[2.2.1]hept-2-enylgruppe oder
eine Cyclohexylgruppe, in der das Kohlenstoffatom in Position 2 mit dem Kohlen- stoffatom in Position 5 über eine -CH2-, -CH2-CH2- oder -CH=CH- Gruppe verbrückt sein kann, bedeuten,
deren Tautomere, Enantiomere, Diastereomere, deren Gemische und deren Salze.
3. Verbindungen der allgemeinen Formel (I) gemäß Anspruch 2, in denen
R1 eine C1-4-Alkylgruppe, die endständig durch ein Fluor-, Chlor-, Brom- oder lodatom oder durch eine Aminogruppe substituiert sein kann, 45
wobei die Wasserstoffatome der Aminogruppe unabhängig voneinander durch eine Ci-3-Alkylgruppe oder eine gegebenenfalls durch eine Ci-3-Alkylgruppe substituierte Phenyl- oder Pyridylgruppe substituiert sein können,
oder eine Phenyl- oder Pyridylgruppe, die jeweils durch ein bis drei Ci-3-Alkylgruppen substiuiert sein können,
R2 ein Fluor-, Chlor-, Brom- oder lodatom oder eine Cyano-, Nitro- oder C-ι-4-Alkyl- oxygruppe und
R3 eine Bicylco[2.2.1]hept-2-enylgruppe oder
eine Cyclohexylgruppe, in der das Kohlenstoffatom in Position 2 mit dem Kohlen¬ stoffatom in Position 5 über eine -CH2-, -CH2-CH2- oder -CH=CH- Gruppe verbrückt sein kann, bedeuten,
deren Tautomere, Enantiomere, Diastereomere, deren Gemische und deren Salze.
4. Verbindungen der allgemeinen Formel (I) gemäß Anspruch 1 , in denen
R1 eine Ci-4-Alkylgruppe, die endständig durch ein Fluor-, Chlor-, Brom- oder lodatom oder durch eine Aminogruppe substituiert sein kann,
wobei die Wasserstoffatome der Aminogruppe unabhängig voneinander durch eine Ci-3-Alkylgruppe oder eine gegebenenfalls durch eine Ci-3-Alkylgruppe substituierte Phenyl- oder Pyridylgruppe substituiert sein können,
oder eine Phenyl- oder Pyridylgruppe, die jeweils durch ein bis drei Ci-3-Alkylgruppen substiuiert sein können,
R2 ein Wasserstoff-, Fluor-, Chlor-, Brom- oder lodatom oder eine Cyano-, Nitro-, Ci- 3-Alykl-, Trifluormethyl- oder Ci-4-Alkyloxygruppe und 46
R3 eine Cyclohexylgruppe, in der das Kohlenstoffatom in Position 2 mit dem Kohlen¬ stoffatom in Position 5 über eine -CH2- oder -CH2-CH2- Gruppe verbrückt sein kann, bedeuten,
deren Tautomere, Enantiomere, Diastereomere, deren Gemische und deren Salze.
5. Verbindungen der allgemeinen Formel (I) gemäß Anspruch 4, in denen
R1 eine Ci-4-Alkylgruppe, die endständig durch ein Fluor-, Chlor-, Brom- oder lod- atom oder durch eine Aminogruppe substituiert sein kann,
wobei die Wasserstoffatome der Aminogruppe unabhängig voneinander durch eine C1-3-Alkylgruppe oder eine gegebenenfalls durch eine Ci-3-Alkylgruppe substituierte Phenyl- oder Pyridylgruppe substituiert sein können,
oder eine Phenyl- oder Pyridylgruppe, die jeweils durch ein oder zwei C-ι-3-Alkyl- gruppen substiuiert sein können,
R2 ein Fluor-, Chlor-, Brom- oder lodatom oder eine Cyano-, Nitro-, Ci-3-AIyW-, Trifluormethyl- oder Ci-4-Alkyloxygruppe und
R3 eine Cyclohexylgruppe, in der das Kohlenstoffatom in Position 2 mit dem Kohlen- stoffatom in Position 5 über eine -CH2- oder -CH2-CH2- Gruppe verbrückt sein kann, bedeuten,
deren Tautomere, Enantiomere, Diastereomere, deren Gemische und deren Salze.
6. Verbindungen der allgemeinen Formel (I) gemäß Anspruch 5, in denen 47
R1 eine C2-4-Alkylgruppe, die endständig durch eine N-Phenyl-N-methyl-aminogruppe substituiert ist,
oder eine Phenylgruppe, die durch ein oder zwei Methylgruppen substiuiert sein kann,
R2 ein Chlor- oder Bromatom oder eine Cyano- oder Nitrogruppe und
R3 eine Cyclohexylgruppe, in der das Kohlenstoffatom in Position 2 mit dem Kohlen- stoffatom in Position 5 über eine -CH2- oder -CH2-CH2- Gruppe verbrückt sein kann, bedeuten,
deren Tautomere, Enantiomere, Diastereomere, deren Gemische und deren Salze.
7. Folgende Verbindungen der allgemeinen Formel (I):
(a) 2-[(1 S,2R,4R)-Bicyclo[2.2.1 ]hept-2-ylcarbonylamino]-4-chlor-3-cyano-5-(2,5- dimethyl-phenyl)-thiophen,
(b) 2-[(2R)-Bicyclo[2.2.2]oct-2-ylcarbonylamino]-4-chlor-3-cyano-5-(2,5-dimethyl- phenyl)-thiophen,
(c) 4-Chlor-3-cyano-2-(cyclohexylcarbonylamino)-5-phenyl-thiophen,
(d) 4-Brom-3-cyano-2-(cyclohexylcarbonylamino)-5-phenyl-thiophen,
(e) 3,4-Dicyano-2-(cyclohexylcarbonylamino)-5-phenyl-thiophen,
(f) S-Cyano^-fcyclohexylcarbonylamino^-nitro-δ-phenyl-thiophen, 48
(g) 2-[(1 S,2R,4R)-Bicyclo[2.2.1]hept-2-ylcarbonylamino]-4-brom-3-cyano-5-(2,5- dimethyl-phenyl)-thiophen,
(h) 2-[(1 S,2R,4R)-Bicyclo[2.2.1 ]hept-2-ylcarbonylamino]-4-brom-3-cyano-5-{3- [methyl-(4-methylphenyl)-amino]-propyl}-thiophen und
(i) 2-[(1 S,2R,4R)-Bicyclo[2.2.1 ]hept-2-ylcarbonylamino]-3,4-dicyano-5-{3-[methyl-(4- methylphenyl)-amino]-propyl}-thiophen
sowie deren Salze.
8. Physiologisch verträgliche Salze der Verbindungen gemäß einem der Ansprüche 1 bis 7 mit anorganischen oder organischen Säuren oder Basen.
9. Arzneimittel, enthaltend eine Verbindung gemäß einem der Ansprüche 1 bis 7 oder ein physiologisch verträgliches Salz gemäß Anspruch 8 neben gegebenenfalls einem oder mehreren inerten Trägerstoffen und/oder Verdünnungsmitteln.
10. Verwendung einer Verbindung nach mindestens einem der Ansprüche 1 bis 9 zur Herstellung eines Arzneimittels, das zur Behandlung von Diabetes geeignet ist.
11. Verfahren zur Herstellung eines Arzneimittels gemäß Anspruch 9, dadurch gekennzeichnet, daß auf nichtchemischen Weg eine Verbindung nach mindestens einem der Ansprüche 1 bis 8 in einen oder mehrere inerte Trägerstoffe und/oder Verdünnungsmittel eingearbeitet wird.
12. Verfahren zur Herstellung der Verbindungen der allgemeinen Formel I gemäß den Ansprüchen 1 bis 8, dadurch gekennzeichnet, dass
a) eine Verbindung der allgemeinen Formel 49
in der R1 und R2 wie in den Ansprüchen 1 bis 8 erwähnt definiert sind,
mit einem Säurechlorid der allgemeinen Formel
R3-COCI (III),
in der R3 wie in den Ansprüchen 1 bis 8 erwähnt definiert ist, umgesetzt wird oder
b) zur Herstellung einer Verbindung der Formel (I), in der R2 ein Fluor-, Chlor-, Brom¬ oder lodatom oder eine Nitrogruppe bedeutet: eine Verbindung der allgemeinen Formel
in der R1 und R3 wie in den Ansprüchen 1 bis 8 erwähnt definiert sind, fluoriert, chloriert, bromiert, jodiert oder nitriert wird, oder
c) zur Herstellung einer Verbindung der Formel (I), in der R2 eine Cyanogruppe bedeutet: eine Verbindung der allgemeinen Formel

in der R1 und R3 wie in den Ansprüchen 1 bis 8 erwähnt definiert sind, mit Cyanid umgesetzt wird, oder
d) zur Herstellung einer Verbindung der Formel (I), in der R1 eine eine Ci-4-Alkyl- gruppe, die endständig durch eine Aminogruppe substituiert sein kann, wobei die Wasserstoffatome der Aminogruppe unabhängig voneinander durch eine Ci-3- Alkylgruppe oder eine gegebenenfalls durch eine Ci-3-Alkylgruppe substituierte Phenyl- oder Pyridylgruppe substituiert sein können, bedeutet: eine Verbindung der allgemeinen Formel
in der R2 und R3 wie in den Ansprüchen 1 bis 8 erwähnt definiert sind und X eine Abgangsgruppe bedeutet, mit einem entsprechenden sekundären Amin umgesetzt wird und anschließend gegebenenfalls die Substituenten an der so eingeführten Aminogruppe umgewandelt werden,
und/oder
anschließend gegegebenenfalls während der Umsetzung verwendete Schutzgruppen abgespalten werden und/oder
die so erhaltenen Verbindungen der allgemeinen Formel I in ihre Enantiomeren und/oder Diastereomeren aufgetrennt werden und/oder
die erhaltenen Verbindungen der Formel I in ihre Salze, insbesondere für die pharmazeutische Anwendung in ihre physiologisch verträglichen Salze mit anorganischen oder organischen Säuren, übergeführt werden.
