Description
NEUES VERFAHREN ZUR HERSTELLUNG VON TIOTROPIUMSALZEN
Die Erfindung betrifft ein neues Verfahren zur Herstellung von Tiotropiumsalzen der allgemeinen Formel 1
worin X " die in den Ansprüchen und in der Beschreibung genannten Bedeutungen haben kann.
Hintergrund der Erfindung Anticholinergika können bei einer Vielzahl von Erkrankungen therapeutisch sinnvoll eingesetzt werden. Hervorzuheben sind hier beispielsweise die Therapie von Asthma oder COPD (chronic obstructive pulmonary disease = chronisch obstruktive Lungenerkrankung). Zur Therapie dieser Erkrankungen werden beispielsweise durch die WO 02/03289 Anticholinergika vorgeschlagen, die ein Scopin-, Tropenol- oder auch Tropin-Grundgerüst aufweisen. Darüberhinaus wird im Stand der Technik insbesondere das Tiotropiumbromid als hoch potentes Anticholinergikum offenbart. Tiotropiumbromid ist beispielsweise aus der EP 418 716 A1 bekannt.
Neben den im vorstehend genannten Stand der Technik offenbarten Syntheseverfahren zur Herstellung von Scopinestern wird insbesondere in der WO03/057694 ein Verfahren zur Herstellung von Estern des Scopins offenbart.
Aufgabe der vorliegenden Erfindung ist es, ein verbessertes technisches Syntheseverfahren bereitzustellen, welches einen einfacheren synthetischen Zugang zu den Verbindungen der allgemeinen Formel 1 in einer gegenüber dem Stand der Technik verbesserten Art und Weise ermöglicht.
Detaillierte Beschreibung der Erfindung
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Tiotropiumsalzen der Formel 1
worin
X " ein einfach negativ geladenes Anion, vorzugsweise ein Anion ausgewählt aus der Gruppe bestehend aus Chlorid, Bromid, Iodid, Methansulfonat oder Trifluormethansulfonat, bedeuten kann,
dadurch gekennzeichnet, daß eine Verbindung der Formel
worin X " die vorstehend genannten Bedeutungen haben kann, in einem Schritt mit einer in situ generierten Verbindung der Formel 3
worin
R ein Rest ausgewählt aus der Gruppe bestehend aus N-Imidazolyl, N-Triazolyl,
-O-C(=NR')-NHR", -O-SO2-Phenyl, -O-SO2-Phenyl-Methyl, -0-SO2-R' -0-CO-C(MeUIyI)3, -O-CO-Phenyl-NO2, Chlor, Brom, -N3 und -0-(P=O)R", ist, wobei
R C1-C4-AIlCyI oder C3-C6-Cycloalkyl;
R" Ci-C4-Alkyl, C3-C6-Cycloalkyl, Ci-C4-Alkylen-N(Ci-C4-Alkyl)2;
R" C1-C4-AIlCyI, -0-C1-C4-AIlCyI, Phenyl oder -O-Phenyl bedeuten
R1 und R2, gleich oder verschieden Methyl, Ethyl, Propyl, Butyl oder Phenyl bedeuten können, wobei Phenyl gegebenenfalls substituiert sein kann durch einen oder mehrere Reste C1-C4-AIlCyI bedeuten, in einem geeigneten Lösemittel unter Zusatz einer geeigneten Base zu einer Verbindung der Formel 4
umgesetzt werden, wobei die Gruppen X", R1 und R2 die vorstehend genannten Bedeutungen haben können, und die Verbindung der Formel 4 ohne Isolierung durch Umsetzung mit einer geeigneten Säure oder einem geeigneten De-Silylierungsreagenz unter Abspaltung der Silylgruppe in die Verbindung der Formel 1 überfuhrt wird.
Bevorzugt betrifft die vorliegende Erfindung ein Verfahren zur Herstellung von Tiotropiumsalzen der Formel 1 , worin X " ein einfach negativ geladenes Anion ausgewählt aus der Gruppe bestehend aus Chlorid, Bromid, Iodid,, Methansulfonat oder Trifluormethansulfonat, bevorzugt Chlorid, Bromid oder Methansulfonat, besonders bevorzugt Bromid, bedeuten kann.
Ein erfindungsgemäß besonders bevorzugtes Verfahren ist dadurch gekennzeichnet, dass die Umsetzung mit einer in situ generierten Verbindung der Formel 3 erfolgt, in der R ein Rest ausgewählt aus der Gruppe bestehend aus N-Imidazolyl, N-Triazolyl, -O-C(=NR')-NHR", -O-SO2-Phenyl-Methyl, -O-CO-C(Methyl)3, und Chlor, ist wobei
R Methyl, Ethyl oder Cyclohexyl; R" Methyl, Ethyl, Cyclohexyl, C2-C3-Alkylen-N(Methyl)2 oder
C2-C3-Alkylen-N(Ethyl)2, bedeuten und R1 und R2, gleich oder verschieden Methyl, Ethyl, Propyl oder Butyl, bedeuten.
Ein erfindungsgemäß besonders bevorzugtes Verfahren ist dadurch gekennzeichnet,
dass die Umsetzung mit einer in situ generierten Verbindung der Formel 3 erfolgt, in der
R ein Rest ausgewählt aus der Gruppe bestehend aus N-Imidazolyl, N-Triazolyl,
-O-C(=N-Cyclohexyl)-NHCyclohexyl, -O-C(=N-Ethyl)-NH-CH2-CH2-CH2-NMe2 und -O-CO-C(Methyl)3, bevorzugt N-Imidazolyl oder N-Triazolyl, besonders bevorzugt N-Imidazolyl ist und
R1 und R2, gleich oder verschieden Methyl, Ethyl, Propyl oder Butyl, bevorzugt Methyl oder Ethyl, besonders bevorzugt Methyl;
R2 Methyl oder Ethyl, bevorzugt Methyl, bedeuten.
Als Alkylgruppen sowie Alkylgruppen, welche Bestandteil anderer Reste sind, werden verzweigte und unverzweigte Alkylgruppen mit 1 bis 4 Kohlenstoffatomen bezeichnet. Beispielsweise werden genannt: Methyl, Ethyl, Propyl, Butyl. Sofern nicht anders genannt, sind von den vorstehend genannten Bezeichnungen Propyl, Butyl sämtliche der möglichen isomeren Formen umfaßt. Beispielsweise umfaßt die Bezeichnung Propyl die beiden isomeren Reste n-Propyl und iso-Propyl, die Bezeichnung Butyl n-Butyl, iso-Butyl, sec. Butyl und tert. -Butyl.
Als Alkylenbrücke oder Alkylengruppe werden, soweit nicht anders angegeben, verzweigte und unverzweigte Alkylgruppen mit 1 bis 4 Kohlenstoffatomen, beispielsweise Methylen, Etyhlen, Propylen, Butylen-Brücken bezeichnet. Besonders bevorzugt sind
Methylen, Etyhlen, Propylen- und Butylen-Brücken. Sofern nicht anders genannt, sind von den vorstehend genannten Bezeichnungen Ethylen, Propylen, Butylen sämtliche der möglichen isomeren Formen umfaßt.
Die Bezeichnungen Phenyl-Methyl und Phenyl-NÜ2 stehen für Phenylringe, die durch
Methyl oder NO2 substituiert sind. Hierbei sind alle möglichen Isomeren (ortho, meta oder para) umfasst, wobei der para- oder meta- Substitution besondere Bedeutung zukommt.
Als Cycloalkylgruppen werden Cycloalkylreste mit 3 - 6 Kohlenstoffatomen beispielsweise Cyclopropyl, Cyclobutyl, Cyclopentyl oder Cyclohexyl bezeichnet.
Zur Durchführung dieses Verfahrens kann wie nachstehend beschrieben vorgegangen werden. Zunächst wird dazu in einem geeigneten Lösemittel die Verbindung der Formel 3 in situ generiert. Der Terminus "in situ" steht dabei für die Darstellung der Verbindung der Formel 3 ohne, dass diese anschließend isoliert wird. Die Darstellung der Formel 3 erfolgt
durch Umsetzung von Dithienylglykolsäure, bevorzugt von Alkalisalzen der Dithienylglykolsäure, besonders bevorzugt von Natriumdithienylglykolat mit einem Kupplungsreagenz ausgewählt aus der Gruppe bestehend aus Carbonyldiimidazol, Carbonyldi-l,2,4-triazol, Dicyclohexylcarbodiimid , Ethyl- dimethylaminopropylcarbodiimid, Toluolsulfonylchlorid, Pivaloylchlorid, Nitrobenzoesäureanhydrid, Oxalylchlorid, Phosgen, Sulfonylchlorid und Phosphorchloriden bevorzugt Carbonyldiimidazol, Carbonyldi-l,2,4-triazol, Dicyclohexylcarbodiimid , Ethyl-dimethylaminopropylcarbodiimid, besonders bevorzugt Carbonyldiimidazol in einem geeigneten Lösemittel, vorzugsweise in einem polaren, aprotischen organischen Lösemittel, besonders bevorzugt in einem Lösemittel ausgewählt aus der Gruppe bestehend aus Acetonitril, Nitromethan, Formamid, Dimethylformamid, N- Methylpyrrolidinon, Dimethylsulfoxid , Dimethylacetamid Tetrahydrofuran, Dioxan und Sulfolan, bevorzugt Tehtrahydrofuran, Dimethylformamid oder N-Methylpyrrolidon bei einer Temperatur von -20°C - 60°C, bevorzugt von -10°C - 45 °C, besonders bevorzugt von -10°C - 25°C und anschließender Zugabe einer Silylverbindung der Formel 5
«* 5, wobei die Reste R1 und R2 die vorstehend genannten Bedeutungen haben können und L für eine Abgangsgruppe steht, die vorzugsweise ausgewählt ist aus der Gruppe bestehend aus Halogenid, Methansulfonat, Trifluormethansulfonat und para-Toluolsulfonat, besonders bevorzugt Methansulfonat, Trifluormethansulfonat, Brom oder Chlor, ferner bevorzugt Brom oder Chlor, wobei Chlor erfindungsgemäß besondere Bedeutung zukommt.
Die Silylverbindung 5 kann entweder der Mischung aus Dithienylglykolsäure bzw. Dithienylglykolsäuresalz mit Kupplungsreagenz im oben genannten Lösemittel, gegebenenfalls in Gegenwart einer Base wie beispielsweise Pyridin, Imidazol oder N- Alkylamin, zugesetzt werden oder aber gemeinsam mit Dithienylglykolsäure bzw. dem Dithienylglykolsäuresalz im oben genannten Lösemittel, gegebenenfalls in Gegenwart einer Base wie beispielsweise Pyridin, Imidazol oder N-Alkylamin, zunächst vorgelegt und anschließend mit oben genanntem Kupplungsreagenz versetzt werden.
Vorzugsweise werden die oben genannten drei Komponenten zur Bildung der Verbindung der Formel 3 in stöchiometrischen Mengen zugesetzt, gegebenenfalls kann die Umsetzung
aber auch bei Vorliegen einer der drei Komponenten im Überschuß (beispielsweise 1,1 bis 1,5 Äquivalent) durchgeführt werden.
Pro Mol in situ generierter Verbindung der Formel 3 gelangen an dieser Stelle vorzugsweise zwischen 0,2 und 1,5 L, besonders bevorzugt zwischen 0,3 und 1 L der genannten Lösemittel zum Einsatz.
Nach Zugabe aller drei Komponenten wird zur Bildung der Verbindung der allgemeinen Formel 3 die erhaltene Lösung bei der oben genannten Temperatur etwa 5 Minuten bis 2 Stunden, vorzugsweise 10 Minuten bis 1 Stunde, besonders bevorzugt 20 - 40 Minuten beispielsweise durch Rühren durchmischt.
Zur so erhaltenen Lösung wird anschließend die Verbindung der Formel 2 zugesetzt. Diese Zugabe kann entweder durch Zugabe einer Lösung oder Suspension der Verbindung der Formel 2 in einem oder mehreren der oben genannten Lösemittel oder durch, vorzugsweise portionsweise, Zugabe der Verbindung der Formel 2 in Substanz erfolgen. Wird die Verbindung der Formel 2 in einem oder mehreren Lösemittel gelöst oder suspendiert zugesetzt, bietet sich die Verwendung derselben Lösemittel an, welches zur in situ Herstellung der Verbindung der Formel 3 zum Einsatz gelangt.
Die Menge, in der die Verbindung der Formel 2 zugesetzt wird, bestimmt sich nach der Menge an in situ generierter Verbindung der Formel 3. Werden zur Bildung der Verbindung der Formel 3 die drei Komponenten Dithienylglykolsäure bzw. Dithienylglykolsäuresalz, Kupplungsreagenz und Verbindung der Formel 5 in stöchiometrischen Mengen eingesetzt, liegt Verbindung der Formel 3 in der molaren Menge vor, wie sie für die drei Komponenten Dithienylglykolsäure bzw. Dithienylglykolsäuresalz, Kupplungsreagenz und Verbindung der Formel 5 gewählt wurde. Werden die drei Komponeten Dithienylglykolsäure bzw. Dithienylglykolsäuresalz, Kupplungsreagenz und Verbindung der Formel 5 nicht in stöchiometrischen Mengen zur Bildung der Verbindung der Formel 3 eingesetzt, liegt die Verbindung der Formel 3 in der molaren Menge der am geringsten portionierten Komponente der drei Ausgangsverbindungen Dithienylglykolsäure bzw. Dithienylglykolsäuresalz, Kupplungsreagenz und Verbindung der Formel 5 vor. Das molare Verhältnis von Verbindung der Formel 2 zu in situ generierter Verbindung der Formel 3 liegt bevorzugt in einem Bereich von 2:1 bis 1:5, bevorzugt 1,5:1 bis 1:3,
besonders bevorzugt 1:1 bis 1:2, wobei einem Verhältnis von 1:1 bis 1:1,5 erfindungsgemäß besondere Bedeutung zukommt.
Nach Zugabe der Verbindung der Formel 3 wird die erhaltene Reaktionsmischung mit in einem geeigneten Lösemittel aufgenommener Base versetzt. Als Lösemittel kommen erfindungsgemäß die vorstehend genannten Lösemittel in Betracht. Vorzugsweise wird an dieser Stelle dasjenige Lösemittel verwendet, welches auch zur Bildung der Verbindung der Formel 3 zum Einsatz gelangt ist. Als Base können organische oder anorganische Basen zum Einsatz gelangen. Als organische Basen werden bevorzugt Alkaliimidazolide verwendet, welche beispielsweise in situ aus den Alkalimetallen und Imidazol oder den Alkalimetallhydriden und Imidazol generiert werden können. Als Akaliimidazolide kommen bevorzugt Imidazolide des Lithiums, Natriums oder Kaliums in Betracht, wobei Natrium- oder Lithiumimidazolid erfindungsgemäß bevorzugt sind. Besonders bevorzugt werden Alkalialkoholate sterisch gehinderter Alkohole (z.B. Kaliumtert.butylat) eingesetzt. Bevorzugte weitere Basen sind erfindungsgemäß ausgewählt aus der Gruppe bestehend aus Lithiumdiisopropylamid (LDA), Lithium oder Natriumhexamethyldisilazan (LiHMDS oder NaHMDS). Als anorganische Base kommen vorzugsweise Hydride des Lithiums, Natriums oder Kaliums in Betracht. Besonders bevorzugt wird als anorganische Base Natriumhydrid eingesetzt.
Pro Mol eingesetzte Verbindung der Formel 2 werden vorzugsweise 0,5-2 Mol, besonders bevorzugt 1-1,5 Mol Base zugesetzt. Im Rahmen des erfindungsgemäßen Verfahrens ist es allerdings zumeist ausreichend, wenn pro Mol eingesetzte Verbindung der Formel 2 lediglich 1 - 1,1 Mol Base zugegeben werden.
Für die Herstellung der Basenlösung oder -Suspension werden pro Mol Base vorzugsweise zwischen 0,2 und 1,5 L, besonders bevorzugt zwischen 0,3 und 1 L des genannten Lösemittels verwendet.
Die Zugabe der Base erfolgt vorzugsweise bei einer Temperatur von -20-60°C, bevorzugt von 0-45°C, besonders bevorzugt von 0-25°C. Nach Zugabe der Base wird die erhaltene Mischung zur Bildung der Verbindung der Formel 4 etwa 10 Minuten bis 6 Stunden, bevorzugt 30 Minuten bis 3 Stunden, besonders bevorzugt 45 Minuten bis 1,5 Stunden bei konstanter Temperatur gerührt.
Zur Freisetzung der Verbindung der Formel 1 aus der in situ generierten Verbindung der Formel 4 erfolgt bevorzugt bei einer Temperatur von unter 10°C, besonders bevorzugt bei etwa 0°C die Zugabe einer geeigneten Säure H-X. Bevorzugt bestimmt die Wahl der Säure sich dabei nach dem Anion X" im gewünschte Endprodukt der allgemeinen Formel 1. Gegebenenfalls kann im Rahmen der vorliegenden Erfindung neben der Säure H-X zusätzlich die Zugabe eines geeigneten Desilylierungsreagenzes erfolgen, welches vorzugsweise ausgewählt ist aus der Gruppe der Ammoniumfluoride, besonders bevorzugt Tetrabutylammoniumfluorid, Tetraethylammoniumfluorid, Benzyltrimethylammoniumfluorid, Tetrahexylammoniumfluorid, Tetraoctylammoniumfluorid oder Fluorwasserstoff frei oder komplexiert wie z. B Pyridiniumfluorid, Triethylamin-HF -Komplex.
Alternativ zum Einsatz einer der vorstehend genannten Säuren kann die Freisetzung der Verbindung der Formel 1 auch ausschließlich mittels vorstehend genannter De- Silylierungsreagentien erfolgen.
Insofern im Rahmen der vorliegenden Erfindung bevorzugt solche Verbindungen der allgemeinen Formel 1 synthetisiert werden, in denen X" für Bromid steht, wird die nachfolgende Vorgehensweise für die Herstellung des erfindungsgemäß bevorzugten Tiotropiumbromids beschrieben. Für den Fachmann ist ersichtlich, daß eine entsprechende Vorgehensweise durch Wahl des geeigneten Reagenzes H-X bzw. Y-F, wobei Y für ein Kation wie ein Proton oder ein Metallkation oder Ammonium, Alkylammonium, Tetraalkylammonium oder Pyridinium oder einen Komplex wie z. B. Aluminiumtrifluorid- HF oder einen anderen Fluorid-Donor wie z.B. Diethylaminoschwefeltrifluorid (DAST) stehen kann, in analoger Art und Weise auch zur Herstellung solcher Verbindungen Verwendung finden kann, in denen X" nicht Bromid bedeutet.
Zur Herstellung von Verbindungen der Formel 1 mit X" = Bromid (= Tiotropiumbromid) werden bezogen auf eingesetzte Verbindung der Formel 2 vorzugsweise 0,2 bis 20 Mol, bevorzugt 0,5 bis 15 Mol, besonders bevorzugt 1 bis 14 Mol Bromwasserstoff bei konstanter Temperatur gegeben. Der verwendete Bromwasserstoff kann hierbei entweder in gasförmiger Form oder in Form, vorzugsweise gesättigter, Lösungen zugegeben werden. Erfindungsgemäß bevorzugt erfolgt die Zugabe an Bromwasserstoff in Eisessig oder Wasser gelöster Form. Besonders bevorzugt gelangt hierbei eine 33%-ige Bromwasserstoff-Lösung in Eisessig zum Einsatz oder besonders bevorzugt als wässrige
62%ige Bromwasserstoffsäure. Die Zugabe der Säure erfolgt vorzugsweise so langsam, dass die Temperatur des Reaktionsgemischs 20°C nicht überschreitet. Nach beendeter Zugabe wird bei konstanter Temperatur, gegebenenfalls auch unter Eiskühlung nachgerührt (zwischen 0,5 und 6 Stunden).
Zur Aufarbeitung kann insbesondere wie in den Beispielen angegeben, nach an sich bekannten Methoden vorgegangen werden. Beispielsweise wird die Reaktionsmischung mit einem protischen Lösemittel, vorzugsweise mit einem Alkohol, besonders bevorzugt mit Methanol oder Ethanol oder Isopropanol versetzt. Pro Mol eingesetzte Verbindung der Formel 2 werden erfindungsgemäß bevorzugt 0,5 bis 20 L, besonders bevorzugt 0,7 bis 13L Alkohol zugesetzt und die erhaltene Mischung bei einer Temperatur von 0-60°C, bevorzugt von 10-45°C, besonders bevorzugt von 15-25°C für einen Zeitraum von etwa 0,5- 6 Stunden, bevorzugt 0,5 - 5 Stunden, besonders bevorzugt 0,5 - 4 Stunden gerührt. Abschließend wird die erhaltene Lösung mit einem unpolareren organischen Lösemittel, vorzugsweise mit einem Lösemittel ausgewählt aus der Gruppe bestehend aus einem Keton (wie beispielsweise Aceton oder Methylethylketon), einem Alkohol (wie beispielsweise Methanol, Ethanol, Propanol, Isopropanol, Butanol oder Amylalkohol), Toluol, Ethylacetat, n-Butylacetat, Dichlormethan, Diethy lether, Methyl-tert.-buyl-Ether, Tetrahydrofuran und Dioxan, besonders bevorzugt Isopropanol, Toluol oder Aceton versetzt.
Nach guter Durchmischung wird das auskristallisierte Produkt abgetrennt und mit dem vorstehend genannten Lösemittel gewaschen. Zur Abtrennung wasserlöslicher Verunreinigungen kann das Rohprodukt mit Wasser oder wässerigen Bromidlösungen z.B. Natrium- oder Kaliumbromidlösung behandelt werden.
Eine weitergehende Reinigung der so erhaltenen Verbindungen der Formel 1 kann, sofern erforderlich, durch Chromatographie über Kieselgel oder mittels Umkristallisation aus geeigneten Lösemitteln wie z.B. niederen Alkoholen, wie beispielsweise Methanol, Ethanol oder Isopropanol, gegebenenfalls unter vorhergehender Behandlung mit Aktivkohle erfolgen.
Aufgrund ihrer zentralen Bedeutung als Intermediate des erfindungsgemäßen Verfahrens zur Herstellung der Verbindungen der Formel 1 betrifft die vorliegende Erfindung ferner Verbindung der Formel 3
worin R, R1 und R2 die vorstehend genannten Bedeutungen haben können, als solche.
Aufgrund ihrer zentralen Bedeutung als Intermediate des erfindungsgemäßen Verfahrens zur Herstellung der Verbindungen der Formel 1 betrifft die vorliegende Erfindung ferner Verbindung der Formel 4
worin X ", R1 und R2 die vorstehend genannten Bedeutungen haben können, als solche.
Ferner betrifft die vorliegende Erfindung die Verwendung der vorstehend genannten Verbindungen der Formel 3 zur Herstellung von Verbindungen der Formel 1.
Ferner betrifft die vorliegende Erfindung die Verwendung der vorstehend genannten Verbindungen der Formel 4 zur Herstellung von Verbindungen der Formel 1.
Die nachfolgenden Beispiele dienen der Illustration exemplarisch durchgeführter Syntheseverfahren. Sie sind lediglich als mögliche, exemplarisch dargestellte Vorgehensweisen zu verstehen, ohne die Erfindung auf deren Inhalt zu beschränken.
Synthcscbcispicl 1:
In eine Mischung aus 13,1g (50mmol) Natriumdithienylglykolat und 8,1g (50mmol) Carbonyldiimidazol in 25ml N-Methylpyrrolidon (NMP) werden bei 20°C 5,43g (50mmol) Chlortrimethylsilan getropft.
Nach 30min Rühren werden 9,38g (37,5mmol) Scopinmethobromid zugegeben und eine Lösung aus 2,59g (38mmol) Imidazol und 1,52g (38mmol) Natriumhydrid (60%ig) in 15ml NMP bei 20°C zugetropft und Ih bei 20°C gerührt.
Nach Abkühlen auf 0°C werden 50ml einer 33%igen Lösung von Bromwasserstoff in Eisessig zugetropft wobei die Temperatur 20°C nicht übersteigt. Anschließend werden 50ml Methanol zugegeben und es wird Ih bei 20°C gerührt. Das Reaktionsgemisch wird zweimal mit je 200ml Toluol extrahiert und nach Abtrennung der Toluolphase mit 150ml Isopropanol bei 0°C kristallisiert. Das Rohprodukt wird abfiltriert, mit 30ml kaltem Isopropanol gewaschen und im Vakuum getrocknet. Ausbeute 15,0g (85%, bzgl. Scopinmethobromid).
Synthcscbcispicl 2:
In eine Mischung aus 13,1g (50mmol) Natriumdithienylglykolat und 8,1g (50mmol) Carbonyldiimidazol in 25ml Dimethylacetamid werden bei 20°C 5,43g (50mmol) Chlortrimethylsilan getropft.
Nach 30min Rühren werden 9,38g (37,5mmol) Scopinmethobromid zugegeben und eine Lösung aus 2,59g (38mmol) Imidazol und 1,52g (38mmol) Natriumhydrid (60%ig) in 30ml Dimethylacetamid bei 20°C zugetropft und Ih bei 20°C gerührt. Nach Abkühlen auf -4°C werden 50ml einer 33%igen Lösung von Bromwasserstoff in Eisessig zugetropft wobei die Temperatur 20°C nicht übersteigt. Anschließend werden 50ml Methanol zugegeben und es wird 3h bei 20°C gerührt. Das Reaktionsgemisch wird mit 500ml Toluol extrahiert und nach Abtrennung der Toluolphase mit 150ml Isopropanol bei 0°C kristallisiert. Das Rohprodukt wird abfiltriert, mit 30ml kaltem Isopropanol gewaschen und im Vakuum getrocknet. Ausbeute 14,1g (80%, bzgl. Scopinmethobromid).
Synthcscbcispicl 3:
In eine Lösung aus 13,1g (50mmol) Natriumdithienylglykolat und 8,1g (50mmol) Carbonyldiimidazol in 25ml Dimethylformamid (DMF) werden bei 20°C 5,43g (50mmol) Chlortrimethylsilan getropft. Nach 30min Rühren werden 12,5g (50mmol) Scopinmethobromid zugegeben und eine Lösung aus 2,59g (38mmol) Imidazol und 1,52g (38mmol) Natriumhydrid (60%ig) in 15ml Dimethylformamid bei 20°C zugetropft und Ih bei 20°C gerührt. Nach Abkühlen auf -5°C werden 50ml einer 33%igen Lösung von Bromwasserstoff in Eisessig zugetropft wobei die Temperatur 20°C nicht übersteigt. Anschließend werden 20ml Methanol zugegeben und es wird Ih bei 20°C gerührt. Das Reaktionsgemisch wird

Das Rohprodukt wird abfiltriert und aus 120ml Methanol unter Zugabe von 5g Aktivkohle umkristallisiert. Nach Abkühlen auf 0°C wird das erhaltene Tiotropiumbromid abfiltriert, mit 5ml kaltem Methanol gewaschen und im Vakuum getrocknet. Ausbeute 15,0g (64% bzgl. Scopinmethobromid).
Synthcscbcispicl 4:
In eine Lösung aus 13,1g (50mmol) Natriumdithienylglykolat und 8,1g (50mmol) Carbonyldiimidazol in 25ml Dimethylformamid werden bei 20°C 5,43g (50mmol) Chlortrimethylsilan getropft.
Nach 30min Rühren werden 12,5g (50mmol) Scopinmethobromid zugegeben und eine Lösung aus 2,59g (38mmol) Imidazol und 1,52g (38mmol) Natriumhydrid (60%ig) in 15ml DMF bei 20°C zugetropft und Ih bei 20°C gerührt. Nach Abkühlen auf -5°C werden 50ml einer 33%igen Lösung von Bromwasserstoff in Eisessig zugetropft wobei die Temperatur 20°C nicht übersteigt. Anschließend werden 20ml Methanol zugegeben und es wird Ih bei 20°C gerührt. Das Reaktionsgemisch wird zweimal mit je 200ml Toluol extrahiert und nach Abtrennung der Toluolphase mit 150ml Isopropanol bei 5°C kristallisiert. Das Rohprodukt wird abfiltriert und aus 120ml Methanol unter Zugabe von 5g Aktivkohle umkristallisiert. Nach Abkühlen auf 0°C wird das erhaltene Tiotropiumbromid abfiltriert, mit 5ml kaltem Methanol gewaschen und im Vakuum getrocknet. Die so erhaltenen Kristalle werden in 20ml Wasser bei 90°C gelöst und das Monohydrat des Tiotropiumbromids durch Abkühlen auf 15°C kristallisiert. Das Produkt wird abfiltriert, mit 7ml Wasser und 8ml Aceton gewaschen und trocken gesaugt. Ausbeute 9,8g (40% bzgl. Scopinmethobromid).
Synthcscbcispicl 5:
In eine Mischung aus 13,1g (50mmol) Natriumdithienylglykolat und 8,1g (50mmol) Carbonyldiimidazol in 25ml Dimethylformamid werden bei 20°C 5,43g (50mmol) Chlortrimethylsilan getropft. Nach 30min Rühren werden 12,5g (50mmol) Scopinmethobromid zugegeben und eine Lösung aus 2,59g (38mmol) Imidazol und 1,52g (38mmol) Natriumhydrid (60%ig) in 15ml Dimethylformamid bei 20°C zugetropft und Ih bei 20°C gerührt. Nach Abkühlen auf 0°C werden 5ml einer 33%igen Lösung von Bromwasserstoff in Eisessig zugetropft wobei die Temperatur 10°C nicht übersteigt. Anschließend werden 120ml Tetrabutylammoniumfluorid IM in THF (0,12mol) zugegeben und es wird Ih bei
Raumtemperatur gerührt. Das Reaktionsgemisch wird mit 800ml Dichlormethan versetzt und Ih bei Raumtemperatur gerührt. Das kristallisierte Rohprodukt wird abfiltriert und aus 120ml Methanol unter Zugabe von 5g Aktivkohle umkristallisiert. Nach Abkühlen auf 0°C wird das erhaltene Tiotropiumbromid abfiltriert, mit kaltem Methanol gewaschen und im Vakuum getrocknet. Ausbeute 9,5g (44% bzgl. Scopinmethobromid).
Synthcscbcispicl 6:
In eine Lösung aus 13,1g (50mmol) Natriumdithienylglykolat in 25ml Dimethylformamid werden bei 20°C 5,43g (50mmol) Chlortrimethylsilan getropft. Nach 30min Rühren bei Raumtemperatur werden portionsweise 8,1g (50mmol) Carbonyldiimidazol zugegeben und es wird weitere 10min gerührt. Anschließend werden 10g (40mmol) Scopinmethobromid zugegeben und eine Lösung aus 2,59g (38mmol) Imidazol und 1,52g (38mmol) Natriumhydrid (60%ig) in 15ml Dimethylformamid bei 20°C zugetropft und Ih bei 20°C gerührt. Nach Abkühlen auf -5°C werden 50ml einer 33%igen Lösung von Bromwasserstoff in Eisessig zugetropft, wobei die Temperatur 20°C nicht übersteigt. Anschließend werden 20ml Methanol zugegeben und es wird 30min bei Raumtemperatur gerührt. Das Reaktionsgemisch wird zweimal mit je 200ml Toluol extrahiert und in 150ml Isopropanol durch Abkühlen auf 5°C kristallisiert. Das kristallisierte Rohprodukt wird abfiltriert und aus 120ml Methanol unter Zugabe von 5g Aktivkohle umkristallisiert. Nach Abkühlen auf 0°C wird das erhaltene Tiotropiumbromid abfiltriert, mit kaltem Methanol gewaschen und im Vakuum getrocknet. Das Produkt wird in 24ml Wasser bei 90°C gelöst und das Monohydrat des Tiotropiumbromids durch Abkühlen auf 15°C kristallisiert. Das Produkt wird abfiltriert und mit 6,5ml Wasser und 10,5ml Aceton gewaschen und getrocknet. Ausbeute 8,1g (42% bzgl. Scopinmethobromid).
Synthcscbcispicl 7:
In eine Lösung aus 13,1g (50mmol) Natriumdithienylglykolat in 25ml Dimethylformamid werden bei 20°C 5,43g (50mmol) Chlortrimethylsilan getropft. Nach 30min Rühren bei Raumtemperatur werden portionsweise 8,1g (50mmol) Carbonyldiimidazol zugegeben und es wird weitere 10min gerührt. Anschließend werden 10g (40mmol) Scopinmethobromid zugegeben und eine Lösung aus 2,59g (38mmol) Imidazol und 1,52g (38mmol) Natriumhydrid (60%ig) in 15ml Dimethylformamid bei 20°C zugetropft und Ih bei 20°C gerührt.
Nach Abkühlen auf 10°C werden 6ml einer 33%igen Lösung von Bromwasserstoff in Eisessig zugetropft wobei die Temperatur 20°C nicht übersteigt. Anschließend werden 120ml Tetrabutylammoniumfluorid IM in THF (0,12mol) zugegeben und es wird 30min bei Raumtemperatur gerührt. Das Reaktionsgemisch wird mit 800ml Dichlormethan versetzt und 15min bei Raumtemperatur gerührt. Das kristallisierte Rohprodukt wird abfiltriert und aus 120ml Methanol unter Zugabe von 2g Aktivkohle umkristallisiert. Nach Abkühlen auf 0°C wird das erhaltene Tiotropiumbromid abfiltriert, mit kaltem Methanol gewaschen und im Vakuum getrocknet. Das Produkt wird in 18ml Wasser bei 90°C gelöst und das Monohydrat des Tiotropiumbromids durch Abkühlen auf 15°C kristallisiert. Das Produkt wird abfiltriert und mit 5ml Wasser und 8ml Aceton gewaschen und getrocknet. Ausbeute 6,5g (34% bzgl. Scopinmethobromid).
Synthcscbcispicl 8:
In eine Lösung von 13,1g (50mmol) Natriumdithienylglykolat in 25ml Tetrahydrofuran werden bei 20-30°C 5,43g (50mmol) Chlortrimethylsilan getropft.
Nach 60min Rühren werden 8,1g (50mmol) Carbonyldiimidazol und nach weiteren 30min 10,01g (40mmol) Scopinmethobromid zugegeben und weitere 30min gerührt. Dann wird eine Lösung aus 2,60g (38mmol) Imidazol und 1,65g (38mmol) Natriumhydrid (55%ig) in 25ml Dimethylformamid bei 20°C zugetropft und Ih bei 20°C gerührt. Nach Abkühlen auf 0°C werden 20ml 62%iger Bromwasserstoffsäure zugetropft wobei die Temperatur 20°C nicht übersteigt. Nach 40min Rühren wird das Reaktionsgemisch bei 20°C in 350ml Isopropanol eingerührt und auf 10°C abgekühlt. Das Rohprodukt wird abfiltriert, mit 50ml kaltem Isopropanol gewaschen und im Vakuum getrocknet. Ausbeute 18,9g rotbraune Kristalle, DC entspricht Vergleich. Das Rohprodukt wird mit 2,2g Aktivkohle in 100ml Methanol unter Rückfluss gelöst und filtriert. Anschließend wird die Lösung auf 30ml eingeengt und auf 3°C abgekühlt. Die Kristalle werden abfiltriert, mit 5ml kaltem Methanol gewaschen und getrocknet. Ausbeute 12,1g weiß-beige Kristalle, DC entspricht Vergleich. Die so erhaltenen Kristalle werden mit 1,2g Aktivkohle in 28ml Wasser bei 80°C gelöst und filtriert. Nach Abkühlen auf 15°C wird das auskristallisierte Tiotropiumbromid
Monohydrat abfiltriert und getrocknet. Ausbeute 9,4g (48% bezogen auf das eingesetzte Scopinmethobromid).
Synthcscbcispicl 9:
In eine Lösung von 39,3g (150mmol) Natriumdithienylglykolat in 117ml Tetrahydrofuran werden bei 0°C 17,9g (165mmol) Chlortrimethylsilan getropft.
Nach 60min Rühren bei 10 - 20°C wird auf 0°C abgekühlt und eine Lösung von 24,3 g
(150mmol) Carbonyldiimidazol in 105ml Dimethylformamid zugetropft. Nach weiteren 30min rühren werden 30,3g (121mmol) Scopinmethobromid zugegeben und weitere 60min bei 10 - 20°C gerührt. Es wird auf 10°C abgekühlt und eine Lösung aus 16,8g (150mmol)
Kaliumtertiärbutylat in 90ml Tetrahydrofuran bei 10 - 20°C zugetropft und 60min bei
20°C gerührt.
Nach Abkühlen auf 0°C werden 60ml 62%iger Bromwasserstoffsäure zugetropft wobei die Temperatur 20°C nicht übersteigt. Nach 40min Rühren wird das Reaktionsgemisch bei
20°C in 1150ml Isopropanol eingerührt und auf 10°C abgekühlt. Das Rohprodukt wird abfiltriert, mit 70ml kaltem Isopropanol gewaschen und im Vakuum getrocknet. Ausbeute
61,5g rotbraune Kristalle, DC entspricht Vergleich.
Das Rohprodukt wird mit 6,15g Aktivkohle in 615ml Methanol unter Rückfluss gelöst und filtriert. Anschließend werden 570ml Methanol abdestilliert und die Lösung auf 10°C abgekühlt. Die Kristalle werden abfiltriert, mit 35ml kaltem Methanol gewaschen und getrocknet. Ausbeute 40,9g weiß-beige Kristalle, DC entspricht Vergleich.
Die so erhaltenen Kristalle werden mit 2,2g Aktivkohle in 94ml Wasser bei 80°C gelöst und filtriert, es wird mit 24ml Wasser nachgewaschen. Nach Abkühlen auf 15°C wird das auskristallisierte Tiotropiumbromid Monohydrat abfiltriert, mit 25ml Wasser und 35ml
Aceton gewaschen und getrocknet. Ausbeute 28,6g (48% bezogen auf das eingesetzte
Scopinmethobromid).
Claims
Patentansprüche
1 ) Verfahren zur Herstellung von Tiotropiumsalzen der Formel 1
woπn
X " ein einfach negativ geladenes Anion, vorzugsweise ein Anion ausgewählt aus der Gruppe bestehend aus Chlorid, Bromid, Iodid, Methansulfonat oder Trifluormethansulfonat, bedeuten kann,
dadurch gekennzeichnet, daß eine Verbindung der Formel 2
worin X " die vorstehend genannten Bedeutungen haben kann, in einem Schritt mit einer in situ generierten Verbindung der Formel 3
worin
R ein Rest ausgewählt aus der Gruppe bestehend aus N-Imidazolyl, N-Triazolyl,
-O-C(=NR')-NHR", -O-SO2-Phenyl, -O-SO2-Phenyl-Methyl, -0-SO2-R'
-O-CO-C(Methyl)3, -O-CO-Phenyl-NO2, Chlor, Brom, -N3 und -0-(P=O)R", ist, wobei R Ci-C4-Alkyl oder C3-C6-Cycloalkyl;
R" Ci-C4-Alkyl, C3-C6-Cycloalkyl, Ci-C4-Alkylen-N(Ci-C4-Alkyl)2;
R" C1-C4-AIkVl, -0-C1-C4-AIkVl, Phenyl oder -O-Phenyl bedeuten R1 und R2, gleich oder verschieden Methyl, Ethyl, Propyl, Butyl oder Phenyl bedeuten können, wobei Phenyl gegebenenfalls substituiert sein kann durch einen oder mehrere Reste C1-C4-AIlCyI bedeuten, in einem geeigneten Lösemittel unter Zusatz einer geeigneten Base zu einer Verbindung der Formel 4
umgesetzt werden, wobei die Gruppen X", R1 und R2 die vorstehend genannten Bedeutungen haben können, und die Verbindung der Formel 4 ohne Isolierung durch Umsetzung mit einer geeigneten Säure oder einem geeigneten De-Silylierungsreagenz unter Abspaltung der Silylgruppe in die Verbindung der Formel 1 überfuhrt wird.
2) Verfahren zur Herstellung von Tiotropiumsalzen der Formel 1 nach Anspruch 1 , worin
X " ein einfach negativ geladenes Anion ausgewählt aus der Gruppe bestehend aus Chlorid, Bromid, Iodid, Methansulfonat oder Trifluormethansulfonat, bevorzugt Chlorid, Bromid oder Methansulfonat, besonders bevorzugt Brom, bedeuten kann.
3) Verfahren zur Herstellung von Tiotropiumsalzen der Formel 1 nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass die Umsetzung mit einer in situ generierten Verbindung der Formel 3 erfolgt, in der
R ein Rest ausgewählt aus der Gruppe bestehend aus N-Imidazolyl, N-Triazolyl, -O-C(=NR')-NHR", -O-SO2-Phenyl-Methyl, -O-CO-C(Methyl)3, und Chlor, ist wobei
R Methyl, Ethyl oder Cyclohexyl;
R" Methyl, Ethyl, Cyclohexyl, C2-C3-Alkylen-N(Methyl)2 oder
C2-C3-Alkylen-N(Ethyl)2, bedeuten und R1 und R2, gleich oder verschieden Methyl, Ethyl, Propyl oder Butyl, bedeuten.
4) Verfahren zur Herstellung von Tiotropiumsalzen der Formel 1 nach Anspruch 1 , 2 oder 3, dadurch gekennzeichnet, dass die Umsetzung mit einer in situ generierten Verbindung der Formel 3 erfolgt, in der R ein Rest ausgewählt aus der Gruppe bestehend aus N-Imidazolyl, N-Triazolyl,
-O-C(=N-Cyclohexyl)-NHCyclohexyl, -O-C(=N-Ethyl)-NH-CH2-CH2-CH2-NMe2 und -O-CO-C(Methyl)3, bevorzugt N-Imidazolyl oder N-Triazolyl, besonders bevorzugt N-Imidazolyl ist und
R1 und R2, gleich oder verschieden Methyl, Ethyl, Propyl oder Butyl, bevorzugt Methyl oder Ethyl, besonders bevorzugt Methyl;
R2 Methyl oder Ethyl, bevorzugt Methyl, bedeuten.
5) Verfahren zur Herstellung von Tiotropiumsalzen der Formel 1 nach einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass die Verbindung der Formel 3 in situ in einem geeigneten Lösemittel generiert wird durch Umsetzung von Dithienylglykolsäure oder Alkalisalzen der Dithienylglykolsäure mit einem Kupplungsreagenz ausgewählt aus der Gruppe bestehend aus Carbonyldiimidazol, Carbonyldi-l,2,4-triazol, Dicyclohexylcarbodiimid , Ethyl-dimethylaminopropylcarbodiimid, Toluolsulfonylchlorid, Pivaloylchlorid, Nitrobenzoesäureanhydrid, Oxalylchlorid, Phosgen, Sulfonylchlorid und Phosphorchloriden, und anschließender Zugabe einer Silylverbindung der Formel 5
R2 R 5 wobei die Reste R1 und R2 die vorstehend genannten Bedeutungen haben können und L für eine Abgangsgruppe steht, die vorzugsweise ausgewählt ist aus der Gruppe bestehend aus Halogenid, Methansulfonat, Trifluormethansulfonat und para-Toluolsulfonat.
6) Verfahren zur Herstellung von Tiotropiumsalzen der Formel 1 nach Anspruch 5, dadurch gekennzeichnet, dass das Lösemittel ausgewählt ist aus der Gruppe bestehend aus Acetonitril, Nitromethan, Formamid, Dimethylformamid, N-Methylpyrrolidinon, Dimethylsulfoxid , Dimethylacetamid Tetrahydrofuran, Dioxan und Sulfolan.
7) Verbindung der Formel 3
worin R, R1 und R2 die in den Ansprüchen 1 bis 5 genannten Bedeutungen haben können.
8) Verbindung der Formel 4
worin X ", R1 und R2 die in den Ansprüchen 1 bis 5 genannten Bedeutungen haben können.
