Description
Undulin und dessen Fragmentierungsprodukte,
Verfahren zu deren Herstellung und ihre Verwendung
Die Erfindung betrifft Undulin und dessen Peptidfragmente, Verfahren zu deren Herstellung und ihre Verwendung zur Produktion spezifischer Antikörper und zur Diagnostik, Prognostik und Therapie von das Bindegewebe betreffenden Erkrankungen und Tumorerkrankungen.
Interstitielles Bindegewebe besteht zur Hauptsache aus Kollagen-I-Fibrillen. Ultrastrukturelle Studien lassen vermuten, daß Makromoleküle den Zusammenhalt der Kollagen- fibrillen "vermitteln. Es wird vermutet, daß neben Fibro- nektin und Proteoglykan eine weitere Strukturkomponente für die supramolekulare Organisation der Kollagen-I- Fibrillen verantwortlich ist.- Bisher sind Versuche, diese Komponente zu isolieren, fehlgeschlagen."
Überraschenderweise wurde nun gefunden, daß bei der Extraktion von Häuten fötaler oder neugeborener Säuge¬ tiere das neue Protein Undulin erhalten wird, das eine wesentliche Rolle in der supramolekularen Organisation der Kollagen-I-Fibrillen spielt und das sich in der Medi¬ zin und Kosmetik verwendet läßt.
Die Erfindung betrifft Undulin und dessen Peptidfragmente, sowie ein Verfahren zu deren Herstellung und ihre Verwen¬ dung.
Unter den interstitiellen Bindegewebsproteinen scheint Undulin neben Kollagen-I und -III eine Hauptkomponente darzustellen. Undulin ist ein stark aggregiertes, makro¬ molekulares Glykoprotein, das hauptsächlich als quer- vernetzter Komplex mit einem Molekulargewicht von >>1 000 000 vorliegt, ermittelt anhand von SDS-Polyacryl- amid-Gel-elektrophorese unter nicht-reduzierenden
l Bedingungen. Unter Verwendung eines Kollagen-I-Standards sind nach Reduktion eine charakteristische Bande bei 270 000 (A-Kette) und ein Dublett bei 175 000/180 000 (Bl- bzw. B2-Kette anfärbbar. Die Mobilität der Dublett- 5 bände ist deutlich größer als die des reduzierten Fibro- nektins.
Figur 1 zeigt Undulin vor (1) und nach (2)-Reduktion in der SDS-Polyacrylamid-Gelelektrophorese. Die Position kollagener α-, ß- und γ-Komponenten ist.durch Pfeile mar- kiert (Standards).
Undulin ist nach Lyophilisation praktisch unlöslich in neutralen nicht-denaturierenden Puffersystemen, z.B. Tris • HC1 Phosphat-Puffer oder verdünnter Essigsäure, g Dagegen ist Undulin in 6 M Guanidin-HCl, pH 7 bis 9, in Gegenwart von Proteaseinhibitoren gut löslich. In Assoziation mit Kollagen-I läßt sich Undulin in 0,5 M Essigsäure/1 % Triton X-100 in Lösung bringen.
Derartige Präparationen lassen sich nach Kegelbedampfung elektronenmikroskopisch untersuchen. Dabei erscheint das Molekül in Form einer bisher unbekannten Struktur, die aus einem globulären Kopfanteil mit einem Durchmesser 5 von ca. 18 nm, einem ca. 70 nm langen Schwanzteil und an dessen anderem Ende z.T. einer kleineren globulären Domäne (Durchmesser ca. 6 nm) besteht. Häufig werden aggregierte Formen, die sich aus zwei, vier oder mehreren dieser Strukturen zusammensetzen und die oft mit Kollagen- I-Molekülen assoziiert sind, beobachtet. Eine elektronen¬ mikroskopische Aufnahme nach Kegelbedampfung, auf der Undulin in isolierter (Pfeile) und aggregierter (Doppel- pfeile)-Form neben Kollagen I zu sehen ist, ist in Fig. 2 abgebildet. 5
In der Aminosäureanalyse (6 M HC1, 24 Stunden, unter Stickstoff bei 110 °C) ergibt Undulin eine für große Glykoproteine typische aber uncharakteristische Amino¬ säurezusammensetzung mit einem hohen Anteil an hydrophoben und sauren Aminosäuren.
Die Aminosäurezusammensetzung, welche sich aber deutlich von der des Basalme branglykoproteins Laminin oder von der des Fibronektins unterscheidet, ist aus der nach¬ folgenden Tabelle ersichtlich.

Der Kohlenhydratanteil von Undulin, ermittelt durch Gaschromatographie der Alditol-Azetate und durch Massen- spektro etrie, liegt mit ca. 2 % (Gewicht pro 100 g Undulin) deutlich unter dem von Laminin (ca. 12 ) und Fibronektin (ca. 5 %). Die Kohlenhydratanalyse zeigt die nachfolgende Tabelle.

Auf unfixierten 4-μm-Kryostatschnitten von Haut oder Leber zeigen Undulinantikörper -in der indirekten Immun- fluoreszeήz ein neuartiges Muster, das sich von allen be-
20 kannten Mustern für Bindegewebsproteine unterscheidet. Wie aus Fig. '3 ersichtlich, besteht das Undulinmuster aus wellenförmigen, parallel angeordneten Fibrillen mit einem einheitlichen Durchmesser (ca. 1 μm) . Diese typi¬ schen wellenförmigen Fibrillenbündel durchziehen das
« Interstitium und sind in Basalmembranen nicht vorhanden. Undulin findet sich vorwiegend im dichten Bindegewebe, wo auch ausgereifte Kollagen (Typ I) Fibrillen zu erwar¬ ten sind, etwa in tieferen Schichten der Dermis (Fig. 3), in der Adventitia größere Blutgefäße oder im Portalfeld
„Q der Leber (Abb. 4), Vergleichbare Verteilungsmuster lassen sich nicht nur im Menschen, sondern auch in der Maus, der Ratte und im Rind. etc. finden. Interessanter¬ weise wird Undulin im Bereich von Tumoren schneller als andere Bindegewebsproteine abgebaut.

Das Verfahren zur Herstellung von Undulin ist dadurch gekennzeichnet, daß man in Gegenwart von Proteaseinhibi- toren
A) Häute von fötalen oder neugeborenen Säugetieren einer Neutralsalzextraktion unterzieht,
B) den Extrakt einer fraktionierten Salzfällung unter¬ zieht,
C) das Präzipitat löst, und durch DEAE-Chromatographie reinigt, wobei man Undulin als Rohprodukt erhält,
D) * das Rohprodukt nach Dialyse und Lyophilisierung weiter durch Extraktion reinigt,
E) das Undulin zur Herstellung der Fragmentierungs¬ produkte mit neutralen oder sauren Proteasen spaltet, bis man Fragmente mit einem Molekulargewicht
<400 000 erhält, und
F) das gemäß Stufe E erhaltene Produkt mittels CM- und DEAE-Cellulose-Chromatographie, Molekularsiebe und/ oder HPLC in einem Puffer von pH 2,5 - 9,0 reinigt.
Vorzugsweise erfolgt die Extraktion gemäß Stufe A) , indem man zunächst eine Vorextraktion mit Neutralpuffer zur Entfernung von Serumproteinen und Lipiden durchführt. Dazu verwendet man eine 10 - 200 mM Neutralpufferbase, beispielsweise 50 - 200 mM Tris • HC1, 4-5 M NaCl vom
pH 7 - 8, 20 - 200 M HEPES-(2-[4-(2-Hydroxyethyl)-l- piperazinyll-ethansulfonsäure)- oder PIPES (Piperazin-N,N'~ bis(2-ethansulfonsäure)-Puffer, zusammen mit 0,1 - 2 % eines nicht-ionischen Detergens, beispielsweise Triton X-100. Man verwendet etwa 2-4 1 Puffer pro kg Haut (Feucht¬ gewicht). Daran schließt sich die eigentliche Extraktion an, die vorzugsweise im gleichen Puffersystem und mit der gleichen Menge erfolgt, jedoch bei niedrigerer Ionen¬ stärke durchgeführt wird.
Der Extrakt wird anschließend gemäß Stufe B) einer • fraktionierten Salzfällung unterzogen. Im ersten Schritt wird der Extrakt durch Zugabe von Ammoniumsulfat .gefällt, bis ein Sättigungsgrad von 20 bis 50 % erreicht ist. Das so erhaltene Präzipitat wird dann erneut in 10 - 200 mM Neutralpufferbase gelöst, und einer fraktionierten Salz¬ fällung mit einem Alkalihalogenid, beispielsweise NaCl, KCl, unterworfen. Die Hauptmenge des Undulins wird zwischen 1,5 M und 4,5 M gefällt.
Zur Reinigung wird das Präzipitat in Neutralpuffer/1-4 M Harnstoff/0,25-0,5 M NaCl gelöst und mit DEAE-Cellulose behandelt. Die ungebundene Fraktion wird gegen den gleichen Puffer, der jedoch nur 0,01 - 0,05 M NaCl ent¬ hält, dialysiert. Das so erhaltene Präzipitat besteht zu ca. 70 % aus Undulin. Der überstand enthält weiteres Undulin, das man durch Chromatographie an DEAE-Cellulose, wobei man einen 'Salzgradienten als Elutionsmittel verwen¬ det, in mehr als 90%iger Reinheit gewinnen kann.
Die Präparate werden durch Dialyse, beispielsweise gegen Essigsäure, von Salzen befreit und lyophilisiert. Das 70%ige Präparat kann durch Extraktion mit Neutralpuffer,
gegebenenfalls in Gegenwart eines nicht-ionischen Detergens, von Verunreinigungen befreit werden. Das so gewonnene Produkt mit einer Reinheit von mehr als 80 % läßt sich in 10 - 100 mM Tris-HCl, 5 - 6 M Guanidin, -pH 8 - 9, lösen und an einer Molekularsiebsäule (etwa Sephacryl 2B, Cl 4B oder 5- 1000) von niedermolekularen Verunreinigungen trennen. Die erhaltene Reinheit beträgt nun nahezu 100 %.
Die Ausbeute beträgt ca. 100 mg/kg Haut (Feuchtgewicht). Weiter läßt sich Undulin noch reinigen, wie unten für die Fragmente des Undulins beschrieben.
Das gesamte Verfahren wird zweckmäßigerweise bei Kühl¬ raumtemperaturen (4 - 10 °C) und in Gegenwart von Proteaseinhibitoren, beispielsweise Phenylmethylsulfonyl- fluorid (PMSF), N-Ethylmaleimid (NEM), Ethylendiamintetra- essigsäure (EDTA), Sojabohnen-T ypsininhibitor' (SBTI) , Trasylol und Benza idinhydrochlorid, durchgeführt.
Undulin ist sehr sensitiv gegenüber neutralen oder sauren Proteasen. Geeignete neutrale Proteasen sind z.B. Trypsin, Thrombin, Plasmin, Elastase, Kathepsin, V8-Staphylokokken- protease, Chymotrypsin, Papain und Thermolysin. Beispiele geeigneter saurer Proteasen sind Pepsin, saures Kathepsin, Chymosin etc.
Bevorzugte Proteasen sind Trypsin, Elastase, Plasmin, Thrombin, V8-Staphylokokkenprotease und Chymotrypsin, sowie Pepsin.
Proteasefragmente von Undulin mit einem Molekulargewicht von <400 000, vorzugsweise <100 000 sind zum Teil in neutralen Puffern löslich, unter Wahrung einiger der anti- genen Merkmale des Undulins.
So liefert beispielsweise der Abbau des Undulins mit Trypsin bei 30 °C über Zwischenstufen mit einem Molekular- 5 gewicht von 300 000 - 400 000, Hauptprodukte mit einem Molekulargewicht von 30.000 - 50 000, 60 000 - 70 000 und 80 000. Das Endprodukt des Pepsinabbaus bei 4 °C besitzt ein Molekulargewicht zwischen 25 000 und 50 000.
10 Die Fragmente lassen sich mittels CM- und DEAE-Cellulose- Chromatographie, Molekularsieb und/oder HPLC in üblichen Puffern mit einem pH von 2,5 - 9,0 reinigen. Geeignete saure Puffer sind beispielsweise Acetat- und Zitratpuffer. Geeignete Neutralpuffer wurden bereits oben erwähnt und ,c geeignete alkalische Puffer sind A moniumbicarbonat, Natriumacetat.
Undulin und seine Fragmente sind stark antigen wirksam. Zum Beispiel genügt eine dreifache Applikation von 0,3 mg Undulin oder seinen Fragmenten zur Herstellung"
20 von hochtitrigen Antiseren, Besonders in Kaninchen und Mäusen können spezifische Antikörper mit Hilfe bekannter
Verfahren, z.B. Äffinitätschromatographie,erhalten werden. Diese Reagenzien reagieren im Western-Blot, im ELISA ©der
25 im Radioimmunoassay in keinerlei Weise kreuz mit anderen Bindegewebsproteinen. Auch mit der monoklonalen Technik lassen sich hochaffine Antikörper gegen Undulin und seine Fragmente herstellen. Derartige Immunreagentien lassen sich in der Immunhistologie, für Zellkulturexperimente, in der Gentechnologie und zum analytischen Nachweis von
30 Undulin und seinen Fragmenten verwenden. So konnte gezeigt werden, daß mit monoklonalen Antikörpern gegen Undulin in der Immunhistologie das gleiche charakteristische Muster wie mit polyklonalen Antikörper erhalten wird.

ELISA = Enzyme - Linked Immuno Sorbent Assay
Darüber hinaus dienen die oben erwähnten Antikörper und Antigene zur quantitativen Bestimmung von Undulin und/oder g seiner Fragmente in Körperflüssigkeiten, Zellmedien und Extrakten mittels Radio-Immuno-Assay, in ELISA- , Fluoreszenz-Immuno-Assay (FIA), Lumineszenz-Immuno-Assay und dergleichen. Es konnte nämlich mittels eines hoch¬ spezifischen und sensitiven Radioimmunoassays gezeigt 0 werden, daß Undulin-Antigenität in Körperflüssigkeiten "zirkuliert und mit Krankheitsbildern, wie das intersti- tielle Bindegewebe betreffen, wie Entzündungen, fibroti- schen Erkrankungen und Tumorleiden, in Form erhöhter Spiegel korreliert. Undulin und seine Fragmente lassen 5 sich auch in kosmetischen Zubereitungen, wie Lotionen, Gelen, Cremes, zur Verbesserung des Aussehens und des Zustandes der Haut verwenden.
Die nachfolgenden Beispiele erläutern die Erfindung. 0

κ Herstellung von Undulin
Die Haut von fötalen oder neugeborenen Affen, Kälbern oder Ratten wird eingeweicht, homogenisiert und in 100 M Tris 4,5 M NaCl vom pH 7,4, das 2 M PMSF, 4 mM NEM und 10 M EDTA als Proteaseinhibitoren enthält, tief¬ gefroren. Alle Arbeiten erfolgen bei 4 °C und in Gegen¬ wart von Proteaseinhibitoren (PI), wenn Puffer verwendet werden, deren pH oberhalb von 4,0 liegt. Das Homogenisat wird zunächst mit 50 mM Tris/PI/4,5 M NaCl, pH 7,4 und 1 % Triton X-100 (31/kg Feuchtgewicht) extrahiert. Der Rückstand wird anschließend mit 50 mM Tris/PI/0,5 M NaCl, pH 7,4 (31/kg Feuchtgewicht) extrahiert. Aus der
überstehenden Flüssigkeit erhält man durch Zugabe von Ammoniumsulfat bis zu einem Sättigungsgrad von 30 % (168 g/1) ein Präzipitat. Dieses wird erneut in Tris/PI/ 0,5 M NaCl (31/kg Feuchtgewicht) gelöst. Die Lösung unter¬ wirft man einer fraktionierten Salzfällung mit NaCl bis zu Endkonzentrationen von 1,7, 2,6 und 4,5 M. Die Fraktion bei 2,6 M NaCl wird in 50 mM Tris, PI,.0,3 M NaCl, 1 - 4 M, pH 7,4 Harnstoff gelöst und gegen dieses Gemisch dialysiert. Einen Niederschlag (Kollagen-I-Aggregat) zentrifugiert man ab und die überstehende Flüssigkeit rührt man mit DEAE-Cellulose. Die ungebundene Fraktion dialysiert man gegen Tris, PI, 0,02 M NaCl, pH 7,4, wobei man ein Präzipitat erhält, das aus Undulin, Kollagen I und einem Protein mit einem Molekulargewicht von 35 000 besteht. Das lösliche Material chro atographiert man an DEAE-Cellulose, wobei man nach Elution mit einem Salzgrar dienten von ungefähr 0,25 M NaCl Undulin mit einer Rein¬ heit von >90 % erhält.
Die Undulin enthaltenen Fraktionen werden gegen 0,1 M Essigsäure dialysiert und lyophilisiert. Zur weiteren Reinigung wird das Lyophilisat mit 50 M Tris-HCl, 2 M Harnstoff, Proteaseinhibitoren, 1 % Triton X-100, pH 8,2, bei 4 °C extrahiert, wobei u.a. verunreinigende Proteine entfernt werden. Das ungelöste ca. 80 % reine Undulin wird dann in 50 M Tris-HCl, 6 M Guanidin, pH 8,2, gelöst und im gleichen Puffer an Sepharose Cl 4B von niedrigmole¬ kularen Verunreinigungen befreit. Auf diese Weise her¬ gestelltes Undulin ist homogen und rein, die Ausbeute beträgt 100 mg/kg eingesetzter Haut (Feuchtgewicht).

5 Trypsinabbau von Undulin
Das lyophilisierte Protein wird zu 0,5 mg/ml in 0,2 M Na.HCO-/Essigsäure, pH 7,0, suspendiert und mit Trypsin- TPCK (Serva, Heidelberg) im Enzym:Substrat-Verhältnis 0 1:50 bei 25 °C unter Schütteln abgebaut. Aliquote werden nach 10, 30, 60 Minuten, 4 und 16 Stunden entnommen, mit Trypsininhibitor (Soybean Trypsin-Inhibitor, Serva, Heidelberg) versetzt und lyophilisiert. Der Abbau nach 16 Stunden enthält hauptsächlich Peptide vom Molekular-
15 gewicht kleiner gleich 80 000.

0 Pepsinabbau von Undulin
Für den Pepsinabbau wird Undulin zu 1 mg/1 ml in 0,5 M Essigsäure suspendiert und bei 4 - 8 °C im Verhältnis 1:50 (Enzym zu Substrat) unter Schütteln mit Pepsin
25 (Sigma, Tauf irchen) abgebaut. Nach 6 bis 10 Stunden er¬ hält man als Hauptprodukt ein Fragment mit dem Molekular¬ gewicht zwischen 25 000 und 50 000.

Herstellung polyvalenter Antiseren und polyklonaler
Antikörper
„ Hochtitrige Antiseren gegen Undulin und seine Fragmente
(von Affen, Kälbern oder Ratten) lassen sich folgender¬ maßen herstellen: g Man verabreicht 0,2 mg Antigen, das in 0,5 ml Neutral¬ puffer suspendiert und in 0,5 ml komplettem Freund's Ad uvans emulgiert ist, subkutan. Daran schließen sich in 3-Wochenintervallen zwei weitere Injektionen unter Verwendung von inkomplettem Adjuvans an. Das Antiserum 1Q gibt man über säulenimmobilisierte Bindegewebsantigene. Nach ausgiebiger Spülung mit Puffer können zuletzt von immobilisiertem Undulin spezifische Antikörper hoher Affi¬ nität eluiert werden.

Herstellung monoklonaler Antikörper
_n 8 - 12 Wochen alte Balb/c Mäuse erhalten s.c. 3 - 4 In¬ jektionen von 100 μg Undulin bzw. Undulinfragmenten im Abstand von jeweils 2 Wochen. Die erste ist im Verhältnis 1:1 mit Freund'sehen Adjuvans (komplett) gemischt, die nächsten sind im Verhältnis 1:1 mit Freund'sehen Adjuvans
25 (inkomplett) gemischt.
Nach positivem Antikörpertiter in der Maus wird deren Milz mit isogenen Myelo zellen (NS1) fusioniert und in Gewebekultur gebracht. Positive Antikörper produzierende Zellklone werden subkloniert, um einzelne Antikörper pro¬ duzierende Zellinien zu erhalten. Der Antikörper kann 0 mit bekannten Methoden aus dem Gewebekulturüberstand bzw. aus der Ascites von als Zellkulturträger dienenden Mäusen gereinigt werden.

Radioimmunoassay
Das in Beispiel 1 nach Chromatographie an Sepharose C14B gereinigte Undulin läßt sich zu 10 μg/50 μl 50 mM Tris- HCl, 6M Guanidin, 0,05 X Triton, pH 8,2, mittels der
125 Chloramin-T Methode mit 1 mCi I radioaktiv markieren
(Markierungsdauer 0,5 - 3 Min. ). Entfernung von niedermole¬ kularer Aktivität erfolgt durch Dialyse gegen 50 mM Tris- HCl, 8 M Harnstoff, 0,05 % Triton x-100, pH 8,2^ bei Rau - temperatur. 125I-Undulin (3 000 - 10 000 cpm/ng Protein) zeigt 90 - 100 * Bindung an sein Antiserum und erlaubt die Durchführung eines sensitiven Flüssigphasen-Radio- immuno-Inhibitionsassays, etwa für die Bestimmung zirku¬ lierender Antigene im Serum oder im Plasma..
Claims
1. Undulin und dessen Fragmentierungsprodukte, dadurch erhältlich, daß man in Gegenwart von Protease- inhibitoren
A) Häute von fötalen oder neugeborenen Säugetieren einer Neutralsalzextraktion unterzieht,
B) den Extrakt einer fraktionierten Salzfällung unter¬ zieht,
C) das Präzipitat löst, und durch DEAE-Chromatographie reinigt, wobei man Undulin als Rohprodukt erhält,
D) das Rohprodukt nach Dialyse und Lyophilisierung weiter durch Extraktion reinigt,
E) das Undulin zur Herstellung der Fragmentierungspro- dukte mit neutralen oder sauren Proteasen spaltet, bis man Fragmente mit einem Molekulargewicht <400 000 erhält, und
F) das gemäß Stufen D und E erhaltene Produkt mittels CM- und DEAE-Cellulose-Chromatographie, Molekular¬ sieb und/oder HPLC in einem Puffer von pH 2,5 - 9,0 reinigt.
2. Verfahren zur Herstellung der Verbindungen nach Anspruch 1, d a d u r c h g e k e n n z e i c h n e t ,
daß man in Gegenwart von Proteaseinhibitoren
A) Häute von fötalen oder neugeborenen Säugetieren einer Neutralsalzextraktion unterzieht,
B) den Extrakt einer fraktionierten Salzfällung unterzieht,
C) das Präzipitat löst, und durch DEAE-Chromato¬ graphie reinigt, wobei man Undulin als Rohprodukt erhält,
D) "das Rohprodukt nach Dialyse und Lyoph-ilisiefung weiter durch Extraktion reinigt,
E) das Undulin zur Herstellung der Fragmentierungs¬ produkte mit neutralen oder sauren Proteasen spaltet, bis man Fragmente mit einem Molekular¬ gewicht < 400 000 erhält, und
F) das gemäß Stufen D und E erhaltene Produkt mittels CM- und DEAE-Cellulose-Chromatographie, Molekular¬ sieb und/oder HPLC in einem Puffer von pH 2,5
9,0 reinigt.
3. Verfahren nach Anspruch 2, dadurch gekennzeichnet, daß die Neutralsalzextraktion gemäß Stufe A in zwei Stufen durchgeführt wird, wobei die erste Stufe mit einer 10 - 200 mM Neutralpufferbase und die zweite Stufe mit einem Neutralpuffer niedrigerer lonenstärke erfolgt.
4. Verfahren nach Anspruch 3, dadurch gekennzeichnet, daß man die Vorextraktion in Gegenwart von 0,1 - 2 % g eines nichtionischen Detergens durchführt.

Verfahren nach einem der Ansprüche 2 - 4, dadurch gekennzeichnet, daß man die fraktionierte Salzfällung gemäß Stufe B durchführt, indem man zu dem aus Q Stufe A erhaltenen Extrakt Ammoniumsulfat gibt, bis ein Sättigungsgrad von 20 - 50 % erreicht ist, das so erhaltene Präzipitat in einer 10 - 200 mM Neutral¬ pufferbase löst und die Lösung einer fraktionierten Salzfällung mit einem Alkalihälogenid unterwirft. 5
Verfahren nach einem der Ansprüche 2 - 6, dadurch gekennzeichnet, daß man die Reinigung gemäß Stufe C durchführt, indem man das aus Stufe B erhaltene Präzipitat in 10 - 200 mM Neutralpufferbase/l - 4 M Harnstoff/0,25 - 0,5 M NaCl löst und mit DEAE- 0 Cellulose behandelt, und die ungebundene Fraktion gegen den gleichen Puffer, der 0,01 - 0,05 M NaCl enthält, dialysiert, wobei man ein Präzipitat von 70%igem Undulin erhält . 5
Verfahren nach Anspruch 6, dadurch gekennzeichnet, daß man den nach der Dialyse erhaltenen Flüssigkeits¬ überstand durch Chromatographie an DEAE-Cellulose unter Verwendung eines Salzgradienten als Elutions- mittel reinigt, wobei man Undulin von mehr als 0 90%iger Reinheit erhält.
Verfahren nach einem der Ansprüche 2 - 7, dadurch gekennzeichnet, daß man die Reinigung gemäß Stufe D durchführt, indem man gegen Essigsäure dialysiert, 5 lyophilisiert und das Lyophilisat mit 10 - 200 mM Neutralpufferbase oder verdünnter Essigsäure, gegebenen¬ falls mehrmals, extrahiert. 1
9. Verfahren nach Anspruch 8, dadurch gekennzeichnet, daß man die Extraktion in Gegenwart von 0,1 - 2 % g eines nichtionischen Detergens durchführt.
10. Verfahren nach Anspruch 2, dadurch gekennzeichnet, daß man als Protease zur Spaltung des Undulins Trypsin, Elastase, Plasmin, Thrombin, V8-Staphylo- 0 kokkenprotease, Chymotrypsin und/oder Pepsin ver¬ wendet.
11. Verfahren nach Anspruch 10, dadurch gekennzeichnet,, daß man Trypsin oder Pepsin verwendet. 5
12. Verwendung der Produkte nach Anspruch 1 zur Her¬ stellung von Antikörpern, .für immunologische Nach¬ weismethoden und zur Diagno.se, Prognose und Verlaufs¬ kontrolle fibrotischer, entzündlicher .und tumoröser Erkrankungen. 0
