Abstract

Изобретение относится к биотехнологии, а именно к получению эндотелиальных клеток из эмбриональных стволовых клеток (ЭСК) человека и их применению. Способ получения эндотелиальных клеток из эмбриональных стволовых клеток (ЭСК) человека предусматривает дифференцировку ЭСК человека на подложке и в синтетической среде, которые обеспечивают направленную прямую дифференцировку ЭСК человека в эндотелиальные клетки на основе смеси DMEM/FI2 с КО заменителем сыворотки с факторами роста VEGF, SCF, ВМР4, bFGF, TGFβ в эффективных количествах, причем TGFβ добавляют на 3-6 день дифференцировки. Также может быть использована среда для дифференцировки на основе смеси DMEM/FI2 с фетальной бычьей сывороткой с факторами роста VEGF, SCF, bFGF. Сепарацию проводят на 3-9 день методом иммунологической селекции с использованием маркеров, специфических для эндотелиальных клеток, после чего полученные эндотелиальные клетки культивируют в указанной среде при плотности посева не менее 50000 клеток на 1 см.
