Claims (10)
1. Modificeret mikroorganisme af Corynebacterium-slægten, som har forbedret evne til at producere putrescin, hvor aktiviteten af et protein med en aminosyresekvens, som repræsenteres af SEQ ID NO: 18 eller SEQ ID NO: 20, er svækket eller fjernet sammenlignet med den endogene aktivitet deraf, og hvor aktiviteten af ornithindecarboxy-lase (ODC) yderligere er indført deri.
2. Mikroorganisme ifølge krav 1, hvor ODC har aminosyresekvensen ifølge SEQ ID NO: 23.
3. Mikroorganisme ifølge krav 1, hvor aktiviteterne af én eller flere, som er valgt fra gruppen bestående af ornithincarbamoyltransferase (ArgF) og glutamateksportør (NCgM 221), er yderligere svækket i sammenligning med den endogene aktivitet deraf.
4. Mikroorganisme ifølge krav 3, hvor ArgF har aminosyresekvensen ifølge SEQ ID NO: 21, og NCgM221 har aminosyresekvensen ifølge SEQ ID NO: 22.
5. Mikroorganisme ifølge krav 1, hvor aktiviteterne af én eller flere, som er valgt fra gruppen bestående af acetyl-gamma-glutamylphosphatreduktase (ArgC), acetyl-glutamatsyntase eller ornithinacetyltransferase (ArgJ), acetylglutamatkinase (ArgB) og acetylornithinaminotransferase (ArgD), er yderligere forøget.
6. Mikroorganisme ifølge krav 5, hvor ArgC, ArgJ, ArgB og ArgD har aminosyresekven-serne ifølge henholdsvis SEQ ID NO: 24, 25, 26 og 27.
7. Mikroorganisme ifølge krav 1, hvor proteinets aktivitet svækkes ved 1) en delvis eller hel deletion af et polynukleotid, som koder for proteinet, 2) en reduktion af polynukleo-tidekspression, 3) en modifikation af polynukleotidsekvensen på kromosomet for at svække proteinets aktivitet eller 4) en kombination deraf.
8. Mikroorganisme ifølge krav 1, som er Corynebacterium glutamicum.
9. Fremgangsmåde til fremstilling af putrescin, hvilken fremgangsmåde omfatter dyrkning, for at fremstille en cellekultur, af en modificeret mikroorganisme af Corynebacte- /rom-slægten, som har forøget evne til at producere putrescin, hvor aktiviteten af et protein med en aminosyresekvens, der repræsenteres af SEQ ID NO: 18 eller SEQ ID NO: 20, er svækket eller fjernet sammenlignet med den endogene aktivitet deraf; og isolering af putrescin fra den opnåede cellekultur.
10. Fremgangsmåde til fremstilling af putrescin ifølge krav 9, hvor mikroorganismen af Corynebacterium-slægten er Corynebacterium glutamicum.
