Claims (18)
- FILAMENTØSE SVAMPE MED EN ÆNDRET VISKOSITETSFÆNOTYPE PATENTKRAV1. Variantstamme af filamentøs svamp, der er afledt af en moderstamme, hvilken variantstamme indbefatter en genetisk ændring, der omfatter en disruption af ips2-genet, der bevirker, at celler af variantstammen producerer en reduceret mængde funktionelt Tps2-protein sammenlignet med celler af moderstammen, hvor cellerne af variantstammen under aerob fermentation i nedsænket kultur producerer en cellebouillon, der (i) kræver en reduceret mængde omrøring for at bevare et forhåndsvalgt opløst oxygenindhold sammenlignet med moderstammens celler, og/eller (ii) bevarer et øget opløst oxygenindhold ved en forhåndsvalgt mængde omrøring, sammenlignet med moderstammens celler, hvor variantstammen endvidere omfatter et gen, der koder for et protein af interesse, til ekspression af proteinet i de filamentøse svampeceller.
- 2. Variantstamme ifølge krav 1, hvor: (a) den genetiske ændring, der omfatter en disruption af ips2-genet, er resultatet af deletering af hele eller en del af ips2-genet; eller (b) den genetiske ændring, der omfatter en disruption af tps2-genet, er resultatet af deletering af en del af genomisk DNA, der omfatter ips2-genet; eller (c) den genetiske ændring, der omfatter en disruption af ips2-genet, er resultatet af mutagenese af ips2-genet.
- 3. Variantstamme ifølge krav 2, hvor disruption af ips2-genet udføres ved anvendelse af stedsspecifik rekombination eller disruption af ips2-genet udføres i kombination med indføring af en valgbar markør ved det genetiske locus for ips2-genet.
- 4. Variantstamme ifølge et hvilket som helst af de foregående krav, hvor variantstammen ikke producerer funktionelt Tps2-protein.
- 5. Variantstamme ifølge et hvilket som helst af de foregående krav, hvor variantstammen ikke producerer Tps2-protein.
- 6. Variantstamme ifølge et hvilket som helst af de foregående krav, der endvidere omfatter en disruption af s/fc3-genet.
- 7. Variantstamme ifølge et hvilket som helst af de foregående krav, hvor variantstammen producerer mere af proteinet af interesse pr. biomassemængdeenhed end moderstammen.
- 8. Variantstamme ifølge et hvilket som helst af de foregående krav, hvor den filamentøse svamp er en Pezizomycotina-ait eller den filamentøse svamp er en Trichoderma spp.
- 9. Variantstamme ifølge krav 8, hvor den filamentøse svamp er Trichoderma reesei.
- 10. Variantstamme ifølge et hvilket som helst af de foregående krav, hvor variantstammen endvidere omfatter en dismption af mindst ét gen, der er udvalgt fra gruppen bestående af sft>3-genet, seW-genet, mpgl-genet, ga,v/-genet og c/z/-genet.
- 11. Fremgangsmåde til at producere en variantstamme af filamentøse svampeceller, hvilken fremgangsmåde omfatter: indføring af en genetisk ændring i en moderstamme af filamentøs svampecelle, hvilken genetisk ændring omfatter disruption af ips2-genet ved anvendelse af genmanipulation til at reducere eller forhindre produktionen af funktionelt Tps2-protein sammenlignet med moderstammens celler, hvorved der produceres en filamentøs variantsvampecelle, der under aerob fermentation i nedsænket kultur producerer en cellebouillon, der (i) kræver en reduceret mængde omrøring for at bevare et forhåndsvalgt opløst oxygenindhold, sammenlignet med moderstammens celler, og/eller (ii) bevarer et øget opløst oxygenindhold ved en forhåndsvalgt mængde omrøring, sammenlignet med moderstammens celler, hvor variantstammen endvidere omfatter et gen, der koder for et protein af interesse, til ekspression af proteinet i de filamentøse svampeceller.
- 12. Fremgangsmåde ifølge krav 11, hvor: (a) den genetiske ændring omfatter deletering af tps2-genet i en filamentøs modersvampecelle ved anvendelse af genmanipulation; eller (b) den genetiske ændring udføres ved anvendelse af stedsspecifik genetisk rekombination.
- 13. Fremgangsmåde ifølge krav 11 eller krav 12, hvor: (a) disruption af tps2-genet udføres i kombination med indføring af en valgbar markør ved det genetiske locus for tps2-genet; eller (b) disruption af tps2-genet udføres i kombination med disruption af sft>3-genet.
- 14. Fremgangsmåde ifølge et hvilket som helst af kravene 11 til 13, hvor variantstammen producerer mere af proteinet af interesse pr. biomassemængdeenhed end moderstammen.
- 15. Fremgangsmåde ifølge et hvilket som helst af kravene 11 til 14, hvor den filamentøse svamp er en Pezizomycotina-ait eller den filamentøse svamp er en Trichoderma spp.
- 16. Fremgangsmåde ifølge krav 15, hvor den filamentøse svamp er Trichoderma reesei.
- 17. Fremgangsmåde ifølge et hvilket som helst af kravene 11 til 16, hvor genet, der koder for proteinet af interesse, er til stede i moderstammen før indføring af den genetiske ændring, der reducerer eller forhindrer produktionen af funktionelt Tps2-protein.
- 18. Fremgangsmåde ifølge et hvilket som helst af kravene 11 til 17, hvor disruption af /p,v2-gcnet udføres i kombination med disruption af mindst ét gen, der er udvalgt fra gruppen bestående af sfb3-genet, sebi-genet, mpgl-genet, gas/-genet og crzl-genet.
